    Diversity and Abundance of Hymenopterous Parasitoids Associated with Anastrepha fraterculus (Diptera: Tephritidae) in Native and Exotic Host Plants in Misiones, Northeastern Argentina

    Some Major host species used by the tephritid fruit flies Anastrepha fraterculus (Wiede-mann) and Ceratitis capitata (Wiedemann), including Acca sellowiana (O. Berg) Burret, Campomanesia xanthocarpa O. Berg, Psidium guajava L., Prunus persica (L.) Batsch, Eriobotrya japonica (Thunb.) Lindl., Citrus reticulata Blanco var. Murcott, C. aurantium L., C. paradisi Macfadyen var. Dalan Dan, and C. paradisi var. Sudashi, were sampled for fruit fly larvae between Feb and Dec 2000 in the northernmost section of the Paranaense forest, in the Province of Misiones, NE Argentina. Both A. fraterculus and C. capitata were obtained from these host plant species, with A. fraterculus accounting for 93% of all tephritid puparia identified. Ten species of larval-pupal parasitoids were recovered from A. fraterculus; Doryctobracon areolatus (Szépligeti), D. brasiliensis (Szépligeti), Utetes anastrephae (Viereck), Opius bellus (Gahan), Diachasmimorpha longicaudata (Ashmead) (Opiinae, raconidae), Odontosema anastrephae Borgmeier, Lopheucoila anastrephae (Rohwer), Aganaspis pelleranoi (Brèthes) (Eucoilinae, Figitidae), Asobara anastrephae (Muessebeck) (Alyssinae, Braconidae), and Aceratoneuromyia indica (Silvestri) (Tetrastichinae, Eulophidae). All these parasitoids, with the exception of D. longicaudata and A. indica, are native to the Neotropical region. No parasitoids were recovered from C. capitata puparia. Asobara anastrephae and O. anastrephae are newly recorded in Argentina, whereas D. brasiliensis, U. anastrephae, and L. anastrephae are newly reported in Misiones. The eucoiline A. pelleranoi wasthe most abundant parasitoid species. Acca sellowiana and P. guajava harbored the highest parasitoid abundance and diversity.Fil: Schliserman, Pablo.     Edmonton Obesity Staging System Prevalence and Association with Weight Loss in a Publicly Funded Referral-Based Obesity Clinic

    Objectives. To determine the distribution of EOSS stages and differences in weight loss achieved according to EOSS stage, in patients attending a referral-based publically funded multisite weight management clinic. Subjects/Methods. 5,787 obese patients were categorized using EOSS staging using metabolic risk factors, medication use, and severity of doctor diagnosis of obesity-related physiological, functional, and psychological comorbidities from electronic patient files. Results. The prevalence of EOSS stages 0 (no risk factors or comorbidities), 1 (mild conditions), 2 (moderate conditions), and 3 (severe conditions) was 1.7%, 10.4%, 84.0%, and 3.9%, respectively. Prehypertension (63%), hypertension (76%), and knee replacement (33%) were the most common obesity-related comorbidities for stages 1, 2, and 3, respectively. In the models including age, sex, initial BMI, EOSS stage, and treatment time, lower EOSS stage and longer treatment times were independently associated with greater absolute (kg) and percentage of weight loss relative to initial body weight P<0.05. Conclusions. Patients attending this publicly funded, referral-based weight management clinic were more likely to be classified in the higher stages of EOSS. Patients in higher EOSS stages required longer treatment times to achieve similar weight outcomes as those in lower EOSS stages

    The Effects of Neutral Inertia on Ionospheric Currents in the High-Latitude Thermosphere Following a Geomagnetic Storm

