    Non-Native Forest Tree Species in Europe: The Question of Seed Origin in Afforestation

    Non-native forest tree species have been introduced in Europe since the 16th century, but only in the second half of the 20th century the significance of the seed source origin for their economic use was recognized, resulting in the establishment of numerous provenance trials at a national, regional, European and International level, as those led by IUFRO. Breeding programs have also been launched in the continent for the most economically important species. Aim of this work is the formulation of provenance recommendations for planting of five non-native tree species in Europe (Douglas fir, grand fir, Sitka spruce, lodgepole pine and black locust), based on the information obtained from twenty countries, in the frame of the EU FP-1403 NNEXT Cost Action. The survey revealed that official and non-official national recommendations, based on provenance research results, have been elaborated and followed at a different level and extend for the above five species, but only for Douglas fir recommendations exist in almost all the participating to the survey countries. The compilation of provenance recommendations across Europe for each species is presented in the current work. Besides the recommended introduced seed sources, European seed sources are also preferred for planting, due to ease of access and high availability of forest reproductive material. European breeding programs yielding genetic material of high productivity and quality constitute currently the seed source of choice for several species and countries. Consolidation of trial data obtained across countries will allow the joint analysis that is urgently needed to draw solid conclusions, and will facilitate the development of ‘Universal-Response-Functions’ for the species of interest, rendering possible the identification of the genetic material suitable for global change. New provenance trial series that will test seed sources from the entire climatic range of the species, established in sites falling within and outside the environmental envelopes of their natural ranges, are urgently needed to pinpoint and understand the species-specific climate constraints, as well as to correlate functional traits to the seed origin and the environmental conditions of the test sites, so that the selection of suitable forest reproductive material of non-native tree species in the face of climate change can be feasible.publishedVersio

    Psychometric testing of a quality of life scale among Swedish people 75 years and above in need of help with activities of daily living or not

    Few Quality of Life (QoL) instruments in use today were originally developed for older populations (75+). Information on their validity and reliability in this age group is sparse. The current study investigated the psychometric properties of the Life Quality Gerontological Centre Scale (LGC), a general QoL instrument designed for use among older people. A further aim was to describe and compare QoL with gender and in people needing help with Activities of Daily Living and those not needing such help. A postal questionnaire surveyed 1093 people (mean age 82.7, SD 5.3). Principal component analysis revealed that LGC was reasonably stable as the construct-and cross validation more or less replicated the suggested QoL factors. The instrument differentiated between needing help and not and between men and women. LGC needs further development with respect to reliability, face and content validity but is potentially useful as a diagnostic or an outcome assessment after interventions among older people. Individuals in need of help and women had lower scores in several QoL areas and in total QoL scores compared to those not needing help and men. Older people’s QoL may be improved by interventions directed to areas beyond health and physical functioning. Men and women might also benefit from different interventions directed towards QoL

    En studie av framtida flödesbelastning på Stockholms huvudavloppssystem

    Denna studie utfördes inom SWEdish research programme on Climate, Impacts and Adaptation (SWECIA), finansierat av Stiftelsen för Miljöstratgisk Forskning (MISTRA), med delfinansiering från Stockholm Vatten AB.Studiens syfte var att bedöma flödesbelastningen på Stockholms huvudavloppssystem under resten av detta sekel mot bakgrund av både klimatförändring och befolkningsökning. Som underlag gjordes flödes-simuleringar med MIKE Urban. Referenssimuleringar för dagens klimat gjordes dels för ett representativt helår (1984), dels för c:a 200 utvalda regnhändelser mellan 1983 och 2007. I framtidssimuleringar beskrevs klimateffekten genom omskalning av indata (temperatur, nederbörd, avdunstning) i enlighet med klimatmodellscenarier och befolknings-effekten genom en ökning i enlighet med officiell bedömning.Resultaten indikerar framför allt att bräddade volymer till Mälaren och Saltsjön kommer att öka kraftigt, men även ett ökat inflöde till reningsverket, och därmed ökat behov av rening, samt en ökad översvämningsrisk.This study was performed within the SWEdish research programme on Climate, Impacts and Adaptation (SWECIA), funded by the Foundation for Strategic Environmental Research (MISTRA), with additional funding from Stockholm Vatten AB. The aim of the study was to assess the discharge load on Stockholm's main sewer system during the rest of this century in light of both climate change and population increase. For this assessment, flow simulations with MIKE Urban were performed. Reference simulations for today's climate were done both for a representative year (1984) and for some 200 selected rainfall events between 1983 and 2007. In future simulations the climate effect was taken into account by rescaling input data (temperature, precipitation, evaporation) in line with climate model scenarios and the population effect by an increase in line with official estimations. The results indicate in particular that the spill volumes to Lake Mälaren and Saltsjön will increase substantially, but also an increased inflow to the treatment plant, and thus an increased need for treatment, and an increased flood risk

