Tissue culture of oil palm : finding the balance between mass propagation and somaclonal variation
The oil palm (Elaeis guineensis Jacq.) is typically propagated in vitro by indirect somatic embryogenesis, a process in which somatic cells of an explant of choice are, via an intermediate phase of callus growth, induced to differentiate into somatic embryos. The architecture of the oil palm, lacking axillary shoots, does not allow for vegetative propagation. Therefore, somatic embryogenesis is the only alternative to seed propagation, which is hampered by long germination times and low germination rates, for the production of planting material. The current oil palm somatic embryogenesis procedure is associated with several difficulties, which are described in this review. The limited availability of explants, combined with low somatic embryo initiation and regeneration rates, necessitate the proliferation of embryogenic structures, increasing the risk for somaclonal variants such as the mantled phenotype. Several ways to improve the efficiency of the tissue culture method and to reduce the risk of somaclonal variation are described. These include the use of alternative explants and propagation techniques, the introduction of specific embryo maturation treatments and the detection of the mantled abnormality in an early stage. These methods have not yet been fully explored and provide interesting research field for the future. The development of an efficient oil palm micropropagation protocol is needed to keep up with the increasing demand for palm oil in a sustainable way. Mass production of selected, high-yielding palms by tissue culture could raise yields on existing plantations, reducing the need for further expansion of the cultivated area, which is often associated with negative environmental impacts
Thermodynamic behavior of short oligonucleotides in microarray hybridizations can be described using Gibbs free energy in a nearest-neighbor model
While designing oligonucleotide-based microarrays, cross-hybridization
between surface-bound oligos and non-intended labeled targets is probably the
most difficult parameter to predict. Although literature describes
rules-of-thumb concerning oligo length, overall similarity, and continuous
stretches, the final behavior is difficult to predict. The aim of this study
was to investigate the effect of well-defined mismatches on hybridization
specificity using CodeLink Activated Slides, and to study quantitatively the
relation between hybridization intensity and Gibbs free energy (Delta G),
taking the mismatches into account. Our data clearly showed a correlation
between the hybridization intensity and Delta G of the oligos over three orders
of magnitude for the hybridization intensity, which could be described by the
Langmuir model. As Delta G was calculated according to the nearest-neighbor
model, using values related to DNA hybridizations in solution, this study
clearly shows that target-probe hybridizations on microarrays with a
three-dimensional coating are in quantitative agreement with the corresponding
reaction in solution. These results can be interesting for some practical
applications. The correlation between intensity and Delta G can be used in
quality control of microarray hybridizations by designing probes and
corresponding RNA spikes with a range of Delta G values. Furthermore, this
correlation might be of use to fine-tune oligonucleotide design algorithms in a
way to improve the prediction of the influence of mismatching targets on

ENT care of children and adolescents in the Brazilian public healthy system in three different municipalities
The data base of ENT care in the Brazilian public health system (Sistema Unico de Saude - SUS) will help organize public health programs. AIM: The following items were investigated in patients aged up to 17 years attended in public health system outpatient units in the city of Mariana, in the ENT screening unit, UNIFESP-EPM, and in CISMISEL: 1) The main otorhinolaryngological diagnoses; 2) The most frequently required exams, drugs, and surgical procedures and their indications; 3) The jobs of parents; the number of siblings; and 4) A statistical analysis and comparison of data in each location. MATERIAL AND METHODS: We undertook a prospective study and a statistical analysis of variables that were gathered during the first visit. RESULTS: The age, the parents' salary, the number of siblings aged below 18 years, the presence of rhinitis, ears diseases, the exams, drugs and otological surgeries that were indicated were all statistically significant. CONCLUSION: The most common diagnosis was mouth breathing. The most common surgery was adenotonsillectomy. The most frequently requested exam was a lateral cranial radiograph. The number of unemployed parents, their poor salaries, and the number of siblings make if difficult for these patients to be treated in any facility other than the public heath system.A criação de um banco de dados sobre as demandas otorrinolaringológicas no SUS poderá fornecer informações para organizar estratégias de saúde e criar ou otimizar recursos do governo ou privados. OBJETIVOS: Determinar em pacientes até 17 anos de idade: 1) As principais doenças otorrinolaringológicas diagnosticadas; 2) Exames complementares mais solicitados; 3) Medicamentos mais prescritos; 4) Procedimentos ambulatoriais realizados e as indicações cirúrgicas; 5) Estado empregatÃcio dos pais