    Colloid chemistry approach to understand the storage stability of fermented carrot juice

    Probiotic-fermented carrot juice (PFCJ) is a functional food, for which colloidal stability during storage is essential. The water-soluble polysaccharides (WSPs) it contains provide this stability as colloidal stabilizers; they are pectinic, mainly containing galacturonic acid, galactose and arabinose as monosaccharide units. The particle-size distribution, ζ potential, centrifugal sediment ratio, soluble solids content, total acid and structural features of these WSPs were evaluated as functions of storage time. The PFCJ displayed pseudoplastic fluid behavior. Molecular weight and solid morphology of WSPs did not change significantly with storage, although the molecular weights of WSPs showed a slight decrease. The storage stability is ascribed to its WSPs acting as electrosteric stabilizers (which are very robust colloidal stabilizers) by adsorbing onto the insoluble plant cell wall polysaccharides which comprise the dispersed phase in the suspension. Viscosity and particle size remained relatively stable when stored at 4 °C and 25 °C, while centrifugal sediment ratio, soluble solids content and total acid all increased significantly; these can also be explained in terms of the structure of the WSPs. This can provide guidance to choosing ingredients which improve storage stability and other properties of importance to consumers by using the precepts of electrosteric colloidal stabilization: e.g. the presence of water-soluble polysaccharides with long hydrophilic moieties

    The HMGB1-RAGE axis modulates the growth of autophagy-deficient hepatic tumors

    Autophagy is an intracellular lysosomal degradative pathway important for tumor surveillance. Autophagy deficiency can lead to tumorigenesis. Autophagy is also known to be important for the aggressive growth of tumors, yet the mechanism that sustains the growth of autophagy-deficient tumors is not unclear. We previously reported that progression of hepatic tumors developed in autophagy-deficient livers required high mobility group box 1 (HMGB1), which was released from autophagy-deficient hepatocytes. In this study we examined the pathological features of the hepatic tumors and the mechanism of HMGB1-mediated tumorigenesis. We found that in liver-specific autophagy-deficient (Atg7ΔHep) mice the tumors cells were still deficient in autophagy and could also release HMGB1. Histological analysis using cell-specific markers suggested that fibroblast and ductular cells were present only outside the tumor whereas macrophages were present both inside and outside the tumor. Genetic deletion of Hmgb1 or one of its receptors, receptor for advanced glycated end product (Rage), retarded liver tumor development. HMGB1 and RAGE enhanced the proliferation capability of the autophagy-deficient hepatocytes and tumors. However, RAGE expression was only found on ductual cells and Kupffer's cells but not on hepatoctyes, suggesting that HMGB1 might promote hepatic tumor growth through a paracrine mode, which altered the tumor microenvironment. Finally, RNAseq analysis of the tumors indicated that HMGB1 induced a much broad changes in tumors. In particular, genes related to mitochondrial structures or functions were enriched among those differentially expressed in tumors in the presence or absence of HMGB1, revealing a potentially important role of mitochondria in sustaining the growth of autophagy-deficient liver tumors via HMGB1 stimulation

    Adequate 25(OH)D moderates the relationship between dietary inflammatory potential and cardiovascular health risk during the second trimester of pregnancy

    BackgroundPro-inflammatory diets play an important role in developing cardiovascular disease (CVD). Vitamin D has been demonstrated to have an anti-inflammatory effect and promote cardiovascular health (CVH). However, it is unclear whether adequate vitamin D during pregnancy protects against poor CVH caused by pro-inflammatory diets.ObjectiveTo investigate the association of pro-inflammatory diets with the cardiovascular risk (CVR) among pregnant women and whether such association was modified by vitamin D status.MethodsThe study was based on a prospective birth cohort that included 3,713 pregnant women between 16 and 23 gestational weeks. In total, 25(OH)D concentrations and high-sensitivity C-reactive protein (hs-CRP) were measured from the collected blood. The dietary inflammatory potential was evaluated using the empirical dietary inflammatory pattern (EDIP) score based on a validated food frequency questionnaire. Gestational CVR was evaluated using the CVR score based on five “clinical” CVR metrics, including body mass index, blood pressure, total cholesterol, glucose levels, and smoking status.ResultsThe proportion of women with a CVR score >0 was 54.3%. We observed a positive association between the EDIP score and CVR score. Compared with the lowest quartile, the CVR score (β = −0.114, 95% CI, −0.217, −0.011) and hs-CRP levels (β = −0.280, 95% CI, −0.495, −0.065) were lower in the highest quartile (P for trend <0.05). Increased CVR connected with high EDIP score was observed only in women with 25(OH)D concentrations <50 nmol/L (RR = 1.85; 95% CI: 1.35, 2.54). Mediation analysis revealed that the proportion of association between the EDIP score and CVR score mediated by 25(OH)D was 28.7%, and the proportion of the association between 25(OH)D and the CVR score mediated by hs-CRP was 21.9%.ConclusionThe higher dietary inflammatory potential was associated with an increased CVR during pregnancy by promoting inflammation. Adequate vitamin D could exert anti-inflammatory effects and modify such association

    Distinct roles of NMB and GRP in itch transmission

    A key question in our understanding of itch coding mechanisms is whether itch is relayed by dedicated molecular and neuronal pathways. Previous studies suggested that gastrin-releasing peptide (GRP) is an itch-specific neurotransmitter. Neuromedin B (NMB) is a mammalian member of the bombesin family of peptides closely related to GRP, but its role in itch is unclear. Here, we show that itch deficits in mice lacking NMB or GRP are non-redundant and Nmb/Grp double KO (DKO) mice displayed additive deficits. Furthermore, both Nmb/Grp and Nmbr/Grpr DKO mice responded normally to a wide array of noxious stimuli. Ablation of NMBR neurons partially attenuated peripherally induced itch without compromising nociceptive processing. Importantly, electrophysiological studies suggested that GRPR neurons receive glutamatergic input from NMBR neurons. Thus, we propose that NMB and GRP may transmit discrete itch information and NMBR neurons are an integral part of neural circuits for itch in the spinal cord

