    Assessing the benthic ecological status in the stressed coastal waters of Yantai, Yellow Sea, using AMBI and M-AMBI.

    The coastal waters around Yantai have been subjected to a variety of anthropogenic pressures over the last two decades. To assess the current benthic ecological health and the recovery process of the benthic ecosystem, four surveys were conducted in 2010 and 2011. The AMBI and M-AMBI were applied to assess the benthic ecological status. The ecological status of the Sishili Bay and Taozi Bay was &quot;moderate&quot; to &quot;good&quot; at most sampling stations during four surveys, but some stations were degraded due to pollution and eutrophication induced by human activities. The ecological status improved after removal of the marine raft culture and minimizing the amount of waste water discharged into the coastal waters of Yantai. The AMBI and M-AMBI could be used as suitable bio-indicator indices to assess the benthic ecological status of coastal waters in Yantai, Shandong Province.&nbsp;</p

    Cyclic axial compressive performance of hybrid double-skin tubular square columns

    This paper presents an experimental study on the cyclic axial compressive behavior of FRP-concrete-steel hybrid double-skin tubular columns. The square column specimens were cast with an external Fiber Reinforced Polymer jackets, inner steel tube and concrete in between. The height of the columns was 500 mm and the side dimension was 150 mm. The effects of loading scheme, void ratio and diameter-thickness ratio on axial compression behavior were investigated. A total of eight columns were tested under monotonic and cyclic axial compression. The experimental results show that the effect of loading scheme on axial stress-strain envelope curve and the peak load were not significant, and the ultimate state of the square columns subjected to cyclic axial compression was very similar to that of specimens subjected to monotonic axial compression. Besides, compared with void ratio, the diameter-thickness ratio of the inner steel tube has significant influence on the peak load of the columns when subjected to cyclic axial compression

    Penaeid shrimp genome provides insights into benthic adaptation and frequent molting

    Crustacea, the subphylum of Arthropoda which dominates the aquatic environment, is of major importance in ecology and fisheries. Here we report the genome sequence of the Pacific white shrimp Litopenaeus vannamei, covering similar to 1.66 Gb (scaffold N50 605.56 Kb) with 25,596 protein-coding genes and a high proportion of simple sequence repeats (>23.93%). The expansion of genes related to vision and locomotion is probably central to its benthic adaptation. Frequent molting of the shrimp may be explained by an intensified ecdysone signal pathway through gene expansion and positive selection. As an important aquaculture organism, L. vannamei has been subjected to high selection pressure during the past 30 years of breeding, and this has had a considerable impact on its genome. Decoding the L. vannamei genome not only provides an insight into the genetic underpinnings of specific biological processes, but also provides valuable information for enhancing crustacean aquaculture

    Sex-Based Differences in Age-Related Changes of the Vertebral Column from a Bronze Age Urban Population in Ancient China

    The health disparities between males and females in bioarchaeological settings are important indicators of gender-based differences in socioeconomic roles. In this study, sex-based differences of the vertebral column in spine pathology were investigated in human skeletons excavated from a Bronze Age cemetery of the Western Zhou Dynasty at the Dahekou site in Shanxi, China. Results demonstrated that females had a higher prevalence of vertebral compressive fractures, with the majority found in those between twenty-five and thirty years old, suggesting that the fractures were a consequence of osteoporosis and its early onset in females. In contrast, males expressed overall more severe ageing in all vertebral divisions compared to females. Males also had a higher prevalence of vertebral facet joint osteoarthritis in cervical and thoracic divisions than females. Likewise, the incidence of facet joint osteoarthritis was more asymmetric between the left and right joints in males than in females. These findings reflect disparities of vertebral health between the two sexes in an urban setting, in which ageing and injuries of the vertebral column might be driven by different mechanisms. Age-related changes in female vertebral columns may have been more influenced by conditions of hormone deficiency such as menopause, while male vertebral columns might have been more prone to age-related changes due to heavy labor-induced physical stressors. Further studies on the differentiation of ageing mechanisms between the two sexes based on physiology, socioeconomic roles, and living conditions are warranted. The studies are necessary in understanding how multiple sociocultural and physiological factors contribute to health disparities in historic and contemporary environments

    Preparation and characterization of a novel composite microporous polymer electrolyte for Li-ion batteries

    A novel composite microporous polymer electrolyte composed of poly(vinylidene fluoride-co-hexafluoropropylene) (PVdF-HFP) and mesoporous SBA-15 was prepared. The composite solid polymer electrolyte (CSPE) exhibits ionic conductivity as high as 0.30 mS-(cm-1) with a composition of SBA-15 : PVdF-HFP=3 : 8 at room temperature. Infrared transmission spectroscopic results suggested that the mechanism of micropore formation is similar to that of the phase inversion. X-ray diffraction (XRD) results demonstrated that the addition of SBA-15 inhibits the crystallization of PVdF-HFP, while the SBA-15 preserves well its ordered mesoporous structure during the course of preparation. The Li/CSPE/MCF of half-cell was assembled, and it showed a good electrochemical and cyclability performance during charge-discharge cycles

