    Elastin Variants in Two Angiographic PCV Subtypes

    Objective To compare the association of elastin (ELN) gene variants between two different angiographic phenotypes of polypoidal choroidal vasculopathy (PCV). Methods We included 411 treatment-naïve PCV patients and 350 controls in the present study. PCV was classified into two phenotypes (152 Type 1 and 259 Type 2) according to the presence or absence of feeding vessels found in indocyanine-green angiography. Single nucleotide polymorphisms (SNPs) in the ELN region including rs868005, rs884843, rs2301995, rs13239907 and rs2856728 were genotyped using TaqMan Genotyping Assays. Results In the allelic association analyses, rs868005 showed the strongest association with Type 2 PCV (allelic odds ratio 1.56; p = 7.4x10-6), while no SNP was significantly associated with Type 1 PCV. Genotype association analyses revealed the significant association of rs868005 with Type 2 PCV in log additive model and predominant model (odds ratio 1.75; p = 1.5x10-6 and odds ratio 1.60; p = 0.0044, respectively), but not with Type 1 PCV. These findings were further corroborated by another control group in the literature. Conclusions There may be significantly different associations in genetic variants of elastin between two angiographic phenotypes of PCV

    In this study, it was divided into 2 groups by the presence of the food experience of the tablet which mixed Capparis Masaikai (MT), and the sensory evaluation of the tablet of improvement (K-MT) were carried out among the young women, and the effect of the improvement was examined. As the result, the group (S-W group) with the food experience of MT showed the tendency in which K-MT was higher than MT for the evaluation of sweetness, refreshingness, moisture sensation, the time in which sweetness and moisture sensation continued were long, and it was shown that bitterness and astringent taste shorten. In addition, the significance could not be accepted on the preference of the taste under the group (F-W group) without the food experience of MT between K-MT and MT, and the time in which sweetness and moisture sensation continued was long, and it was shown that bitterness and astringent taste shorten. But the total evaluation was also low on tasting and after for the K-MT. From this result, there seemed to be a necessity of improving evaluation of the preference of taste of K~MT. It was shown that sweetness and moisture sensation were long and that the duration of bitterness and astringent taste shortened as an effect of the improvement of MT. The time in which sweetness and moisture sensation continued was long, and it was shown that bitterness and astringent taste shorten as an effect of the improvement of MT

    The E3 Ubiquitin Ligase Activity of Trip12 Is Essential for Mouse Embryogenesis

    Protein ubiquitination is a post-translational protein modification that regulates many biological conditions [1], [2], [3], [4]. Trip12 is a HECT-type E3 ubiquitin ligase that ubiquitinates ARF and APP-BP1 [5], [6]. However, the significance of Trip12 in vivo is largely unknown. Here we show that the ubiquitin ligase activity of Trip12 is indispensable for mouse embryogenesis. A homozygous mutation in Trip12 (Trip12mt/mt) that disrupts the ubiquitin ligase activity resulted in embryonic lethality in the middle stage of development. Trip12mt/mt embryos exhibited growth arrest and increased expression of the negative cell cycle regulator p16 [7], [8], [9], [10]. In contrast, Trip12mt/mt ES cells were viable. They had decreased proliferation, but maintained both the undifferentiated state and the ability to differentiate. Trip12mt/mt ES cells had increased levels of the BAF57 protein (a component of the SWI/SNF chromatin remodeling complex) and altered gene expression patterns. These data suggest that Trip12 is involved in global gene expression and plays an important role in mouse development

    精神科においては精神障害者の生活の質の向上のために退院促進が求められている.そのためにはチームアプローチが不可欠であり,それを実践するためには,各専門職者の持っている能力を統合した活動を展開しなければならない.学際的連携チームケアモデルInterdisciplinary collaborative teamcare model (ICTCM)とは,チームケアを実践するための具体的な方法(教育,理論,実践,研究)を示すものである.そこで,ICTCMを使用して,チームケアサービスの質の改善活動の一環として,看護サービスをレベルアップすることを目的とした多職種参加による看護診断学習会を行ってきた.その結果,チームケア運営体制の整備,職種間の役割理解及び職種間で積極的に情報を共有しようとする姿勢が改善したと考えられた.This article describes workshop on nursing diagnosis that aimed improving an interdisciplinary collaboration in the psychiatric hospital. Psychiatric hospitals are required the promotion of discharge a patients with chronic mental disorders in order to enhance the quality of life of the people with mental disorders. For that purpose, team approach is absolutely imperative. In order to practice team approach, we have to unify the competency of each professional. Interdisciplinary Collaborative Team Care Model (ICTCM)shows the concrete method(education, theory, practice, research)for practicing team care. Our hospitals’ staffs are working based on this model in order to assure continuous improvement of providing efficient and high-quality of team care services. Nursing diagnosis workshop based on this model has been performed which aims to improve nursing services involve the participation of various health care professional. The overall conclusion of the report was that the management system of the team care is well-organized, also understanding of the role of other professions, the active and positive attitude for sharing information between inter-health care providers have improved

