A retrospective clinicopathological study on oral lichen planus and malignant transformation: analysis of 518 cases
Objective: To investigate the epidemiological and clinical characteristics of a relatively large cohort of patients
with oral lichen planus (OLP) from eastern China.
Study design: A total of 518 patients with histologically confirmed OLP in a long-term follow-up period (6 months-
21.5 years) were retrospectively reviewed in our clinic.
Results: Of the 518 patients, 353 females and 165 males were identified. The average age at diagnosis was 46.3
years (range 9-81 years) with the buccal mucosa being the most common site (87.8%). At initial presentation, white
lichen and red lichen was seen in 52.3% and 47.7% patients, respectively. Of these, 5 (0.96%) patients previously
diagnosed clinically and histopathologically as OLP developed oral cancer. All of them were the females with no
a history of smoking or alcohol use.
Conclusions: Clinical features of eastern Chinese OLP patients were elucidated. Notably, approximately 1% of
OLP developed into cancer, which provides further evidence of potentially malignant nature of OLP
The Photometric System of Tsinghua-NAOC 80-cm Telescope at NAOC Xinglong Observatory
Tsinghua-NAOC (National Astronomical Observatories of China) Telescope
(hereafter, TNT) is an 80-cm Cassegrain reflecting telescope located at
Xinglong bservatory of NAOC, with main scientific goals of monitoring various
transients in the universe such as supernovae, gamma-ray bursts, novae,
variable stars, and active galactic nuclei. We present in this paper a
systematic test and analysis of the photometric performance of this telescope.
Based on the calibration observations on twelve photometric nights, spanning
the period from year 2004 to year 2012, we derived an accurate transformation
relationship between the instrumental magnitudes and standard Johnson
and Cousins magnitudes. In particular, the color terms and the
extinction coefficients of different passbands are well determined. With these
data, we also obtained the limiting magnitudes and the photometric precision of
TNT. It is worthwhile to point out that the sky background at Xinglong
Observatory may become gradually worse over the period from year 2005 to year
2012 (e.g., 21.4 mag vs. 20.1 mag in the V band).Comment: 12 pages,9 figures, accepted by RA

A regulatory mutant on TRIM26 conferring the risk of nasopharyngeal carcinoma by inducing low immune response.
The major histocompatibility complex (MHC) is most closely associated with nasopharyngeal carcinoma (NPC), but the complexity of its genome structure has proven challenging for the discovery of causal MHC loci or genes. We conducted a targeted MHC sequencing in 40 Cantonese NPC patients followed by a two-stage replication in 1065 NPC cases and 2137 controls of Southern Chinese descendent. Quantitative RT-PCR analysis (qRT-PCR) was used to detect gene expression status in 108 NPC and 43 noncancerous nasopharyngeal (NP) samples. Luciferase reporter assay and chromatin immunoprecipitation (ChIP) were used to assess the transcription factor binding site. We discovered that a novel SNP rs117565607_A at TRIM26 displayed the strongest association (OR = 1.909, Pcombined = 2.750 × 10-19 ). We also observed that TRIM26 was significantly downregulated in NPC tissue samples with genotype AA/AT than TT. Immunohistochemistry (IHC) test also found the TRIM26 protein expression in NPC tissue samples with the genotype AA/AT was lower than TT. According to computational prediction, rs117565607 locus was a binding site for the transcription factor Yin Yang 1 (YY1). We observed that the luciferase activity of YY1 which is binding to the A allele of rs117565607 was suppressed. ChIP data showed that YY1 was binding with T not A allele. Significance analysis of microarray suggested that TRIM26 downregulation was related to low immune response in NPC. We have identified a novel gene TRIM26 and a novel SNP rs117565607_A associated with NPC risk by regulating transcriptional process and established a new functional link between TRIM26 downregulation and low immune response in NPC

Effect of 17α-methyltestosterone (MT) on osmoregulatory responses and apoptosis in genetically improved farmed tilapia (GIFT), Oreochromis niloticus (L.)
361-368Androgenic compounds can affect osmoregulatory response and apoptosis in fish. In the present study, we exposed genetically improved farmed tilapia (GIFT) Oreochromis niloticus (L.) to 17α-methyltestosterone (MT, 0.5 and 5 mg/L) for 7, 14 and 21 days for understanding the phenomenon. The activities of Na+/K+ ATPase (NKA) and Ca2+/Mg2+ ATPase (CMA) were measured in the gill, kidney and intestine to evaluate the change in osmoregulation of GIFT, and genotoxicity was also detected. Results showed that organic NKA were significantly decreased in 5 mg/L MT exposure groups. The intestine NKA was significantly increased (0.5 mg/L MT). MT exposures increased the CMA of kidney and intestine (0.5 mg/L), together with gill CMA (5 mg/L MT). The results of genotoxicity assay showed gill atp1a1a and nkcc2 transcripts significantly increased, while intestine atp1a1a and fxyd7 transcripts revealed significant increases for MT exposure groups. Caspases proteins demonstrated significant increases at 7th and 21st day, and their transcripts were enhanced in 0.5 mg/L MT exposure groups. The results have evidently demonstrated that chronic exposure of MT could result in organic osmoregulatory response and hepatic apoptosis in GIFT O. niloticus

Phosphoantigen/IL2 Expansion and Differentiation of Vγ2Vδ2 T Cells Increase Resistance to Tuberculosis in Nonhuman Primates
Dominant Vγ2Vδ2 T-cell subset exist only in primates, and recognize phosphoantigen from selected pathogens including M. tuberculosis(Mtb). In vivo function of Vγ2Vδ2 T cells in tuberculosis remains unknown. We conducted mechanistic studies to determine whether earlier expansion/differentiation of Vγ2Vδ2 T cells during Mtb infection could increase immune resistance to tuberculosis in macaques. Phosphoantigen/IL-2 administration specifically induced major expansion and pulmonary trafficking/accumulation of phosphoantigen-specific Vγ2Vδ2 T cells, significantly reduced Mtb burdens and attenuated tuberculosis lesions in lung tissues compared to saline/BSA or IL-2 controls. Expanded Vγ2Vδ2 T cells differentiated into multifunctional effector subpopulations capable of producing anti-TB cytokines IFNγ, perforin and granulysin, and co-producing perforin/granulysin in lung tissue. Mechanistically, perforin/granulysin-producing Vγ2Vδ2 T cells limited intracellular Mtb growth, and macaque granulysin had Mtb-bactericidal effect, and inhibited intracellular Mtb in presence of perforin. Furthermore, phosphoantigen/IL2-expanded Vγ2Vδ2 T effector cells produced IL-12, and their expansion/differentiation led to enhanced pulmonary responses of peptide-specific CD4+/CD8+ Th1-like cells. These results provide first in vivo evidence implicating that early expansion/differentiation of Vγ2Vδ2 T effector cells during Mtb infection increases resistance to tuberculosis. Thus, data support a rationale for conducting further studies of the γδ T-cell-targeted treatment of established TB, which might ultimately help explore single or adjunctive phosphoantigen expansion of Vγ2Vδ2 T-cell subset as intervention of MDR-tuberculosis or HIV-related tuberculosis
