    Changing cultural pathways through gender role and sexual development: A theoretical framework

    Greenfield's theory linking sociodemographic change to dynamic cultural values for family interdependence versus individual independence is applied to sexual and gender role socialization and development. The theory explains how cultural pathways for sexual and gender-role development transform in concert with sociodemographic changes: urbanization, formal schooling, capitalism, and communication technologies. As environments become more urban, commercial, and technological, with more opportunities for formal education, sexual development moves away from the ideals of procreation and family responsibility and toward the ideals of personal pleasure and personal responsibility. At the same time, gender-role development moves away from the ideals of complementary and ascribed gender roles and toward chosen and equal gender roles. We present psychological, anthropological, and sociological evidence for these trends in a variety of communities undergoing social and ecological change. © 2014 by the American Anthropological Association

    Pulsed-Field Gradient NMR Self Diffusion and Ionic Conductivity Measurements for Liquid Electrolytes Containing LiBF₄ and Propylene Carbonate

    Liquid electrolytes have been prepared using lithium tetrafluoroborate (LiBF₄) and propylene carbonate (PC). Pulsed-field gradient nuclear magnetic resonance (PFG-NMR) measurements were taken for the cation, anion and solvent molecules using lithium (⁷Li), fluorine (¹⁹F) and hydrogen (¹H) nuclei, respectively. It was found that lithium diffusion was slow compared to the much larger fluorinated BF₄ anion likely resulting from a large solvation shell of the lithium. Ionic conductivity and viscosity have also been measured for a range of salt concentrations and temperatures. By comparing the measured conductivity with a ideal predicted conductivity derived from the Nernst-Einstein equation and self diffusion coefficients the degree of ionic association of the anion and cation was determined and was observed to increase with salt concentration and temperature. Using the measured viscosity and self diffusion coefficients the effective radius of each of the species was determined for various salt concentrations

    Pleistocene uplift and palaeoenvironments of Macquarie Island: evidence from palaeobeaches and sedimentary deposits

    Macquarie Island (54°30'S, 159°00'E) is an emergent part of the Macquarie Ridge Complex composed of ocean-floor rocks of Miocene age now 4000 m above the ocean floor. A number of landforms, including palaeobeaches now above sea level (a.s.l.)on Macquarie Island, were formed by marine erosion during uplift of the island. During the last Pleistocene period of low sea level (c. 20 ka) the island was three times larger than now. Thermoluminescence (TL) dating of two palaeobeaches indicates Pleistocene ages: 172 ± 40 ka for one at 100 m a.s.l. and 340 ± 80 ka for another at 263 m a.s.l. Matching the altitude sequence of palaeobeaches on Macquarie Island with the pattern of peaks in world sea level determined from deep sea cores allows an independent estimate of beach ages. Comparison of the altitude and sea level sequences most plausibly places the 100 m palaeobeach in Oxygen Isotope Stage 5e (130-125 ka) and the 263 m palaeobeach in Stage 9 (340-330 ka), matching reasonably with the TL dates. Other palaeobeaches at about 50 m and 170-190 m a.s.l. then correlate with high sea levels. We calculate an average rate of uplift forthe island of 0.8 mma-I . At this rate, 4000 m of Macquarie Ridge uplift would have taken about five million years and the top of the island may first have emerged some 700 to 600 ka. During the six Pleistocene glacial-interglacial cycles since then, there has been periglacial rather than glacial activity on cold uplands, but conditions suitable for vegetation of the present type persisted close to sea level

    Clinical expression of plakophilin-2 mutations in familial arrhythmogenic right ventricular cardiomyopathy

    Background - Arrhythmogenic right ventricular cardiomyopathy (ARVC) is an inherited cardiac disorder characterized by loss of cardiomyocytes and their replacement by adipose and fibrous tissue. It is considered a disease of cell adhesion because mutations in desmosomal genes, desmoplakin and plakoglobin, have been implicated in the pathogenesis of ARVC. In a recent report, mutations in plakophilin-2, a gene highly expressed in cardiac desmosomes, have been shown to cause ARVC.Methods and Results - We investigated 100 white patients with ARVC for mutations in plakophilin-2. Nine different mutations were identified by direct sequencing in 11 cases. Five of these mutations are novel (A733fsX740, L586fsX658, V570fsX576, R413X, and P533fsX561) and predicted to cause a premature truncation of the plakophilin-2 protein. Family studies showed incomplete disease expression in mutation carriers and identified a number of individuals who would be misdiagnosed with the existing International Task Force and modified diagnostic criteria for ARVC.Conclusions - In this study, we provide new evidence that mutations in the desmosomal plakophilin-2 gene can cause ARVC. A systematic clinical evaluation of mutation carriers within families demonstrated variable phenotypic expression, even among individuals with the same mutation, and highlighted the need for a more accurate set of diagnostic criteria for ARVC

    The effect of time constraint on anticipation, decision making, and option generation in complex and dynamic environments