    Results of an experimental and theoretical investigation into the effects of the time dependent neutral wind flywheel on high-latitude ionospheric electrodynamics are presented. The results extend our previous work which used the National Center for Atmospheric Research Thermosphere/Ionosphere General Circulation Model (NCAR TIGCM) to theoretically simulate flywheel effects in the aftermath of a geomagnetic storm. The previous results indicated that the neutral circulation, set up by ion-neutral momentum coupling in the main phase of a geomagnetic storm, is maintained for several hours after the main phase has ended and may dominate height-integrated Hall currents and field-aligned currents for up to 4-5 hours. We extend the work of Deng et al. to include comparisons between the calculated time-dependent ionospheric Hall current system in the storm-time recovery period and that measured by instruments on board the Dynamics Explorer 2 (DE 2) satellite. Also, comparisons are made between calculated field-aligned currents and those derived from DE 2 magnetometer measurements. These calculations also allow us to calculate the power transfer rate (sometimes called the Poynting flux) between the magnetosphere and ionosphere. The following conclusions have been drawn: (1) Neutral winds can contribute significantly to the horizontal ionospheric current system in the period immediately following the main phase of a geomagnetic storm, especially over the magnetic polar cap and in regions of ion drift shear. (2) Neutral winds drive Hall currents that flow in the opposite direction to those driven by ion drifts. (3) The overall morphology of the calculated field-aligned current system agrees with previously published observations for the interplanetary magnetic field (IMF) B(sub Z) southward conditions, although the region I and region 2 currents are smeared by the TI(ICM model grid resolution. (4) Neutral winds can make significant contributions to the field-aligned current system when B(sub Z) northward conditions prevail following the main phase of a storm, but can account for only a fraction of the observed currents. (5) DE 2 measurements provide a demonstration of "local" (satellite-altitude) flywheel effects. (6) On the assumption that the magnetosphere acts as an insulator, we calculate neutral-wind-induced polarization electric fields of approx. 20-30 kV in the period immediately following the geomagnetic storm

    Calibrated BOLD using direct measurement of changes in venous oxygenation

    Calibration of the BOLD signal is potentially of great value in providing a closer measure of the underlying changes in brain function related to neuronal activity than the BOLD signal alone, but current approaches rely on an assumed relationship between cerebral blood volume (CBV) and cerebral blood flow (CBF). This is poorly characterised in humans and does not reflect the predominantly venous nature of BOLD contrast, whilst this relationship may vary across brain regions and depend on the structure of the local vascular bed. This work demonstrates a new approach to BOLD calibration which does not require an assumption about the relationship between cerebral blood volume and cerebral blood flow. This method involves repeating the same stimulus both at normoxia and hyperoxia, using hyperoxic BOLD contrast to estimate the relative changes in venous blood oxygenation and venous CBV. To do this the effect of hyperoxia on venous blood oxygenation has to be calculated, which requires an estimate of basal oxygen extraction fraction, and this can be estimated from the phase as an alternative to using a literature estimate. Additional measurement of the relative change in CBF, combined with the blood oxygenation change can be used to calculate the relative change in CMRO2 due to the stimulus. CMRO2 changes of 18 ± 8% in response to a motor task were measured without requiring the assumption of a CBV/CBF coupling relationship, and are in agreement with previous approaches

    Is late-life dependency increasing or not? A comparison of the Cognitive Function and Ageing Studies (CFAS)

    Background: Little is known about how dependency levels have changed between generational cohorts of older people. We estimated years lived in different care states at age 65 in 1991 and 2011 and new projections of future demand for care. Methods: Two population-based studies of older people in defined geographical areas conducted two decades apart (the Cognitive Function and Ageing Studies) provided prevalence estimates of dependency in four states: high (24-hour care); medium (daily care); low (less than daily); independent. Years in each dependency state were calculated by Sullivan’s method. To project future demand, the proportions in each dependency state (by age group and sex) were applied to the 2014 England population projections. Findings: Between 1991 and 2011 there were significant increases in years lived from age 65 with low (men:1·7 years, 95%CI 1·0-2·4; women:2·4 years, 95%CI 1·8-3·1) and high dependency (men:0·9 years, 95%CI 0·2-1·7; women:1·3 years, 95%CI 0·5-2·1). The majority of men’s extra years of life were independent (36%) or with low dependency (36%) whilst for women the majority were spent with low dependency (58%), only 5% being independent. There were substantial reductions in the proportions with medium and high dependency who lived in care homes, although, if these dependency and care home proportions remain constant in the future, further population ageing will require an extra 71,000 care home places by 2025. Interpretation: On average older men now spend 2.4 years and women 3.0 years with substantial care needs (medium or high dependency), and most will live in the community. These findings have considerable implications for older people’s families who provide the majority of unpaid care, but the findings also supply valuable new information for governments and care providers planning the resources and funding required for the care of their future ageing populations

    Gene transfer into hepatocytes using asialoglycoprotein receptor mediated endocytosis of DNA complexed with an artificial tetra-antennary galactose ligand

