    Damage mechanics based predictions of creep crack growth in 316 stainless steel

    This paper describes a novel modelling process for creep crack growth prediction of a 316 stainless steel using continuum damage mechanics, in conjunction with finite element (FE) analysis. A damage material behaviour model, proposed by Liu and Murakami [1], was used which is believed to have advantages in modelling components with cracks. The methods used to obtain the material properties in the multiaxial form of the creep damage and creep strain equations are described, based on uniaxial creep and creep crack growth test data obtained at 600 °C. Most of the material constants were obtained from uniaxial creep test data. However, a novel procedure was developed to determine the tri-axial stress state parameter in the damage model by use of creep crack growth data obtained from testing of compact tension (CT) specimens. The full set of material properties derived were then used to model the creep crack growth for a set of thumbnail crack specimen creep tests which were also tested at 600 °C. Excellent predictions have been achieved when comparing the predicted surface profiles to those obtained from experiments. The results obtained clearly show the validity and capability of the continuum damage modelling approach, which has been established, in modelling the creep crack growth for components with complex initial crack shapes

    Long-term trends in tropical cyclone tracks around Korea and Japan in late summer and early fall

    This study investigates long-term trends in tropical cyclones (TCs) over the extratropical western North Pacific (WNP) over a period of 35 years (1982-2016). The area analyzed extended across 30-45 degrees N and 120-150 degrees E, including the regions of Korea and Japan that were seriously affected by TCs. The northward migration of TCs over the WNP to the mid-latitudes showed a sharp increase in early fall. In addition, the duration of TCs over the WNP that migrated northwards showed an increase, specifically in early to mid-September. Therefore, more recently, TC tracks have been observed to significantly extend into the mid-latitudes. The recent northward extension of TC tracks over the WNP in early fall was observed to be associated with changes in environmental conditions that were favorable for TC activities, including an increase in sea surface temperature (SST), decrease in vertical wind shear, expansion of subtropical highs, strong easterly steering winds, and an increase in relative vorticity. In contrast, northward migrations of TCs to Korea and Japan showed a decline in late August, because of the presence of unfavorable environmental conditions for TC activities. These changes in environmental conditions, such as SST and vertical wind shear, can be partially associated with the Pacific decadal oscillation

    Monitoring Chemical Pollution in Europe’s Seas: Programmes, practices and priorities for research

    This report has been produced by the Marine Board Working Group on Existing and Emerging Chemical Pollutants (WGPOL) first convened in 2008 and tasked to examine the assessment and monitoring of existing and emerging chemicals in the European marine and coastal environment. The Working Group considered (i) existing monitoring/assessment frameworks; (ii) current monitoring practices; and (iii) new and emerging chemicals of concern and the mechanisms used to include them in current monitoring programmes. The primary conclusions and recommendations of this position paper are: 1. Fully implement state of the art environmental risk assessment procedures (combining exposure and effect assessment) to evaluate the full impact of chemical substances on the different compartments of coastal and open sea systems. 2. Further improve the coordination, cooperation and harmonization between existing monitoring efforts and those under development, to avoid duplication of effort, loss of expertise and a reduced willingness to fulfil the obligations towards regional conventions. 3. Ensure that the development and implementation of monitoring programmes for the assessment of chemicals in marine and coastal environment are based on a science-based and dynamic process. 4. Apply more resources targeted at developing appropriate approaches, tools and practices (education and training) to improve the acquisition and management of monitoring data. In addition to the above main recommendations, two further recommendations have been identified on the basis of two specific case studies which form part of this paper and which focus on the release, effects and monitoring of (i) hydrophobic and insoluble chemicals in the marine environment from merchant shipping; and (ii) chemicals released by the offshore oil-industry in the North Sea. These case studies highlighted the need to: 5. Develop a consistent, pan-European or regional (legal) framework/regulation which covers the activities of the oil and gas industry at sea. At the same time, more information and research is needed on the release and the effects of chemicals arising from offshore oil and gas activities. 6. Develop and apply state-of-the-art environmental risk assessment procedures (combining exposure and effect assessments, including on human health) to evaluate the impact of noxious liquid substances listed under MARPOL Annex II on the different compartments in coastal and open sea ecosystems