    Mitochonic Acid 5 (MA-5) Facilitates ATP Synthase Oligomerization and Cell Survival in Various Mitochondrial Diseases

    Mitochondrial dysfunction increases oxidative stress and depletes ATP in a variety of disorders. Several antioxidant therapies and drugs affecting mitochondrial biogenesis are undergoing investigation, although not all of them have demonstrated favorable effects in the clinic. We recently reported a therapeutic mitochondrial drug mitochonic acid MA-5 (Tohoku J. Exp. Med., 2015). MA-5 increased ATP, rescued mitochondrial disease fibroblasts and prolonged the life span of the disease model “Mitomouse” (JASN, 2016). To investigate the potential of MA-5 on various mitochondrial diseases, we collected 25 cases of fibroblasts from various genetic mutations and cell protective effect of MA-5 and the ATP producing mechanism was examined. 24 out of the 25 patient fibroblasts (96%) were responded to MA-5. Under oxidative stress condition, the GDF-15 was increased and this increase was significantly abrogated by MA-5. The serum GDF-15 elevated in Mitomouse was likewise reduced by MA-5. MA-5 facilitates mitochondrial ATP production and reduces ROS independent of ETC by facilitating ATP synthase oligomerization and supercomplex formation with mitofilin/Mic60. MA-5 reduced mitochondria fragmentation, restores crista shape and dynamics. MA-5 has potential as a drug for the treatment of various mitochondrial diseases. The diagnostic use of GDF-15 will be also useful in a forthcoming MA-5 clinical trial

    Heterogeneous fibroblasts underlie age-dependent tertiary lymphoid tissues in the kidney

    Acute kidney injury (AKI) is a common clinical condition defined as a rapid decline in kidney function. AKI is a global health burden, estimated to cause 2 million deaths annually worldwide. Unlike AKI in the young, which is reversible, AKI in the elderly often leads to end-stage renal disease, and the mechanism that prevents kidney repair in the elderly is unclear. Here we demonstrate that aged but not young mice developed multiple tertiary lymphoid tissues (TLTs) in the kidney after AKI. TLT size was associated with impaired renal function and increased expression of proinflammatory cytokines and homeostatic chemokines, indicating a possible contribution of TLTs to sustained inflammation after injury. Notably, resident fibroblasts from a single lineage diversified into p75 neurotrophin receptor(+) (p75NTR(+)) fibroblasts and homeostatic chemokine-producing fibroblasts inside TLTs, and retinoic acid-producing fibroblasts around TLTs. Deletion of CD4(+) cells as well as late administration of dexamethasone abolished TLTs and improved renal outcomes. Importantly, aged but not young human kidneys also formed TLTs that had cellular and molecular components similar to those of mouse TLTs. Therefore, the inhibition of TLT formation may offer a novel therapeutic strategy for AKI in the elderly.</p

    日本語能力試験の実施機関である国際交流基金と日本国際教育支援協会は「日本語能力試験の改善に関する検討会」を2005年に発足させたが、2009年からの新試験開始を目指して各分科会は日々調査研究を重ねている。本稿は、分科会の一つである出題基準分科会漢字表・語彙表部会が行っている語彙表作成作業の2007年9月現在の中間報告である。 部会の活動は、この2年間で4つの段階を経た。作成方針と選別方針を決定し、データベースに関する調査および整備をする第一段階、語彙の選別をする第二段階、語彙の再選別及び記述方法の検討をする第三段階、語彙の再選別と初出級の検討をする第四段階である。実際にどのようなデータベースを使い、どのような検討を重ね、どのような選別作業をしたのか。本稿ではそれぞれの段階ごとに具体的な例を挙げながら報告をする。また、それとともに今後考えていかなければならない課題についても言及する。The Japan Foundation and Japan Educational Exchanges and Services inaugurated an "Examination Committee on Improving Japanese Language Proficiency Test" in 2005, and have been conducting research on the New Test to be incorporated in 2009. This is an interim report as of September 2007, on the working of vocabulary list which has been made by the kanji and vocabulary list subdivision of the Test Content Specifications division. The activities of the subdivision have been divided into four stages during these two years. The first stage is for decisions on the policy for composition and selection, and research on the database. The second stage is for the selection of vocabulary. The third stage is for discussion on the re-selection and description of vocabulary. The forth stage is for re-selection of vocabulary and for each level to be decided
