    Acquisition of photoautotrophy in kleptoplastic dinoflagellates-Nusuttodinium aeruginosum/acidotum as a case study

    The integration of stable plastids by endosymbiosis led to an enormous diversity of photosynthetic eukaryotic organisms on earth. However, the steps during the establishment of a stable endosymbiosis are poorly understood. To address this early step of evolution, many studies have been performed on organisms with transient plastids. Originally heterotrophic, are these organisms able to acquire photoautotrophy through the uptake of photosynthetic prey. In contrast to being digested, the prey preserves its photosynthetic ability and supplies the host with photosynthetic products. The prey can either be reduced to an endosymbiont or even to a single plastid. In the latter case the plastids are stolen from photosynthetic prey and therefore called ‘kleptoplasts’. Kleptoplasts are widespread among eukaryotes and occur in multicellular as well as in unicellular organisms. One of the most prominent groups harboring kleptoplasts is the dinoflagellates. This study focuses on freshwater isolates of the kleptoplastic genus Nusuttodinium with a special focus on the species N. aeruginosum/acidotum that acquires its kleptoplasts from the blue-green cryptophyte genus Chroomonas. Stable axenic co-cultures from different locations were established for N. aeruginosum/acidotum and for another species N. amphidinoides and their live cycles were studied. In contrast to all other systems described until now, N. aeruginosum/acidotum is able to harbor kleptoplasts of different morphology and stability making it an ideal model system to study possible steps during the acquisition of stable permanent plastids. Depending on the phylogenetic position of the prey either ‘granular’ or ‘U-shaped’ kleptoplasts are formed. Granular kleptoplasts are characterized morphologically by their multiple number and small size. Each kleptoplast is associated with a small cryptophyte nucleus, the ‘kleptokaryon’ and resembles recently ingested prey. Kleptokarya are stable for up to 10 days and kleptoplasts for up to 14 days. In the granular stage cells took up prey permanently, especially just after cell division that takes place during the dark phase. In contrast, the U-shaped situation is characterized by just one large U-shaped kleptoplast filling out the cell and a single large kleptokaryon located antapically in the curve of the U. Compared to the free living prey and the granular situation, the kleptoplast increases up to 20 fold in size and the kleptokaryon up to 16 fold. Whereas the kleptoplast is distributed equally to both daughter cells during cell division, just one inherits the kleptokaryon. A stability of up to 50 days was recorded for the kleptokaryon and up to 60 days for the kleptoplast. DNA fluorescence measurements and a PCR based approach with primers specific to the cryptophyte nucleus indicate polyploidization of the cryptophyte nucleus after ingestion by the dinoflagellate. In addition enlarged kleptokarya increase kleptoplast stability and influence the behavior of N.acidotum/aeruginosum. Prey uptake is inhibited by intact kleptokarya and dinoflagellates mostly show straight swimming behavior that clearly differs from the behavior in the granular situation. If prey availability is very high N. aeruginosum/acidotum does not form U-shaped kleptoplasts but instead forms granular kleptoplasts that are digested if prey availability remains high or, if prey availability decreases, are distributed equally to the daughter cells in which they develop into stable U-shaped kleptoplasts. In contrast to N. aeruginosum/acidotum, N. amphidinoides represents a more primitive stage during plastid acquisition as kleptokarya are digested shortly after ingestion and kleptoplasts are less modified and stable than the U-shaped plastids. Comparing the ‘primitive’ granular with the ‘advanced’ U-shaped situation with respect to literature allows the formulation of a new hypothesis after which prey scarcity is a trigger and polyploidization one of the most important steps towards the acquisition of stable plastids

    Additive Manufacturing and Vulcanization of Carbon Black Filled Natural Rubber Based Components

    Additive manufacturing of thermoplastics or metals is a well-approved sustainable process for obtaining rapidly precise and individual technical components. Except for crosslinked silicone rubber or thermoplastic elastomers, there is no method of additive manufacturing of elastomers. Based on the development of the additive manufacturing of elastomers (AME) process, the material group of rubber-based cured elastomers may gain first access to the process field of three-dimensional (3D) printing. Printing and crosslinking of rubber is separated into two steps. In the first step, printing is realized by extrusion of the rubber by using a twin-screw extruder, which works according to the derived fused-filament-fabrication principle. In the second step, the component is vulcanized in a high-pressure hot-air autoclave. Because of the plastic flow behavior of non–crosslinked rubber materials, a thermoplastic shell is probably needed to maintain the geometry and position of the additively manufactured rubber. In this way, one layer of thermoplastic and one layer of rubber are printed alternatingly until the component is finished. Afterward, the manufactured binary component is placed in an autoclave to obtain the elastomer after vulcanization under a hot-air and high-pressure atmosphere. Then, the thermoplastic shell is removed from the elastomer and can subsequently be recycled. As compared with conventional thermoplastics, the high viscosity of rubber during processing and its instable shape after extrusion are challenging factors in the development of the AME. This contribution will show a modified 3D printer; explain the printing process from the designed component, via shell generation, to the vulcanized component; and show first printed components

