    Comparison of bungee-aided and free-bouncing accelerations on trampoline

    Trampolines remain the single best apparatus for the training of aerial acrobatics skills. Trampoline use has led to catastrophic injuries from poor landings. Passive injury prevention countermeasures such as specialized matting have been largely ineffective. Active injury countermeasures such as hand spotting, “throw-in” mats, and overhead spotting rigs provide the most effective methods. The recent addition of several bungee cords between the ropes and the gymnast’s spotting harness has resulted in altered teaching and coaching of trampoline-related acrobatics. Bungee cords have eliminated the need for a coach/spotter to manage the ropes during skill learning. The purpose of this study was to assess the influence of the addition of bungee cords with a traditional rope-based overhead spotting rig. There is a paucity of any research involving trampoline injury countermeasures. Ten experienced trampoline acrobatic athletes (5 males, 5 females) from the U.S. Ski and Snowboard Association Aerials National Team performed 10 bounces as high as they could control. A triaxial accelerometer (200 Hz) characterized 10 bungee cord aided bounces and 10 freebounces on a trampoline from each athlete. Bed contact times, peak accelerations, and average accelerations were obtained. The results supported our hypotheses that the bungeeaided bounces achieved only 40% (average) to 70% (peak) of the free-bouncing accelerations (all ρ 0.092). The bed contact time was approximately 65% longer during the bungee-aided bounces (ρ < 0.001). Bungee cords may reduce the harshness of landings on trampoline

    Absolute integrated cross sections for some O2 Herzberg I transitions near 248–249 nm

    A frequency doubled tunable dye laser system with ∌0.4 cm−1 resolution was used to measure the integrated absorption cross sections of more than 20 rotational transitions in the O2 Herzberg I (A 3ÎŁ+u←X 3Σ−g) 8‐0 and 9‐0 vibrational bands near 248 nm and 249 nm. Oxygen pressures from 200 to 800 Torr and path lengths from 5 to 25 m were employed. The measured absorbances were fitted using a nonlinear least squares analysis and Beer’s Law to obtain absolute values for the individual transition integrated cross sections in good agreement with a recent spectral simulation and experimental data. By using the spectral simulation in conjunction with the present experimental results, total oscillator strengths in reasonable agreement with literature values were estimated for the 8‐0 and 9‐0 vibrational bands. © 1995 American Institute of Physics.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/70777/2/JCPSA6-103-1-6-1.pd

    The still present belief some 40 years ago that British politics was both exceptional and superior has been replaced by more theoretically sophisticated analyses based on a wider and more rigorously deployed range of research techniques, although historical analysis appropriately remains important. The American influence on the study of British politics has declined, but the European Union dimension has not been fully integrated. The study of interest groups has been in some respects a fading paradigm, but important questions related to democratic health have still to be addressed. Public administration has been supplanted by public policy, but economic policy remains under-studied. A key challenge for the future is the study of the management of expectations

    Birthweight, Maternal Weight Trajectories and Global DNA Methylation of LINE-1 Repetitive Elements

    Low birthweight, premature birth, intrauterine growth retardation, and maternal malnutrition have been related to an increased risk of cardiovascular disease, type 2 diabetes mellitus, obesity, and neuropsychiatric disorders later in life. Conversely, high birthweight has been linked to future risk of cancer. Global DNA methylation estimated by the methylation of repetitive sequences in the genome is an indicator of susceptibility to chronic diseases. We used data and biospecimens from an epigenetic birth cohort to explore the association between trajectories of fetal and maternal weight and LINE-1 methylation in 319 mother-child dyads. Newborns with low or high birthweight had significantly lower LINE-1 methylation levels in their cord blood compared to normal weight infants after adjusting for gestational age, sex of the child, maternal age at delivery, and maternal smoking during pregnancy (p = 0.007 and p = 0.036, respectively), but the magnitude of the difference was small. Infants born prematurely also had lower LINE-1 methylation levels in cord blood compared to term infants, and this difference, though small, was statistically significant (p = 0.004). We did not find important associations between maternal prepregnancy BMI or gestational weight gain and global methylation of the cord blood or fetal placental tissue. In conclusion, we found significant differences in cord blood LINE-1 methylation among newborns with low and high birthweight as well as among prematurely born infants. Future studies may elucidate whether chromosomal instabilities or other functional consequences of these changes contribute to the increased risk of chronic diseases among individuals with these characteristics

