    Plasticity of GABA(B) receptor-mediated heterosynaptic interactions at mossy fibers after status epilepticus

    Several neurotransmitters, including GABA acting at presynaptic GABAB receptors, modulate glutamate release at synapses between hippocampal mossy fibers and CA3 pyramidal neurons. This phenomenon gates excitation of the hippocampus and may therefore prevent limbic seizure propagation. Here we report that status epilepticus, triggered by either perforant path stimulation or pilocarpine administration, was followed 24 hr later by a loss of GABAB receptor-mediated heterosynaptic depression among populations of mossy fibers. This was accompanied by a decrease in the sensitivity of mossy fiber transmission to the exogenous GABAB receptor agonist baclofen. Autoradiography revealed a reduction in GABAB receptor binding in the stratum lucidum after status epilepticus. Failure of GABAB receptor-mediated modulation of mossy fiber transmission at mossy fibers may contribute to the development of spontaneous seizures after status epilepticus

    This tutorial is devoted to review the modern tools of quantum mechanics, which are suitable to describe states, measurements, and operations of realistic, not isolated, systems in interaction with their environment, and with any kind of measuring and processing devices. We underline the central role of the Born rule and and illustrate how the notion of density operator naturally emerges, together the concept of purification of a mixed state. In reexamining the postulates of standard quantum measurement theory, we investigate how they may formally generalized, going beyond the description in terms of selfadjoint operators and projective measurements, and how this leads to the introduction of generalized measurements, probability operator-valued measures (POVM) and detection operators. We then state and prove the Naimark theorem, which elucidates the connections between generalized and standard measurements and illustrates how a generalized measurement may be physically implemented. The "impossibility" of a joint measurement of two non commuting observables is revisited and its canonical implementations as a generalized measurement is described in some details. Finally, we address the basic properties, usually captured by the request of unitarity, that a map transforming quantum states into quantum states should satisfy to be physically admissible, and introduce the notion of complete positivity (CP). We then state and prove the Stinespring/Kraus-Choi-Sudarshan dilation theorem and elucidate the connections between the CP-maps description of quantum operations, together with their operator-sum representation, and the customary unitary description of quantum evolution. We also address transposition as an example of positive map which is not completely positive, and provide some examples of generalized measurements and quantum operations.Comment: Tutorial. 26 pages, 1 figure.     Movement of cerebrospinal fluid tracer into brain parenchyma and outflow to nasal mucosa is reduced at 24 h but not 2 weeks post-stroke in mice

    Background Recent data indicates that cerebrospinal fluid (CSF) dynamics are disturbed after stroke. Our lab has previously shown that intracranial pressure rises dramatically 24 h after experimental stroke and that this reduces blood flow to ischaemic tissue. CSF outflow resistance is increased at this time point. We hypothesised that reduced transit of CSF through brain parenchyma and reduced outflow of CSF via the cribriform plate at 24 h after stroke may contribute to the previously identified post-stroke intracranial pressure elevation. Methods Using a photothrombotic permanent occlusion model of stroke in C57BL/6 adult male mice, we examined the movement of an intracisternally infused 0.5% Texas Red dextran throughout the brain and measured tracer efflux into the nasal mucosa via the cribriform plate at 24 h or two weeks after stroke. Brain tissue and nasal mucosa were collected ex vivo and imaged using fluorescent microscopy to determine the change in CSF tracer intensity in these tissues. Results At 24 h after stroke, we found that CSF tracer load was significantly reduced in brain tissue from stroke animals in both the ipsilateral and contralateral hemispheres when compared to sham. CSF tracer load was also reduced in the lateral region of the ipsilateral hemisphere when compared to the contralateral hemisphere in stroke brains. In addition, we identified an 81% reduction in CSF tracer load in the nasal mucosa in stroke animals compared to sham. These alterations to the movement of CSF-borne tracer were not present at two weeks after stroke. Conclusions Our data indicates that influx of CSF into the brain tissue and efflux via the cribriform plate are reduced 24 h after stroke.     Direct measurement of optical quasidistribution functions: multimode theory and homodyne tests of Bell's inequalities

    We develop a multimode theory of direct homodyne measurements of quantum optical quasidistribution functions. We demonstrate that unbalanced homodyning with appropriately shaped auxiliary coherent fields allows one to sample point-by-point different phase space representations of the electromagnetic field. Our analysis includes practical factors that are likely to affect the outcome of a realistic experiment, such as non-unit detection efficiency, imperfect mode matching, and dark counts. We apply the developed theory to discuss feasibility of observing a loophole-free violation of Bell's inequalities by measuring joint two-mode quasidistribution functions under locality conditions by photon counting. We determine the range of parameters of the experimental setup that enable violation of Bell's inequalities for two states exhibiting entanglement in the Fock basis: a one-photon Fock state divided by a 50:50 beam splitter, and a two-mode squeezed vacuum state produced in the process of non-degenerate parametric down-conversion.Comment: 18 pages, 7 figure

