    In the Fall of 2015, the HIR Fellows for Vocation entered an exploration of the Vocation Learning Outcome outlined in the Habits of Inquiry and Reflection (2006), the document undergirding the development of the Common Academic Program. The Fellows took the following steps in developing our collective understanding of vocation, the learning outcome, the University’s current approaches to addressing vocation via curricular and co-curricular offerings, and opportunities ahead. We studied David S. Cunningham’s (2015) At This Time and in This Place: Vocation and Higher Education\u27, discussed our collective understanding of vocation; studied Habits of Inquiry and Reflection; reviewed curricular offerings approved through the CAP process that included vocation as a learning outcome; reviewed co-curricular offerings related to vocation; conducted focus group interviews with students, faculty and staff who are already interested and engaged in vocation-related activities on campus; and conducted focus group interviews with undergraduate students to better understand their experiences at the University of Dayton related to vocation and vocational discernment. In this white paper, we share the results of our research and discussions and identify some of the challenges and opportunities facing our campus in meeting the University’s Vocation Learning Outcome, as articulated in Habits of Inquiry and Reflection

    Portable Rabies Virus Sequencing in Canine Rabies Endemic Countries Using the Oxford Nanopore MinION

    As countries with endemic canine rabies progress towards elimination by 2030, it will become necessary to employ techniques to help plan, monitor, and confirm canine rabies elimination. Sequencing can provide critical information to inform control and vaccination strategies by identifying genetically distinct virus variants that may have different host reservoir species or geographic distributions. However, many rabies testing laboratories lack the resources or expertise for sequencing, especially in remote or rural areas where human rabies deaths are highest. We developed a low-cost, high throughput rabies virus sequencing method using the Oxford Nanopore MinION portable sequencer. A total of 259 sequences were generated from diverse rabies virus isolates in public health laboratories lacking rabies virus sequencing capacity in Guatemala, India, Kenya, and Vietnam. Phylogenetic analysis provided valuable insight into rabies virus diversity and distribution in these countries and identified a new rabies virus lineage in Kenya, the first published canine rabies virus sequence from Guatemala, evidence of rabies spread across an international border in Vietnam, and importation of a rabid dog into a state working to become rabies-free in India. Taken together, our evaluation highlights the MinION’s potential for low-cost, high volume sequencing of pathogens in locations with limited resources

    Increased Virulence of an Epidemic Strain of Mycobacterium massiliense in Mice

    Chronic pulmonary disease and skin/soft tissue infections due to non-tuberculous mycobacteria (NTM) of the Mycobacterium chelonae-abscessus-massiliense group is an emerging health problem worldwide. Moreover, the cure rate for the infections this group causes is low despite aggressive treatment. Post-surgical outbreaks that reached epidemic proportions in Brazil recently were caused by M. massiliense isolates resistant to high-level disinfection with glutaraldehyde (GTA). Understanding the differences in the virulence and host immune responses induced by NTM differing in their sensitivity to disinfectants, and therefore their relative threat of causing outbreaks in hospitals, is an important issue.We compared the replication and survival inside macrophages of a GTA-susceptible reference Mycobacterium massiliense clinical isolate CIP 108297 and an epidemic strain from Brazil, CRM-0019, and characterized the immune responses of IFNγ knockout mice exposed to a high dose aerosol with these two isolates. CRM-0019 replicated more efficiently than CIP 108297 inside mouse bone marrow macrophages. Moreover, the animals infected with CRM-0019 showed a progressive lung infection characterized by a delayed influx of CD4+ and CD8+ T cells, culminating in extensive lung consolidation and demonstrated increased numbers of pulmonary CD4+ Foxp3+ regulatory T cells compared to those infected with the reference strain. Immunosuppressive activity of regulatory T cells may contribute to the progression and worsening of NTM disease by preventing the induction of specific protective immune responses.These results provide the first direct evidence of the increased virulence in macrophages and mice and pathogenicity in vivo of the Brazilian epidemic isolate and the first observation that NTM infections can be associated with variable levels of regulatory T cells which may impact on their virulence and ability to persist in the host

    Genome-Wide Association Analysis of Oxidative Stress Resistance in Drosophila melanogaster

