Aptamers in Diagnostics and Treatment of Viral Infections
Aptamers are in vitro selected DNA or RNA molecules that are capable of binding a wide range of nucleic and non-nucleic acid molecules with high affinity and specificity. They have been conducted through the process known as SELEX (Systematic Evolution of Ligands by Exponential Enrichment). It serves to reach specificity and considerable affinity to target molecules, including those of viral origin, both proteins and nucleic acids. Properties of aptamers allow detecting virus infected cells or viruses themselves and make them competitive to monoclonal antibodies. Specific aptamers can be used to interfere in each stage of the viral replication cycle and also inhibit its penetration into cells. Many current studies have reported possible application of aptamers as a treatment or diagnostic tool in viral infections, e.g., HIV (Human Immunodeficiency Virus), HBV (Hepatitis B Virus), HCV (Hepatitis C Virus), SARS (Severe Acute Respiratory Syndrome), H5N1 avian influenza and recently spread Ebola. Streszczenie
Wprowadzenie: Proces degradacji białek zależny od retikulum endoplazmatycznego (ang. endoplasmic-reticulum-associated protein degradation - ERAD) odgrywa istotną rolę w utrzymaniu stanu homeostazy wewnątrzkomórkowej i dotyczy przede wszystkim białek, które nie uzyskały posttranslacyjnie prawidłowej konformacji. Mechanizm ten może zostać wykorzystany przez wirusy, zwykle celem uniknięcia detekcji przez układ odpornościowy gospodarza. Zależnie od typu wirusa, rodzaj i sposób degradacji białek bywa odmienny.
Cel pracy: Celem pracy jest przedstawienie mechanizmu ERAD oraz jego znaczenia dla cyklu infekcyjnego wybranych cząsteczek wirusowych. Dokładne poznanie tego procesu może istotnie przyczynić się do wskazania potencjalnie skutecznych, nowych przeciwwirusowych rozwiązań terapeutycznych.
Skrócony opis stanu wiedzy: W pracy omówiono mechanizm ERAD i jego wykorzystanie przez: Poliomawirusy (BK i SV-40), Herpeswirusy (Cytomegalowirus), Retrowirusy (HIV-I) oraz Hepadnawirusy (HBV).
Podsumowanie: Znaczenie ERAD w cyklu infekcyjnym wirusów jest niezwykle istotne w przebiegu zakażenia. Należy dalej badać możliwość wykorzystywania ERAD przez inne rodzaje wirusów. Warto również monitorować skuteczność inhibitorów proteasomu jako potencjalnych leków w walce z chorobami wirusowymi.

Summary
Introduction: The endoplasmic-reticulum-associated protein degradation (ERAD) pathway plays an important role in maintaining intracellular homeostasis and primarily affects proteins that have not achieved correct post-translational conformation. ERAD can be also exploited by viruses, usually in order to avoid detection by the host's immune system. Depending on the type of virus, the character and manner of protein degradation may be different.
Aim of the study: The aim of this paper is to present the ERAD mechanism and its importance for the infection cycle of select viral particles. Gaining knowledge about this process can help to identify potentially effective new antiviral therapies.
Short description of state of knowledge: The ERAD mechanism and its utilization by: Poliomaviruses (BK and SV-40), Herpesviruses (Cytomegalovirus), Retroviruses (HIV-I) and Hepadnaviruses (HBV) are discussed in the paper.
Summation: The ERAD process in the viral infection cycle is extremely important. The possibility of using ERAD by differnet viruses should be further investigated. It could be also useful to exploit proteasome inhibitors as potential drugs in the fight against viral diseases.

Enhanced expression of Fas Ligand (FasL) in the lower airways of patients with fibrotic interstitial lung diseases (ILDs)
The exact role of FasL, and particularly its soluble and membrane-bound forms, in the development of
chronic ILDs and lung fibrosis has not been extensively explored. We aimed at analyzing membrane-bound FasL
expression on alveolar macrophages (AM) and lymphocytes (AL) as well as soluble FasL (sFasL) levels in bronchoalveolar
lavage (BAL) from ILDs patients, incl. pulmonary sarcoidosis (PS), hypersensitivity pneumonitis
(HP), silicosis, asbestosis, idiopathic pulmonary fibrosis (IPF), nonspecific interstitial pneumonia (NSIP), and
healthy subjects (n = 89, 12, 7, 8, 23, 6, 17, respectively). In IPF, significantly increased percentage of AM FasL+
and CD8+FasL+ cells as well as sFasL levels in BAL were found. Increased sFasL levels were also observed in
HP. NSIP and asbestosis were characterized by higher AM FasL+ relative number; CD8+FasL+ population was
expanded in asbestosis only. There was a significant decline in AL FasL+ percentage in PS and HP. Vital capacity
was negatively correlated with sFasL levels, AM FasL+ and CD8+FasL+ cell relative count. CD4+FasL+ and
CD8+FasL+ percentage strongly correlated with BAL neutrophilia, an unfavorable prognostic factor in lung
fibrosis. The concurrent comparative BAL analysis of FasL expression indicates that FasL+ AM and AL (mainly
Tc cells) comprise an important element of the fibrotic process, mostly in IPF. FasL might play a crucial role in
other fibrosis-complicated ILDs, like NSIP and asbestosis. (Folia Histochemica et Cytobiologica 2011; Vol. 49,