e o número de irmãos dependentes; 6) Comparar e analisar estatisticamente os dados nos locais estudados. MATERIAL E MÉTODO: Estudo prospectivo e análise estatÃstica das variáveis obtidas nas primeiras-consultas na PoliclÃnica de Mariana, na sede Consórcio Intermunicipal de Saúde da Microrregião de Sete Lagoas e na Triagem da Otorrinolaringologia da UNIFESP-EPM. RESULTADOS: Há diferenças estatisticamente significativas quanto à s variáveis idade, média salarial dos pais empregados, número de irmãos até 17 anos, rinite, doenças otológicas, exames solicitados, prescrições e indicações de cirurgias micro-otológicas. CONCLUSÕES: O diagnóstico mais encontrado foi respiração oral. O exame complementar e a cirurgia mais indicados foram a radiografia de cavum e a adenoamigdalectomia. Clinical and videofluoroscopic evaluation of deglutition in children with tetraparetic spastic cerebral palsy
Cerebral palsy is a condition that may be associated with swallowing disorders, that is, oropharingeal dysphagia. The aim of this study was to characterize and compare the swallowing processes of 32 children with this condition, by clinical and videofluoroscopic evaluation, with special focus on tracheal aspiration detectability. Results show an important compromise of oral phase. The most important findings during the pharyngeal phase were velopharingeal incompetence and residuals on the pharyngeal recesses. Aspiration was more common with liquids, before and after deglutition. On clinical and videofluoroscopic evaluation, cervical hyperextension was the commonest postural abnormality. Videofluoroscopy confirmed the occurrence of aspiration on most of the cases that presented suggestive signs of aspiration during clinical evaluation. We conclude that clinical and videofluoroscopic evaluations are complementary on deglutition evaluation and together may point to the most specific rehabilitation procedure.A paralisia cerebral é doença que pode cursar com distúrbios da deglutição (disfagias orofarÃngeas). O objetivo deste artigo foi caracterizar e comparar a deglutição em 32 crianças com esta doença, pela avaliação clÃnica fonoaudiológica e videofluoroscópica, com ênfase na aspiração traqueal, Os resultados mostraram importante comprometimento da fase oral. Na fase farÃngea a aspiração, a incompetência velofarÃngea e o resÃduo em recessos farÃngeos, foram os achados mais encontrados. A aspiração foi encontrada mais frequentemente com lÃquidos, antes e durante a deglutição. Na avaliação clÃnica e na videofluoroscopia, a hiperextensão cervical foi a anormalidade postural mais encontrada. A videofluoroscopia confirmou a presença de aspiração na maioria dos casos que apresentaram sinais sugestivos de aspiração na avaliação clÃnica da deglutição. Suggestion of topographic staging for laryngeal papillomatosis
Laryngeal Papillomatosis is a disease that most often affect the true vocal folds, epiglottis, false vocal folds and may extend in the entire larynx. There isn't a consensus about a classification of the site and intensive of the lesions. AIM: To propose a staging based on the classification of the site and intensity of the lesions in the larynx that can have easily application for the physician. STUDY DESIGN: Clinical prospective. MATERIAL AND METHOD: We divided the larynx in three sites: supraglottis (S), glottis (G) and infraglottis (I); and in four degree of extension. Classification: S1 (focal lesion), S2 (less than 2/3 of extension of the lumen), S3 (more than 2/3 of extension) and S4 (obstructive lesion or tracheotomy); G1 (focal lesion on the fold or on the anterior or posterior commissure - less than 1/3 of extension), G1a (only one vocal fold) G1b (both vocal folds), G2 (more than 1/3 extension of the vocal fold), G2a (only one vocal fold), G2b (both vocal folds), G3 (more than 2/3 extension of the vocal fold) and G4 (obstructive lesion or tracheotomy); I1 (focal lesion), I2 (less than 2/3 extension of the lumen), I3 (more than 2/3 extension of the lumen), and I4 (obstructive lesion or tracheotomy). Based on this topographic classification we obtain 4 stages: I, II, III e IV. We evaluated 74 laryngoscopes of 10 patients with laryngeal papillomatosis before and after the treatment with cidofovir to establish the viability of the staging. RESULTS: All the exams could be submitted to the classification and staging. CONCLUSION: We concluded that the staging proposed was viable on the studied population.A papilomatose larÃngea (PL) é uma doença que acomete principalmente as pregas vocais, epiglote e pregas vestibulares, mas pode atingir a laringe em toda sua extensão. Até o momento não existe consenso sobre como estadiar o local e a intensidade das lesões papilomatosas. OBJETIVO: Propor um estadiamento a partir da classificação do local e intensidade das lesões na laringe, que seja abrangente, de fácil entendimento e aplicação clÃnica pelo otorrinolaringologista. FORMA DE ESTUDO: ClÃnico prospectivo. MATERIAL E MÉTODO: Partimos da divisão da laringe em três nÃveis: supraglote (S), glote (G) e infraglote (I); e da adoção de quatro graus de acometimento, de 1 (leve) a 4 (mais intenso). Classificação proposta: S1 (lesão focal), S2 (menos de 2/3 de extensão da luz), S3 (mais de 2/3 da luz), S4 (obstrutiva ou traqueotomia); G1 (focal em prega vocal ou com.anterior ou posterior - menos de 1/3 de extensão da prega), G1a (em 1 prega vocal) G1b (nas duas pregas), G2 (mais de 1/3 de extensão), G2a (em 1 prega vocal), G2b (nas duas pregas), G3 (mais de 2/3 extensão), G3a (1 prega vocal), G3b (nas duas pregas), G4 (lesão obstrutiva ou traqueotomia); I1(focal), I2 (menos 2/3 extensão da luz), I3 (mais de 2/3 extensão) e I4 (obstrutiva ou traqueotomia). Baseando-se nesta classificação topográfica, obtivemos 4 estádios: I, II, III e IV. Realizamos avaliação de 74 laringoscopias de 10 pacientes com PL antes e após o tratamento com cidofovir para constatar a viabilidade desta proposta de estadiamento. RESULTADOS: Todos os 74 exames analisados puderam ser submetidos à classificação e estadiamento. Oral breathing and speech disorders in children