First published December 15, 2009; doi:10.1152/ajpendo.00395.2009.-Many members of the forkhead genes family of transcription factors have been implicated as important regulators of metabolism, in particular, glucose homeostasis, e. g., Foxo1, Foxa3, and Foxc2. The purpose of this study was to exploit the possibility that yet unknown members of this gene family play a role in regulating glucose tolerance in adipocytes. We identified Foxf2 in a screen for adipose-expressed forkhead genes. In vivo overexpression of Foxf2 in an adipose tissue-restricted fashion demonstrated that such mice display a significantly induced insulin secretion in response to an intravenous glucose load compared with wild-type littermates. In response to increased Foxf2 expression, insulin receptor substrate 1 (IRS1) mRNA and protein levels are significantly downregulated in adipocytes; however, the ratio of serine vs. tyrosine phosphorylation of IRS1 seems to remain unaffected. Furthermore, adipocytes over-expressing Foxf2 have a significantly lower insulin-mediated glucose uptake compared with wild-type adipocytes. These findings argue that Foxf2 is a previously unrecognized regulator of cellular and systemic whole body glucose tolerance, at least in part, due to lower levels of IRS1. Foxf2 and its downstream target genes can provide new insights with regard to identification of novel therapeutic targets

    The Human Adenovirus 2 Transcriptome : An Amazing Complexity of Alternatively Spliced mRNAs

    We have used the Nanopore long-read sequencing platform to demonstrate how amazingly complex the human adenovirus type 2 (Ad2) transcriptome is with a flexible splicing machinery producing a range of novel mRNAs both from the early and late transcription units. In total we report more than 900 alternatively spliced mRNAs produced from the Ad2 transcriptome whereof more than 850 are novel mRNAs. A surprising finding was that more than 50% of all E1A transcripts extended upstream of the previously defined transcriptional start site. The novel start sites mapped close to the inverted terminal repeat (ITR) and within the E1A enhancer region. We speculate that novel promoters or enhancer driven transcription, so-called eRNA transcription, is responsible for producing these novel mRNAs. Their existence was verified by a peptide in the Ad2 proteome that was unique for the E1A ITR mRNA. Although we show a high complexity of alternative splicing from most early and late regions, the E3 region was by far the most complex when expressed at late times of infection. More than 400 alternatively spliced mRNAs were observed in this region alone. These mRNAs included extended L4 mRNAs containing E3 and L5 sequences and readthrough mRNAs combining E3 and L5 sequences. Our findings demonstrate that the virus has a remarkable capacity to produce novel exon combinations, which will offer the virus an evolutionary advantage to change the gene expression repertoire and protein production in an evolving environment. IMPORTANCE Work in the adenovirus system led to the groundbreaking discovery of RNA splicing and alternative RNA splicing in 1977. These mechanisms are essential in mammalian evolution by increasing the coding capacity of a genome. Here, we have used a long-read sequencing technology to characterize the complexity of human adenovirus pre-mRNA splicing in detail. It is mindboggling that the viral genome, which only houses around 36,000 bp, not being much larger than a single cellular gene, generates more than 900 alternatively spliced mRNAs. Recently, adenoviruses have been used as the backbone in several promising SARS-CoV-2 vaccines. Further improvement of adenovirus-based vaccines demands that the virus can be tamed into an innocent carrier of foreign genes. This requires a full understanding of the components that govern adenovirus replication and gene expression

    Cumbersome but desirable—Breaking the code of everyday cycling

    Introduction Cycling for transport could integrate physical activity (PA) into daily routines and potentially increase total PA levels. However, for parents with young children, most factors affecting transport mode choice tend to facilitate car use. Greater insight is necessary into reasons for (not) using sustainable transport modes in parents with young children. Therefore, the objective of this study was to explore the experiences, including motives, perceptions, attitudes, and norms, of parents of young children by using an e-bike, a longtail bike, and a traditional bike for everyday travel to the workplace, kindergarten, and the grocery store during the autumn, winter, and spring, in nine months. Methods Semistructured focus group interviews were conducted with 18 parents of young children residing in southern Norway. Parents were recruited through Facebook announcements and direct contact with kindergartens, selected organisations, and companies in the Kristiansand municipality. Data were analysed by systematic text condensation by using NVivo V.11. Results Participants’ experiences were summarised by three main themes: ‘cycling is cumbersome’, ‘cycling reflects the desirable me’, and ‘breaking the cycling code’. Time use, planning, logistics, wet and cold weather, long distances, and no cycling habit were frequently mentioned barriers, and the most notable facilitator was the children’s attitude towards cycling. In general, children loved to cycle and preferred cycling to driving. Additionally, the freedom and independence of cycling were emphasised and valued. Conclusion In challenging weather conditions, parents of young children may experience cycling as cumbersome but desirable, and bike access could increase the feasibility of daily cycling