The Latest Achievements in the Construction of Influenza Virus Detection Aptasensors
Aptamers are short fragments of nucleic acids, DNA or RNA that have the ability to bind selected proteins with high specificity and affinity. These properties allow them to be used as an element of biosensors for the detection of specific proteins, including viral ones, which makes it possible to design valuable diagnostic tools. The influenza virus causes a huge number of human and animal deaths worldwide every year, and contributes to remarkable economic losses. In addition, in 2020, a new threat appeared—the SARS-Cov-2 pandemic. Both disease entities, especially in the initial stage of infection, are almost identical in terms of signs and symptoms. Therefore, a diagnostic solution is needed that will allow distinguishing between both pathogens, with high sensitivity and specificity; it should be cheap, quick and possible to use in the field, for example, in a doctor’s office. All the mentioned properties are met by aptasensors in which the detection elements are specific aptamers. We present here the latest developments in the construction of various types of aptasensors for the detection of influenza virus. Aptasensor operation is based on the measurement of changes in electric impedance, fluorescence or electric signal (impedimetric, fluorescence and electrochemical aptasensors, respectively); it allows both qualitative and quantitative determinations. CD83 – niezidentyfikowany marker ucieczki nowotworu spod nadzoru immunologicznego gospodarza
Komórki dendrytyczne (DC) odgrywają znaczącą rolę w pobudzaniu limfocytów,
zwłaszcza dziewiczych, gdyż stanowią najsilniejszą frakcję komórek prezentujących
antygen (APC, ang. Antigen Presenting Cells). Znaczącą rolę w dojrzewaniu
DC pełni cząsteczka CD83, która jest jednocześnie ich markerem dojrzałości. Jej
ekspresję odkryto, zarówno na innych prawidłowych komórkach APC, jak i na
komórkach nowotworowych, co skłania do refleksji nad pytaniem, czy cząsteczka
CD83 może brać udział w patogenezie tego rodzaju schorzeń.
Celem pracy była ocena częstości występowania cząsteczki CD83, w wybranych
modelowych liniach ustalonych ludzkich nowotworów złośliwych (MDA-231
- rak sutka, TOV-112D - rak jajnika).
Wyniki otrzymano dzięki wykorzystaniu techniki immunofluorescencji bezpośredniej
w połączeniu z odczytem w cytometrze przepływowym.
W wyniku przeprowadzonych badań odnotowano znaczną ekspresję formy CD83
zakotwiczonej w błonie na powierzchni komórek badanych linii ustalonych.
Przeprowadzone doświadczenia wskazują na możliwą rolę cząsteczki CD83 w patogenezie
nowotworów. Jej wysoka ekspresja na powierzchni komórek, w połączeniu
z możliwością jej „złuszczenia” z powierzchni komórki w formie rozpuszczalnej
do mikrośrodowiska nowotworu może nasilać zjawisko immunosupresji
w układzie immunologicznym i tym samym powodować ucieczkę nowotworu
spod nadzoru immunologicznego organizmu gospodarza. W celu określenia dokładnej
roli tytułowej cząsteczki niezbędne są dalsze badania

Wprowadzenie:
W ostatnich latach potwierdzono, że większość substancji znajdujących się w surowicy krwi, obecnych jest również w ślinie. Wykazano także korelację pomiędzy poziomem substancji występujących w ślinie, ze zmianami zachodzącymi w surowicy krwi. Coraz bardziej podkreśla się liczne zalety śliny jako materiału do badań diagnostycznych, w porównaniu z innymi płynami ustrojowymi. W ostatnich dekadach wskaźniki zachorowalności i umieralności z powodu chorób wirusowych uległy znaczącej poprawie. Jednakże zbyt późna wykrywalność zakażeń wirusowych jest nadal poważnym problemem.
Cel pracy:
Celem pracy jest przedstawienie możliwości wykorzystania śliny w screeningowych badaniach chorób wirusowych.
Skrócony opis stanu wiedzy:
W pracy przytoczono rezultaty badań, do których włączono pacjentów z wirusem RSV, HIV, HAV, HBV, HCV. Potwierdzają one obecność markerów diagnostycznych poszczególnych zakażeń w ślinie, wskazując tym samym na możliwość wykorzystania śliny w diagnostyce chorób wirusowych.
Podsumowanie:
Wysoka czułość, dokładność, silna korelacja ze stopniem zaawansowania choroby, to bez wątpienia cechy, jakie wykazują badania wykonywane w surowicy krwi. Wymagają one jednak zaawansowanej aparatury, są kosztowne, dość inwazyjne, trudne w przechowywaniu. Ważne jest zatem, aby poszukiwać nowych metod, możliwych do wykorzystania w badaniach screeningowych. Badania takie powinny łączyć technologie szybkiego wykrywania z zastosowaniem łatwego do pobrania materiału. Coraz częściej podkreśla się potencjał śliny, jako materiału diagnostycznego do tego rodzaju badań.

