    Copper fractionation in plasma, muscle and liver of Nile tilapia was performed after protein separation by 2D-PAGE. SR XRF analysis indicated the presence of copper in three protein spots of plasma, and in two protein spots of muscle and liver, respectively. Copper ions were found to be distributed mostly in proteins that had a molar mass of less than 54 kDa and greater than 13 kDa and a pI in the 5.3-9.3 range. The copper concentration bound to these proteins was determined by GFAAS which showed concentrations in the 1.20-4.82 mg g-1 range

    Rational Design of Pathogen-Mimicking Amphiphilic Materials as Nanoadjuvants

    An opportunity exists today for cross-cutting research utilizing advances in materials science, immunology, microbial pathogenesis, and computational analysis to effectively design the next generation of adjuvants and vaccines. This study integrates these advances into a bottom-up approach for the molecular design of nanoadjuvants capable of mimicking the immune response induced by a natural infection but without the toxic side effects. Biodegradable amphiphilic polyanhydrides possess the unique ability to mimic pathogens and pathogen associated molecular patterns with respect to persisting within and activating immune cells, respectively. The molecular properties responsible for the pathogen-mimicking abilities of these materials have been identified. The value of using polyanhydride nanovaccines was demonstrated by the induction of long-lived protection against a lethal challenge of Yersinia pestis following a single administration ten months earlier. This approach has the tantalizing potential to catalyze the development of next generation vaccines against diseases caused by emerging and re-emerging pathogens

    pyam: Analysis and visualisation of integrated assessment and macro-energy scenarios [version 2; peer review: 3 approved]

    The open-source Python package pyam provides a suite of features and methods for the analysis, validation and visualization of reference data and scenario results generated by integrated assessment models, macro-energy tools and other frameworks in the domain of energy transition, climate change mitigation and sustainable development. It bridges the gap between scenario processing and visualisation solutions that are "hard-wired" to specific modelling frameworks and generic data analysis or plotting packages. The package aims to facilitate reproducibility and reliability of scenario processing, validation and analysis by providing well-tested and documented methods for working with timeseries data in the context of climate policy and energy systems. It supports various data formats, including sub-annual resolution using continuous time representation and "representative timeslices". The pyam package can be useful for modelers generating scenario results using their own tools as well as researchers and analysts working with existing scenario ensembles such as those supporting the IPCC reports or produced in research projects. It is structured in a way that it can be applied irrespective of a user's domain expertise or level of Python knowledge, supporting experts as well as novice users. The code base is implemented following best practices of collaborative scientific-software development. This manuscript describes the design principles of the package and the types of data which can be handled. The usefulness of pyam is illustrated by highlighting several recent applications

    Epithelial Neutrophil-Activating Peptide (ENA-78), Acute Coronary Syndrome Prognosis, and Modulatory Effect of Statins

    Endothelial inflammation with chemokine involvement contributes to acute coronary syndromes (ACS). We tested the hypothesis that variation in the chemokine gene CXCL5, which encodes epithelial neutrophil-activating peptide (ENA-78), is associated with ACS prognosis. We also investigated whether statin use, a potent modulator of inflammation, modifies CXCL5's association with outcomes and characterized the in vitro effect of atorvastatin on endothelial ENA-78 production. Using a prospective cohort of ACS patients (n = 704) the association of the CXCL5 −156 G>C polymorphism (rs352046) with 3-year all-cause mortality was estimated with hazard ratios (HR). Models were stratified by genotype and race. To characterize the influence of statins on this association, a statin*genotype interaction was tested. To validate ENA-78 as a statin target in inflammation typical of ACS, endothelial cells (HUVECs) were treated with IL-1ÎČ and atorvastatin with subsequent quantification of CXCL5 expression and ENA-78 protein concentrations. C/C genotype was associated with a 2.7-fold increase in 3-year all-cause mortality compared to G/G+G/C (95%CI 1.19–5.87; p = 0.017). Statins significantly reduced mortality in G/G individuals only (58% relative risk reduction; p = 0.0009). In HUVECs, atorvastatin dose-dependently decreased IL-1ÎČ-stimulated ENA-78 concentrations (p<0.0001). Drug effects persisted over 48 hours (p<0.01). CXCL5 genotype is associated with outcomes after ACS with potential statin modification of this effect. Atorvastatin lowered endothelial ENA-78 production during inflammation typical of ACS.     Greater Number of Larger Pieces: A Strategy to Promote Prospective Teachers’ Fraction Number Sense Development

    Prospective teachers (PTs) need opportunities to develop fraction number sense, yet little research has explicated how this development occurs. Our research team collaboratively designed a task targeted at helping PTs develop fraction number sense through an exploration of fraction comparison strategies. This paper focuses on developing one particular strategy, which we call Greater Number of Larger Pieces (GLP). We argue that understanding this strategy has the potential to support PTs’ number sense, particularly in regards to the measure interpretation of fractions. Analysis of data from two iterations of this task (implemented by five mathematics teacher educators at five US institutions with 124 PTs) showed an improvement in the task’s ability to naturally elicit the GLP strategy from PTs. We share our task, results from each iteration, and discuss modifications that we believe led to increased usage of the GLP strategy
