    Linking Models of Land Use, Resources, and Economy to Simulate the Development of Mountain Regions (ALPSCAPE)

    We present a framework of a scenario-based model that simulates the development of the municipality of Davos (Swiss Alps). We illustrate our method with the calculation of the scenario for 2050 "Decrease in subsidies for mountain agriculture and liberalization of markets.” The main objective was to link submodels of land-use allocation (regression-based approach), material and energy flows submodels (Material and Energy Flux Analysis), and economic submodels (Input-Output Analysis). Letting qualitative and quantitative information flow from one submodel to the next, following the storyline describing a scenario, has proven to be suitable for linking submodels. The succession of the submodels is then strongly dependent on the scenario. Qualitative information flows are simulated with microsimulations of actor choices. Links between the submodels show different degrees of robustness: although the links involving microsimulations are the weakest, the uncertainty introduced by the land-use allocation model is actually advantageous because it allows one possible change in the landscape in the future to be simulated. The modeling results for the scenario here presented show that the disappearance of agriculture only marginally affects the region's factor income, but that the consequences for the self-sufficiency rate, for various landscape-related indicators and ecosystem services, and for the economy in the long term may be considerable. These benefits compensate for agriculture's modest direct economic value. The framework presented can potentially be applied to any region and scenario. This framework provides a basis for a learning package that allows potential detrimental consequences of regional development to be anticipated at an early stag

    Die Dynamik der Lohnungleichheit in deutschen Betrieben: Sonderbericht

    Lohnungleichheit ist ein breit diskutiertes Thema nicht nur in Deutschland. Dieser Forschungsbericht konzentriert sich auf die innerbetriebliche Lohndispersion und zeigt neben der Entwicklung in LPP Betrieben seit 2012 Unterschiede zwischen Branchen und Hoch- und Niedriglohnbetrieben auf. Außerdem werden relevante Einflussfaktoren identifiziert. Abschließend zeigt der Bericht, dass die hohe Einkommenszufriedenheit von Beschäftigten in LPP-Betrieben hauptsächlich ausschließlich mit dem eigenen Lohn und dem betrieblichen Durchschnittslohn korreliert und nicht mit der betrieblichen Lohndispersion.Wage inequality is a widely discussed topic not only in Germany. This research report focuses on intra-firm wage dispersion and shows differences between industries and high- and low-wage firms in addition to the development in LPP firms since 2012. It also identifies relevant influencing factors. Finally, the report shows that the high income satisfaction of employees in LPP establishments is solely correlated with employee’s own wage and the establishment average wage and not with the establishment wage dispersion

    Orally bioavailable CDK9/2 inhibitor shows mechanism-based therapeutic potential in MYCN-driven neuroblastoma

    The undruggable nature of oncogenic Myc transcription factors poses a therapeutic challenge in neuroblastoma, a pediatric cancer in which MYCN amplification is strongly associated with unfavorable outcome. Here, we show that CYC065 (fadraciclib), a clinical inhibitor of CDK9 and CDK2, selectively targeted MYCN-amplified neuroblastoma via multiple mechanisms. CDK9 — a component of the transcription elongation complex P-TEFb — bound to the MYCN-amplicon superenhancer, and its inhibition resulted in selective loss of nascent MYCN transcription. MYCN loss led to growth arrest, sensitizing cells for apoptosis following CDK2 inhibition. In MYCN-amplified neuroblastoma, MYCN invaded active enhancers, driving a transcriptionally encoded adrenergic gene expression program that was selectively reversed by CYC065. MYCN overexpression in mesenchymal neuroblastoma was sufficient to induce adrenergic identity and sensitize cells to CYC065. CYC065, used together with temozolomide, a reference therapy for relapsed neuroblastoma, caused long-term suppression of neuroblastoma growth in vivo, highlighting the clinical potential of CDK9/2 inhibition in the treatment of MYCN-amplified neuroblastoma

