    Caracterización de los procesos cognoscitivos y competencias involucrados en los niveles de comprensión lectora en Estudiantes Universitarios

    La presente investigación de carácter descriptivo tuvo como objetivo caracterizar los procesos cognoscitivos involucrados en la lectura y su relación con los distintos niveles de comprensión lectora. Para ello, se estudió una muestra de 124 estudiantes universitarios, quienes cursaban primer semestre de Psicología; los participantes fueron hombres y mujeres, con edades de entre 16 y 30 años de edad, residentes en la ciudad Bogotá. Para caracterizar las operaciones cognoscitivas se usó el Cuestionario de Evaluación de la Comprensión Lectora de Arenas (2007). Los resultados con respecto a los niveles de comprensión fueron analizados con el modelo de Rasch. Además, se usó la metodología Angoff para la especificación de los niveles de competencias. Se concluyó que existen cinco niveles de comprensión lectora. Así mismo, se encontró que la mayor parte de los estudiantes se clasifican en los niveles de lectura literal e inferencial.     Multi-omics in vitro study of the salivary modulation of the goat rumen microbiome

    Ruminants are able to produce large quantities of saliva which enter into the rumen and salivary components exert different physiological functions. Although previous research has indicated that salivary immunoglobulins can partially modulate the rumen microbial activity, the role of the salivary components other than ions on the rumen microbial ecosystem has not been thoroughly investigated in ruminants. To investigate this modulatory activity, a total of 16 semi-continuous in vitro cultures with oats hay and concentrate were used to incubate rumen fluid from four donor goats with autoclaved saliva (AUT) as negative control, saliva from the same rumen fluid donor (OWN) as positive control, and either goat (GOAT) or sheep (SHEEP) saliva as experimental interventions. Fermentation was monitored throughout 7 days of incubation and the microbiome and metabolome were analysed at the end of this incubation by Next-Generation sequencing and liquid chromatography coupled with mass spectrometry, respectively. Characterisation of the proteome and metabolome of the different salivas used for the incubation showed a high inter-animal variability in terms of metabolites and proteins, including immunoglobulins. Incubation with AUT saliva promoted lower fermentative activity in terms of gas production (−9.4%) and highly divergent prokaryotic community in comparison with other treatments (OWN, GOAT and SHEEP) suggesting a modulatory effect derived from the presence of bioactive salivary components. Microbial alpha-diversity at amplicon sequence variant (ASV) level was unaffected by treatment. However, some differences were found in the microbial communities across treatments, which were mostly caused by a greater abundance of Proteobacteria and Rikenellacea in the AUT treatment and lower of Prevotellaceae. These bacteria, which are key in the rumen metabolism, had greater abundances in GOAT and SHEEP treatments. Incubation with GOAT saliva led to a lower protozoal concentration and propionate molar proportion indicating a capacity to modulate the rumen microbial ecosystem. The metabolomics analysis showed that the AUT samples were clustered apart from the rest indicating different metabolic pathways were promoted in this treatment. These results suggest that specific salivary components contribute to host-associated role in selecting the rumen commensal microbiota and its activity. These findings could open the possibility of developing new strategies to modulate the saliva composition as a way to manipulate the rumen function and activity

    Natural and artificial feeding management before weaning promote different rumen microbial colonization but not differences in gene expression levels at the rumen epithelium of newborn goats

    The aim of this work was to evaluate the effect of feeding management during the first month of life (natural with the mother, NAT, or artificial with milk replacer, ART) on the rumen microbial colonization and the host innate immune response. Thirty pregnant goats carrying two fetuses were used. At birth one kid was taken immediately away from the doe and fed milk replacer (ART) while the other remained with the mother (NAT). Kids from groups received colostrum during first 2 days of life. Groups of four kids (from ART and NAT experimental groups) were slaughtered at 1, 3, 7, 14, 21 and 28 days of life. On the sampling day, after slaughtering, the rumen content was sampled and epithelial rumen tissue was collected. Pyrosequencing analyses of the bacterial community structure on samples collected at 3, 7, 14 and 28 days showed that both systems promoted significantly different colonization patterns (P = 0.001). Diversity indices increased with age and were higher in NAT feeding system. Lower mRNA abundance was detected in TLR2, TLR8 and TLR10 in days 3 and 5 compared to the other days (7, 14, 21 and 28). Only TLR5 showed a significantly different level of expression according to the feeding system, presenting higher mRNA abundances in ART kids. PGLYRP1 showed significantly higher abundance levels in days 3, 5 and 7, and then experienced a decline independently of the feeding system. These observations confirmed a highly diverse microbial colonisation from the first day of life in the undeveloped rumen, and show that the colonization pattern substantially differs between pre-ruminants reared under natural or artificial milk feeding systems. However, the rumen epithelial immune development does not differentially respond to distinct microbial colonization patterns.publishersversionPeer reviewe

