    We show theoretically that a quantum-defect below 1% is possible in tandem-pumped Yb-doped aluminosilicate fibers operating off the gain peak. Experimentally, we reach a quantum defect of 2% and a slope efficiency of 90% or more

    Kinematics and helicity evolution of a loop-like eruptive prominence

    We aim at investigating the morphology, kinematic and helicity evolution of a loop-like prominence during its eruption. We use multi-instrument observations from AIA/SDO, EUVI/STEREO and LASCO/SoHO. The kinematic, morphological, geometrical, and helicity evolution of a loop-like eruptive prominence are studied in the context of the magnetic flux rope model of solar prominences. The prominence eruption evolved as a height expanding twisted loop with both legs anchored in the chromosphere of a plage area. The eruption process consists of a prominence activation, acceleration, and a phase of constant velocity. The prominence body was composed of left-hand (counter-clockwise) twisted threads around the main prominence axis. The twist during the eruption was estimated at 6pi (3 turns). The prominence reached a maximum height of 526 Mm before contracting to its primary location and partially reformed in the same place two days after the eruption. This ejection, however, triggered a CME seen in LASCO C2. The prominence was located in the northern periphery of the CME magnetic field configuration and, therefore, the background magnetic field was asymmetric with respect to the filament position. The physical conditions of the falling plasma blobs were analysed with respect to the prominence kinematics. The same sign of the prominence body twist and writhe, as well as the amount of twisting above the critical value of 2pi after the activation phase indicate that possibly conditions for kink instability were present. No signature of magnetic reconnection was observed anywhere in the prominence body and its surroundings. The filament/prominence descent following the eruption and its partial reformation at the same place two days later suggest a confined type of eruption.     Humidity responsivity of poly(methyl methacrylate)-based optical fiber Bragg grating sensors

    The humidity response of poly(methyl methacrylate) (PMMA)-based optical fiber Bragg gratings (POFBGs) has been studied. The characteristic wavelength of the grating is modulated by water absorption-induced swelling and refractive index change in the fiber. This work indicates that anisotropic expansion may exist in PMMA optical fiber, reducing the humidity responsivity of the grating and introducing uncertainty in the responsivity from fiber to fiber. By pre-straining a grating, one can get rid of this uncertainty and simultaneously improve the POFBG response time

    Broken unitarity and phase measurements in Aharonov-Bohm interferometers

    Aharonov-Bohm mesoscopic solid-state interferometers yield a conductance which contains a term cos(ϕ+β)\cos(\phi+\beta), where ϕ\phi relates to the magnetic flux. Experiments with a quantum dot on one of the interfering paths aim to relate β\beta to the dot's intrinsic Friedel transmission phase, α1\alpha_1. For closed systems, which conserve the electron current (unitarity), the Onsager relation requires that β=0\beta=0. For open systems, we show that β\beta depends in general on the details of the broken unitarity. Although it gives information on the resonances of the dot, β\beta is generally not equal to α1\alpha_1.     WebTraceMiner: a web service for processing and mining EST sequence trace files

    Expressed sequence tags (ESTs) remain a dominant approach for characterizing the protein-encoding portions of various genomes. Due to inherent deficiencies, they also present serious challenges for data quality control. Before GenBank submission, EST sequences are typically screened and trimmed of vector and adapter/linker sequences, as well as polyA/T tails. Removal of these sequences presents an obstacle for data validation of error-prone ESTs and impedes data mining of certain functional motifs, whose detection relies on accurate annotation of positional information for polyA tails added posttranscriptionally. As raw DNA sequence information is made increasingly available from public repositories, such as NCBI Trace Archive, new tools will be necessary to reanalyze and mine this data for new information. WebTraceMiner (www.conifergdb.org/software/wtm) was designed as a public sequence processing service for raw EST traces, with a focus on detection and mining of sequence features that help characterize 3′ and 5′ termini of cDNA inserts, including vector fragments, adapter/linker sequences, insert-flanking restriction endonuclease recognition sites and polyA or polyT tails. WebTraceMiner complements other public EST resources and should prove to be a unique tool to facilitate data validation and mining of error-prone ESTs (e.g. discovery of new functional motifs)

    Serum Amyloid A Is Not Incorporated into HDL during HDL Biogenesis

    Liver-derived serum amyloid A (SAA) is present in plasma where it is mainly associated with HDL and from which it is cleared more rapidly than are the other major HDL-associated apolipoproteins. Although evidence suggests that lipid-free and HDL-associated forms of SAA have different activities, the pathways by which SAA associates and disassociates with HDL are poorly understood. In this study, we investigated SAA lipidation by hepatocytes and how this lipidation relates to the formation of nascent HDL particles. We also examined hepatocyte-mediated clearance of lipid-free and HDL-associated SAA. We prepared hepatocytes from mice injected with lipopolysaccharide or an SAA-expressing adenoviral vector. Alternatively, we incubated primary hepatocytes from SAA-deficient mice with purified SAA. We analyzed conditioned media to determine the lipidation status of endogenously produced and exogenously added SAA. Examining the migration of lipidated species, we found that SAA is lipidated and forms nascent particles that are distinct from apoA-I-containing particles and that apoA-I lipidation is unaltered when SAA is overexpressed or added to the cells, indicating that SAA is not incorporated into apoA-I-containing HDL during HDL biogenesis. Like apoA-I formation, generation of SAA-containing particles was dependent on ABCA1, but not on scavenger receptor class B type I. Hepatocytes degraded significantly more SAA than apoA-I. Taken together, our results indicate that SAA\u27s lipidation and metabolism by the liver is independent of apoA-I and that SAA is not incorporated into HDL during HDL biogenesis

    Serum Amyloid A3 is a High Density Lipoprotein-Associated Acute-Phase Protein

    Serum amyloid A (SAA) is a family of acute-phase reactants. Plasma levels of human SAA1/SAA2 (mouse SAA1.1/2.1) can increase ≥ 1,000-fold during an acute-phase response. Mice, but not humans, express a third relatively understudied SAA isoform, SAA3. We investigated whether mouse SAA3 is an HDL-associated acute-phase SAA. Quantitative RT-PCR with isoform-specific primers indicated that SAA3 and SAA1.1/2.1 are induced similarly in livers (∼2,500-fold vs. ∼6,000-fold, respectively) and fat (∼400-fold vs. ∼100-fold, respectively) of lipopolysaccharide (LPS)-injected mice. In situ hybridization demonstrated that all three SAAs are produced by hepatocytes. All three SAA isoforms were detected in plasma of LPS-injected mice, although SAA3 levels were ∼20% of SAA1.1/2.1 levels. Fast protein LC analyses indicated that virtually all of SAA1.1/2.1 eluted with HDL, whereas ∼15% of SAA3 was lipid poor/free. After density gradient ultracentrifugation, isoelectric focusing demonstrated that ∼100% of plasma SAA1.1 was recovered in HDL compared with only ∼50% of SAA2.1 and ∼10% of SAA3. Thus, SAA3 appears to be more loosely associated with HDL, resulting in lipid-poor/free SAA3. We conclude that SAA3 is a major hepatic acute-phase SAA in mice that may produce systemic effects during inflammation
