    Xenogeneic, extracorporeal liver perfusion in primates improves the ratio of branched-chain amino acids to aromatic amino acids (Fischer's ratio)

    In fulminant hepatic failure (FHF), the development of hepatic encephalopathy is associated with grossly abnormal concentrations of plasma amino acids (PAA). Normalization of the ratio of branched-chain amino acids to aromatic amino acids (Fischer's ratio) correlates with clinical improvement. This study evaluated changes in PAA metabolism during 4 h of isolated, normothermic extracorporeal liver perfusion using a newly designed system containing human blood and a rhesus monkey liver. Bile and urea production were within the physiological range. Release of the transaminases AST, ALT and LDH were minimal. The ratio of branched (valine, leucine, isoleucine) to aromatic (tyrosine, phenylalanine) amino acids increased significantly. These results indicate that a xenogeneic extracorporeal liver perfusion system is capable of significantly increasing Fischer's ratio and may play a role in treating and bridging patients in FHF in the future

    Alfvén waves in the near-PSBL lobe: Cluster observations

    Electromagnetic low-frequency waves in the magnetotail lobe close to the PSBL (Plasma Sheet Boundary Layer) are studied using the Cluster spacecraft. The lobe waves show Alfvénic properties and transport their wave energy (Poynting flux) on average toward the Earth along magnetic field lines. Most of the wave events are rich with oxygen (O+) ion plasma. The rich O+ plasma can serve to enhance the magnetic field fluctuations, resulting in a greater likelihood of observation, but it does not appear to be necessary for the generation of the waves. Taking into account the fact that all events are associated with auroral electrojet enhancements, the source of the lobe waves might be a substorm-associated instability, i.e. some instability near the reconnection site, or an ion beam-related instability in the PSBL

    The stigma of obesity in the general public and its implications for public health - a systematic review

    <p>Abstract</p> <p>Background</p> <p>Up to this date, prevalence rates of obesity are still rising. Aside from co-morbid diseases, perceived discrimination and stigmatization leads to worsen outcomes in obese individuals. Higher stigmatizing attitudes towards obese individuals may also result in less support of preventive and interventive measures. In light of the immense burden of obesity on health care systems and also on the individuals' quality of life, accepted and subsidized preventive measures are needed. Policy support might be determined by views of the lay public on causes of obesity and resulting weight stigma. This study seeks to answer how representative samples of the lay public perceive people with obesity or overweight status (stigmatizing attitudes); what these samples attribute obesity to (causal attribution) and what types of interventions are supported by the lay public and which factors determine that support (prevention support).</p> <p>Methods</p> <p>A systematic literature search was conducted. All studies of representative samples reporting results on (a) stigmatizing attitudes towards overweight and obese individuals, (b) causal beliefs and (c) prevention support were included.</p> <p>Results</p> <p>Only 7 articles were found. One study reported prevalence rates of stigmatizing attitudes. About a quarter of the population in Germany displayed definite stigmatizing attitudes. Other studies reported causal attributions. While external influences on weight are considered as well, it seems that internal factors are rated to be of higher importance. Across the studies found, regulative prevention is supported by about half of the population, while childhood prevention has highest approval rates. Results on sociodemographic determinants differ substantially.</p> <p>Conclusions</p> <p>Further research on public attitudes toward and perception of overweight and obesity is urgently needed to depict the prevailing degree of stigmatization. Introducing a multidimensional concept of the etiology of obesity to the lay public might be a starting point in stigma reduction.</p

    ‘Do i care?’ young adults' recalled experiences of early adolescent overweight and obesity: a qualitative study

    &lt;p&gt;Objective: Individual behaviour change to reduce obesity requires awareness of, and concern about, weight. This paper therefore describes how young adults, known to have been overweight or obese during early adolescence, recalled early adolescent weight-related awareness and concerns. Associations between recalled concerns and weight-, health- and peer-related survey responses collected during adolescence are also examined.&lt;/p&gt; &lt;p&gt;Design: Qualitative semi-structured interviews with young adults; data compared with responses to self-report questionnaires obtained in adolescence.&lt;/p&gt; &lt;p&gt;Participants: A total of 35 participants, purposively sub-sampled at age 24 from a longitudinal study of a school year cohort, previously surveyed at ages 11, 13 and 15. Physical measures during previous surveys allowed identification of participants with a body mass index (BMI) indicative of overweight or obesity (based on British 1990 growth reference) during early adolescence. Overall, 26 had been obese, of whom 11 had BMI99.6th centile, whereas 9 had been overweight (BMI=95th–97.9th centile).&lt;/p&gt; &lt;p&gt;Measures: Qualitative interview responses describing teenage life, with prompts for school-, social- and health-related concerns. Early adolescent self-report questionnaire data on weight-worries, self-esteem, friends and victimisation (closed questions).&lt;/p&gt; &lt;p&gt;Results: Most, but not all recalled having been aware of their overweight. None referred to themselves as having been obese. None recalled weight-related health worries. Recollection of early adolescent obesity varied from major concerns impacting on much of an individual's life to almost no concern, with little relation to actual severity of overweight. Recalled concerns were not clearly patterned by gender, but young adult males recalling concerns had previously reported more worries about weight, lower self-esteem, fewer friends and more victimisation in early adolescence; no such pattern was seen among females. Conclusion: The popular image of the unhappy overweight teenager was not borne out. Many obese adolescents, although well aware of their overweight recalled neither major dissatisfaction nor concern. Weight-reduction behaviours are unlikely in such circumstances.&lt;/p&gt

