    The work of making Malta : the Council of Europe's Archaeology and Planning Committee 1988-1996

    This article began life as a paper in the session ‘Opening Doors for Archaeologists: Making Malta Work’ at the 2006 EAA Annual Meeting in Kracow, Poland. It explores the background of the European Convention on the Protection of the Archaeological Heritage (Revised), also known as the Valletta Convention or the Convention of Malta. The article examines some of the major issues that were discussed and describes the drafting process of the Convention from the author’s personal perspective as one of the members of the committee responsible. It concludes with a brief consideration of some subsequent developments and a plea for a more active role for the EAA at Strasbourg and Brussels.     Clinical problems with the performance of euthanasia and physician-assisted suicide in The Netherlands

    BACKGROUND AND METHODS: The characteristics and frequency of clinical problems with the performance of euthanasia and physician-assisted suicide are uncertain. We analyzed data from two studies of euthanasia and physician-assisted suicide in The Netherlands (one conducted in 1990 and 1991 and the other in 1995 and 1996), with a total of 649 cases. We categorized clinical problems as technical problems, such as difficulty inserting an intravenous line; complications, such as myoclonus or vomiting; or problems with completion, such as a longer-than-expected interval between the administration of medications and death. RESULTS: In 114 cases, the physician's intention was to provide assistance with suicide, and in 535, the intention was to perform euthanasia. Problems of any type were more frequent in cases of assisted suicide than in cases of euthanasia. Complications occurred in 7 percent of cases of assisted suicide, and problems with completion (a longer-than-expected time to death, failure to induce coma, or induction of coma followed by awakening of the patient) occurred in 16 percent of the cases; complications and problems with completion occurred in 3 percent and 6 percent of cases of euthanasia, respectively.     De Zuid-Limburgse hellingschraallanden behoren tot de meest soortenrijke graslandtypen in ons land. Ze liggen op een gradiënt van zure heischrale graslanden bovenaan de helling en basische kalkgraslanden in het middendeel van de helling. Onderaan de helling komen voedselrijkere en dus meer ruige graslandtypen voor. Het oppervlak aan goed ontwikkelde hellingschraallanden is in de 20e eeuw sterk achteruit gegaan en ook de kwaliteit ervan. Om na te gaan wat de oorzaken zijn voor de achteruitgang van flora als fauna en hoe verder herstelbeheer moet plaatsvinden, is in 2005 in het kader van OBN een vierjarig onderzoek gestart. Hieruit blijkt dat er zeker nog perspectief is voor herstel van soortenrijke hellingsschraallanden. Het beheer binnen de hellingschraallanden kan verder geoptimaliseerd worden, zodat meer afvoer van nutriënten plaatsvindt en een meer heterogene vegetatiestructuur ontstaat. Ook moet de sterke mate van versnippering en isolatie van de hellingschraallanden worden aangepakt door het vergroten en onderling verbinden van de huidige reservaten

    AbstractIsolated complex I deficiency is the most common enzymatic defect of the oxidative phosphorylation (OXPHOS) system, causing a wide range of clinical phenotypes. We reported before that the rates at which reactive oxygen species (ROS)-sensitive dyes are converted into their fluorescent oxidation products are markedly increased in cultured skin fibroblasts of patients with nuclear-inherited isolated complex I deficiency. Using video-imaging microscopy we show here that these cells also display a marked increase in NAD(P)H autofluorescence. Linear regression analysis revealed a negative correlation with the residual complex I activity and a positive correlation with the oxidation rates of the ROS-sensitive dyes 5-(and-6)-chloromethyl-2′,7′-dichlorodihydrofluorescein and hydroethidine for a cohort of 10 patient cell lines. On the other hand, video-imaging microscopy of cells expressing reduction–oxidation sensitive GFP1 in either the mitochondrial matrix or cytosol showed the absence of any detectable change in thiol redox state. In agreement with this result, neither the glutathione nor the glutathione disulfide content differed significantly between patient and healthy fibroblasts. Finally, video-rate confocal microscopy of cells loaded with C11-BODIPY581/591 demonstrated that the extent of lipid peroxidation, which is regarded as a measure of oxidative damage, was not altered in patient fibroblasts.     Regulation and quantification of cellular mitochondrial morphology and content

    Mitochondria play a key role in signal transduction, redox homeostasis and cell survival, which extends far beyond their classical functioning in ATP production and energy metabolism. In living cells, mitochondrial content (“mitochondrial mass”) depends on the cell-controlled balance between mitochondrial biogenesis and degradation. These processes are intricately linked to changes in net mitochondrial morphology and spatiotemporal positioning (“mitochondrial dynamics”), which are governed by mitochondrial fusion, fission and motility. It is becoming increasingly clear that mitochondrial mass and dynamics, as well as its ultrastructure and volume, are mechanistically linked to mitochondrial function and the cell. This means that proper quantification of mitochondrial morphology and content is of prime importance in understanding mitochondrial and cellular physiology in health and disease. This review first presents how cellular mitochondrial content is regulated at the level of mitochondrial biogenesis, degradation and dynamics. Next we discuss how mitochondrial dynamics and content can be analyzed with a special emphasis on quantitative live-cell microscopy strategies.acceptedVersio