    Tailoring the Curing Kinetics of NBR-Based Rubber Compounds for Additive Manufacturing of Rod Seals

    The additive manufacturing (AM) of elastomeric parts based on high-viscosity reinforced rubbers has increasingly become a topic of scientific research in recent years. In addition to the viscosity, which is several decades higher during processing than the viscosities of thermoplastics, the flowability of the compound after the printing process and the necessary chemical crosslinking of the printed component play a decisive role in producing an elastic, high-quality, and geometrically stable part. After the first technological achievements using the so-called additive manufacturing of elastomers (AME) process, the knowledge gained has to be transferred first to concrete industrial parts. Therefore, in this study, the cure kinetics of a conventional rubber compound are tailored to match the specific requirements for scorch safety in the additive manufacturing of an industrial 2-component rod seal based on an acrylonitrile butadiene rubber O-ring in combination with a thermoplastic polyurethane as the base body. Experimental tests on a test rig for rod seals demonstrate the functionality of this additively manufactured 2-component rod seal

    Unchaining Collective Intelligence for Science, Research and Technology Development by Blockchain-Boosted Community Participation

    Since its launch just over a decade ago by the cryptocurrency Bitcoin, the distributed ledger technology (DLT) blockchain has followed a breathtaking trajectory into manifold application spaces. This paper analyses how key factors underpinning the success of this ground-breaking “internet of value” technology, such as staking of collateral (“skin in the game”), competitive crowdsourcing, crowdfunding, and prediction markets, can be applied to substantially innovate the legacy organization of science, research and technology development (RTD). Here, we elaborate a highly integrative, community-based strategy where a token-based crypto-economy supports finding best possible consensus, trust and truth through adding unconventional elements known from reputation systems, betting, secondary markets and social networking. These tokens support the holder’s formalized reputation, and are used in liquid-democracy style governance and arbitration within projects or community-driven initiatives. This participatory research model serves as a solid basis for comprehensively leveraging collective intelligence by effectively incentivizing contributions from the crowd, such as intellectual property (IP), work, validation, assessment, infrastructure, education, assessment, governance, publication, and promotion of projects. On the analogy of its current blockbusters like peer-to-peer structured decentralized finance (“DeFi”), blockchain technology can seminally enhance the efficiency of science and RTD initiatives, even permitting to fully stage operations as a chiefless Decentralised Autonomous Organization (DAOs)

    Genome analysis of the pico-eukaryotic marine green algaPrasinoderma colonialeCCMP 1413 unveils the existence of a novel phylum within green plants (Viridiplantae), the Prasinodermophyta, which diverged before the split of Chlorophyta and Streptophyta. Structural features of the genome and gene family comparisons revealed an intermediate position of theP. colonialegenome (25.3 Mb) between the extremely compact, small genomes of picoplanktonic Mamiellophyceae (Chlorophyta) and the larger, more complex genomes of early-diverging streptophyte algae. Reconstruction of the minimal core genome of Viridiplantae allowed identification of an ancestral toolkit of transcription factors and flagellar proteins. Adaptations ofP. colonialeto its deep-water, oligotrophic environment involved expansion of light-harvesting proteins, reduction of early light-induced proteins, evolution of a distinct type of C(4)photosynthesis and carbon-concentrating mechanism, synthesis of the metal-complexing metabolite picolinic acid, and vitamin B-1, B(7)and B(12)auxotrophy.     In plants, two independent serine biosynthetic pathways, the photorespiratory and glycolytic phosphoserine (PS) pathways, have been postulated. Although the photorespiratory pathway is well characterized, little information is available on the function of the PS pathway in plants. Here, we present a detailed characterization of phosphoglycerate dehydrogenases (PGDHs) as components of the PS pathway in Arabidopsis thaliana. All PGDHs localize to plastids and possess similar kinetic properties, but they differ with respect to their sensitivity to serine feedback inhibition. Furthermore, analysis of pgdh1 and phosphoserine phosphatase mutants revealed an embryo-lethal phenotype and PGDH1-silenced lines were inhibited in growth. Metabolic analyses of PGDH1-silenced lines grown under ambient and high CO2 conditions indicate a direct link between PS biosynthesis and ammonium assimilation. In addition, we obtained several lines of evidence for an interconnection between PS and tryptophan biosynthesis, because the expression of PGDH1 and PHOSPHOSERINE AMINOTRANSFERASE1 is regulated by MYB51 and MYB34, two activators of tryptophan biosynthesis. Moreover, the concentration of tryptophan-derived glucosinolates and auxin were reduced in PGDH1-silenced plants.