OBJECTIVE: To assess speech alterations in mouth-breathing children, and to correlate Mouth breathing; them with the respiratory type, etiology, gender, and age. METHOD: A total of 439 mouth-breathers were evaluated, aged between 4 and 12 years. The presence of speech alterations in children older than 5 years was considered delayed speech development. The observed alterations were tongue interposition (TI), frontal lisp (FL), articulatory disorders (AD), sound omissions (SO), and lateral lisp (LL). The etiology of mouth breathing, gender, age, respiratory type, and speech disorders were correlated. RESULTS: Speech alterations were diagnosed in 31.2% of patients, unrelated to the respiratory type: oral or mixed. Increased frequency of articulatory disorders and more than one speech disorder were observed in males. TI was observed in 53.3% patients, followed by AD in 26.3%, and by FL in 21.9%. The co-occurrence of two or more speech alterations was observed in 24.8% of the children. CONCLUSION: Mouth breathing can affect speech development, socialization, and school performance. Early detection of mouth breathing is essential to prevent and minimize its negative effects on the overall development of individuals.OBJETIVO: Verificar alterações na fala em crianças respiradoras orais e relacioná-las com o tipo respiratório, a etiologia, o gênero e a idade. MÉTODO: Foram avaliados 439 respiradores orais com idade entre quatro e 12 anos. Considerou-se atraso no desenvolvimento de fala a presença de alterações em crianças acima de cinco anos de idade. As alterações observadas foram interposição de lÃngua (IL), ceceio frontal (CF), troca articulatória (TA), omissões (OM) e ceceio lateral (CL). Relacionou-se etiologia da respiração oral, gênero, idade, tipo respiratório e alterações de fala. RESULTADOS: Alterações de fala foram diagnosticadas em 31,2% dos pacientes sem relação com o tipo respiratório: oral ou misto. Maior frequência de trocas articulatórias e mais de uma alteração de fala ocorreram no gênero masculino. IL foi documentada em 53,3% pacientes, seguida por TA em 26,3% e CF em 21,9%. Concomitância de duas ou mais alterações de fala ocorreu em 24,8% das crianças. CONCLUSÃO: Respirar pela boca pode afetar o desenvolvimento da fala, a socialização e o desempenho escolar. Adenotonsilectomia em pacientes na fase de crescimento facial: efeitos dentários espontâneos
Children with hypertrophic tonsils and adenoids may have adverse effects on dental occlusion, which tend to worsen during the growth period. Diagnosis and early treatment is essential. AIM: Prospective clinical study to compare the cephalometric measurements before and after adenotonsillectomy in mouth breathing patients. MATERIAL AND METHOD: We had 38 patients of both genders, aged between 7 and 11 years in our sample, broken down into: oral group, 18 patients with obstructive hypertrophy of pharyngeal tonsil and/or palate grade 3 or 4; control group, 20 patients with normal breathing. Angular and linear dental measurements were compared between the groups in a 14 months interval. We used the t Student and Wilcoxon tests for unpaired samples, at 5% significance, for statistical purposes. RESULTS: The sagittal position and axial angle of the lower incisors increased significantly in the group with oral breathing, the sagittal position of the upper incisors increased significantly in the oral group, which still had a significant increase in overbite. CONCLUSION: Adenotonsillectomy was very effective in improving some dental measurements, with benefits to growing patients preventing malocclusions from becoming difficult to treat or permanent.Crianças com tonsilas e adenoides hipertróficas podem apresentar efeitos desfavoráveis na oclusão dentária, que tendem a agravar no perÃodo de crescimento, tornando imprescindÃvel seu diagnóstico e tratamento precoce. OBJETIVO: Este estudo clÃnico prospectivo comparou medidas cefalométricas dos incisivos antes e após a adenotonsilectomia, em respiradores orais. MATERIAL E MÉTODO: A amostra foi de 38 pacientes de ambos os gêneros, entre 7 e 11 anos, dividida em: grupo oral, com 18 pacientes com hipertrofia obstrutiva da tonsila farÃngea e/ou palatinas grau 3 ou 4; grupo controle, com 20 pacientes com respiração nasal. Medidas dentárias angulares e lineares foram comparadas entre os grupos, antes e após adenotonsilectomia, em um intervalo de 14 meses. A análise estatÃstica utilizou os testes t-Student e Wilcoxon para amostras não pareadas, ao nÃvel de significância de 5%. RESULTADOS: A inclinação axial e a posição sagital dos incisivos inferiores aumentaram significativamente no grupo com respiração oral; o posicionamento sagital dos incisivos superiores aumentou significativamente no grupo oral, que ainda obteve aumento significativo de sobremordida.
