    Cavity-induced temperature control of a two-level system

    We consider a two-level atom interacting with a single mode of the electromagnetic field in a cavity within the Jaynes-Cummings model. Initially, the atom is thermal while the cavity is in a coherent state. The atom interacts with the cavity field for a fixed time. After removing the atom from the cavity and applying a laser pulse the atom will be in a thermal state again. Depending on the interaction time with the cavity field the final temperature can be varied over a large range. We discuss how this method can be used to cool the internal degrees of freedom of atoms and create heat baths suitable for studying thermodynamics at the nanoscale

    Mesoscopic Rydberg Gate based on Electromagnetically Induced Transparency

    We demonstrate theoretically a parallelized C-NOT gate which allows to entangle a mesoscopic ensemble of atoms with a single control atom in a single step, with high fidelity and on a microsecond timescale. Our scheme relies on the strong and long-ranged interaction between Rydberg atoms triggering Electromagnetically Induced Transparency (EIT). By this we can robustly implement a conditional transfer of all ensemble atoms among two logical states, depending on the state of the control atom. We outline a many body interferometer which allows a comparison of two many-body quantum states by performing a measurement of the control atom.Comment: published versio

    Genetic Mechanisms Underlying the Pathogenicity of Cold-Stressed Salmonella Enterica Serovar Typhimurium in Cultured Intestinal Epithelial Cells

    Salmonella encounters various stresses in the environment and in the host during infection. The effects of cold (5 C, 48 h), peroxide (5 mM H2O2, 5 h) and acid stress (pH 4.0, 90 min) were tested on pathogenicity of Salmonella. Prior exposure of Salmonella to cold stress significantly (P \u3c 0.05) increased adhesion and invasion of cultured intestinal epithelial (Caco-2) cells. This increased Salmonella-host cell association was also correlated with significant induction of several virulence-associated genes, implying an increased potential of cold-stressed Salmonella to cause an infection. In Caco-2 cells infected with cold-stressed Salmonella, genes involved in the electron transfer chain were significantly induced, but no simultaneous significant increase in expression of antioxidant genes that neutralize the effect of superoxide radicals or reactive oxygen species was observed. Increased production of caspase 9 and caspase 3/7 was confirmed during host cell infection with cold-stressed Salmonella. Further, a prophage gene, STM2699, induced in cold-stressed Salmonella and a spectrin gene, SPTAN1, induced in Salmonella-infected intestinal epithelial cells were found to have a significant contribution in increased adhesion and invasion of cold-stressed Salmonella in epithelial cells

    Alfalfa use by dairy cattle has decreased in recent years because of excessive nonprotein nitrogen and low fiber digestibility. Ideal attributes for plant modification of alfalfa may include those that increase milk potential per acre and/or per ton, enhance digestible NDF, improve protein content and amino acid balance, improve agronomic traits for insect protection (safer forage supply), herbicide tolerance, virus resistance, drought tolerance, cold tolerance, improved mineral availability and enhanced yield. Progress in attaining these attributes will accelerate with the use of biotechnology. Livestock and hay enterprises will benefit from alfalfa that is less prone to contain mycotoxins or toxic weeds, or to induce bloat; have improved nutrient utilization for milk and meat production; and produce less animal wastes resulting in improved efficiency, profitability, and a better environment. Value-added traits of alfalfa are needed to provide farmers new high value profitable products. Processing alfalfa to obtain value added products includes three different fractionation methods: 1) wet fractionation; separation into juice fraction and a fiber fraction, 2) dry fractionation; separation into leaves and stems, and 3) fractionation by passage of the whole herbage through the digestive systems of ruminant animals, leaving a high fiber residue. Phytase from transgenic alfalfa has been tested in poultry and swine rations. Alfalfa hay can be fractionated to yield stems and leaf meal. Alfalfa leaf meal has been shown to be acceptable supplement to replace a portion of alfalfa hay and soybean meal in diets of lactating dairy cattle, replace protein supplement in beef cow diets, finishing steer diets and diets of growing turkeys. The fiber portion of alfalfa can produce lactic acid, ethanol or a bioadhesives for use in plywood

    In vitro gas production as a surrogate measure of the fermentability of cellulosic biomass to ethanol

