    In this final part of this extensive study, a new systematic data-driven fuzzy modelling approach has been developed, taking into account both the modelling accuracy and its interpretability (transparency) as attributes. For the first time, a data-driven modelling framework has been proposed designed and implemented in order to model the intricate FSW behaviours relating to AA5083 aluminium alloy, consisting of the grain size, mechanical properties, as well as internal process properties. As a result, ‘Pareto-optimal’ predictive models have been successfully elicited which, through validations on real data for the aluminium alloy AA5083, have been shown to be accurate, transparent and generic despite the conservative number of data points used for model training and testing. Compared with analytically based methods, the proposed data-driven modelling approach provides a more effective way to construct prediction models for FSW when there is an apparent lack of fundamental process knowledge

    Wideband circularly polarised antenna array with isoflux pattern

    The authors present a wideband circularly polarised antenna array consisting of a 10-element ellipse dipole antenna and a feed network. A metal pillar is vertically loaded at the end of each ellipse dipole arm to extend the current path, thereby reducing the lateral dimension of the antenna element. For uniform coverage over the Earth's surface, antenna with an isoflux pattern is required due to its uniform power density over a wide coverage area. To obtain wideband and isoflux characteristics, the amplitude and phase distributions along the network are optimised by applying the hybrid of the genetic algorithm and particle swarm optimisation. Particularly, the feed network with broadband phase shifting performance is designed to attain a broad bandwidth requirement. The proposed antenna can achieve excellent impedance matching and axial ratio characteristics from 1.1 to 1.6 GHz. In addition, the isoflux radiation pattern is obtained with the beamwidth of ±21° where the gain is higher than 0 dBi in the elevation plane (xoz-Plane). The realised gain is better than 10.2 dBi and the side lobes are below −14.2dB across the operating frequency. Finally, the antenna array is manufactured and measured, and good agreement is achieved between the simulation and measurement results

    Tuning the vertical location of helical surface states in topological insulator heterostructures via dual-proximity effects

    In integrating topological insulators (TIs) with conventional materials, one crucial issue is how the topological surface states (TSS) will behave in such heterostructures. We use first-principles approaches to establish accurate tunability of the vertical location of the TSS via intriguing dual-proximity effects. By depositing a conventional insulator (CI) overlayer onto a TI substrate (Bi2Se3 or Bi2Te3), we demonstrate that, the TSS can float to the top of the CI film, or stay put at the CI/TI interface, or be pushed down deeper into the otherwise structurally homogeneous TI substrate. These contrasting behaviors imply a rich variety of possible quantum phase transitions in the hybrid systems, dictated by key material-specific properties of the CI. These discoveries lay the foundation for accurate manipulation of the real space properties of TSS in TI heterostructures of diverse technological significance

    Minimum Sensitivity Based Robust Beamforming with Eigenspace Decomposition

    An enhanced eigenspace-based beamformer (ESB) derived using the minimum sensitivity criterion is proposed with significantly improved robustness against steering vector errors. The sensitivity function is defined as the squared norm of the appropriately scaled weight vector and since the sensitivity function of an array to perturbations becomes very large in the presence of steering vector errors, it can be used to find the best projection for the ESB, irrespective of the distribution of additive noises. As demonstrated by simulation results, the proposed method has a better performance than the classic ESBs and the previously proposed uncertainty set based approach

    Down-regulated CK8 expression in human intervertebral disc degeneration

    As an intermediate filament protein, cytokeratin 8 (CK8) exerts multiple cellular functions. Moreover, it has been identified as a marker of notochord cells, which play essential roles in human nucleus pulposus (NP). However, the distribution of CK8 positive cells in human NP and their relationship with intervertebral disc degeneration (IDD) have not been clarified until now. Here, we found the percentage of CK8 positive cells in IDD (25.7+/-4.14%) was significantly lower than that in normal and scoliosis NP (51.9+/-9.73% and 47.8+/-5.51%, respectively, p<0.05). Western blotting and qRT-PCR results confirmed the down-regulation of CK8 expression in IDD on both of protein and mRNA levels. Furthermore, approximately 37.4% of cell clusters were CK8 positive in IDD. Taken together, this is the first study to show a down-regulated CK8 expression and the percentage of CK8 positive cell clusters in IDD based upon multiple lines of evidence. Consequently, CK8 positive cells might be considered as a potential option in the development of cellular treatment strategies for NP repair.published_or_final_versio

    Screening of Diagnostic Biomarkers and Immune Infiltration Characteristics Linking Rheumatoid Arthritis and Rosacea Based on Bioinformatics Analysis