    Researchers interested in performance in complex and dynamic situations have focused on how individuals predict their opponent(s) potential courses of action (i.e., during assessment) and generate potential options about how to respond (i.e., during intervention). When generating predictive options, previous research supports the use of cognitive mechanisms that are consistent with long-term working memory (LTWM) theory (Ericsson and Kintsch in Phychol Rev 102(2):211–245, 1995; Ward et al. in J Cogn Eng Decis Mak 7:231–254, 2013). However, when generating options about how to respond, the extant research supports the use of the take-the-first (TTF) heuristic (Johnson and Raab in Organ Behav Hum Decis Process 91:215–229, 2003). While these models provide possible explanations about how options are generated in situ, often under time pressure, few researchers have tested the claims of these models experimentally by explicitly manipulating time pressure. The current research investigates the effect of time constraint on option-generation behavior during the assessment and intervention phases of decision making by employing a modified version of an established option-generation task in soccer. The results provide additional support for the use of LTWM mechanisms during assessment across both time conditions. During the intervention phase, option-generation behavior appeared consistent with TTF, but only in the non-time-constrained condition. Counter to our expectations, the implementation of time constraint resulted in a shift toward the use of LTWM-type mechanisms during the intervention phase. Modifications to the cognitive-process level descriptions of decision making during intervention are proposed, and implications for training during both phases of decision making are discussed

    The development and application of audit criteria for assessing knowledge exchange plans in health research grant applications.

    Background: Research funders expect evidence of end user engagement and impact plans in research proposals. Drawing upon existing frameworks, we developed audit criteria to help researchers and their institutions assess the knowledge exchange plans of health research proposals. Findings: Criteria clustered around five themes: problem definition; involvement of research users; public and patient engagement; dissemination and implementation; and planning, management and evaluation of knowledge exchange. We applied these to a sample of grant applications from one research institution in the United Kingdom to demonstrate feasibility. Conclusion: Our criteria may be useful as a tool for researcher self-assessment and for research institutions to assess the quality of knowledge exchange plans and identify areas for systematic improvement

    Distinct Effects of Respiratory Viral Infection Models on miR-149-5p, IL-6 and p63 Expression in BEAS-2B and A549 Epithelial Cells

    \ua9 2024 by the authors. Respiratory viruses cause airway inflammation, resulting in epithelial injury and repair. miRNAs, including miR-149-5p, regulate different pathological conditions. We aimed to determine how miR-149-5p functions in regulating pro-inflammatory IL-6 and p63, key regulators of airway epithelial wound repair, in response to viral proteins in bronchial (BEAS-2B) and alveolar (A549) epithelial cells. BEAS-2B or A549 cells were incubated with poly (I:C, 0.5 \ub5g/mL) for 48 h or SARS-CoV-2 spike protein-1 or 2 subunit (S1 or S2, 1 μg/mL) for 24 h. miR-149-5p was suppressed in BEAS-2B challenged with poly (I:C), correlating with IL-6 and p63 upregulation. miR-149-5p was down-regulated in A549 stimulated with poly (I:C); IL-6 expression increased, but p63 protein levels were undetectable. miR-149-5p remained unchanged in cells exposed to S1 or S2, while S1 transfection increased IL-6 expression in BEAS-2B cells. Ectopic over-expression of miR-149-5p in BEAS-2B cells suppressed IL-6 and p63 mRNA levels and inhibited poly (I:C)-induced IL-6 and p63 mRNA expressions. miR-149-5p directly suppressed IL-6 mRNA in BEAS-2B cells. Hence, BEAS-2B cells respond differently to poly (I:C), S1 or S2 compared to A549 cells. Thus, miR-149-5p dysregulation may be involved in poly (I:C)-stimulated but not S1- or S2-stimulated increased IL-6 production and p63 expression in BEAS-2B cells

    Abnormal M1/M2 macrophage phenotype profiles in the small airway wall and lumen in smokers and chronic obstructive pulmonary disease (COPD)

We explore potential dysregulation of macrophage phenotypes in COPD pathogenesis through integrated study of human small airway tissue, bronchoalveolar lavage (BAL) and an experimental murine model of COPD. We evaluated human airway tissue and BAL from healthy controls, normal lung function smokers (NLFS), and COPD subjects. Both small airways and BAL cells were immunohistochemically stained with anti-CD68 for total macrophages and with anti-CD163 for M2, and anti-iNOS for M1 macrophages. Multiplex ELISA measured BAL cytokines. Comparable cigarette smoke-induced experimental COPD mouse model was assessed for relevant mRNA profiles. We found an increase in pro-inflammatory M1s in the small airways of NLFS and COPD compared to controls with a reciprocal decrease in M2 macrophages, which remained unchanged among pathological groups. However, luminal macrophages showed a dominant M2 phenotype in both NLFS and COPD subjects. BAL cytokine skewed towards an M2 profile with increase in CCL22, IL-4, IL-13, and IL-10 in both NLFS and COPDs. The mouse-model of COPD showed similar increase in mRNA for M2 markers. Our finding suggests abnormal macrophage switching in both mucosal and luminal areas of COPD patients, that strongly associated with cytokine balance. There may be potential for beneficial therapeutic cytokine manipulation of macrophage phenotypes in COPD