    Differential prognostic utility of adiposity measures in chronic kidney disease

    Objective Adipose tissue contributes to adverse outcomes in chronic kidney disease (CKD), but there is uncertainty regarding the prognostic relevance of different adiposity measures. We analyzed the associations of neck circumference (NC), waist circumference (WC), and body mass index (BMI) with clinical outcomes in patients with mild to severe CKD. Methods The German Chronic Kidney Disease (GCKD) study is a prospective cohort study, which enrolled Caucasian adults with mild to severe CKD, defined as estimated glomerular filtration rate (eGFR): 30–60 mL/min/1.73 m2, or >60 mL/min/1.73 m2 in the presence of overt proteinuria. Associations of NC, WC and BMI with all-cause death, major cardiovascular events (MACE: a composite of non-fatal stroke, non-fatal myocardial infarction, peripheral artery disease intervention, and cardiovascular death), kidney failure (a composite of dialysis or transplantation) were analyzed using multivariable Cox proportional hazards regression models adjusted for confounders and the Akaike information criteria (AIC) were calculated. Models included sex interactions with adiposity measures. Results A total of 4537 participants (59% male) were included in the analysis. During a 6.5-year follow-up, 339 participants died, 510 experienced MACE, and 341 developed kidney failure. In fully adjusted models, NC was associated with all-cause death in women (HR 1.080 per cm; 95% CI 1.009–1.155), but not in men. Irrespective of sex, WC was associated with all-cause death (HR 1.014 per cm; 95% CI 1.005–1.038). NC and WC showed no association with MACE or kidney failure. BMI was not associated with any of the analyzed outcomes. Models of all-cause death including WC offered the best (lowest) AIC. Conclusion In Caucasian patients with mild to severe CKD, higher NC (in women) and WC were significantly associated with increased risk of death from any cause, but BMI was not

    A Theoretical Analysis of the Geography of Schistosomiasis in Burkina Faso Highlights the Roles of Human Mobility and Water Resources Development in Disease Transmission

    We study the geography of schistosomiasis across Burkina Faso by means of a spatially explicit model of water-based disease dynamics. The model quantitatively addresses the geographic stratification of disease burden in a novel framework by explicitly accounting for drivers and controls of the disease, including spatial information on the distributions of population and infrastructure, jointly with a general description of human mobility and climatic/ecological drivers. Spatial patterns of disease are analysed by the extraction and the mapping of suitable eigenvectors of the Jacobian matrix subsuming the stability of the disease-free equilibrium. The relevance of the work lies in the novel mapping of disease burden, a byproduct of the parametrization induced by regional upscaling, by model-guided field validations and in the predictive scenarios allowed by exploiting the range of possible parameters and processes. Human mobility is found to be a primary control at regional scales both for pathogen invasion success and the overall distribution of disease burden. The effects of water resources development highlighted by systematic reviews are accounted for by the average distances of human settlements from water bodies that are habitats for the parasite's intermediate host. Our results confirm the empirical findings about the role of water resources development on disease spread into regions previously nearly disease-free also by inspection of empirical prevalence patterns. We conclude that while the model still needs refinements based on field and epidemiological evidence, the proposed framework provides a powerful tool for large-scale public health planning and schistosomiasis management