    Sorting Nexin 24 is required for α-granule biogenesis and cargo delivery in megakaryocytes

    Germline defects affecting the DNA-binding domain of the transcription factor FLI1 are associated with a bleeding disorder that is characterised by the presence of large, fused α-granules in platelets. We investigated whether the genes showing abnormal expression in FLI1-deficient platelets could be involved in platelet α-granule biogenesis by undertaking transcriptome analysis of control platelets and platelets harbouring a DNA-binding variant of FLI1. Our analysis identified 2276 transcripts that were differentially expressed in FLI1- deficient platelets. Functional annotation clustering of the coding transcripts revealed significant enrichment for gene annotations relating to protein transport, and identified Sorting nexin 24 (SNX24) as a candidate for further investigation. Using an iPSC-derived megakaryocyte model, SNX24 expression was found to be increased during the early stages of megakaryocyte differentiation and downregulated during proplatelet formation, indicating tight regulatory control during megakaryopoiesis. CRISPR-Cas9 mediated knockout (KO) of SNX24 led to decreased expression of immature megakaryocyte markers, CD41 and CD61, and increased expression of the mature megakaryocyte marker CD42b (p=0.0001), without affecting megakaryocyte polyploidisation, or proplatelet formation. Electron microscopic analysis revealed an increase in empty membrane-bound organelles in SNX24 KO megakaryocytes, a reduction in α-granules and an absence of immature and mature multivesicular bodies, consistent with a defect in the intermediate stage of α-granule maturation. Co-localisation studies showed that SNX24 associates with each compartment of α-granule maturation. Reduced expression of CD62P and VWF was observed in SNX24 KO megakaryocytes. We conclude that SNX24 is required for α-granule biogenesis and intracellular trafficking of α-granule cargo within megakaryocytes

    Tune in to your emotions: a robust personalized affective music player

    The emotional power of music is exploited in a personalized affective music player (AMP) that selects music for mood enhancement. A biosignal approach is used to measure listeners’ personal emotional reactions to their own music as input for affective user models. Regression and kernel density estimation are applied to model the physiological changes the music elicits. Using these models, personalized music selections based on an affective goal state can be made. The AMP was validated in real-world trials over the course of several weeks. Results show that our models can cope with noisy situations and handle large inter-individual differences in the music domain. The AMP augments music listening where its techniques enable automated affect guidance. Our approach provides valuable insights for affective computing and user modeling, for which the AMP is a suitable carrier application

    Black Hole Spin via Continuum Fitting and the Role of Spin in Powering Transient Jets

    The spins of ten stellar black holes have been measured using the continuum-fitting method. These black holes are located in two distinct classes of X-ray binary systems, one that is persistently X-ray bright and another that is transient. Both the persistent and transient black holes remain for long periods in a state where their spectra are dominated by a thermal accretion disk component. The spin of a black hole of known mass and distance can be measured by fitting this thermal continuum spectrum to the thin-disk model of Novikov and Thorne; the key fit parameter is the radius of the inner edge of the black hole's accretion disk. Strong observational and theoretical evidence links the inner-disk radius to the radius of the innermost stable circular orbit, which is trivially related to the dimensionless spin parameter a_* of the black hole (|a_*| < 1). The ten spins that have so far been measured by this continuum-fitting method range widely from a_* \approx 0 to a_* > 0.95. The robustness of the method is demonstrated by the dozens or hundreds of independent and consistent measurements of spin that have been obtained for several black holes, and through careful consideration of many sources of systematic error. Among the results discussed is a dichotomy between the transient and persistent black holes; the latter have higher spins and larger masses. Also discussed is recently discovered evidence in the transient sources for a correlation between the power of ballistic jets and black hole spin.Comment: 30 pages.