Results in recent years have confirmed that most of substances contained in the blood serum also exist in the saliva. Significant correlations were found between salivary and serum levels. Aim of the study:
The aim of the study is to present the possibilities of using saliva in the screening of viral diseases.
Short description of state of knowledge:
The study presents results of studies which included patients with RSV, HIV, HAV, HBV, HCV. These confirms the presence of the diagnostic markers in saliva, indicating the possibility of using saliva in the diagnosis of viral diseases.
Summation:
Analysis made in the blood serum display characteristics such as high sensitivity, precision and a strong correlation with stadium of disease. However, they require the advanced apparatus, are expensive, very invasive, difficult to store. It is important to looking for a new methods that can be used in screening test which should connect rapid detection technology with using easy-to-sample collection. Increasingly, emphasizes potential of saliva as a diagnostic material for this kind of research.

EGFR potencjalnym celem dla terapii genowej nowotworów
Receptor dla naskórkowego czynnika wzrostu (ang. epidermal growth factor
receptor; EGFR) to jeden z protoonkogenów, które bardzo często ulegają nadekspresji
w komórkach nowotworowych. Podwyższoną ekspresję tego receptora
zauważono w przypadku raka trzustki, jelita grubego, wątroby, jajników, piersi
i płuc. Zwiększona ilość EGFR w błonie komórkowej komórek nowotworowych
jest szczególnie niepokojąca, ponieważ receptor ten jest zdolny do wiązania wielu
ligandów, nie tylko EGF (ang. epidermal growth factor), ale także innych cząsteczek,
takich jak na przykład TGF-α (ang. transforming growth factor-α), czy
też naskórkowy czynnik wzrostowy wiążący heparynę (HB‑EGF; heparyne-
-binding EGF‑like growth factor). Związanie się liganda ze wspomnianym wyżej
receptorem skutkuje zwiększoną proliferacją komórek nowotworowych, ich
unieśmiertelnieniem i zmniejszeniem wrażliwości na sygnały proapoptotyczne, co
w efekcie przekłada się na większą agresywność nowotworu i gorsze rokowanie.
Z tego powodu możliwość wyciszenia ekspresji EGFR w komórkach nowotworowych
wydaje się niezwykle atrakcyjną strategią w terapii nowotworów. Niniejsza
praca skupia się na zahamowaniu szlaków sygnałowych, w które zaangażowany
jest EGFR, za pomocą plazmidu kodującego wstawki shRNA (ang. short hairpin
RNA) anty-EGFR i anty-HER2. Potencjalne wykorzystanie plazmidu jako nośnika
wstawki wyciszającej wydaje się skuteczną metodą, co potwierdzono późniejszym
rozdziałem elektroforetycznym cDNA uzyskanego w reakcji RT-PCR z wyizolowanego
z komórek linii A549 RNA. Badanie stanu apoptozy w komórkach
przeprowadzono wykorzystując barwienie jodkiem propidyny (ang. propidium
iodide; PI) oraz aneksyną V. Ukazało ono znamienny statystycznie wzrost odsetka
komórek w późnej apoptozie po transfekcji plazmidem ze wstawką shRNA anty-
EGFR. Wyniki badań pozwalają sądzić, że hamowanie genu dla EGFR może
mieć znaczenie w ewentualnych próbach immunoterapii i terapii genowej niedrobnokomórkowego
raka płuca
Ekspresja transformującego czynnika wzrostu-β1 oraz jego receptora w dolnych drogach oddechowych u pacjentów z wybranymi chorobami śródmiąższowymi płuc
Transformujący czynnik wzrostu-β jest endogenną substancją uczestniczącą w regulacji
wzrostu, a także różnicowania komórek. Cytokina ta wykazuje początkowo działanie
prozapalne, później zaś immunosupresyjne. Uznaje się ją za stymulator prawidłowej
regeneracji tkanek oraz włóknienia narządów. TGF-β oddziałuje za pośrednictwem receptorów
typu I i II oraz cząsteczki pomocniczej - CD105. Celem niniejszej pracy była
ocena ekspresji TGF-β oraz CD105 w dolnych drogach oddechowych w warunkach
prawidłowych i w chorobach śródmiąższowych płuc (ang. interstitial lung diseases, ILD).
Stężenie izoformy TGF-β1 oznaczano metodą ELISA, wykorzystując materiał z płukania
oskrzelowo-pęcherzykowego (ang. bronchoalveolar lavage, BAL), pochodzący od
chorych z sarkoidozą (ang. pulmonary sarcoidosis, PS), samoistnym włóknieniem płuc
(ang. idiopathic pulmonary fibrosis, IPF), alergicznym zewnątrzpochodnym zapaleniem
pęcherzyków płucnych (ang. extrinsic allergic alveolitis, EAA) i w grupie kontrolnej.
Ekspresję CD105 zbadano dokonując znakowania immunofluorescencyjnego,
połączonego z odczytem cytometrycznym we wszystkich wymienionych grupach.
W przypadku chorych z PS i EAA oraz w grupie kontrolnej wykazano zbliżony poziom
ekspresji TGF-β1. Znaczący wzrost stężenia TGF-β1 wykazano natomiast w IPF
(w porównaniu z grupą kontrolną). Stwierdzono też, w tej jednostce chorobowej,
znacząco wyższy odsetek limfocytów BAL CD105+. Zaobserwowano także znacznie
obniżoną ekspresję CD105 na limfocytach pęcherzykowych w PS. Podsumowując,
TGF-β wykryto w całej przebadanej grupie osób. Wzmożona ekspresja tej cytokiny
cechuje chorobę IPF. Cząsteczka CD105 jest powszechnie obecna w dolnych drogach
oddechowych na makrofagach, jednocześnie wykazuje niską ekspresję na limfocytach
pęcherzykowych. Wyniki badań potwierdzają niekorzystny udział TGF-β w procesie
włóknienia płuc oraz znaczenie reaktywności komórek zapalnych na tę cytokinę