    DNA binding of Myc and Miz1 and transcriptional regulation of their target genes

    Die Deregulation des Transkriptionsfaktors Myc ist ein charakteristisches Merkmal für eine Vielzahl von humanen Tumoren. Durch die transkriptionelle Aktivierung von Genen, die im Zusammenhang mit Metabolismus, Translation und Proliferation stehen, wird dadurch das Tumorwachstum begünstigt. Myc bildet zudem mit dem Zinkfinger-Protein Miz1 einen Komplex, der hemmend auf die Transkription von Zielgenen wirkt. Bisher sind nur wenige Myc/Miz1-reprimierte Zielgene bekannt. In der vorliegenden Arbeit konnten genomweit die DNA-Bindestellen von Myc und Miz1 durch Chromatin-Immunpräzipitationen gefolgt von Hochdurchsatzsequenzierung in einer Zervixkarzinomzelllinie bestimmt werden. Es konnte gezeigt werden, dass Myc an Promotoren aller drei RNA-Polymerasen sowie in enhancer-Regionen bindet, während Miz1 Kernpromotoren von RNA-Polymerase II- und III-transkribierten Genen besetzt. reChIP-Experimente zeigten, dass Myc und Miz1 als Komplex an Promotoren von Zielgenen binden. Zudem wurde ein Miz1-DNA-Bindemotiv identifiziert und der transaktivierende Einfluss von Miz1 auf Gene mit diesem Motiv nachgewiesen. Das überwiegende Vorhandensein von Myc/Max-Komplexen führt zu einer Transaktivierung von E-Box-haltigen Promotoren. Andererseits erfolgt die transkriptionelle Repression von Myc/Miz1-Zielgenen an Promotoren, an denen der Myc/Miz1-Komplex vorherrscht. In aktuellen Publikationen konnte gezeigt werden, dass nach mitogener Stimulation von Lymphozyten es zu einer Erhöhung der Myc-Expression kommt, wodurch Myc als ein genereller Transkriptionsaktivator fungiert, der alle Gene gleichermaßen induziert. Trotz hoher Myc-Mengen in Tumorzellen konnte die generelle Myc-vermittelte Transaktivierung nicht nachgewiesen werden. Zusätzlich zur Myc-abhängigen Transaktivierung von E-Box-haltigen Genen, z. B. beteiligt an Translation und RNA-Prozessierung, und der Miz1-vermittelten transkriptionellen Aktivierung von Genen mit Miz1-Motiv (z. B. involviert in Autophagie), konnte entgegen dem Modell der generellen Genamplifikation durch Myc eine Myc/Miz1-abhängige Repression von Zielgenen belegt werden. Die neu gewonnenen Erkenntnisse des Bindeverhaltens des Myc/Miz1-Komplexes und der daraus resultierenden transkriptionellen Regulation von Myc/Miz1-Zielgenen ermöglichen ein besseres Verständnis der Myc-Funktion in Tumorzellen und könnte zur Verbesserung von Tumortherapien führen.Deregulation of the transcription factor Myc is a characteristic feature of a variety of human tumors. The Myc-dependent transcriptional activation of genes involved in metabolism, translation and proliferation therefor promotes tumor growth. Additionally, Myc forms a complex with the zinc finger protein Miz1, which represses transcription of target genes. So far, only a limited number of Myc/Miz1-repressed genes is known. Within the present thesis DNA binding sites of Myc and Miz1 were mapped genome-wide using chromatin immunoprecipitations followed by high-throughput sequencing in a cervical cancer cell line. It could be shown that Myc binds to promoters of all three RNA polymerases as well as to enhancer regions, whereas Miz1 binding sites could be found only in core promoters of RNA polymerase II and III transcribed genes. reChIP experiments illustrated binding of Myc and Miz1 as a complex on DNA. Additionally, a DNA binding motif of Miz1 was identified and furthermore it was possible to verify the transctivating influence of Miz1 on genes carrying that motif in the promoter. On E-box containing promoters the predominantly existence of Myc/Max complexes resulted in transactivation of the respective genes. Otherwise, transcriptional repression of Myc/Miz1 target genes occured at promoters where the Myc/Miz1 complex dominates. Recent publications have illustrated that after mitogenic stimulation of primary lymphocytes, Myc expression is enhanced, whereby Myc serves as a general transcriptional activator that induces the expression of virtually all genes. Although Myc levels are high in tumor cells that general mechanism of Myc-mediated transactivation could not be verified. Additionally to the Myc-dependent transactivation of E-box-containing genes, e. g. involved in translation and RNA processing, and Miz1-mediated transcriptional activation of genes containing a Miz1 binding motif (e. g. autophagy-related genes), and in opposition to the general amplifier model a Myc/Miz1-dependent repression of target genes could be proven. The obtained evidences concerning DNA binding properties of the Myc/Miz1 complex as well as the resulting transcriptional regulation of Myc/Miz1 target genes facilitates a better understanding of Myc function in tumor cells and could leed to better anti-tumor therapies
