    Experiences with and benefits of the inspection of air-assisted sprayers from the fruit- and winegrowers’ point of view

    Until the 1960s pesticides were applied with a spray gun connected to a long hose or directly to the tank and operated by a worker standing on a platform. This technique was gradually replaced by airassisted sprayers.The purchase of three sprayer test facilities in 1991 signified the start of a new era in pesticide application:• The ejection rate of the individual nozzles as well as the distribution pattern of the liquid could be measured and optimized on a vertical test stand by adjusting the nozzles or adding additional ones. • The ceramic nozzles were replaced by hollow-cone nozzles, which allowed for the first time to calculate the exact amount of liquid per hectare and tree height based on various parameters such as driving speed, spray pressure, number of open nozzles and nozzle output volume. The amount of water was reduced from 500 l/m tree height and hectare to 166 – 100 l. • The efficacy of the pesticides was increased and spray drift reduced, which brings about economic and ecological benefits. • Up to the year 1997 sprayer calibration was optional. Since then, a sprayer calibration in 5-year intervals has become a condition for participation in the South Tyrolean IP-programme. This also facilitates the observance of the GobalGAP control points and Compliance Criteria regarding application equipment.Since 2011 the South Tyrolean sprayer manufacturers and fruit- and winegrowers have been able to measure and optimize the air blast and emission pattern. Thereby we hope to further reduce spray drift and increase the efficacy of the pesticides. A properly calibrated sprayer enables the fruit- and winegrowers to observe more easily the regulations for the distances to residential and public buildings as well as pastures enacted by the South Tyrolean Provincial Government and AGRIOS in 2011. We trust that the South Tyrolean fruit- and winegrowers will have no difficulties in complying with the Directive 2009/128/EC on the sustainable use of pesticides with regard to application techniques and sprayer calibration

    Heat-shock mediated overexpression of HNF1β mutations has differential effects on gene expression in the Xenopus pronephric kidney.

    The transcription factor HNF1B, encoded by the TCF2 gene, plays an important role in the organogenesis of vertebrates. In humans, heterozygous mutations of HNF1B are associated with several diseases, such as pancreatic β-cell dysfunction leading to maturity-onset diabetes of the young (MODY5), defective kidney development, disturbed liver function, pancreas atrophy, and malformations of the genital tract. The African claw frog Xenopus laevis is an excellent model to study the processes involved in embryogenesis and organogenesis, as it can be manipulated easily with a series of methods. In the present study, we overexpressed HNF1β mutants in the developing Xenopus embryo to assess their roles during organogenesis, particularly in the developing pronephric kidney. Towards this goal, we developed a heat-shock inducible binary Cre/loxP system with activator and effector strains. Heat-shock activation of the mutant HNF1B variants P328L329del and A263insGG resulted in malformations of various organs and the affected larvae developed large edemas. Defects in the pronephros were primarily confined to malformed proximal tubules. Furthermore, the expression of the proximal tubule marker genes tmem27 and slc3a1, both involved in amino acid transport, was affected. Both P328L329del and A263insGG downregulated expression of slc3a1. In addition, P328L329del reduced tmem27 expression while A263insGG overexpression decreased expression of the chloride channel clcnk and the transcription factor pax2. Overexpression of two mutant HNF1B derivatives resulted in distinct phenotypes reflected by either a reduction or an enlargement of pronephros size. The expression of selected pronephric marker genes was differentially affected upon overexpression of HNF1B mutations. Based on our findings, we postulate that HNF1B mutations influence gene regulation upon overexpression in specific and distinct manners. Furthermore, our study demonstrates that the newly established Cre/loxP system for Xenopus embryos is an attractive alternative to examine the gene regulatory potential of transcription factors in developing pronephric kidney as exemplified here for HNF1B