    Development of Quantitative Structure-Property Relationship Models for Self-Emulsifying Drug Delivery System of 2-Aryl Propionic Acid NSAIDs

    We developed the quantative structure-property relationships (QSPRs) models to correlate the molecular structures of surfactant, cosurfactant, oil, and drug with the solubility of poorly water-soluble 2-aryl propionic acid nonsteroidal anti-inflammatory drugs (2-APA-NSAIDs) in self-emulsifying drug delivery systems (SEDDSs). The compositions were encoded with electronic, geometrical, topological, and quantum chemical descriptors. To obtain reliable predictions, we used multiple linear regression (MLR) and artificial neural network (ANN) methods for model development. The obtained equations were validated using a test set of 42 formulations and showed a great predictive power, and linear models were found to be better than nonlinear ones. The obtained QSPR models would greatly facilitate fast screening for the optimal formulations of SEDDS at the early stage of drug development and minimize experimental effort

    Probiotic fermentation modifies the structures of pectic polysaccharides from carrot pulp

    Previous studies have suggested that water-soluble polysaccharides from fermented carrot pulp (WSP-p) have stronger anti-diabetic effects than those from un-fermented carrot pulp (WSP-n). This study aimed to improve understanding of these functional differences by comparing their molecular structures. Weight-average molecular weights of WSP-p fractions were lower than those of the corresponding WSP-n fractions. While both WSPs had similar functional groups, more fragmented particles were observed on the surface of large particles of WSP-n than WSP-p. Monosaccharide composition and methylation analysis confirmed that both WSP-p and WSP-n were pectic polysaccharides, containing rhamnogalacturonan-I-type polysaccharides with 1,4-linked α-d-galacturonic acid residues and homogalacturonan regions with 1,4-GalpA linkages. 1H and 13C NMR showed that they had similar linkage patterns. These findings suggested that probiotic fermentation of WSP mainly cleaved the linkages between repeating units, and resulted in less polydisperse molecular size distributions

    Prevalence of syphilis infection in different tiers of female sex workers in China: implications for surveillance and interventions

    <p>Abstract</p> <p>Background</p> <p>Syphilis has made a dramatic resurgence in China during the past two decades and become the third most prevalent notifiable infectious disease in China. Female sex workers (FSWs) have become one of key populations for the epidemic. In order to investigate syphilis infection among different tiers of FSWs, a cross-sectional study was conducted in 8 sites in China.</p> <p>Methods</p> <p>Serum specimens (n = 7,118) were collected to test for syphilis and questionnaire interviews were conducted to obtain socio-demographic and behavioral information among FSWs recruited from different types of venues. FSWs were categorized into three tiers (high-, middle- and low-tier FSWs) based on the venues where they solicited clients. Serum specimens were screened with enzyme-linked immunosorbent assay (ELISA) for treponemal antibody followed by confirmation with non-treponemal toluidine red unheated serum test (TRUST) for positive ELISA specimens to determine syphilis infection. A logistic regression model was used to determine factors associated with syphilis infection.</p> <p>Results</p> <p>Overall syphilis prevalence was 5.0% (95%CI, 4.5-5.5%). Low-tier FSWs had the highest prevalence (9.7%; 95%CI, 8.3-11.1%), followed by middle-tier (4.3%; 95%CI, 3.6-5.0%, <it>P </it>< 0.001) and high-tier FSWs (2.2%; 95%CI, 1.6-2.9%, <it>P </it>< 0.001). Factors independently associated with syphilis infection included older age, lower education level, geographic location, lower tier of typology, and injection drug use.</p> <p>Conclusions</p> <p>This multi-site survey showed a high prevalence of syphilis infection among FSWs and substantial disparities in syphilis prevalence by the tier of FSWs. The difference in syphilis prevalence is substantial between different tiers of FSWs, with the highest rate among low-tier FSWs. Thus, current surveillance and intervention activities, which have low coverage in low-tier FSWs in China, should be further examined.</p

    Effects of oxytetracycline and sulfachloropyridazine residues on the reductive activity of Shewanella decolorationis S12

    Effects of oxytetracycline (OTC) and sulfachloropyridazine (SCP), two of the widely used antibiotics in livestock production, on beneficial environmental microorganisms were studied. Shewanella decolorationis S12 was selected as the target bacteria for the role in reduction of Fe(III) and dye under anaerobic conditions. The results showed that the antibiotics significantly inhibited Fe(III) reduction and dye decoloration in the reduction system. The rates of Fe(II) formed (−r) were 3.6 and 0.2 mg/L/day for the OTC concentrations of 0−1 mg/L and 1−50 mg/L, respectively, with 1 mg/L as the turning point of the inhibition effect. The turning point of inhibition effect was much higher for SCP treatments, at 4 mg/L. The results also showed higher production values for adsorbed Fe(II) than soluble Fe(II) in OTC treatments, but the reverse occurred in the SCP treatments. The difference between the treatments could be due to higher sorption coefficients of OTC as compared to SCP. Transmission electron micrographs showed changes in cell structures of S. decolorationis S12 grown in medium with OTC. Detached cell walls and large vacuoles in internal cell contents were found in OTC-treated cells. The results of the present study indicated that the inhibition of antibiotic on the reduction activity of S. decolorationis S12 may be due to a decrease in live S. decolorationis S12 population and/or damages of their cell structure in this reduction system