    The future of sovereignty in multilevel governance Europe: a constructivist reading

    Multilevel governance presents a depiction of contemporary structures in EU Europe as consisting of overlapping authorities and competing competencies. By focusing on emerging non-anarchical structures in the international system, hence moving beyond the conventional hierarchy/anarchy dichotomy to distinguish domestic and international arenas, this seems a radical transformation of the familiar Westphalian system and to undermine state sovereignty. Paradoxically, however, the principle of sovereignty proves to be resilient despite its alleged empirical decline. This article argues that social constructivism can explain the paradox, by considering sovereign statehood as a process-dependent institutional fact, and by showing that multilevel governance can feed into this process

    Emergence and spread of predominantly community-onset Clostridium difficile PCR ribotype 244 infection in Australia, 2010 to 2012

    We describe an Australia-wide Clostridium difficile outbreak in 2011 and 2012 involving the previously uncommon ribotype 244. In Western Australia, 14 of 25 cases were community-associated, 11 were detected in patients younger than 65 years, 14 presented to emergency/outpatient departments, and 14 to non-tertiary/community hospitals. Using whole genome sequencing, we confirm ribotype 244 is from the same C. difficile clade as the epidemic ribotype 027. Like ribotype 027, it produces toxins A, B, and binary toxin, however it is fluoroquinolone-susceptible and thousands of single nucleotide variants distinct from ribotype 027. Fifteen outbreak isolates from across Australia were sequenced. Despite their geographic separation, all were genetically highly related without evidence of geographic clustering, consistent with a point source, for example affecting the national food chain. Comparison with reference laboratory strains revealed the outbreak clone shared a common ancestor with isolates from the United States and United Kingdom (UK). A strain obtained in the UK was phylogenetically related to our outbreak. Follow-up of that case revealed the patient had recently returned from Australia. Our data demonstrate new C. difficile strains are an on-going threat, with potential for rapid spread. Active surveillance is needed to identify and control emerging lineages

    A novel genetic variant of Streptococcus pneumoniae serotype 11A discovered in Fiji.

    OBJECTIVES: As part of annual cross-sectional Streptococcus pneumoniae carriage surveys in Fiji (2012-2015), we detected pneumococci in over 100 nasopharyngeal swabs that serotyped as '11F-like' by microarray. We examined the genetic basis of this divergence in the 11F-like capsular polysaccharide (cps) locus compared to the reference 11F cps sequence. The impact of this diversity on capsule phenotype, and serotype results using genetic and serologic methods were determined. METHODS: Genomic DNA from representative 11F-like S. pneumoniae isolates obtained from the nasopharynx of Fijian children was extracted and subject to whole genome sequencing. Genetic and phylogenetic analyses were used to identify genetic changes in the cps locus. Capsular phenotypes were evaluated using the Quellung reaction and latex agglutination. RESULTS: Compared to published 11F sequences, the wcwC and wcrL genes of the 11F-like cps locus are phylogenetically divergent, and the gct gene contains a single nucleotide insertion within a homopolymeric region. These changes within the DNA sequence of the 11F-like cps locus have modified the antigenic properties of the capsule, such that 11F-like isolates serotype as 11A by Quellung reaction and latex agglutination. CONCLUSIONS: This study demonstrates the ability of molecular serotyping by microarray to identify genetic variants of S. pneumoniae and highlights the potential for discrepant results between phenotypic and genotypic serotyping methods. We propose that 11F-like isolates are not a new serotype but rather are a novel genetic variant of serotype 11A. These findings have implications for invasive pneumococcal disease surveillance as well as studies investigating vaccine impact

    Omnivoria e repartição de recursos em åguas pobres em nutrientes da Bacia do Rio Negro

    Amazonian biodiversity is notorious, this is also valid for the fauna of the mineral-deficient waters of the Rio Negro System. Some 25 years of research on the benthic fauna of Central Amazonian streams resulted in species-rich foodwebs with a high degree of omnivory within dense animal communities. To exemplify the taxonomic range of omnivorous consumers, the detailed resource spectra of 18 consumer species, including Protozoa (2 species), Platyhelminthes (1 species), insects (2 species), fish (6 species) and shrimps (Decapoda, 7 species), associated primarily with the benthic habitats of Rio Negro tributaries, are presented. Special features of omnivory are characterized, and the importance of litter-decomposing fungi as essential energy input into the foodwebs is documented. It is shown that general omnivory -diverse omnivore consumers sharing most of the resource types- is a prevalent feature.