    Das klinische Bild der akuten Pankreatitis wird entscheidend durch die sekundäre Schädigung von Herz-Kreislauf-System, Lunge und Niere bestimmt. Ziel der vorliegenden Untersuchung war es, durch Messungen in venösem Pankreasblut, Pankreaslymphe und Peritonealexsudat die Kompartimente zu bestimmen, über die die systemischen Schädigungen vermittelt werden. An anästhesierten Schweinen wurden die systemischen, hämodynamischen Parameter durch gesteuerte Volumentherapie konstant gehalten. Die Schweine wurden randomisiert der Kontrollgruppe (n = 9) oder einer der Pankreatitisgruppen zugeteilt (jeweils n = 10). Die Pankreatitis wurde durch Infusion von freier Fettsäure in die Pankreasarterien (FFS) oder durch Infusion einer 5%igen Natrium-Taurocholat-Lösung retrograd in den Pankreasgang (NaT) ausgelöst. Nach Isolation des Pankreas wurde venöses Pankreasblut, Pankreaslymphe und Peritonealexsudat gewonnen und die Aktivität von Lipase, Phospholipase A und Plasmaprokallikrein sowie die Konzentration von Organkallikrein und Kininogen bestimmt. In beiden Pankreatitismodellen fand sich ein Anstieg der Enzymaktivitäten. Die höchsten Aktivitäten fanden sich im Peritonealexsudat (Phospholipase A nach 40 min: Kontrolle 10,0 U/1, NaT 72,2 U/1). In beiden Pankreatitismodellen fanden sich außerdem Hinweise für eine Aktivierung des Organkallikrein-Kinin-Systems durch den Anstieg der Organkallikreinkonzentration und den Abfall der Gesamtkininogenkonzentration. Die stärksten Veränderungen fanden sich wieder im Peritonealexsudat (Organkallikrein nach 40 min: Kontrolle 14,7 ng/ml, NaT 452 ng/ml).The clinical course of acute pancreatitis is strongly influenced by secondary cardiac, pulmonary and renal damage. The aim of the present study was to gather information about the compartment promoting the systemic damage. Therefore the activity of lipase, phospholipase A and plasmaprokallikrein and the concentration of tissue kallikrein and kininogen were measured in portal venous blood, pancreatic lymph and peritoneal exudate. Anaesthetized pigs were subjected to fluid resuscitation to keep systemic haemodynamic parameters constant. The pancreas was isolated in situ. The pigs were randomly assigned to a control group (n = 9) or one of the two pancreatitis groups (n = 10 each). Pancreatitis was induced by i.a. infusion of free fatty acid (FFS) or retrograde infusion of 5 % sodium taurocholate intraductally (NaT). In both pancreatitis groups the activity of lipase and phospholipase A increased. The most pronounced changes were seen in the peritoneal exsudate (phospholipase A activity 40 min after induction: control 10.0 U/1, NaT 72.2 U/1). In both pancreatitis groups there was evidence for activation of the tissue kallikreinkinin system in the form of an increase in the kallikrein concentration and a decrease in the kininogen concentration. Again the changes were most pronounced in the peritoneal exsudate (tissue kallikrein 40 min after induction: control 14.7 ng/ml, NaT 452 ng/ml)

    Identification of a penta- and hexapeptide of islet amyloid polypeptide (IAPP) with amyloidogenic and cytotoxic properties.

    Pancreatic arnyloid is found in more than 95 % of type II diabetes patients. Pancreatic amyloid is formed by the aggregation of islet amyloid polypeptide (hIAPP or amylin), which is a 37-residue peptide. Because pancreatic amyloid is cytotoxic, it is believed that its formation is directly associated with the development of the disease. We recently showed that hIAPP amyloid formation follows the nucleation-dependent polymerization mechanism and proceeds via a conformational transition of soluble hIAPP into aggregated beta-sheets. Here, we report that the penta- and hexapeptide sequences, hIAPP(23-27) (FGAIL) and hIAPP(22-27) (NFGAIL) of hIAPP are sufficient for the formation of beta-sheet- containing amyloid fibrils. Although these two peptides differ by only one amino acid residue, they aggregate into completely different fibrillar assemblies. hIAPP(23-27) (FGAIL) fibrils self-assemble laterally into unusually broad ribbons, whereas hIAPP(22-27) (NFGAIL) fibrils coil around each other in a typical arnyloid fibril morphology. hIAPP(20-27) (SNNFGAIL) also aggregates into beta-sheet- containing fibrils, whereas no arnyloidogenicity is found for hIAPP(24-27) (GAIL), indicating that hIAPP(23-27) (FGAIL) is the shortest fibrillogenic sequence of hIAPP. Insoluble amyloid formation by the partial hIAPP sequences followed kinetics that were consistent with a nucleation-dependent polymerization mechanism. hIAPP(22-27) (NFGAIL), hIAPP(20-27) (SNNFGAIL), and also the known fibrillogenic sequence, hIAPP(20-29) (SNNFGAILSS) exhibited significantly lower kinetic and thermodynamic solubilities than the pentapeptide hIAPP(23-27) (FGAIL). Fibrils formed by all short peptide sequences and also by hIAPP(20-29) were cytotoxic towards the pancreatic cell line RIN5fm, whereas no cytotoxicity was observed for the soluble form of the peptides, a notion that is consistent with hIAPP cytotoxicity. Our results suggest that a penta- and hexapeptide sequence of an appropriate amino acid composition can be sufficient for beta-sheet and amyloid fibril formation and cytotoxicity and may assist in the rational design of inhibitors of pancreatic amyloid formation or other arnyloidosis-related diseases