    A practical Java tool for small-molecule compound appraisal

    The increased use of small-molecule compound screening by new users from a variety of different academic backgrounds calls for adequate software to administer, appraise, analyse and exchange information obtained from screening experiments. While software and spreadsheet solutions exist, there is a need for software that can be easily deployed and is convenient to use.The Java application cApp addresses this need and aids in the handling and storage of information on small-molecule compounds. The software is intended for the appraisal of compounds with respect to their physico-chemical properties, analysis in relation to adherence to likeness rules as well as recognition of pan-assay interference components and cross-linking with identical entries in the PubChem Compound Database. Results are displayed in a tabular form in a graphical interface, but can also be written in an HTML or PDF format. The output of data in ASCII format allows for further processing of data using other suitable programs. Other features include similarity searches against user-provided compound libraries and the PubChem Compound Database, as well as compound clustering based on a MaxMin algorithm.cApp is a personal database solution for small-molecule compounds which can handle all major chemical formats. Being a standalone software, it has no other dependency than the Java virtual machine and is thus conveniently deployed. It streamlines the analysis of molecules with respect to physico-chemical properties and drug discovery criteria; cApp is distributed under the GNU Affero General Public License version 3 and available from http://www.structuralchemistry.org/pcsb/.     Development of a lung slice preparation for recording ion channel activity in alveolar epithelial type I cells

    BACKGROUND: Lung fluid balance in the healthy lung is dependent upon finely regulated vectorial transport of ions across the alveolar epithelium. Classically, the cellular locus of the major ion transport processes has been widely accepted to be the alveolar type II cell. Although evidence is now emerging to suggest that the alveolar type I cell might significantly contribute to the overall ion and fluid homeostasis of the lung, direct assessment of functional ion channels in type I cells has remained elusive. METHODS: Here we describe a development of a lung slice preparation that has allowed positive identification of alveolar type I cells within an intact and viable alveolar epithelium using living cell immunohistochemistry. RESULTS: This technique has allowed, for the first time, single ion channels of identified alveolar type I cells to be recorded using the cell-attached configuration of the patch-clamp technique. CONCLUSION: This exciting new development should facilitate the ascription of function to alveolar type I cells and allow us to integrate this cell type into the general model of alveolar ion and fluid balance in health and disease

    Validating TDP1 as an Inhibition Target for the Development of Chemosensitizers for Camptothecin-Based Chemotherapy Drugs.

    Cancer chemotherapy sensitizers hold the key to maximizing the potential of standard anticancer treatments. We have a long-standing interest in developing and validating inhibitors of the DNA repair enzyme tyrosyl-DNA phosphodiesterase 1 (TDP1) as chemosensitizers for topoisomerase I poisons such as topotecan. Herein, by using thieno[2,3-b]pyridines, a class of TDP1 inhibitors, we showed that the inhibition of TDP1 can restore sensitivity to topotecan, results that are supported by TDP1 knockout cell experiments using CRISPR/Cas9. However, we also found that the restored sensitivity towards topoisomerase I inhibitors is likely regulated by multiple complementary DNA repair pathways. Our results showed that one of these pathways is likely modulated by PARP1, although it is also possible that other redundant and partially overlapping pathways may be involved in the DNA repair process. Our work thus raises the prospect of targeting multiple DNA repair pathways to increase the sensitivity to topoisomerase I inhibitors

    First observations of separated atmospheric nu_mu and bar{nu-mu} events in the MINOS detector

    The complete 5.4 kton MINOS far detector has been taking data since the beginning of August 2003 at a depth of 2070 meters water-equivalent in the Soudan mine, Minnesota. This paper presents the first MINOS observations of nu” and [overline nu ]” charged-current atmospheric neutrino interactions based on an exposure of 418 days. The ratio of upward- to downward-going events in the data is compared to the Monte Carlo expectation in the absence of neutrino oscillations, giving Rup/downdata/Rup/downMC=0.62-0.14+0.19(stat.)±0.02(sys.). An extended maximum likelihood analysis of the observed L/E distributions excludes the null hypothesis of no neutrino oscillations at the 98% confidence level. Using the curvature of the observed muons in the 1.3 T MINOS magnetic field nu” and [overline nu ]” interactions are separated. The ratio of [overline nu ]” to nu” events in the data is compared to the Monte Carlo expectation assuming neutrinos and antineutrinos oscillate in the same manner, giving R[overline nu ][sub mu]/nu[sub mu]data/R[overline nu ][sub mu]/nu[sub mu]MC=0.96-0.27+0.38(stat.)±0.15(sys.), where the errors are the statistical and systematic uncertainties. Although the statistics are limited, this is the first direct observation of atmospheric neutrino interactions separately for nu” and [overline nu ]”