    A Novel Candidate Gene Associated With Body Weight in the Pacific White Shrimp Litopenaeus vannamei

    Improvements of growth traits are always the focus in selective breeding programs for the Pacific white shrimp Litopenaeus vannamei (L. vannamei). Identification of growth-related genes or markers can contribute to the application of modern breeding technologies, and thus accelerate the genetic improvement of growth traits. The aim of this study was to identify the genes and molecular markers associated with the growth traits of L. vannamei. A population of 200 individuals was genotyped using 2b-RAD techniques for genome-wide linkage disequilibrium (LD) analysis and genome-wide association study (GWAS). The results showed that the LD decayed fast in the studied population, which suggest that it is feasible to fine map the growth-related genes with GWAS in L. vannamei. One gene designated as LvSRC, encoding the class C scavenger receptor (SRC), was identified as a growth-related candidate gene by GWAS. Further targeted sequencing of the candidate gene in another population of 322 shrimps revealed that several non-synonymous mutations within LvSRC were significantly associated with the body weight (P &lt; 0.01), and the most significant marker (SRC_24) located in the candidate gene could explain 13% of phenotypic variance. The current results provide not only molecular markers for genetic improvement in L. vannamei, but also new insights for understanding the growth regulation mechanism in penaeid shrimp

    Genome-wide identification and expression analysis of Dmrt genes in bivalves

    Abstract In recent years, some common themes in the development of sex-specific traits in different animal lineages have started to emerge since the discovery of the Dmrt (doublesex-mab3-related transcription factor gene) genes. Bivalves are characterized by a diversity of sexual systems, including simultaneous hermaphroditism, sequential hermaphroditism, and strict gonochorism. However, to date, no research has focused on the genome-wide characterization and analysis of Dmrt genes in bivalves. In this study, the identification and analysis of Dmrt genes in 15 bivalves were performed using bioinformatics methods. A total of 55 Dmrt genes were retrieved in the studied bivalve genomes. The number of Dmrt genes in different species ranged from 3 to 5. The phylogenetic tree showed that Dmrt genes in bivalves can be subdivided into 5 classes: the Dmrt2-like class, Dmrt3-like class, Dmrt4/5-like class, Dsx-like class, and scallop-specific Dmrt class. The Ka/Ks ratios suggested that all Dmrt classes underwent purifying selection pressure. Furthermore, the spatiotemporal expression of Dmrt genes in four bivalve species suggested that different Dmrt genes may have different functions, and scallop-specific Dmrt genes may play a key role in sex determination/differentiation. In general, this study provides a molecular basis for in-depth examination of the functions of Dmrt genes and phylogenomic analyses in bivalves

    The lysosome-phagosome pathway mediates immune regulatory mechanisms in Mesocentrotus nudus against Vibrio coralliilyticus infection

    Sea urchins are a popular model species for studying invertebrate diseases. The immune regulatory mechanisms of the sea urchin Mesocentrotus nudus during pathogenic infection are currently unknown. This study aimed to reveal the potential molecular mechanisms of M. nudus during resistance to Vibrio coralliilyticus infection by integrative transcriptomic and proteomic analyses. Here, we identified a total of 135,868 unigenes and 4,351 proteins in the four infection periods of 0 h, 20 h, 60 h and 100 h in M. nudus. In the I20, I60 and I100 infection comparison groups, 10,861, 15,201 and 8,809 differentially expressed genes (DEGs) and 2,188, 2,386 and 2,516 differentially expressed proteins (DEPs) were identified, respectively. We performed an integrated comparative analysis of the transcriptome and proteome throughout the infection phase and found very a low correlation between transcriptome and proteome changes. KEGG pathway analysis revealed that most upregulated DEGs and DEPs were involved in immune strategies. Notably, "lysosome" and "phagosome" activated throughout the infection process, could be considered the two most important enrichment pathways at the mRNA and protein levels. The significant increase in phagocytosis of infected M. nudus coelomocytes further demonstrated that the lysosome-phagosome pathway played an important immunological role in M. nudus resistance to pathogenic infection. Key gene expression profiles and protein-protein interaction analysis revealed that cathepsin family and V-ATPase family genes might be key bridges in the lysosome-phagosome pathway. In addition, the expression patterns of key immune genes were verified using qRT-PCR, and the different expression trends of candidate genes reflected, to some extent, the regulatory mechanism of immune homeostasis mediated by the lysosome-phagosome pathway in M. nudus against pathogenic infection. This work will provide new insights into the immune regulatory mechanisms of sea urchins under pathogenic stress and help identify key potential genes/ proteins for sea urchin immune responses
