    Characterisation of the bacterial flora associated with the grey field slug Deroceras reticulatum and assessment of its suitability as a target for biological control

    The field slug Deroceras reticulatum is a major pest in UK agriculture and amidst growing concern and regulatory pressures surrounding chemical molluscicides, innovation is required to advance the current repertoire of slug controls. This study set out to investigate the bacteria associated with D. reticulatum to assess their importance to the slug and potential as a target for biological control. Slug gut bacterial isolates identified using the phenotypical API system (BioMérieux) and 16S rRNA gene sequencing, were mainly soil-dwelling organisms of the phyla Proteobacteria and Bacteriodetes some of which may be important in human or plant disease. A ribosomal intergenic spacer analysis (RISA) was developed to study microbial communities in the slug gut. Slugs had an average species richness of 12 and comparing the bacterial communities in slugs from different locations yielded a mean similarity of 0.159 (Jaccard index) which was significantly lower than similarity indices of slugs collected within a single location (Mean Jaccard index 0.205, p<0.001, ANOVA). Cloning and sequencing of RISA bands common to slugs and slug eggs, but absent from the surrounding soil and plants identified bacteria for future investigation as potential beneficial symbionts. Bacteria extracted from the slug gut were tested for sensitivity to 16 antibiotics and greatest inhibition of growth was observed for chloramphenicol, gentamicin and tetracycline. These antibiotics administered to slugs by feeding and injection caused a reduction in gut-associated bacteria in plate counts, and in bacterial 16S rDNA quantities estimated by real-time quantitative PCR. Field collected D. reticulatum has a large transient gut bacterial population which is reduced upon starvation to a low background level. No significant detrimental effect of antibiotic treatment on the fitness and survival of the slugs was seen, in some instances control slugs suffered greater mortality than slugs that had been injected with antibiotic. Slugs that died during bioassays had a significantly greater amount of bacterial 16S rDNA in their gut than slugs that were sacrificed as healthy individuals suggesting the presence of a bacterial pathogen. This study has found little evidence that a bacterial symbiont may exist and be important for optimal fitness and survival of D. reticulatum, but insight into slug associated bacteria will be valuable in the direction of future studies in this field

    New Limits on Sterile Neutrinos from Suzaku Observations of the Ursa Minor Dwarf Spheroidal Galaxy

    We present results of our search for X-ray line emission associated with the radiative decay of the sterile neutrino, a well-motivated dark matter candidate, in Suzaku Observatory spectra of the Ursa Minor dwarf spheroidal galaxy. These data represent the first deep observation of one of these extreme mass-to-light systems and the first dedicated dark matter search using an X-ray telescope. No such emission line is positively detected, and we place new constraints on the combination of the sterile neutrino mass and the active-sterile neutrino oscillation mixing angle. Line flux upper limits are derived using a maximum-likelihood-based approach that, along with the lack of intrinsic X-ray emission, enables us to minimize systematics and account for those that remain. The limits we derive match or approach the best previous results over the entire 1--20 keV mass range from a single Suzaku observation. These are used to place constraints on the existence of sterile neutrinos with given parameters in the general case and in the case where they are assumed to constitute all of the dark matter. The range allowed implies that sterile neutrinos remain a viable candidate to make up some -- or all -- of the dark matter and also explain pulsar kicks and various other astrophysical phenomena.Comment: revised to closely match version to be published in ApJ v. 69

    Novel octaketide macrolides related to 6-deoxyerythronolide B provide evidence for iterative operation of the erythromycin polyketide synthase

    AbstractBackground: The macrolide antibiotic erythromycin A, like other complex aliphatic polyketides, is synthesised by a bacterial modular polyketide synthase (PKS). Such PKSs, in contrast to other fatty acid and polyketide synthases which work iteratively, contain a separate set or module of enzyme activities for each successive cycle of polyketide chain extension, and the number and type of modules together determine the structure of the polyketide product. Thus, the six extension modules of the erythromycin PKS (DEBS) together catalyse the production of the specific heptaketide 6-deoxyerythronolide B.Results: A mutant strain of the erythromycin producer Saccharopolyspora erythraea, which accumulates the aglycone intermediate erythronolide B, was found unexpectedly to produce two novel octaketides, both 16-membered macrolides. These compounds were detectable in fermentation broths of wild-type S. erythraea, but not in a strain from which the DEBS genes had been specifically deleted. From their structures, both of these octaketides appear to be aberrant products of DEBS in which module 4 has ‘stuttered’, that is, has catalysed two successive cycles of chain extension.Conclusions: The isolation of novel DEBS-derived octaketides provides the first evidence that an extension module in a modular PKS has the potential to catalyse iterative rounds of chain elongation like other type I FAS and PKS systems.     Flat-spectrum symmetric objects with ~1 kpc sizes I. The candidates