Recent research indicates a heightened RA risk among individuals with rosacea. However, the molecular mechanisms linking these diseases remain largely unknown. This study aims to uncover shared molecular regulatory networks and immune cell infiltration patterns in both rosacea and RA.Methods: The gene expression profiles of RA (GSE12021, GSE55457), and the rosacea gene expression profile (GSE6591), were downloaded from Gene Expression Omnibus (GEO) databases, and obtained to screen differentially expressed genes (DEGs) by using “limma” package in R software. Various analyses including GO, KEGG, protein–protein interaction (PPI) network, and weighted gene co-expression network analyses (WGCNA) were conducted to explore potential biological functions and signaling pathways. CIBERSORT was used to assess the abundance of immune cells. Pearson coefficients were used to calculate the correlations between overlapped genes and the leukocyte gene signature matrix. Flow cytometry (FCM) analysis confirmed the most abundant immune cells detected in rheumatoid arthritis and rosacea. Receiver operator characteristic (ROC) analysis, enzyme-linked immunosorbent assay (ELISA), and qRT-PCR were used to confirm biomarkers and functions.Results: Two hundred seventy-seven co-expressed DEGs were identified from these datasets. Functional enrichment analysis indicated that these DEGs were associated with immune processes and chemokine-mediated signaling pathways. Fourteen and 17 hub genes overlapped between cytoHubba and WGCNA were identified in RA and rosacea, respectively. Macrophages and dendritic cells were RA and rosacea’s most abundant immune cells, respectively. The ROC curves demonstrated potential diagnostic values of CXCL10 and CCL27, showing higher levels in the serum of patients with RA or rosacea, and suggesting possible regulation in the densities and functions of macrophages and dendritic cells from RA and rosacea, which were validated by FCM and qRT-PCR.Conclusion: Importantly, our findings may contribute to the scientific basis for biomarkers and therapeutic targets for patients with RA and rosacea in the future.Keywords: autoimmune disease, immune infiltration, macrophage, chemokine, diagnostic biomarker

    Oxidative stress-dependent cyclooxygenase-2-derived prostaglandin F2α impairs endothelial function in renovascular hypertensive rats

    Abstract Aims: The role of endothelium-derived contracting factors (EDCFs) in regulating renovascular function is yet to be elucidated in renovascular hypertension (RH). The current study investigated whether oxidative stress-dependent cyclooxygenase (COX)-2-derived prostaglandin F(2alpha) (PGF(2alpha)) impairs endothelial function in renal arteries of renovascular hypertensive rats (RHR). Results: Renal hypertension was induced in rats by renal artery stenosis of both kidneys using the 2-kidney 2-clip model. Acute treatment with reactive oxygen species (ROS) scavengers, COX-2 inhibitors, and thromboxane-prostanoid receptor antagonists, but not COX-1 inhibitors, improved endothelium-dependent relaxations and eliminated endothelium-dependent contractions in RHR renal arteries. Five weeks of treatment with celecoxib or tempol reduced blood pressure, increased renal blood flow, and restored endothelial function in RHRs. Increased ROS production in RHR arteries was inhibited by ROS scavengers, but unaffected by COX-2 inhibitors; whereas increased PGF(2alpha) release was reduced by both ROS scavengers and COX-2 inhibitors. ROS also induced COX-2-dependent contraction in RHR renal arteries, which was accompanied by the release of COX-2-derived PGF(2alpha). Further, chronic tempol treatment reduced COX-2 and BMP4 upregulation, p38MAPK phosphorylation, and the nitrotyrosine level in RHR renal arteries. Conclusion: These findings demonstrate the functional importance of oxidative stress, which serves as an initiator of increased COX-2 activity, and that COX-2-derived PGF(2alpha) plays an important role in mediating endothelial dysfunction in RH.     Improvement in photovoltaic performance of rutile-phased TiO2 nanorod/nanoflower-based dye-sensitized solar cell

    An improved dye-sensitized solar cell (DSC) of rutile-phased titanium dioxide (TiO2) electrode with increased power conversion efficiency was successfully fabricated. Rutile-phased TiO2 nanorods and nanoflowers were grown directly on fluorine-doped SnO2 (FTO) by simple aqueous chemical growth technique using one-step hydrothermal process. The solution was prepared by mixing hydrochloric acid, deionized water, and titanium butoxide used as precursor. In the preparation of DSC, both TiO2 nanorods and nanoflowers, platinum (Pt), ruthenium dye N719, and DPMII electrolyte were used as photoelectrode, counter electrode, dye solution, and liquid electrolyte, respectively. The prepared rutile-phased TiO2 nanorods and nanoflowers samples were characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM). The DSCs were fabricated based on the rutile-phased titanium dioxide nanorod and nanoflower photoelectrodes. For their energy conversion efficiency, I-V characteristics and electrochemical impedance spectroscopy were studied. We also investigated the effect of cetyltrimethylammonium bromide (CTAB) reaction times 2, 5, and 10 h in the preparation of rutile-phased TiO2 nanoflowers for DSC. CTAB is one of the capping agents that cover the refine surface of nanoparticles and prevent them from coagulation or aggregation. In our final result, the combination of rutile-phased TiO2 nanorod- and nanoflower-based DSCs showed best efficiency at approximately 3.11% due to its good electron transport of TiO2 nanorods and increased surface area by the TiO2 nanoflowers that had increased dye absorption