    A model for collisions in granular gases

    We propose a model for collisions between particles of a granular material and calculate the restitution coefficients for the normal and tangential motion as functions of the impact velocity from considerations of dissipative viscoelastic collisions. Existing models of impact with dissipation as well as the classical Hertz impact theory are included in the present model as special cases. We find that the type of collision (smooth, reflecting or sticky) is determined by the impact velocity and by the surface properties of the colliding grains. We observe a rather nontrivial dependence of the tangential restitution coefficient on the impact velocity.Comment: 11 pages, 2 figure

    Four-spacecraft determination of magnetopause orientation, motion and thickness: comparison with results from single-spacecraft methods

    In this paper, we use Cluster data from one magnetopause event on 5 July 2001 to compare predictions from various methods for determination of the velocity, orientation, and thickness of the magnetopause current layer. We employ established as well as new multi-spacecraft techniques, in which time differences between the crossings by the four spacecraft, along with the duration of each crossing, are used to calculate magnetopause speed, normal vector, and width. The timing is based on data from either the Cluster Magnetic Field Experiment (FGM) or the Electric Field Experiment (EFW) instruments. The multi-spacecraft results are compared with those derived from various single-spacecraft techniques, including minimum-variance analysis of the magnetic field and deHoffmann-Teller, as well as Minimum-Faraday-Residue analysis of plasma velocities and magnetic fields measured during the crossings. In order to improve the overall consistency between multi- and single-spacecraft results, we have also explored the use of hybrid techniques, in which timing information from the four spacecraft is combined with certain limited results from single-spacecraft methods, the remaining results being left for consistency checks. The results show good agreement between magnetopause orientations derived from appropriately chosen single-spacecraft techniques and those obtained from multi-spacecraft timing. The agreement between magnetopause speeds derived from single- and multi-spacecraft methods is quantitatively somewhat less good but it is evident that the speed can change substantially from one crossing to the next within an event. The magnetopause thickness varied substantially from one crossing to the next, within an event. It ranged from 5 to 10 ion gyroradii.     Abstract PhyA from Selenomonas ruminantium (PhyAsr), is a bacterial protein tyrosine phosphatase (PTP)-like inositol polyphosphate phosphatase (IPPase) that is distantly related to known PTPs. PhyAsr has a second substrate binding site referred to as a standby site and the P-loop (HCX 5 R) has been observed in both open (inactive) and closed (active) conformations. Site-directed mutagenesis and kinetic and structural studies indicate PhyAsr follows a classical PTP mechanism of hydrolysis and has a broad specificity toward polyphosphorylated myo-inositol substrates, including phosphoinositides. Kinetic and molecular docking experiments demonstrate PhyAsr preferentially cleaves the 3-phosphate position of Ins P 6 and will produce Ins(2)P via a highly ordered series of sequential dephosphorylations: D-Ins(1,2,4,5,6)P 5 , Ins(2,4,5,6)P 4 , D-Ins(2,4,5)P 3 , and D-Ins(2,4)P 2 . The data support a distributive enzyme mechanism and suggest the PhyAsr standby site is involved in the recruitment of substrate. Structural studies at physiological pH and high salt concentrations demonstrate the &apos;&apos;closed&apos;&apos; or active P-loop conformation can be induced in the absence of substrate. These results suggest PhyAsr should be reclassified as a D-3 myo-inositol hexakisphosphate phosphohydrolase and suggest the PhyAsr reaction mechanism is more similar to that of PTPs than previously suspected.     Ising model on 3D random lattices: A Monte Carlo study

    We report single-cluster Monte Carlo simulations of the Ising model on three-dimensional Poissonian random lattices with up to 128,000 approx. 503 sites which are linked together according to the Voronoi/Delaunay prescription. For each lattice size quenched averages are performed over 96 realizations. By using reweighting techniques and finite-size scaling analyses we investigate the critical properties of the model in the close vicinity of the phase transition point. Our random lattice data provide strong evidence that, for the available system sizes, the resulting effective critical exponents are indistinguishable from recent high-precision estimates obtained in Monte Carlo studies of the Ising model and \phi^4 field theory on three-dimensional regular cubic lattices.Comment: 35 pages, LaTex, 8 tables, 8 postscript figure

    The Electron Drift Instrument on Cluster: overview of first results

    International audienceEDI measures the drift velocity of artificially injected electron beams. From this drift velocity, the perpendicular electric field and the local magnetic field gradients can be deduced when employing different electron energies. The technique requires the injection of two electron beams at right angles to the magnetic field and the search for those directions within the plane that return the beams to their associated detectors after one or more gyrations. The drift velocity is then derived from the directions of the two beams and/or from the difference in their times-of-flight, measured via amplitude-modulation and coding of the emitted electron beams and correlation with the signal from the returning electrons. After careful adjustment of the control parameters, the beam recognition algorithms, and the onboard magnetometer calibrations during the commissioning phase, EDI is providing excellent data over a wide range of conditions.
