    Inhibitory actions of electrolysed water on food spoilage fungi

    Chemical sanitisers are needed for controlling microbial contamination and growth in various settings, including healthcare, domestic settings and the food industry. One promising sanitiser with proposed applications in the food industry is the oxidising sanitiser electrolysed water (EW). For EW applications to be effective, a more in-depth understanding of EW mode-of-action is needed, together with the identification of limiting factors associated with the chemical matrices in relevant EW applications. This study investigated the reactivity of EW with different organic substances. For this purpose, the common food spoilage fungus Aspergillus niger was treated with EW in the presence or absence of the organic materials. The sanitiser retained partial fungicidal activity at high levels of added soil, commonly associated with freshly harvested produce. By narrowing down the reactivity of EW from complex matrices to specific organic molecules, proteins and amino acids were found to strongly suppress EW fungicidal activity. The potential implications of EW reactivity with proteins and amino acids within cells were investigated in the fungal model organism Saccharomyces cerevisiae. This capitalised on the advanced understanding of the uptake and metabolism of diverse compounds and the availability of convenient genetic tools in this model organism. Pre-culture with methionine but not with other tested amino acids increased yeast resistance to subsequent treatments (in the absence of methionine) with sanitisers (EW, sodium hypochlorite, ozonated water). Further tests suggested a direct role of the methionine molecule itself, as opposed to downstream products of methionine metabolism or methionine misincorporation in proteins, in increasing yeast EW resistance. Intracellular methionine oxidation can disturb FeS cluster proteins and this study found that EW treatment impairs FeS cluster protein activity. Because active species in EW can be inactivated by organic compounds, microorganisms may be exposed to low effective EW doses during EW treatments where organic matter is present. When low, sub-lethal EW doses were applied to A. niger, early germination events and colony growth post-treatment were delayed and there was increased variation in size among resultant colonies. The delay and increased variation were non-heritable and were observed with chlorine-containing sanitisers (EW, sodium hypochlorite) but not the chlorine-free oxidant ozonated water. The collective findings led to a hypothesis that chloramine formation within spores during EW treatment may contribute to the observed phenotypes. This study increases the understanding of limiting chemical factors for EW applications and sheds light onto the cellular mode-of-action of EW and the fungal response mechanisms to sanitiser treatments. Such insights can improve the rational development of EW application processes and contribute to understanding and prediction of antimicrobial efficacy in diverse settings

    Top-Down Characterisation of an Antimicrobial Sanitiser, Leading from Quenchers of Efficacy to Mode of Action

    We developed a top-down strategy to characterise an antimicrobial, oxidising sanitiser, which has diverse proposed applications including surface-sanitisation of fresh foods, and with benefits for water resilience. The strategy involved finding quenchers of antimicrobial activity then antimicrobial mode of action, by identifying key chemical reaction partners starting from complex matrices, narrowing down reactivity to specific organic molecules within cells. The sanitiser electrolysed-water (EW) retained partial fungicidal activity against the food-spoilage fungus Aspergillus niger at high levels of added soils (30–750 mg mL-1), commonly associated with harvested produce. Soil with high organic load (approx. 98 mg g-1) gave stronger EW inactivation. Marked inactivation by a complex organics mix (YEPD medium) was linked to its protein-rich components. Addition of pure proteins or amino acids (≀1 mg mL-1) fully suppressed EW activity. Mechanism was interrogated further with the yeast model, corroborating marked suppression of EW action by the amino acid methionine. Pre-culture with methionine increased resistance to EW, sodium hypochlorite, or chlorine-free ozonated water. Overexpression of methionine sulfoxide reductases (which reduce oxidised methionine) protected against EW. Fluoroprobe-based analyses indicated that methionine and cysteine inactivate free chlorine species in EW. Intracellular methionine oxidation can disturb cellular FeS-clusters and we showed that EW treatment impairs FeS-enzyme activity. The study establishes the value of a top-down approach for multi-level characterisation of sanitiser efficacy and action. The results reveal proteins and amino acids as key quenchers of EW activity and, among the amino acids, the importance of methionine oxidation and FeS-cluster damage for antimicrobial mode-of-action

    Acute cytomegalovirus infection modulates the intestinal microbiota and targets intestinal epithelial cells

    Primary and recurrent cytomegalovirus (CMV) infections frequently cause CMV colitis in immunocompromised as well as inflammatory bowel disease (IBD) patients. Additionally, colitis occasionally occurs upon primary CMV infection in patients who are apparently immunocompetent. In both cases, the underlying pathophysiologic mechanisms are largely elusive - in part due to the lack of adequate access to specimens. We employed the mouse cytomegalovirus (MCMV) model to assess the association between CMV and colitis. During acute primary MCMV infection of immunocompetent mice, the gut microbial composition was affected as manifested by an altered ratio of the Firmicutes to Bacteroidetes phyla. Interestingly, these microbial changes coincided with high-titer MCMV replication in the colon, crypt hyperplasia, increased colonic pro-inflammatory cytokine levels, and a transient increase in the expression of the antimicrobial protein Regenerating islet-derived protein 3 gamma (Reg3Îł). Further analyses revealed that murine and human intestinal epithelial cell lines, as well as primary intestinal crypt cells and organoids represent direct targets of CMV infection causing increased cell death. Accordingly, in vivo MCMV infection disrupted the intestinal epithelial barrier and increased apoptosis of intestinal epithelial cells. In summary, our data show that CMV transiently induces colitis in immunocompetent hosts by altering the intestinal homeostasis

