    Salah satu operasi di dalam analisis citra adalah segmentasi citra, yaitu memisahkan objek dari latar belakangnya atau dari objek lain yang tidak menjadi perhatian. Metode sementasi yang sederhana adalah dengan operasi pengambangan (thresholding). Operasi pengambangan menghasilkan citra biner, yang dalam hal ini objek yang diacu di-set berwarna putih sedangkan latar belakangnya di-set berwarna hitam (atau sebaliknya bergantung kasusnya). Makalah ini mempresentasikan penggunaan operasi pengambangan untuk melakukan segmentasi objek. Eksperimen dilakukan dengan menggunakan kakas MATLAB.     目的  针对护理操作技能培训中临床情境不足、易机械性、习惯性模仿的问题,探索提高培训师临床设置下操作技能培训效果的有效教学方法,使操作技能培训更贴近临床。方法  改变管理角色,变教学督导、检查为“对口辅导”的模式对培训过程、教学方法及考核过程进行现场辅导,并举办培训师经验分享、建立培训交流空间及改革质量监控方法等措施;对辅导型管理前后的考核结果进行自身前后对照。结果  对口辅导前后考核平均成绩具有统计学意义(P<0.05),辅导型管理前后科室考核与护理部抽考成绩差异性>±10%的发生例数、临床结合问题不足的例数差异均具有显著性(P<0.001);辅导型管理前后不及格例数差异无显著性(P>0.05)。结论  辅导型管理模式能有效提高培训师临床设置下护理技能培训效果。Objective: In view of the problem of shortage of clinical situation and easy mechanically imitation in nursing skills training , to explore an effective teaching method to improve the trainers’training effect of nursing skills under clinical settings,To make nursing skills more close to clinical. Methods: Change management role from Supervision and inspection to “counterpart counseling”, by performing the teaching process guidance and the examine guidance , establishing the space to share information , reforming the quality control method. to compare the results by setting a self-control between before and after the management of counterpart counseling. Results: Average marks before and after the management of counterpart counseling had a difference (P < 0.05 ),the number of the difference over ±10% between the scores assesmented by trainers in their department and the scores assessed by nursing department had an obvious difference(P < 0.001), The number of the shortage correlating clinical condition had an obvious difference(P < 0.001).The number of failing in the exam before and after the teaching guidance had no differences(P > 0.05). Conclusion: The management coach can effectively improve the effect of training under clinical settings

    Development of a EST dataset and characterization of EST-SSRs in a traditional Chinese medicinal plant, Epimedium sagittatum (Sieb. Et Zucc.) Maxim

    <p>Abstract</p> <p>Background</p> <p><it>Epimedium sagittatum </it>(Sieb. Et Zucc.) Maxim, a traditional Chinese medicinal plant species, has been used extensively as genuine medicinal materials. Certain <it>Epimedium </it>species are endangered due to commercial overexploition, while sustainable application studies, conservation genetics, systematics, and marker-assisted selection (MAS) of <it>Epimedium </it>is less-studied due to the lack of molecular markers. Here, we report a set of expressed sequence tags (ESTs) and simple sequence repeats (SSRs) identified in these ESTs for <it>E. sagittatum</it>.</p> <p>Results</p> <p>cDNAs of <it>E. sagittatum </it>are sequenced using 454 GS-FLX pyrosequencing technology. The raw reads are cleaned and assembled into a total of 76,459 consensus sequences comprising of 17,231 contigs and 59,228 singlets. About 38.5% (29,466) of the consensus sequences significantly match to the non-redundant protein database (E-value < 1e-10), 22,295 of which are further annotated using Gene Ontology (GO) terms. A total of 2,810 EST-SSRs is identified from the <it>Epimedium </it>EST dataset. Trinucleotide SSR is the dominant repeat type (55.2%) followed by dinucleotide (30.4%), tetranuleotide (7.3%), hexanucleotide (4.9%), and pentanucleotide (2.2%) SSR. The dominant repeat motif is AAG/CTT (23.6%) followed by AG/CT (19.3%), ACC/GGT (11.1%), AT/AT (7.5%), and AAC/GTT (5.9%). Thirty-two SSR-ESTs are randomly selected and primer pairs are synthesized for testing the transferability across 52 <it>Epimedium </it>species. Eighteen primer pairs (85.7%) could be successfully transferred to <it>Epimedium </it>species and sixteen of those show high genetic diversity with 0.35 of observed heterozygosity (<it>Ho</it>) and 0.65 of expected heterozygosity (<it>He</it>) and high number of alleles per locus (11.9).</p> <p>Conclusion</p> <p>A large EST dataset with a total of 76,459 consensus sequences is generated, aiming to provide sequence information for deciphering secondary metabolism, especially for flavonoid pathway in <it>Epimedium</it>. A total of 2,810 EST-SSRs is identified from EST dataset and ~1580 EST-SSR markers are transferable. <it>E. sagittatum </it>EST-SSR transferability to the major <it>Epimedium </it>germplasm is up to 85.7%. Therefore, this EST dataset and EST-SSRs will be a powerful resource for further studies such as taxonomy, molecular breeding, genetics, genomics, and secondary metabolism in <it>Epimedium </it>species.</p