    NMR evaluation of total statin content and HMG-CoA reductase inhibition in red yeast rice (Monascus spp.) food supplements

    Background Red yeast rice (i.e., rice fermented with Monascus spp.), as a food supplement, is claimed to be blood cholesterol-lowering. The red yeast rice constituent monacolin K, also known as lovastatin, is an inhibitor of the hydroxymethylglutaryl-CoA (HMG-CoA) reductase. This article aims to develop a sensitive nuclear magnetic resonance (NMR) method to determine the total statin content of red yeast rice products. Methods The total statin content was determined by a 400 MHz 1H NMR spectroscopic method, based on the integration of the multiplet at δ 5.37-5.32 ppm of a hydrogen at the hexahydronaphthalene moiety in comparison to an external calibration with lovastatin. The activity of HMG-CoA reductase was measured by a commercial spectrophotometric assay kit. Results The NMR detection limit for total statins was 6 mg/L (equivalent to 0.3 mg/capsule, if two capsules are dissolved in 50 mL ethanol). The relative standard deviations were consistently lower than 11%. The total statin concentrations of five red yeast rice supplements were between 1.5 and 25.2 mg per specified daily dose. A dose-dependent inhibition of the HMG-CoA reductase enzyme activity by the red yeast rice products was demonstrated. Conclusion A simple and direct NMR assay was developed to determine the total statin content in red yeast rice. The assay can be applied for the determination of statin content for the regulatory control of red yeast rice products

    Methods and Processes of Developing the Strengthening the Reporting of Observational Studies in Epidemiology – Veterinary (STROBE-Vet) Statement

    BACKGROUND Reporting of observational studies in veterinary research presents challenges that often are not addressed in published reporting guidelines. OBJECTIVE To develop an extension of the STROBE (Strengthening the Reporting of Observational Studies in Epidemiology) statement that addresses unique reporting requirements for observational studies in veterinary medicine related to health, production, welfare, and food safety. DESIGN Consensus meeting of experts. SETTING Mississauga, Canada. PARTICIPANTS Seventeen experts from North America, Europe, and Australia. METHODS Experts completed a pre-meeting survey about whether items in the STROBE statement should be modified or added to address unique issues related to observational studies in animal species with health, production, welfare, or food safety outcomes. During the meeting, each STROBE item was discussed to determine whether or not rewording was recommended and whether additions were warranted. Anonymous voting was used to determine consensus. RESULTS Six items required no modifications or additions. Modifications or additions were made to the STROBE items 1 (title and abstract), 3 (objectives), 5 (setting), 6 (participants), 7 (variables), 8 (data sources/measurement), 9 (bias), 10 (study size), 12 (statistical methods), 13 (participants), 14 (descriptive data), 15 (outcome data), 16 (main results), 17 (other analyses), 19 (limitations), and 22 (funding). CONCLUSION The methods and processes used were similar to those used for other extensions of the STROBE statement. The use of this STROBE statement extension should improve reporting of observational studies in veterinary research by recognizing unique features of observational studies involving food-producing and companion animals, products of animal origin, aquaculture, and wildlife

    Internal Modes and Magnon Scattering on Topological Solitons in 2d Easy-Axis Ferromagnets

    We study the magnon modes in the presence of a topological soliton in a 2d Heisenberg easy-axis ferromagnet. The problem of magnon scattering on the soliton with arbitrary relation between the soliton radius R and the "magnetic length" Delta_0 is investigated for partial modes with different values of the azimuthal quantum numbers m. Truly local modes are shown to be present for all values of m, when the soliton radius is enough large. The eigenfrequencies of such internal modes are calculated analytically on limiting case of a large soliton radius and numerically for arbitrary soliton radius. It is demonstrated that the model of an isotropic magnet, which admits an exact analytical investigation, is not adequate even for the limit of small radius solitons, R<<Delta_0: there exists a local mode with nonzero frequency. We use the data about local modes to derive the effective equation of soliton motion; this equation has the usual Newtonian form in contrast to the case of the easy-plane ferromagnet.
