Duplication of Aldolase and Esterase Loci in Cicer (Cicereae Alef.)
The genetic control of fructose bisphosphate aldolase (ALDO, EC 4.1.2.13) and esterase (EST, EC 3.1.1.2) isozymes in Cicer was studied by starch gel electrophoresis. Fixed heterozygote enzyme phenotypes were observed in homozygous lines for both Aldo-1, Aldo-2 and Est-4, Est-5. Crosses between the individuals carrying different alleles of the duplicated genes gave rise to asymmetrically staining bands for both enzyme systems. Subcellular localization studies demonstrated that the products of duplicated aldolase loci are present in the plastids, whereas duplicated esterase isozymes were found in the cytosolic compartment. Selfing and crossing experiments revealed that there are two nuclear genes encoding the plastid specific ALDO isozymes (Aldo-1 and Aldo-2). Similarly, EST-4 and EST-5 isozymes are specified by two nuclear genes (Est-4 and Est-5). No linkage was found between any of the duplicated genes and the other isozyme loci examined in this study. Taxonomic distribution of both duplications was examined in the electrophoretic survey of the related species. Present evidence suggests that these duplications are unique and probably occurred only in this monophyletic tribe, Cicereae, since no duplication was reported in the related genera. No evidence for mutations silencing any of the duplicated copies was detected in the genus. Although the mechanism for duplications is not known, evidence for translocations in Cicer and the existence of a similar linkage between ALDO and EST isozymes in related genera indicate that both duplications may have arisen simultaneously via duplication of a chromosomal segment carrying the ancestral state of the gene
Low Shear in Short-Term Impacts Endothelial Cell Traction and Alignment in Long-Term
Within the vascular system, endothelial cells (ECs) are exposed to fluid shear stress (FSS), a mechanical force exerted by blood flow that is critical for regulating cellular tension and maintaining vascular homeostasis. The way ECs react to FSS varies significantly; while high, laminar FSS supports vasodilation and suppresses inflammation, low or disturbed FSS can lead to endothelial dysfunction and increase the risk of cardiovascular diseases. Yet, the adaptation of ECs to dynamically varying FSS remains poorly understood. This study focuses on the dynamic responses of ECs to brief periods of low FSS, examining its impact on endothelial tractionâa measure of cellular tension that plays a crucial role in how endothelial cells respond to mechanical stimuli. By integrating traction force microscopy (TFM) with a custom-built flow chamber, we analyzed how human umbilical vein endothelial cells (HUVECs) adjust their traction in response to shifts from low to high shear stress. We discovered that initial exposure to low FSS prompts a marked increase in traction force, which continues to rise over 10 hours before slowly decreasing. In contrast, immediate exposure to high FSS causes a quick spike in traction followed by a swift reduction, revealing distinct patterns of traction behavior under different shear conditions. Importantly, the direction of traction forces and the resulting cellular alignment under these conditions indicate that the initial shear experience dictates long-term endothelial behavior. Our findings shed light on the critical influence of short-lived low-shear stress experiences in shaping endothelial function, indicating that early exposure to low FSS results in enduring changes in endothelial contractility and alignment, with significant consequences for vascular health and the development of cardiovascular diseases

Several scientific papers on this subject are appearing in image processing, computer vision and pattern analysis contexts, or use techniques of these areas. Individual differences in reproductive strategy are related to views about recreational drug use in Belgium, the Netherlands and Japan
Individual differences in moral views are often explained as the downstream effect of ideological commitments, such as political orientation and religiosity. Recent studies in the U.S. suggest that moral views about recreational drug use are also influenced by attitudes towards sex and that this relationship cannot be explained by ideological commitments. In this study, we investigate student samples from Belgium, the Netherlands, and Japan. We find that, in all samples, sexual attitudes are strongly related to views about recreational drug use, even after controlling for various ideological variables. We discuss our results in the light of reproductive strategies as determinants of moral views
Patient survival after D 1 and D 2 resections for gastric cancer: long-term results of the MRC randomized surgical trial
Controversy still exists on the optimal surgical resection for potentially curable gastric cancer. Much better long-term survival has been reported in retrospective/non-randomized studies with D 2 resections that involve a radical extended regional lymphadenectomy than with the standard D 1 resections. In this paper we report the long-term survival of patients entered into a randomized study, with follow-up to death or 3 years in 96% of patients and a median follow-up of 6.5 years. In this prospective trial D 1 resection (removal of regional perigastric nodes) was compared with D 2 resection (extended lymphadenectomy to include level 1 and 2 regional nodes). Self-evaluation as a tool in developing environmental responsibility
The purpose of the paper is to share the findings of an action research project aimed at exploring the impact of transformative pedagogies on pre-service teachers following an environmental education programme (EEP), offered by the University of Malta. Assessment and evaluation practices of environmental education (EE) and education for sustainable education (ESD) programmes tend to cater just for knowledge content and skills, usually failing to target the development of attitudes and values that promote sustainable lifestyles. The EEP was specifically designed to target the development of pro-environmental values by actively involving students in their learning mainly and providing opportunities for reflection and self-evaluation. The paper analyses qualitative research data obtained from evaluation questionnaires about every study unit in the programme; reflective questionnaires drawing upon the studentsĂ reflective journals; a focus group interview and in depth one-to-one interviews with individual students. The paper provides studentsĂ evaluations about the course design and effectiveness that should provide insights for course developers and evaluators seeking to develop EE/ESD programmes that address individual needs through learner centred pedagogies.peer-reviewe
Early immune pressure initiated by tissue-resident memory T cells sculpts tumor evolution in non-small cell lung cancer
Tissue-resident memory T (TRM) cells provide immune defense against local infection and can inhibit cancer progression. However, it is unclear to what extent chronic inflammation impacts TRM activation and whether TRM cells existing in tissues before tumor onset influence cancer evolution in humans. We performed deep profiling of healthy lungs and lung cancers in never-smokers (NSs) and ever-smokers (ESs), finding evidence of enhanced immunosurveillance by cells with a TRM-like phenotype in ES lungs. In preclinical models, tumor-specific or bystander TRM-like cells present prior to tumor onset boosted immune cell recruitment, causing tumor immune evasion through loss of MHC class I protein expression and resistance to immune checkpoint inhibitors. In humans, only tumors arising in ES patients underwent clonal immune evasion, unrelated to tobacco-associated mutagenic signatures or oncogenic drivers. These data demonstrate that enhanced TRM-like activity prior to tumor development shapes the evolution of tumor immunogenicity and can impact immunotherapy outcomes
Using social and behavioural science to support COVID-19 pandemic response
The COVID-19 pandemic represents a massive global health crisis. Because the crisis requires large-scale behaviour change and places significant psychological burdens on individuals, insights from the social and behavioural sciences can be used to help align human behavior with the recommendations of epidemiologists and public health experts. Here we review experimental and correlational data from a selection of research topics relevant to pandemics, including work on navigating threats, social and cultural influences on behaviour, science communication, moral decision-making, leadership, and stress and coping. In each section, we note the nature and quality of prior research, including uncertainty and unsettled issues. We identify several insights for effective response to the COVID-19 pandemic, and also highlight important gaps researchers should move quickly to fill in the coming weeks and months