    Observación de oportunidad de un Giro argelino al sur de Cabo de Palos (Mediterráneo sudoccidental)

    Large anticyclonic eddies can detach from the Algerian Current, forming open-sea Algerian Eddies. These mesoscale structures have been intensively studied by means of sea surface temperature and altimetry data, and using numerical models. However, few studies describe an in situ sampling of their whole vertical structure. Furthermore, the area extending from Cape La Nao (western edge of the Balearic Channels) to the Almería-Orán Front has received very little attention, and it could be considered that there is a gap in our present oceanographic knowledge of this part of the western Mediterranean. An Algerian Eddy lasting for several months was detected in December 2021 to the south of Cape Palos. In order to analyse this eddy, an opportunity sampling was designed taking advantage of the periodic monitoring campaign RADMED 0222. This sampling revealed that the eddy had a baroclinic character, affecting the whole water column. These results suggest that this eddy was generated at the Algerian Current, finally affecting an area close to the eastern Spanish coast. The presence of these structures in this region of the western Mediterranean could alter the southward progression of the Northern Current and even the presence and structure of the Almería-Orán Front.Giros anticiclónicos de gran tamaño pueden desprenderse de la Corriente Argelina, llegando a formar giros en mar abierto. Estas estructuras de mesoescala han sido estudiadas intensamente mediante datos de temperatura superficial del mar, datos de altimetría, y modelos numéricos. Sin embargo, hay pocos trabajos que describan mediante medidas in situ la estructura vertical de estos giros. Al margen de esta circunstancia, la zona que se extiende desde el Cabo La Nao (en el extremo occidental de los Canales Baleares) hasta el Frente Almería-Orán, ha recibido poca atención, pudiéndose considerar que existe una laguna en nuestro conocimiento sobre la oceanografía de esta zona. Un giro anticiclónico fue detectado en diciembre de 2021 al sur de Cabo de Palos, pudiéndose observar durante varios meses. Para analizar este giro se diseñó un muestreo de oportunidad, aprovechando la campaña rutinaria RADMED0222. Este muestreo mostró la estructura baroclina del giro, la cual afectaba a toda su extensión vertical. Los resultados obtenidos también sugieren que el giro se formó en la Corriente Argelina, afectando finalmente a una zona próxima a la costa española. La presencia de este tipo de estructuras en esta región del Mediterráneo Occidental podría afectar a la progresión hacia el sur de la Corriente Septentrional, e incluso a la presencia y estructura del Frente Almería-Orán

    Influencia del bienestar psicológico y subjetivo en el afrontamiento, los hábitos de estudio y uso de TICS en estudiantes universitarios durante la pandemia por COVID-19

    Introduction. Salutogenic and biopsychosocial models encourage promoting wellbeing. Studies recognize that they favor study habits. In light of the COVID-19 pandemic, changes led to researching if wellbeing influenced coping, study habits and the use of Information and Communications Technology (ICT) in university students during the COVID-19 pandemic. Methodology. A quantitative, correlational and cross-sectional study was carried out with undergraduate students of different socioeconomic levels from two private institutions in Bogotá, Colombia. The data was analyzed through the partial least squares approach of structural equations modeling. Results. 212 students participated, whose ages were between 18 and 30 (A=22.74 and SD= 5.55). The model yielded a Standardized Root Mean Squared of 0.079. Path coefficient β and statistical coefficient t demonstrated positive and significant relations between psychological wellbeing and proper coping (β=0.50, t=6.75, P=0.000), study habits (β= 0.49, t=5.49, P=0.000) and the use of ICT (β=0.29, t=2.70, P=0.007), and negative relations to improper coping (β=-0.35, t=4.28, P=0.000). The same applies to the relationship between subjective wellbeing and improper coping (β=-0.23, t=2.76, P=0.006) and study habits (β=-0.21, t=2.09, P=0.037). Discussion. The findings of other research on the influence of wellbeing on favoring mental health were validated. Conclusions. Positive and significant relationships were demonstrated between wellbeing and proper coping, study habits and the use of ICT, and negative relationships were observed with improper coping.Introducción. Los modelos salutogénico y biopsicosocial fomentan la promoción del bienestar. Estudios reconocen que favorece los hábitos de estudio. Ante la pandemia por COVID-19, los cambios llevaron a investigar si el bienestar influye en el afrontamiento, los hábitos de estudio y el uso de las Tecnologías de la Información y la Comunicación (TIC) en estudiantes universitarios durante la pandemia por COVID-19. Metodología. Se realizó una investigación cuantitativa, correlacional y transversal con estudiantes de pregrado de diferentes niveles socioeconómicos, de dos instituciones privadas de Bogotá, Colombia. Los datos se analizaron a través del enfoque de mínimos cuadrados parciales del modelo de ecuaciones estructurales. Resultados. Participaron 212 estudiantes cuyas edades estaban entre 18 y 30 años (M=22.74 y DE= 5.55). Se cumplió el modelo con una Raíz Cuadrada Media Estandarizada de 0.079. Los coeficientes de ruta β y el estadístico t evidenciaron relaciones positivas y significativas entre el bienestar psicológico y el afrontamiento adecuado (β=0.50, t=6.75, P=0.000), los hábitos de estudio (β= 0.49, t=5.49, P=0.000) y el uso de las TIC (β=0.29, t=2.70, P=0.007), y negativas con el afrontamiento inadecuado (β=-0.35, t=4.28, P=0.000). También entre el bienestar subjetivo y el afrontamiento inadecuado (β=-0.23, t=2.76, P=0.006) y los hábitos de estudio (β=-0.21, t=2.09, P=0.037). Discusión. Se validan hallazgos de otras investigaciones sobre la influencia del bienestar para favorecer la salud mental. Conclusiones. Se evidenciaron relaciones positivas y significativas entre el bienestar y el afrontamiento adecuado, los hábitos de estudio y el uso de las TIC, y negativas con el afrontamiento inadecuado