    In order to understand the origin and evolution of radio galaxies, searches for the youngest such sources have been conducted. Compact-medium symmetric objects (CSO-MSOs) are thought to be the earliest stages of radio sources, with possible ages of <10^3 yrs for CSOs (<1 kpc in size) and 10^4-10^5 yrs for MSOs (1-15 kpc). From a literature selection in heterogeneous surveys, we have established a sample of 37 confirmed CSOs. In addition, we only found three confirmed flat-spectrum MSOs in the literature. The typical CSO resides on a z<0.5 galaxy, has a flat radio spectrum (a_thin<0.5; S_v proportional to v^-a), is <0.3 kpc in size, has an arm length ratio <2, and well-aligned (theta<20 deg) opposite lobes with a flux density ratio <10. In order to populate the 0.3-1 kpc size range (large CSOs) and also in order to find more flat-spectrum MSOs, we have built a sample of 157 radio sources with a_{1.40}^{4.85}<0.5 that were resolved with the VLA-A 8.4 GHz. As first results, we have 'rediscovered' nine of the known CSO/MSOs while identifying two new ~14 kpc MSOs and two candidate CSO/MSOs (which only lack redshifts for final classification). We were able to reject 61 of the remaining 144 objects from literature information alone. In the series of papers that starts with this one we plan to classify the remaining 83 CSO/MSO candidates (thanks to radio and optical observations) as well as characterize the physical properties of the (likely) many 0.3-15 kpc flat-spectrum CSO/MSOs to be found.Comment: 19 pages, 14 figures, 6 tables (note that Table 2, in landscape format, has a separate file); accepted by MNRA

    Reducing alcohol harms whilst minimising impact on hospitality businesses: ‘Sweetspot’ policy options

    Background During COVID-19, hospitality businesses (e.g. bars, restaurants) were closed/restricted whilst off-sales of alcohol increased, with health consequences. Post-covid, governments face lobbying to support such businesses, but many health services remain under pressure. We appraised ‘sweetspot’ policy options: those with potential to benefit public services and health, whilst avoiding or minimising negative impact on the hospitality sector. Methods We conducted rapid non-systematic evidence reviews using index papers, citation searches and team knowledge to summarise the literature relating to four possible ‘sweetspot’ policy areas: pricing interventions (9 systematic reviews (SR); 14 papers/reports); regulation of online sales (1 SR; 1 paper); place-shaping (2 SRs; 18 papers/reports); and violence reduction initiatives (9 SRs; 24 papers/reports); and led two expert workshops (n = 11). Results Interventions that raise the price of cheaper shop-bought alcohol appear promising as ‘sweetspot’ policies; any impact on hospitality is likely small and potentially positive. Restrictions on online sales such as speed or timing of delivery may reduce harm and diversion of consumption from on-trade to home settings. Place-shaping is not well-supported by evidence and experts were sceptical. Reduced late-night trading hours likely reduce violence; evidence of impact on hospitality is scant. Other violence reduction initiatives may modestly reduce harms whilst supporting hospitality, but require resources to deliver multiple measures simultaneously in partnership. Conclusions Available evidence and expert views point to regulation of pricing and online sales as having greatest potential as ‘sweetspot’ alcohol policies, reducing alcohol harm whilst minimising negative impact on hospitality businesses

    Fine-mapping identifies multiple prostate cancer risk loci at 5p15, one of which associates with TERT expression

    Associations between single nucleotide polymorphisms (SNPs) at 5p15 and multiple cancer types have been reported. We have previously shown evidence for a strong association between prostate cancer (PrCa) risk and rs2242652 at 5p15, intronic in the telomerase reverse transcriptase (TERT) gene that encodes TERT. To comprehensively evaluate the association between genetic variation across this region and PrCa, we performed a fine-mapping analysis by genotyping 134 SNPs using a custom Illumina iSelect array or Sequenom MassArray iPlex, followed by imputation of 1094 SNPs in 22 301 PrCa cases and 22 320 controls in The PRACTICAL consortium. Multiple stepwise logistic regression analysis identified four signals in the promoter or intronic regions of TERT that independently associated with PrCa risk. Gene expression analysis of normal prostate tissue showed evidence that SNPs within one of these regions also associated with TERT expression, providing a potential mechanism for predisposition to disease

    Vitamin D accelerates resolution of inflammatory responses during tuberculosis treatment

    Calcidiol, the major circulating metabolite of vitamin D, supports induction of pleiotropic antimicrobial responses in vitro. Vitamin D supplementation elevates circulating calcidiol concentrations, and thus has a potential role in the prevention and treatment of infection. The immunomodulatory effects of administering vitamin D to humans with an infectious disease have not previously been reported. To characterize these effects, we conducted a detailed longitudinal study of circulating and antigen-stimulated immune responses in ninety-five patients receiving antimicrobial therapy for pulmonary tuberculosis who were randomized to receive adjunctive high-dose vitamin D or placebo in a clinical trial, and who fulfilled criteria for per-protocol analysis. Vitamin D supplementation accelerated sputum smear conversion and enhanced treatment-induced resolution of lymphopaenia, monocytosis, hypercytokinaemia, and hyperchemokinaemia. Administration of vitamin D also suppressed antigen-stimulated proinflammatory cytokine responses, but attenuated the suppressive effect of antimicrobial therapy on antigen-stimulated secretion of IL-4, CC chemokine ligand 5, and IFN-α. We demonstrate a previously unappreciated role for vitamin D supplementation in accelerating resolution of inflammatory responses during tuberculosis treatment. Our findings suggest a potential role for adjunctive vitamin D supplementation in the treatment of pulmonary infections to accelerate resolution of inflammatory responses associated with increased risk of mortality
