Numerical Simulation of Decomposition of Polymer Nano-composites: Investigation of the Influence of the Char Structure
AbstractIn recent years, nano-particles such as nano-clays, carbon nanotubes and graphenes have been extensively used in flame-retardant polymeric materials. The surface char layer formed in combustion acts as protective barriers that limit the heat transfer into the unpyrolysed polymer and volatilization of combustible degradation products and diffusion of oxygen into the material. A numerical simulation tool Thermakin is used to simulate the thermal decomposition of the neat polymers (polypropylene (PP), Acrylonitrile Butadiene Styrene (ABS)) and corresponding nano-composites (PP/multi-walled carbon nanotube (PP/MWCNT) and ABS/ graphene nano-sheets /NiFe-layered double hydroxide hybrid (ABS/GNS-LDH) in cone calorimetry experiments. PP/MWCNT forms a porous network while ABS/GNS-LDH forms a compact, dense char layer during combustion. With appropriate input parameters, the heat release rates (or mass loss rates) are predicted very well. Finally, the effect of input parameters on model outputs are discussed
Involvement of Lysosome Membrane Permeabilization and Reactive Oxygen Species Production in the Necrosis Induced by Chlamydia muridarum Infection in L929 Cells
Chlamydiae, obligate intracellular bacteria, are associated with a variety of human diseases. The chlamydial life cycle undergoes a biphasic development: replicative reticulate bodies (RBs) phase and infectious elementary bodies (EBs) phase. At the end of the chlamydial intracellular life cycle, EBs have to be released to the surrounded cells. Therefore, the interactions between Chlamydiae and cell death pathways could greatly influence the outcomes of Chlamydia infection. However, the underlying molecular mechanisms remain elusive. Here, we investigated host cell death after Chlamydia infection in vitro, in L929 cells, and showed that Chlamydia infection induces cell necrosis, as detected by the propidium iodide (PI)-Annexin V double-staining flow-cytometric assay and Lactate dehydrogenase (LDH) release assay. The production of reactive oxygen species (ROS), an important factor in induction of necrosis, was increased after Chlamydia infection, and inhibition of ROS with specific pharmacological inhibitors, diphenylene iodonium (DPI) or butylated hydroxyanisole (BHA), led to significant suppression of necrosis. Interestingly, live-cell imaging revealed that Chlamydia infection induced lysosome membrane permeabilization (LMP). When an inhibitor upstream of LMP, CA-074-Me, was added to cells, the production of ROS was reduced with concomitant inhibition of necrosis. Taken together, our results indicate that Chlamydia infection elicits the production of ROS, which is dependent on LMP at least partially, followed by induction of host-cell necrosis. To our best knowledge, this is the first live-cell-imaging observation of LMP post Chlamydia infection and report on the link of LMP to ROS to necrosis during Chlamydia infection. </p
An infectious clone of enterovirus 71(EV71) that is capable of infecting neonatal immune competent mice without adaptive mutations
Enterovirus 71 (EV71) is a major pathogen that causes hand, foot and mouth disease (HFMD), which is a life threatening disease in certain children. The pathogenesis of EV71-caused HFMD is poorly defined due to the lack of simple and robust animal models with severe phenotypes that recapitulate symptoms observed in humans. Here, we generated the infectious clone of a clinical isolate from a severe HFMD patient. Virus rescued from the cDNA clone was infectious in cell lines. When administrated intraperitoneally to neonatal ICR, BALB/c and C57 immune competent mice at a dosage of1.4 × 104 pfu per mouse, the virus caused weight loss, paralysis and death in the infected mice after 4-5 days of infection. In the infected mice, detectable viral replication was detected in various tissues such as heart, liver, brain, lung, kidney, small intestine, leg skeletal muscle and medulla oblongata. The histology of the infected mice included massive myolysis, glomerular atrophy, villous blunting in small intestine, widened alveolar septum, diminished alveolar spaces and lymphocytes infiltration into the lung. By using the UV-inactivated virus as a control, we elucidated that the virus first amplified in the leg skeletal muscle tissue and the muscle tissue served as a primary viral replication site. In summary, we generated a stable EV71 infectious clone that is capable of infecting neonatal immune competent mice without adaptive mutations and provide a simple, valuable animal model for the studies of EV71pathogenesis and therapy.</p

Previous studies suggested that both the frequency and the mean fluorescence intensity (MFI) of cytokine secreting T cells could be of great value for immunogenicity evaluation of a vaccine. In this study, by constructing epitope-based DNA vaccines encoding a previously identified CD8+ T cell epitope, we investigated the influence of multiplying epitope copies on both the frequency and the MFI of specific IFN-γ secreting CD8+ T cells. We found that frequencies of specific CD8+ T cell could be improved by multiplying epitope copies, while the MFI of IFN-γ secreted by epitope-specific CD8+ T cells decreased synchronously. And further analysis showed that the decrease of MFI was not caused by the functional avidity variation of CD8+ T cell receptor
Determining the Quantitative Threshold of High-Frequency Oscillation Distribution to Delineate the Epileptogenic Zone by Automated Detection
Objective: We proposed an improved automated high frequency oscillations (HFOs) detector that could not only be applied to various intracranial electrodes, but also automatically remove false HFOs caused by high-pass filtering. We proposed a continuous resection ratio of high order HFO channels and compared this ratio with each patient's post-surgical outcome, to determine the quantitative threshold of HFO distribution to delineate the epileptogenic zone (EZ).Methods: We enrolled a total of 43 patients diagnosed with refractory epilepsy. The patients were used to optimize the parameters for SEEG electrodes, to test the algorithm for identifying false HFOs, and to calculate the continuous resection ratio of high order HFO channels. The ratio can be used to determine a quantitative threshold to locate the epileptogenic zone.Results: Following optimization, the sensitivity, and specificity of our detector were 66.84 and 73.20% (ripples) and 69.76 and 66.13% (fast ripples, FRs), respectively. The sensitivity and specificity of our algorithm for removing false HFOs were 76.82 and 94.54% (ripples) and 72.55 and 94.87% (FRs), respectively. The median of the continuous resection ratio of high order HFO channels in patients with good surgical outcomes, was significantly higher than in patients with poor outcome, for both ripples and FRs (P < 0.05 ripples and P < 0.001 FRs).Conclusions: Our automated detector has the advantage of not only applying to various intracranial electrodes but also removing false HFOs. Based on the continuous resection ratio of high order HFO channels, we can set the quantitative threshold for locating epileptogenic zones
Toward the shell biorefinery: processing crustacean shell waste using hot water and carbonic acid
Biomass fractionation is a prerequisite for almost any biorefinery process. Yet, a cost-effective and environmentally benign approach to separate biomass feedstock into valuable fractions remain a challenge. Herein we introduce a new fractionation method to extract high-value chitin from crustacean shell (e.g., shrimp shell) using hot water for deproteinization and carbonic acid for demineralization (termed as the HOW-CA process). This method features high deproteinization and demineralization efficiencies (>90%), and the whole process is accomplished within hours. The desired final product chitin exhibits a high purity. This work addresses the major problems associated with the current industrial practice including the employment of corrosive reagents, the destructive removal of a useful component, and the generation of a large amount of waste. Economic and life cycle analyses imply that the HOW-CA process is superior to the conventional method, offering both economic and environmental benefit
