    Systematic process of waste management for sustainable operations of hotel industry

    Hotel industry contributes to Gross Domestic Product of Sri Lanka by 12.5% (2018) which is a considerable value. Hotel industry contributes for a considerable amount of daily wastes since there are number of hotel networks which are spread throughout the country. Determining the sustainability adaptation of the Sri Lankan hotel industry is crucial since there is not a strict legal background that specifically governs the hotel industry waste, except for a few regulations related to tourism. Addressing gaps in between such requirements and waste management systems in hotels from the perspective of the Circular Economy concept, this research identifies match making opportunities for hotel industry waste. This ensures that waste is not thrown away and is used in the production process of other goods or providing services. Although many studies on the concept of CE have been carried out globally, it is a novel topic in the Sri Lankan context. Further, research that adapted the CE concept for managing hotel industry waste is scarce. This research therefore investigates on applying the CE concept for managing hotel sector waste by analyzing, the 6R principles associated with the CE concept, the waste match making process and its cost implications. A case study strategy with semi structured interviews as the data collection tool was used in the study. Case studies were preceded by a brief pilot study to set the context for the main study. Manual content analysis method was used for analyzing the collected data. Through the research findings, it was revealed that the possibility of adapting waste exchanging program for hotel industry mainly depends on the category and the location of the hotel. Even though the barriers exist, those can be mitigated through proper vendor selection and waste match making. This research identified possible waste matches, pros and cons of such matches and the enablers, barriers and favorable outcomes of such match making exercises.     Sintered Bottom and Vitrified Silica Ashes Derived from Incinerated Municipal Solid Waste as Circular Economy-Friendly Partial Replacements for Cement in Mortars

    Sintered bottom ash (SBA) and vitrified ash (VA) derived from the incineration of municipal solid waste (MSW) at 1200 °C were used in this study as replacements for Type 2 Portland cement (CEM-II) based mortars. This approach negates the need to send them to landfill, benefits the circular economy and contributes towards the decarbonisation of cementitious construction materials in response to international net zero carbon emission agendas. The material (physico-chemical) characteristics of VA and SBA were analysed before being used as partial replacements for CEM-II in mortars, whereby compressive strength (CS) was the primary criterion for assessing engineering performance. VA and SBA replaced CEM-II at dosages of 10%, 25% and 50% based on their high inorganic and pozzolanic contents; whereby the 10% and 25% replacements did not compromise mortar strength. The alkalinity and pozzolanic properties of SBA collectively indicated it has greater potential as a cementitious material over VA, which possessed a neutral pH. The 28-day CS recorded for mixtures containing 25% VA and 10% SBA were 13.74 MPa and 11.77 MPa, respectively compared with 17.06 MPa for CEM-II control samples. The use of 2% additional water in 25% SBA mortar designs improved strength further, indicating that SBA’s water retention properties permitted further hydration and strength development with curing. Microstructural, mineralogical and infrared spectroscopy analyses indicated that these strengths were owed to the formation of silicate-based hydration products. The outcomes from this study highlight that SBA has potential for replacing CEM-II and VA as a filler in cementitious mortar. Graphical Abstract: [Figure not available: see fulltext.]</p

    Accessibility and Usage of Digital Technologies among Academics for Research: A Case of Selected Humanities and Social Sciences Faculties in Sri Lankan Universities

    The Humanities and Social Sciences (HSS) involve understanding the human experience and the relationships between individuals and groups in society. The adoption of digital technologies has challenged the discipline of HSS, creating an entirely new environment for the study of human activities. This research aims to explore the employment of digital tools, resources and services in HSS research. Further, the use of digital methods (DM) throughout the research process; the impact of COVID-19 on the use of DM in research; the constraints of using DM in research were tested. Both quantitative and qualitative data were collected from Colombo, Kelaniya and Sri Jayewardenepura universities targeting academics in Economics, English, English Language Teaching, Geography, History and Archeology, Buddhist Studies, Political Science, Sinhala, and Sociology. Considering the digital infrastructure facilities, most of the academics rated email (45.6%) and LMS services (46.7%) as excellent but maintenance provided by the institute was not adequate. Most academics rated good on access to data storage (37.9%); reference management software (27.5%); plagiarism detection software (29.1%); institutional repository (35.2%); and support to online publications (39.6%) provided by their institutes. 55.7% of surveyed academics in SS often use digital data collection methods while in the Humanities it was 43.4%. Online publishing was most often used by SS (50.9%) and only 39.5% by the Humanities. 53.8% of SS academics and 43.4% of humanities academics often use cloud storage. Findings confirmed the expansion of using digital research methods during the pandemic compared to the early pandemic situation. Collaborative research works, virtual conferences, citation databases and digital indexing were identified as popular trends. DOI: http://doi.org/10.31357/fhss/vjhss.v08i02.0

    Investigation of Daily Macronutrient intakes by Sri Lankan Managerial Level Employees working in the Private Sector

    With changes of lifestyles and social values in the food culture, many individuals working as managerial level employees in the as private sector are seemingly selecting more improper daily meal combinations. This study was designed to determine whether this occurrence can have a severe impact to imbalance the daily nutrient intakes by the individuals in the mentioned social segment, which increase the tendency of having nutrition related chronic diseases. In a hierarchy range from junior executives to CEOs of private sector organizations, 800 individuals were selected by disproportionate stratified random sampling. Selected individuals are interviewed using a structured questionnaire to assess their daily food combinations and their consumed portion sizes. Frequently consumed meal combinations were then analyzed for their macronutrient composition, to compare with world Health organizations&rsquo; (WHO) Reference Dietary Intake (RDI) levels of nutrients. The results reveal of significantly (p&lt;0.05) higher daily fat (45.3 &plusmn;1.7 g/day) and protein (65.2 &plusmn;1.4 g/day) intakes than the WHO recommendation levels and significantly (p&lt;0.05) lower in dietary fibre (22.3 &plusmn;1.1 g/day) contents by selected participants. Carbohydrate intake (133.1 &plusmn;2.2 g/day) was higher than reference levels but was not significant (p&gt;0.05). This indicates of a considerable risk for many individuals in the concerned social segment, of having non-communicable diseases, if observed dietary patterns are continued

    The association of circulating amylin with β-amyloid in familial Alzheimer's disease.