    Current methods for measuring ethanol yields from lignocellulosic biomass are relatively slow and are not well geared for analyzing large numbers of samples generated by feedstock management and breeding research. The objective of this study was to determine if an in vitro ruminal fermentation assay used in forage quality research was predictive of results obtained using a conventional biomass-to-ethanol conversion assay. In the conventional assay, herbaceous biomass samples were converted to ethanol by Saccharomyces cerevisiae cultures in the presence of cellulase enzymes. Cultures were grown in sealed serum bottles and gas production monitored by measuring increasing head space pressure. Gas accumulation as calculated from the pressure measurements was highly correlated (r2\u3e0.9) with ethanol production measured by gas chromatography at 24 h or 7 days. The same feedstocks were also analyzed by in vitro ruminal digestion, as also measured by gas accumulation. Good correlations (r2∼0.63–0.82) were observed between ethanol production during simultaneous saccharification and fermentation and gas accumulation in parallel in vitro ruminal fermentations. Because the in vitro ruminal fermentation assay can be performed without sterilization of the medium and does not require aseptic conditions, this assay may be useful for biomass feedstock agronomic and breeding research

    Local effective dynamics of quantum systems: A generalized approach to work and heat

    By computing the local energy expectation values with respect to some local measurement basis we show that for any quantum system there are two fundamentally different contributions: changes in energy that do not alter the local von Neumann entropy and changes that do. We identify the former as work and the latter as heat. Since our derivation makes no assumptions on the system Hamiltonian or its state, the result is valid even for states arbitrarily far from equilibrium. Examples are discussed ranging from the classical limit to purely quantum mechanical scenarios, i.e. where the Hamiltonian and the density operator do not commute.Comment: 5 pages, 1 figure, published versio

    Bacterial communities in the rumen of Holstein heifers differ when fed orchardgrass as pasture vs. hay

    The rich and diverse microbiota of the rumen provides ruminant animals the capacity to utilize highly fibrous feedstuffs as their energy source, but there is surprisingly little information on the composition of the microbiome of ruminants fed all-forage diets, despite the importance of such agricultural production systems worldwide. In three 28-d periods, three ruminally-cannulated Holstein heifers sequentially grazed orchardgrass pasture (OP), then were fed orchardgrass hay (OH), then returned to OP. These heifers displayed greater shifts in ruminal bacterial community composition (determined by automated ribosomal intergenic spacer analysis and by pyrotag sequencing of 16S rRNA genes) than did two other heifers maintained 84 d on the same OP. Phyla Firmicutes and Bacteroidetes dominated all ruminal samples, and quantitative PCR indicated that members of the genus Prevotella averaged 23 % of the 16S rRNA gene copies, well below levels previously reported with cows fed total mixed rations. Differences in bacterial community composition and ruminal volatile fatty acid (VFA) profiles were observed between the OP and OH despite similarities in gross chemical composition. Compared to OP, feeding OH increased the molar proportion of ruminal acetate (P = 0.02) and decreased the proportion of ruminal butyrate (P < 0.01), branched-chain VFA (P < 0.01) and the relative population size of the abundant genus Butyrivibrio (P < 0.001), as determined by pyrotag sequencing. Despite the low numbers of animals examined, the observed changes in VFA profile in the rumens of heifers on OP vs. OH are consistent with the shifts in Butyrivibrio abundance and its known physiology as a butyrate producer that ferments both carbohydrates and proteins

    <p>Abstract</p> <p>Background</p> <p>Improvement of biofeedstock quality for cellulosic ethanol production will be facilitated by inexpensive and rapid methods of evaluation, such as those already employed in the field of ruminant nutrition. Our objective was to evaluate whether forage quality and compositional measurements could be used to estimate ethanol yield of maize stover as measured by a simplified pretreatment and simultaneous saccharification and fermentation assay. Twelve maize varieties selected to be diverse for stover digestibility and composition were evaluated.</p> <p>Results</p> <p>Variation in ethanol yield was driven by glucan convertibility rather than by glucan content. Convertibility was highly correlated with ruminal digestibility and lignin content. There was no relationship between structural carbohydrate content (glucan and neutral detergent fiber) and ethanol yield. However, when these variables were included in multiple regression equations including convertibility or neutral detergent fiber digestibility, their partial regression coefficients were significant and positive. A regression model including both neutral detergent fiber and its ruminal digestibility explained 95% of the variation in ethanol yield.</p> <p>Conclusion</p> <p>Forage quality and composition measurements may be used to predict cellulosic ethanol yield to guide biofeedstock improvement through agronomic research and plant breeding.</p

    Dressing of Ultracold Atoms by their Rydberg States in a Ioffe-Pritchard Trap

    We explore how the extraordinary properties of Rydberg atoms can be employed to impact the motion of ultracold ground state atoms. Specifically, we use an off-resonant two-photon laser dressing to map features of the Rydberg states on ground state atoms. It is demonstrated that the interplay between the spatially varying quantization axis of the considered Ioffe-Pritchard field and the fixed polarizations of the laser transitions provides the possibility of substantially manipulating the ground state trapping potential.Comment: 11 pages, 4 figure