Streszczenie
Wprowadzenie:
Skóra jako największy i najłatwiej dostępny narząd stanowi atrakcyjny cel dla terapii genowej, która od wielu lat budzi ogromne nadzieje środowiska naukowego. Jednakże, próby terapii przeprowadzone z wykorzystaniem wektorów wirusowych wykazały szereg wad i ograniczeń m.in. obserwowano indukcję odpowiedzi immunologicznej, losową integrację transgenu z genomem gospodarza i/lub niską wydajność jego ekspresji. Dlatego, wciąż poszukuje się alternatywnych, skuteczniejszych i jednocześnie bezpieczniejszych metod transferu genów. Atrakcyjnej alternatywy upatruje się w metodach niewirusowych.
Cel pracy:
Przedstawienie wybranych metod niewirusowego transferu genów wykorzystywanych w terapii genowej chorób skóry.
Skrócony opis stanu wiedzy:
Terapia genowa chorób skóry obejmuje wykorzystanie wektorów plazmidowych jako nośnika genów terapeutycznych, a także metod ich dostarczania do komórek takich jak: elektroporacja, mikroiniekcja, sonikacja, wykorzystanie nośników lipidowych i polimerów kationowych.
Podsumowanie:
Niewirusowe metody transferu genów oferują pewne zalety włączając niską toksyczność, brak infekcyjności oraz łatwość i niskie koszty produkcji w porównaniu z technikami wirusowymi. Niewirusowe metody wydają się być obiecującym narzędziem terapii genowej chorób skóry w szczególności nowotworów tego narządu.

Summary
Introduction:
Skin, the largest and most accessible organ of the human body is considered as an ideal gene therapy target. However, various types of viral vectors used in classical gene therapy have a number of disadvantages, such as possibility of immune response induction, random integration of inserts into the host genome or low expression efficiency. Therefore, there is an urgent need for alternative, non-viral methods of gene transfer.
Aim of the study:
To present methods for non-viral gene transfer used in gene therapy of skin diseases.
Short description of knowledge state:
Gene therapy for skin diseases include the usage of plasmid vectors as a carrier for therapeutic genes and different methods for their delivery into cells such as: electroporation, microinjection, sonication, lipid carriers and cationic polymers.
Summary:
Non-viral gene transfer methods offer some advantages including lower toxicity, non-infectious properties, ease of production and low costs as compared to viral techniques. Non-viral approaches are the promising tool in gene therapy of skin diseases, in particular in skin cancer ceases.