    Evolving challenges and strategies for fungal control in the food supply chain

    Fungi that spoil foods or infect crops can have major socioeconomic impacts, posing threats to food security. The strategies needed to manage these fungi are evolving, given the growing incidence of fungicide resistance, tightening regulations of chemicals use and market trends imposing new food-preservation challenges. For example, alternative methods for crop protection such as RNA-based fungicides, biocontrol, or stimulation of natural plant defences may lessen concerns like environmental toxicity of chemical fungicides. There is renewed focus on natural product preservatives and fungicides, which can bypass regulations for ‘clean label’ food products. These require investment to find effective, safe activities within complex mixtures such as plant extracts. Alternatively, physical measures may be one key for fungal control, such as polymer materials which passively resist attachment and colonization by fungi. Reducing or replacing traditional chlorine treatments (e.g. of post-harvest produce) is desirable to limit formation of disinfection by-products. In addition, the current growth in lower sugar food products can alter metabolic routing of carbon utilization in spoilage yeasts, with implications for efficacy of food preservatives acting via metabolism. The use of preservative or fungicide combinations, while involving more than one chemical, can reduce total chemicals usage where these act synergistically. Such approaches might also help target different subpopulations within heteroresistant fungal populations. These approaches are discussed in the context of current challenges for food preservation, focussing on pre-harvest fungal control, fresh produce and stored food preservation. Several strategies show growing potential for mitigating or reversing the risks posed by fungi in the food supply chain

This study increases the understanding of limiting chemical factors for EW applications and sheds light onto the cellular mode-of-action of EW and the fungal response mechanisms to sanitiser treatments. Such insights can improve the rational development of EW application processes and contribute to understanding and prediction of antimicrobial efficacy in diverse settings

    3,5-Diiodo-L-Thyronine (3,5-T2) Exerts Thyromimetic Effects on Hypothalamus-Pituitary-Thyroid Axis, Body Composition, and Energy Metabolism in Male Diet-Induced Obese Mice

    Effective and safe antiobesity drugs are still needed in face of the obesity pandemic worldwide. Recent interventions in rodents revealed 3,5-diiodo-L-thyronine (3,5-T2) as a metabolically active iodothyronine affecting energy and lipid metabolism without thyromimetic side effects typically associated with T3 administration. Accordingly, 3,5-T2 has been proposed as a potential hypolipidemic agent for treatment of obesity and hepatic steatosis. In contrast to other observations, our experiments revealed dose-dependent thyromimetic effects of 3,5-T2 akin to those of T3 in diet-induced obese male C57BL/6J mice. 3,5-T2 treatment exerted a negative feedback regulation on the hypothalamus-pituitary-thyroid axis, similar to T3. This is demonstrated by decreased expression of genes responsive to thyroid hormones (TH) in pituitary resulting in a suppressed thyroid function with lower T4 and T3 concentrations in serum and liver of 3,5-T2-treated mice. Analyses of hepatic TH target genes involved in lipid metabolism revealed T3-like changes in gene expression and increased type I-deiodinase activity after application of 3,5-T2 (2.5 ÎŒg/g body weight). Reduced hepatic triglyceride and serum cholesterol concentrations reflected enhanced lipid metabolism. Desired increased metabolic rate and reduction of different fat depots were, however, compromised by increased food intake preventing significant body weight loss. Moreover, enlarged heart weights indicate potential cardiac side effects of 3,5-T2 beyond hepatic thyromimetic actions. Altogether, the observed thyromimetic effects of 3,5-T2 in several mouse TH target tissues raise concern about indiscriminate administration of 3,5-T2 as powerful natural hormone for the treatment of hyperlipidemia and pandemic obesity

    Metal budget and origin of aqueous brines depositing deep-seated Zn-Pb mineralization linked to hydrocarbon reservoirs, North German Basin

    The origin, evolution, and interplay of brine and hydrocarbon fluid systems play a crucial role in the formation of deep sediment-hosted base metal ore deposits. Here we investigate ratios of halogens, noble gases, stable C and S isotopes, and metal budgets of aqueous brines, which deposited deep-seated and near-surface hydrothermal Zn-Pb mineralization hosted by Zechstein carbonates in the Lower Saxony Basin (North German Basin), by studies of fluid inclusions in sphalerite and quartz. Major and trace element geochemistry and noble gas isotopic signatures of brine inclusions revealed that the ore-forming fluids were highly reactive and experienced prolonged interactions with host rocks in the constricted, over-pressured metal source regions and consequently evolved from near-neutral, oxidized brines towards more reduced, acidic high-salinity brines. Quartz-hosted halite-saturated fluid inclusions with T-h <200 degrees C contain Zn and Pb concentrations up to ca. 9400 mu g g(-1) and 5200 mu g g(-1), respectively, and indicate the efficiency of metal scavenging processes. The interactions with Westphalian coals and C-org-rich shales influenced the redox state as well as the trace and critical element budget of the sphalerite-hosted fluid inclusions, with enrichment in Ge, Pd, Sb, Tl, Bi, and Ag. The salinities of metalliferous fluids originated primarily from seawater evaporation, however in addition a significant halite-dissolution component is present in the southern part of the Lower Saxony Basin. High concentrations of radiogenic noble gases and potassium in the sphalerite-hosted fluid inclusions are ascribed to strong interactions with the Paleozoic siliciclastic sedimentary pile and crystalline basement rocks. Reflux of the strongly modified, sulfur-poor, Zn-Pb-bearing acidic brines, proceeded via re-activated structurally controlled pathways into sour gas or gas-saturated brine pools in the Zechstein Ca2 carbonate unit. Here, mixing of the ascending metal-rich brines with H2S derived from thermochemical sulfate reduction (TSR), resulted in the deposition of deep-seated Zn-Pb ores in the Lower Saxony Basin. The overall timing of the Zn-Pb ore formation can be constrained to the Upper Cretaceous basin inversion.ISSN:0026-4598ISSN:1432-186
