Increased expression of pigment epithelium-derived factor in aged mesenchymal stem cells impairs their therapeutic efficacy for attenuating myocardial infarction injury.

AIMS: Mesenchymal stem cells (MSCs) can ameliorate myocardial infarction (MI) injury. However, older-donor MSCs seem less efficacious than those from younger donors, and the contributing underlying mechanisms remain unknown. Here, we determine how age-related expression of pigment epithelium-derived factor (PEDF) affects MSC therapeutic efficacy for MI. METHODS AND RESULTS: Reverse transcriptase-polymerized chain reaction and enzyme-linked immunosorbent assay analyses revealed dramatically increased PEDF expression in MSCs from old mice compared to young mice. Morphological and functional experiments demonstrated significantly impaired old MSC therapeutic efficacy compared with young MSCs in treatment of mice subjected to MI. Immunofluorescent staining demonstrated that administration of old MSCs compared with young MSCs resulted in an infarct region containing fewer endothelial cells, vascular smooth muscle cells, and macrophages, but more fibroblasts. Pigment epithelium-derived factor overexpression in young MSCs impaired the beneficial effects against MI injury, and induced cellular profile changes in the infarct region similar to administration of old MSCs. Knocking down PEDF expression in old MSCs improved MSC therapeutic efficacy, and induced a cellular profile similar to young MSCs administration. Studies in vitro showed that PEDF secreted by MSCs regulated the proliferation and migration of cardiac fibroblasts. CONCLUSIONS: This is the first evidence that paracrine factor PEDF plays critical role in the regulatory effects of MSCs against MI injury. Furthermore, the impaired therapeutic ability of aged MSCs is predominantly caused by increased PEDF secretion. These findings indicate PEDF as a promising novel genetic modification target for improving aged MSC therapeutic efficacy

Banxia Baizhu Tianma Decoction for Essential Hypertension: A Systematic Review of Randomized Controlled Trials

Objectives. To assess the current clinical evidence of Banxia Baizhu Tianma Decoction (BBTD) for essential hypertension (EH). Search Strategy. Electronic databases were searched until July 2012. Inclusion Criteria. We included randomized clinical trials testing BBTD against placebo, antihypertensive drugs, or combined with antihypertensive drugs against antihypertensive drugs. Data Extraction and Analyses. Study selection, data extraction, quality assessment, and data analyses were conducted according to Cochrane standards. Results. 16 randomized trials were included. Methodological quality of the included trials was evaluated as generally low. 2 trials compared prescriptions based on BBTD using alone with antihypertensive drugs. Meta-analysis showed no significant effect of modified BBTD compared with captopril in systolic blood pressure (MD: −0.75 (−5.77, 4.27); P=0.77) and diastolic blood pressure (MD: −0.75 (−2.89, 1.39); P=0.49). 14 trials compared the combination of BBTD or modified BBTD plus antihypertensive drugs with antihypertensive drugs. Meta-analysis showed there are significant beneficial effect on systolic blood pressure in the combination group compare to the antihypertensive drugs (MD: −4.33 (−8.44, −0.22); P=0.04). The safety of BBTD is uncertain. Conclusions. There is encouraging evidence of BBTD for lowering SBP, but evidence remains weak. Rigorously designed trials are warranted to confirm these results

Soil Moisture Retrieval Using BuFeng-1 A/B Based on Land Surface Clustering Algorithm

    Get PDF
A new land surface clustering algorithm is developed to retrieve soil moisture (SM) using the Global Navigation Satellite System reflectometry (GNSS-R) technique. Data from the BuFeng-1 (BF-1) twin satellites A/B, a pilot mission for the Chinese GNSS-R constellation, is used for SM retrieval. The core concept of the algorithm is to cluster global land areas into different types according to the land properties and calculate the SM type by type, based on the linear relationship between equivalent specular reflectivity and SM. The global comparison between the results and SM product from the Soil Moisture Active Passive mission shows the correlation coefficient (R) is 0.82, and unbiased root mean square error (ubRMSE) is 0.070 cm3·cm-3. The results also show good agreement compared with in situ SM measurements with the mean ubRMSE of 0.036 cm3·cm-3. This study proves that the global SM can be retrieved successfully from the BF-1 mission with the land surface clustering algorithm. By taking full advantage of the similarity of land surface physical properties in different regions, the algorithm provides a practical approach for global SM retrieval using spaceborne GNSS-R data.10.13039/501100001809-National Natural Science Foundation of China (Grant Number: 41971377). Ponencia expuesta online en el 2021 Dragon Symposium celebrado el 22 de julio de 2021Respect to the objectives and schedule of our project, the first-year report will include on-going activities and results of Bufeng-1 data processing, calibration workflow, and validation of the calibrated results on hurricane winds, soil moisture, and sea level measurements. The presentation has three parts. Firstly, a short introduction will be given about Bufeng-1 twin satellites that carry the Chinese first generation spaceborne GNSS-R instruments started using reflected GNSS signals to perform earth observation. Secondly, by utilizing the Bufeng-1 Normalized Bistatic Radar Cross Section (NBRCS), earth reflectivity, and range measurements, the preliminary results show that BuFeng-1 has a high agreement compared with other observations on severe sea surface winds, soil moisture, and sea level. In this presentation, the measurements of Bufeng-1 will be aligned with SFMR collected hurricanes, SMAP derived soil moisture, and DTU10 sea level models. Then, the validations of the accuracy and correlation coefficients will be analyzed to discuss the limitations and issues for the future research. For the last part, we will give the outlook about our future works of the objectives and the future plan of Bufeng missions

Identifying remaining oil distribution is an essential study at the Third District in North Saertu of the West Block in Daqing Oilfield. This field is known as a flooding fine potential tapping demonstration zone, characterized by a long-developing history and complex well history. Based on tectonic features and sedimentary characteristics of the study area, the methods of facies controlled reservoir 3D geological modeling and numerical simulation are used in the process of establishing the geological 3D static model. In this paper, we summarized the causes and distribution law of remaining oil in the study area by using the method of fine reservoir numerical simulation to provide a reliable basis for the development and adjustment of the oil field. In combination with fine exploration such as water drive fracturing, water plugging, reperforating and injection-production segment, the recoverable reserves recovery rate could be effectively increased.
