    Vegetation history of Laysan Island, Northwestern Hawaiian Islands

    Paleoenvironmental investigations were undertaken on Laysan Island in the remote Northwestern Hawaiian Islands to investigate its flora before historical observations. Substantial impacts occurred to the island as a result of late nineteenth- and early twentieth-century guano mining, commercial feather collecting, and denudation of vegetation by feral rabbits. An account of Laysan's historically known vegetation is presented, followed by discussion of results from the investigation of a 16.41-m sediment core froin Laysan's central hypersaline lake. The 7,000-year pollen and seed record, besides indicating the former importance of Pritchardia palms on Laysan, showed the former presence of Seven Previously unkown taxa, only four of which could be identified. Diatom analysis indicated fresh to brackish lake water during the early Holocene, a finding supported by the mollusk assemblage. Diatom diversity gradually decreased over time until there is a near monoculture, with types indicating a gradual increase of salinity. Hypersaline conditions were first recognizable near the top of the sequence with the appearance of Artemia zooplankton. Generally wetter Conditions Seem to have characterized the island before about 5,150 yr B.P., with drier conditions thereafter. The pollen record also suggests two possibly very brief periods of much drier conditions, conceivably related to El Nino-Southern Oscillation episodes

    Gene expression patterns in heterozygous Plk4 murine embryonic fibroblasts

    <p>Abstract</p> <p>Background</p> <p>The polo-like kinases (Plks) are a group of serine/threonine kinases which have roles in many aspects of cellular function including the regulation of mitotic activity and cellular stress responses. This study focuses on Plk4, the most divergent member of the Plk family, which is necessary for proper cellular proliferation. More specifically, alterations in Plk4 levels cause significantly adverse mitotic defects including abnormal centrosome duplication and aberrant mitotic spindle formation. We sought to clarify the effect of reduced Plk4 levels on the cell by examining transcript profiles of <it>Plk4 </it>wild-type and heterozygous mouse embryonic fibroblasts (MEFs). Subsequently, the levels of several key proteins involved in the DNA damage response were examined.</p> <p>Results</p> <p>143 genes were found to be significantly up-regulated in the heterozygous MEFs compared to their wild-type counterparts, while conversely, 9 genes were down-regulated. Numerous genes with increased transcript levels in heterozygous MEFs were identified to be involved in p53-dependent pathways. Furthermore, examination of the promoter regions of all up- and down-regulated genes revealed that the majority contained putative p53 responsive elements.</p> <p>An analysis of transcript levels in MEFs after exposure to either ionizing or ultraviolet radiation revealed a significant change between wild type and heterozygous MEFS for Plk4 transcript levels upon only UV exposure. Furthermore, changes in protein levels of several important cell check-point and apoptosis regulators were examined, including p53, Chk1, Chk2, Cdc25C and p21. In heterozygous MEFs, p53, p21 and Chk2 protein levels were at significantly higher levels. Furthermore, p53 activity was increased 5 fold in the <it>Plk4 </it>heterozygous MEFs.</p> <p>Conclusion</p> <p>Global transcript profiles and levels of key proteins involved in cellular proliferation and DNA damage pathways were examined in wild-type and <it>Plk4 </it>heterozygous MEFs. It was determined that Plk4 haploinsufficiency leads to changes in the levels of RNA accumulation for a number of key cellular genes as well as changes in protein levels for several important cell cycle/DNA damage proteins. We propose a model in which reduced Plk4 levels invoke an increase in p53 levels that leads to the aforementioned changes in global transcription profiles.</p

    Bicyclic or tricyclic nitrogen-containing heterocyclic scaffolds were constructed rapidly by intramolecular nucleophilic aromatic substitution of metallated nitriles tethered by a urea linkage to a series of electronically unactivated heterocyclic precursors. The substitution reaction constitutes a ring expansion, enabled by the conformationally constrained tether between the nitrile and the heterocycle. Attack of the metallated urea leaving group on the nitrile generates a hydantoin that bridges the polycyclic products. X-ray crystallography reveals ring-dependant strain within the hydantoin

    Proposal for a low cost close air support aircraft for the year 2000: The Raptor

    The Raptor is a proposed low cost Close Air Support (CAS) aircraft for the U.S. Military. The Raptor incorporates a 'cranked arrow' wing planform, and uses canards instead of a traditional horizontal tail. The Raptor is designed to be capable of responsive delivery of effective ordnance in close proximity to friendly ground forces during the day, night, and 'under the weather' conditions. Details are presented of the Raptor's mission, configuration, performance, stability and control, ground support, manufacturing, and overall cost to permit engineering evaluation of the proposed design. A description of the design process and analysis methods used is also provided

    Microbial mats in the Turks and Caicos Islands reveal diversity and evolution of phototrophy in the Chloroflexota order Aggregatilineales

    Genome-resolved metagenomic sequencing approaches have led to a substantial increase in the recognized diversity of microorganisms; this included the discovery of novel metabolic pathways in previously recognized clades, and has enabled a more accurate determination of the extant distribution of key metabolisms and how they evolved over Earth history. Here, we present metagenome-assembled genomes of members of the Chloroflexota (formerly Chloroflexi or Green Nonsulfur Bacteria) order Aggregatilineales (formerly SBR1031 or Thermofonsia) discovered from sequencing of thick and expansive microbial mats present in an intertidal lagoon on Little Ambergris Cay in the Turks and Caicos Islands. These taxa included multiple new lineages of Type 2 reaction center-containing phototrophs that were not closely related to previously described phototrophic Chloroflexota—revealing a rich and intricate history of horizontal gene transfer and the evolution of phototrophy and other core metabolic pathways within this widespread phylum