    Background: Aerobic organisms are susceptible to damage by reactive oxygen species. Oxidative stress resistance is a quantitative trait with population variation attributable to the interplay between genetic and environmental factors. Drosophila melanogaster provides an ideal system to study the genetics of variation for resistance to oxidative stress. Methods and Findings: We used 167 wild-derived inbred lines of the Drosophila Genetic Reference Panel for a genomewide association study of acute oxidative stress resistance to two oxidizing agents, paraquat and menadione sodium bisulfite. We found significant genetic variation for both stressors. Single nucleotide polymorphisms (SNPs) associated with variation in oxidative stress resistance were often sex-specific and agent-dependent, with a small subset common for both sexes or treatments. Associated SNPs had moderately large effects, with an inverse relationship between effect size and allele frequency. Linear models with up to 12 SNPs explained 67–79 % and 56–66 % of the phenotypic variance for resistance to paraquat and menadione sodium bisulfite, respectively. Many genes implicated were novel with no known role in oxidative stress resistance. Bioinformatics analyses revealed a cellular network comprising DNA metabolism and neuronal development, consistent with targets of oxidative stress-inducing agents.     Citizenship Norms in Eastern Europe

    Research on Eastern Europe stresses the weakness of its civil society and the lack of political and social involvement, neglecting the question: What do people themselves think it means to be a good citizen? This study looks at citizens’ definitions of good citizenship in Poland, Slovenia, the Czech Republic and Hungary, using 2002 European Social Survey data. We investigate mean levels of civic mindedness in these countries and perform regression analyses to investigate whether factors traditionally associated with civic and political participation are also correlated with citizenship norms across Eastern Europe. We show that mean levels of civic mindedness differ significantly across the four Eastern European countries. We find some support for theories on civic and political participation when explaining norms of citizenship, but also demonstrate that individual-level characteristics are differently related to citizenship norms across the countries of our study. Hence, our findings show that Eastern Europe is not a monolithic and homogeneous bloc, underscoring the importance of taking the specificities of countries into account

    Waveform lidar over vegetation : An evaluation of inversion methods for estimating return energy

    Full waveform lidar has a unique capability to characterise vegetation in more detail than any other practical method. The reflectance, calculated from the energy of lidar returns, is a key parameter for a wide range of applications and so it is vital to extract it accurately. Fifteen separate methods have been proposed to extract return energy (the amount of light backscattered from a target), ranging from simple to mathematically complex, but the relative accuracies have not yet been assessed. This paper uses a simulator to compare all methods over a wide range of targets and lidar system parameters. For hard targets the simplest methods (windowed sum, peak and quadratic) gave the most consistent estimates. They did not have high accuracies, but low standard deviations show that they could be calibrated to give accurate energy. This may be why some commercial lidar developers use them, where the primary interest is in surveying solid objects. However, simulations showed that these methods are not appropriate over vegetation. The widely used Gaussian fitting performed well over hard targets (0.24% root mean square error, RMSE), as did the sum and spline methods (0.30% RMSE). Over vegetation, for large footprint (15 m) systems, Gaussian fitting performed the best (12.2% RMSE) followed closely by the sum and spline (both 12.7% RMSE). For smaller footprints (33 cm and 1 cm) over vegetation, the relative accuracies were reversed (0.56% RMSE for the sum and spline and 1.37% for Gaussian fitting). Gaussian fitting required heavy smoothing (convolution with an 8 m Gaussian) whereas none was needed for the sum and spline. These simpler methods were also more robust to noise and far less computationally expensive than Gaussian fitting. Therefore it was concluded that the sum and spline were the most accurate for extracting return energy from waveform lidar over vegetation, except for large footprint (15 m), where Gaussian fitting was slightly more accurate. These results suggest that small footprint (≪ 15 m) lidar systems that use Gaussian fitting or proprietary algorithms may report inaccurate energies, and thus reflectances, over vegetation. In addition the effect of system pulse length, sampling interval and noise on accuracy for different targets was assessed, which has implications for sensor design

    The Science Performance of JWST as Characterized in Commissioning

    This paper characterizes the actual science performance of the James Webb Space Telescope (JWST), as determined from the six month commissioning period. We summarize the performance of the spacecraft, telescope, science instruments, and ground system, with an emphasis on differences from pre-launch expectations. Commissioning has made clear that JWST is fully capable of achieving the discoveries for which it was built. Moreover, almost across the board, the science performance of JWST is better than expected; in most cases, JWST will go deeper faster than expected. The telescope and instrument suite have demonstrated the sensitivity, stability, image quality, and spectral range that are necessary to transform our understanding of the cosmos through observations spanning from near-earth asteroids to the most distant galaxies.Comment: 5th version as accepted to PASP; 31 pages, 18 figures; https://iopscience.iop.org/article/10.1088/1538-3873/acb29