    We have constructed an artificial ligand for the hepatocyte-specific asialoglycoprotein receptor for the purpose of generating a synthetic delivery system for DNA. This ligand has a tetra-antennary structure, containing four terminal galactose residues on a branched carrier peptide. The carbohydrate residues of this glycopeptide were introduced by reductive coupling of lactose to the alpha- and epsilon-amino groups of the two N-terminal lysines on the carrier peptide. The C-terminus of the peptide, containing a cysteine separated from the branched N-terminus by a 10 amino acid spacer sequence, was used for conjugation to 3-(2-pyridyldithio)propionate-modified polylysine via disulfide bond formation. Complexes containing plasmid DNA bound to these galactose-polylysine conjugates have been used for asialoglycoprotein receptor-mediated transfer of a luciferase gene into human (HepG2) and murine (BNL CL.2) hepatocyte cell lines. Gene transfer was strongly promoted when amphipathic peptides with pH-controlled membrane-disruption activity, derived from the N-terminal sequence of influenza virus hemagglutinin HA-2, were also present in these DNA complexes. Thus, we have essentially borrowed the small functional domains of two large proteins, asialoglycoprotein and hemagglutinin, and assembled them into a supramolecular complex to generate an efficient gene-transfer system

    The repeating Fast Radio Burst FRB 121102: Multi-wavelength observations and additional bursts

    We report on radio and X-ray observations of the only known repeating Fast Radio Burst (FRB) source, FRB 121102. We have detected six additional radio bursts from this source: five with the Green Bank Telescope at 2 GHz, and one at 1.4 GHz at the Arecibo Observatory for a total of 17 bursts from this source. All have dispersion measures consistent with a single value (559\sim559 pc cm3^{-3}) that is three times the predicted maximum Galactic value. The 2-GHz bursts have highly variable spectra like those at 1.4 GHz, indicating that the frequency structure seen across the individual 1.4 and 2-GHz bandpasses is part of a wideband process. X-ray observations of the FRB 121102 field with the Swift and Chandra observatories show at least one possible counterpart; however, the probability of chance superposition is high. A radio imaging observation of the field with the Jansky Very Large Array at 1.6 GHz yields a 5σ\sigma upper limit of 0.3 mJy on any point-source continuum emission. This upper limit, combined with archival WISE 22-μ\mum and IPHAS Hα\alpha surveys, rules out the presence of an intervening Galactic HII region. We update our estimate of the FRB detection rate in the PALFA survey to be 1.11.0+3.7×104^{+3.7}_{-1.0} \times 10^4 FRBs sky1^{-1} day1^{-1} (95% confidence) for peak flux density at 1.4 GHz above 300 mJy. We find that the intrinsic widths of the 12 FRB 121102 bursts from Arecibo are, on average, significantly longer than the intrinsic widths of the 13 single-component FRBs detected with the Parkes telescope.Comment: 18 pages, 5 figures.     A mechanism for morphogen-controlled domain growth

    Many developmental systems are organised via the action of graded distributions of morphogens. In the Drosophila wing disc, for example, recent experimental evidence has shown that graded expression of the morphogen Dpp controls cell proliferation and hence disc growth. Our goal is to explore a simple model for regulation of wing growth via the Dpp gradient: we use a system of reaction-diffusion equations to model the dynamics of Dpp and its receptor Tkv, with advection arising as a result of the flow generated by cell proliferation. We analyse the model both numerically and analytically, showing that uniform domain growth across the disc produces an exponentially growing wing disc

    Encapsulation of drugs within nanocarriers that selectively target malignant cells promises to mitigate side effects of conventional chemotherapy and to enable delivery of the unique drug combinations needed for personalized medicine. To realize this potential, however, targeted nanocarriers must simultaneously overcome multiple challenges, including specificity, stability and a high capacity for disparate cargos. Here we report porous nanoparticle-supported lipid bilayers (protocells) that synergistically combine properties of liposomes and nanoporous particles. Protocells modified with a targeting peptide that binds to human hepatocellular carcinoma exhibit a 10,000-fold greater affinity for human hepatocellular carcinoma than for hepatocytes, endothelial cells or immune cells. Furthermore, protocells can be loaded with combinations of therapeutic (drugs, small interfering RNA and toxins) and diagnostic (quantum dots) agents and modified to promote endosomal escape and nuclear accumulation of selected cargos. The enormous capacity of the high-surface-area nanoporous core combined with the enhanced targeting efficacy enabled by the fluid supported lipid bilayer enable a single protocell loaded with a drug cocktail to kill a drug-resistant human hepatocellular carcinoma cell, representing a 10(6)-fold improvement over comparable liposomes