    INTRODUCTION: This study assessed the hypothesis that circulating human amylin (amyloid-forming) cross-seeds with amyloid beta (Aβ) in early Alzheimer's disease (AD). METHODS: Evidence of amylin-AD pathology interaction was tested in brains of 31 familial AD mutation carriers and 20 cognitively unaffected individuals, in cerebrospinal fluid (CSF) (98 diseased and 117 control samples) and in genetic databases. For functional testing, we genetically manipulated amylin secretion in APP/PS1 and non-APP/PS1 rats. RESULTS: Amylin-Aβ cross-seeding was identified in AD brains. High CSF amylin levels were associated with decreased CSF Aβ42 concentrations. AD risk and amylin gene are not correlated. Suppressed amylin secretion protected APP/PS1 rats against AD-associated effects. In contrast, hypersecretion or intravenous injection of human amylin in APP/PS1 rats exacerbated AD-like pathology through disruption of CSF-brain Aβ exchange and amylin-Aβ cross-seeding. DISCUSSION: These findings strengthened the hypothesis of circulating amylin-AD interaction and suggest that modulation of blood amylin levels may alter Aβ-related pathology/symptoms

    Insulin Storage and Glucose Homeostasis in Mice Null for the Granule Zinc Transporter ZnT8 and Studies of the Type 2 Diabetes–Associated Variants

    International audienceObjective. Zinc ions are essential for the formation of hexameric insulin and hormone crystallisation. Correspondingly, a non-synonymous single nucleotide polymorphism rs13266634 in the SLC30A8 gene, encoding the secretory granule zinc transporter ZnT8, is associated with type 2 diabetes. Here, we describe the effects of deleting the ZnT8 gene in mice and explore the action of the at-risk allele. Research Design and Methods. Slc30a8 null mice were generated and backcrossed at least twice onto a C57BL/6J background. Glucose and insulin tolerance were measured by intraperitoneal injection, or euglycemic clamp, respectively. Insulin secretion, electrophysiology, imaging, and the generation of adenoviruses encoding the low- (W325) or elevated- (R325) risk ZnT8 alleles, were undertaken using standard protocols. Results. ZnT8(-/-) mice displayed age, sex and diet-dependent abnormalities in glucose tolerance, insulin secretion and body weight. Islets isolated from null mice had reduced granule zinc content, and showed age-dependent changes in granule morphology, with markedly fewer dense cores but more rod-like crystals. Glucose-stimulated insulin secretion, granule fusion and insulin crystal dissolution, as assessed by total internal reflection fluorescence microscopy, were unchanged or enhanced in ZnT8(-/-) islets. Insulin processing was normal. Molecular modelling revealed that residue-325 was located at the interface between ZnT8 monomers. Correspondingly, the R325 variant displayed lower apparent Zn(2+) transport activity than W325 ZnT8 by fluorescence-based assay. Discussion and conclusions. ZnT8 is required for normal insulin crystallisation and insulin release in vivo but not, remarkably, in vitro. Defects in the former processes in carriers of the R allele may increase type 2 diabetes risk

    Pten (phosphatase and tensin homologue gene) haploinsufficiency promotes insulin hypersensitivity

    AIMS/HYPOTHESIS: Insulin controls glucose metabolism via multiple signalling pathways, including the phosphatidylinositol 3-kinase (PI3K) pathway in muscle and adipose tissue. The protein/lipid phosphatase Pten (phosphatase and tensin homologue deleted on chromosome 10) attenuates PI3K signalling by dephosphorylating the phosphatidylinositol 3,4,5-trisphosphate generated by PI3K. The current study was aimed at investigating the effect of haploinsufficiency for Pten on insulin-stimulated glucose uptake. MATERIALS AND METHODS: Insulin sensitivity in Pten heterozygous (Pten(+/−)) mice was investigated in i.p. insulin challenge and glucose tolerance tests. Glucose uptake was monitored in vitro in primary cultures of myocytes from Pten(+/−) mice, and in vivo by positron emission tomography. The phosphorylation status of protein kinase B (PKB/Akt), a downstream signalling protein in the PI3K pathway, and glycogen synthase kinase 3β (GSK3β), a substrate of PKB/Akt, was determined by western immunoblotting. RESULTS: Following i.p. insulin challenge, blood glucose levels in Pten(+/−) mice remained depressed for up to 120 min, whereas glucose levels in wild-type mice began to recover after approximately 30 min. After glucose challenge, blood glucose returned to normal about twice as rapidly in Pten(+/−) mice. Enhanced glucose uptake was observed both in Pten(+/−) myocytes and in skeletal muscle of Pten(+/−) mice by PET. PKB and GSK3β phosphorylation was enhanced and prolonged in Pten(+/−) myocytes. CONCLUSIONS/INTERPRETATION: Pten is a key negative regulator of insulin-stimulated glucose uptake in vitro and in vivo. The partial reduction of Pten due to Pten haploinsufficiency is enough to elicit enhanced insulin sensitivity and glucose tolerance in Pten(+/−) mice