    Loop-Mediated Isothermal Amplification Assay Targeting the MOMP Gene for Rapid Detection of Chlamydia psittaci Abortus Strain

    For rapid detection of the Chlamydia psittaci abortus strain, a loop-mediated isothermal amplification (LAMP) assay was developed and evaluated in this study. The primers for the LAMP assay were designed on the basis of the main outer membrane protein (MOMP) gene sequence of C. psittaci. Analysis showed that the assay could detect the abortus strain of C. psittaci with adequate specificity. The sensitivity of the test was the same as that of the nested-conventional PCR and higher than that of chick embryo isolation. Testing of 153 samples indicated that the LAMP assay could detect the genome of the C. psittaci abortus strain effectively in clinical samples. This assay is a useful tool for rapid diagnosis of C. psittaci infection in sheep, swine and cattle

    Demonstration of Adiabatic Variational Quantum Computing with a Superconducting Quantum Coprocessor

    Adiabatic quantum computing enables the preparation of many-body ground states. This is key for applications in chemistry, materials science, and beyond. Realisation poses major experimental challenges: Direct analog implementation requires complex Hamiltonian engineering, while the digitised version needs deep quantum gate circuits. To bypass these obstacles, we suggest an adiabatic variational hybrid algorithm, which employs short quantum circuits and provides a systematic quantum adiabatic optimisation of the circuit parameters. The quantum adiabatic theorem promises not only the ground state but also that the excited eigenstates can be found. We report the first experimental demonstration that many-body eigenstates can be efficiently prepared by an adiabatic variational algorithm assisted with a multi-qubit superconducting coprocessor. We track the real-time evolution of the ground and exited states of transverse-field Ising spins with a fidelity up that can reach about 99%.Comment: 12 pages, 4 figure

    Integration of the metabolome and transcriptome reveals the metabolites and genes related to nutritional and medicinal value in Coriandrum sativum

    Coriandrum sativum (Coriander) or Chinese parsley is a culinary herb with multiple medicinal effects, which is widely used in cooking and traditional medicine. It is enriched with essential oils and anti-oxidant compounds with unknown significance. To explore the untapped reservoir of Coriander, we studied the transcriptome and metabolic profiles from three developmental stages. Here, we identified 10 tyrosine metabolic pathway-related genes (TMPRGs), six porphyrins and chlorophyll metabolic pathway-related genes (PCMPRGs), and five Vitamin E metabolic pathway-related genes (VEMPRGs). These genes were associated with the early development of Coriander. Our analysis suggests that these pathways are involved in the production of critical phenolic metabolites. Furthermore, we constructed the interaction network between these pathway-related genes and transcription factors (TFs), which supported the regulatory pathways for phenolic metabolites. Interestingly, we identified several nutritional or medicinally relevant metabolites, including 59 phenols, two polyamines, 12 alkaloids, and one terpenoid. The higher concentrations of metabolites were from caffeic acid, agmatine, and its derivatives. We found higher levels of caffeic acid and agmatine at 30 days compared to 60 or 90 days. This study provides evidence to stimulate further investigation of the role of these metabolites in medicinal and nutritional research
