    Clinical efficacy and safety of xiaoyao pill in post-stroke depression: A systematic review and meta-analysis of randomized controlled trials

    Purpose: To review the efficacy and safety of the xiaoyao pill in the treatment of post-stroke depression. Methods: A meta-analysis was conducted using eligible studies found in relevant electronic databases [e.g., Embase, Baidu Scholar, Google Scholar, PubMed, Cochrane Library, Science and Technology Periodical Database (VIP) in China, Chinese Biomedical Database, Wanfang, and China National Knowledge Infrastructure]. Statistical analyses were performed using Stata (version 12) and Review (version 5.3). Results: Eleven articles with a total of 1007 patients were included in this study. Overall, the results of the published studies show that xiaoyao pill combined with conventional drug therapy increases clinical response by 20 %. In contrast, Hamilton Depression Scale score and Scandinavian Stroke Scale score were significantly (p < 0.05) lower in xiaoyao pill treatment group than in control group. As an adjuvant therapy, xiaoyao pill reduces potential adverse reactions, suggesting that it can be used as a supplementary therapy in the management of post-stroke depression patients. Conclusion: The review and meta-analysis provide preliminarily proof that xiaoyao pill can improve the clinical symptoms of patients with post-stroke depression and has a higher safety profile than conventional drug therapy. These findings suggest that xiaoyao pill can be used as an alternative or complementary drug for the management of post-stroke depression

    Rapid label-free identification of mixed bacterial infections by surface plasmon resonance

    <p>Abstract</p> <p>Background</p> <p>Early detection of mixed aerobic-anaerobic infection has been a challenge in clinical practice due to the phenotypic changes in complex environments. Surface plasmon resonance (SPR) biosensor is widely used to detect DNA-DNA interaction and offers a sensitive and label-free approach in DNA research.</p> <p>Methods</p> <p>In this study, we developed a single-stranded DNA (ssDNA) amplification technique and modified the traditional SPR detection system for rapid and simultaneous detection of mixed infections of four pathogenic microorganisms (<it>Pseudomonas aeruginosa</it>, <it>Staphylococcus aureus</it>, <it>Clostridium tetani </it>and <it>Clostridium perfringens</it>).</p> <p>Results</p> <p>We constructed the circulation detection well to increase the sensitivity and the tandem probe arrays to reduce the non-specific hybridization. The use of 16S rDNA universal primers ensured the amplification of four target nucleic acid sequences simultaneously, and further electrophoresis and sequencing confirmed the high efficiency of this amplification method. No significant signals were detected during the single-base mismatch or non-specific probe hybridization (<it>P </it>< 0.05). The calibration curves of amplification products of four bacteria had good linearity from 0.1 nM to 100 nM, with all R<sup>2 </sup>values of >0.99. The lowest detection limits were 0.03 nM for <it>P. aeruginosa</it>, 0.02 nM for <it>S. aureus</it>, 0.01 nM for <it>C. tetani </it>and 0.02 nM for <it>C. perfringens</it>. The SPR biosensor had the same detection rate as the traditional culture method (<it>P </it>< 0.05). In addition, the quantification of PCR products can be completed within 15 min, and excellent regeneration greatly reduces the cost for detection.</p> <p>Conclusions</p> <p>Our method can rapidly and accurately identify the mixed aerobic-anaerobic infection, providing a reliable alternative to bacterial culture for rapid bacteria detection.</p

    XAF1 as a modifier of p53 function and cancer susceptibility

    Cancer risk is highly variable in carriers of the common TP53-R337H founder allele, possibly due to the influence of modifier genes. Whole-genome sequencing identified a variant in the tumor suppressor XAF1 (E134*/Glu134Ter/rs146752602) in a subset of R337H carriers. Haplotype-defining variants were verified in 203 patients with cancer, 582 relatives, and 42,438 newborns. The compound mutant haplotype was enriched in patients with cancer, conferring risk for sarcoma (P = 0.003) and subsequent malignancies (P = 0.006). Functional analyses demonstrated that wild-type XAF1 enhances transactivation of wild-type and hypomorphic TP53 variants, whereas XAF1-E134* is markedly attenuated in this activity. We propose that cosegregation of XAF1-E134* and TP53-R337H mutations leads to a more aggressive cancer phenotype than TP53-R337H alone, with implications for genetic counseling and clinical management of hypomorphic TP53 mutant carriers.Fil: Pinto, Emilia M..     Genome-wide homozygosity and risk of four non-Hodgkin lymphoma subtypes

    AIM: Recessive genetic variation is thought to play a role in non-Hodgkin lymphoma (NHL) etiology. Runs of homozygosity (ROH), defined based on long, continuous segments of homozygous SNPs, can be used to estimate both measured and unmeasured recessive genetic variation. We sought to examine genome-wide homozygosity and NHL risk. METHODS: We used data from eight genome-wide association studies of four common NHL subtypes: 3061 chronic lymphocytic leukemia (CLL), 3814 diffuse large B-cell lymphoma (DLBCL), 2784 follicular lymphoma (FL), and 808 marginal zone lymphoma (MZL) cases, as well as 9374 controls. We examined the effect of homozygous variation on risk by: (1) estimating the fraction of the autosome containing runs of homozygosity (FROH); (2) calculating an inbreeding coefficient derived from the correlation among uniting gametes (F3); and (3) examining specific autosomal regions containing ROH. For each, we calculated beta coefficients and standard errors using logistic regression and combined estimates across studies using random-effects meta-analysis. RESULTS: We discovered positive associations between FROH and CLL (β = 21.1, SE = 4.41, P = 1.6 × 10(-6)) and FL (β = 11.4, SE = 5.82, P = 0.02) but not DLBCL (P = 1.0) or MZL (P = 0.91). For F3, we observed an association with CLL (β = 27.5, SE = 6.51, P = 2.4 × 10(-5)). We did not find evidence of associations with specific ROH, suggesting that the associations observed with FROH and F3 for CLL and FL risk were not driven by a single region of homozygosity. CONCLUSION: Our findings support the role of recessive genetic variation in the etiology of CLL and FL; additional research is needed to identify the specific loci associated with NHL risk

    Normal manipulation for bas-relief modeling

    We introduce a normal-based modeling framework for bas-relief generation and stylization which is motivated by the recent advancement in this topic. Creating bas-relief from normal images has successfully facilitated bas-relief modeling in image space. However, the use of normal images in previous work is restricted to the cut-and-paste or blending operations of layers. These operations simply treat a normal vector as a pixel of a general color image. This paper is intended to extend normal-based methods by processing the normal image from a geometric perspective. Our method can not only generate a new normal image by combining various frequencies of existing normal images and details transferring, but also build bas-reliefs from a single RGB image and its edge-based sketch lines. In addition, we introduce an auxiliary function to represent a smooth base surface or generate a layered global shape. To integrate above considerations into our framework, we formulate the bas-relief generation as a variational problem which can be solved by a screened Poisson equation. One important advantage of our method is that it can generate more styles than previous methods and thus it expands the bas-relief shape space. We experimented our method on a range of normal images and it compares favorably to other popular classic and state-of-the-art methods