    Analysis of the rumen microbiome and metabolome to study the effect of an antimethanogenic treatment applied in early life of kid goats

    This work aimed to gain insight into the transition from milk to solid feeding at weaning combining genomics and metabolomics on rumen contents from goat kids treated with a methanogenic inhibitor (bromochloromethane, BCM). Sixteen goats giving birth to two kids were used. Eight does were treated (D+) with BCM after giving birth and over 2 months. One kid per doe in both groups was treated with BCM (k+) for 3 months while the other was untreated (k–). Rumen samples were collected from kids at weaning (W) and 1 (W + 1) and 4 (W + 4) months after and from does at weaning and subjected to 16S pyrosequencing and metabolomics analyses combining GC/LC-MS. Results from pyrosequencing showed a clear effect of age of kids, with more diverse bacterial community as solid feed becomes more important after weaning. A number of specific OTUs were significantly different as a result of BCM treatment of the kid at W while at W + 1 and W + 4 less OTUs were significantly changed. At W + 1, Prevotella was increased and Butyrivibrio decreased in BCM treated kids. At W + 4 only the effect of treating mothers resulted in significant changes in the abundance of some OTUs: Ruminococcus, Butyrivibrio and Prevotella. The analysis of the OTUs shared by different treatments revealed that kids at weaning had the largest number of unique OTUs compared with kids at W + 1 (137), W + 4 (238), and does (D) (23). D + k+ kids consistently shared more OTUs with mothers than the other three groups at the three sampling times. The metalobomic study identified 473 different metabolites. In does, lipid super pathway included the highest number of metabolites that were modified by BCM, while in kids all super-pathways were evenly affected. The metabolomic profile of samples from kids at W was different in composition as compared to W + 1 and W + 4, which may be directly ascribed to the process of rumen maturation and changes in the solid diet. This study shows the complexity of the bacterial community and metabolome in the rumen before weaning, which clearly differ from that after weaning and highlight the importance of the dam in transmitting the primary bacterial community after birth.</p

    An Influenza A/H1N1/2009 Hemagglutinin Vaccine Produced in Escherichia coli

    The A/H1N1/2009 influenza pandemic made evident the need for faster and higher-yield methods for the production of influenza vaccines. Platforms based on virus culture in mammalian or insect cells are currently under investigation. Alternatively, expression of fragments of the hemagglutinin (HA) protein in prokaryotic systems can potentially be the most efficacious strategy for the manufacture of large quantities of influenza vaccine in a short period of time. Despite experimental evidence on the immunogenic potential of HA protein constructs expressed in bacteria, it is still generally accepted that glycosylation should be a requirement for vaccine efficacy.We expressed the globular HA receptor binding domain, referred to here as HA(63-286)-RBD, of the influenza A/H1N1/2009 virus in Escherichia coli using a simple, robust and scalable process. The recombinant protein was refolded and purified from the insoluble fraction of the cellular lysate as a single species. Recombinant HA(63-286)-RBD appears to be properly folded, as shown by analytical ultracentrifugation and bio-recognition assays. It binds specifically to serum antibodies from influenza A/H1N1/2009 patients and was found to be immunogenic, to be capable of triggering the production of neutralizing antibodies, and to have protective activity in the ferret model.Projections based on our production/purification data indicate that this strategy could yield up to half a billion doses of vaccine per month in a medium-scale pharmaceutical production facility equipped for bacterial culture. Also, our findings demonstrate that glycosylation is not a mandatory requirement for influenza vaccine efficacy
