    Maternal Rest/Nrsf Regulates Zebrafish Behavior through snap25a/b

    During embryonic development, regulation of gene expression is key to creating the many subtypes of cells that an organism needs throughout its lifetime. Recent work has shown that maternal genetics and environmental factors have lifelong consequences on diverse processes ranging from immune function to stress responses. The RE1-silencing transcription factor (Rest) is a transcriptional repressor that interacts with chromatin-modifying complexes to repress transcription of neural specific genes during early development. Here we show that in zebrafish, maternally supplied rest regulates expression of target genes during larval development and has lifelong impacts on behavior. Larvae deprived of maternal rest are hyperactive and show atypical spatial preferences. Adult male fish deprived of maternal rest present with atypical spatial preferences in a novel environment assay. Transcriptome sequencing revealed 158 genes that are repressed by maternal rest in blastula stage embryos. Furthermore, we found that maternal rest is required for target gene repression until at least 6 dpf. Importantly, disruption of the RE1 sites in either snap25a or snap25b resulted in behaviors that recapitulate the hyperactivity phenotype caused by absence of maternal rest. Both maternal rest mutants and snap25a RE1 site mutants have altered primary motor neuron architecture that may account for the enhanced locomotor activity. These results demonstrate that maternal rest represses snap25a/b to modulate larval behavior and that early Rest activity has lifelong behavioral impacts

    The SKA view of the Neutral Interstellar Medium in Galaxies

    Two major questions in galaxy evolution are how star-formation on small scales leads to global scaling laws and how galaxies acquire sufficient gas to sustain their star formation rates. HI observations with high angular resolution and with sensitivity to very low column densities are some of the important observational ingredients that are currently still missing. Answers to these questions are necessary for a correct interpretation of observations of galaxy evolution in the high-redshift universe and will provide crucial input for the sub-grid physics in hydrodynamical simulations of galaxy evolutions.     Adsorption of Mn(II) and Co(II) ions from aqueous solution using Maize cob activated carbon: Kinetics and Thermodynamics Studies

    The adsorption of Mn(II) and Co(II) ions from aqueous solution was investigated using batch adsorption experiment at room temperature. The effect of pH, contact time, metal ion concentration and temperature were evaluated. The residual concentrations of the metal ions were determined by atomic absorption spectrophotometer. The results showed that maximum removal of Mn(II) and Co(II) ions occurred at pH 9. Some physico-chemical and spectroscopic characterization of the adsorbents were done such as moisture content, ash content, bulk density, volatile matter, iodine number, point of zero charge (PZC), XRF, and FTIR. The moisture and ash content of MCAC adsorbent were found to be 9.85±0.06 and 5.5±0.1% respectively. The bulk density was found to be 0.37±0.01g/m3, iodine number of 367.66±30 mg/g and PZC of 6.8. The SEM micrograph shows particle grains and jelly like rough surfaces, FTIR analysis results show different functional group in the MCAB adsorbent such as O-H, C=O, and C=C stretching. Kinetic study shows that the pseudo-second order kinetic model best described the adsorption of metal ions. The equilibrium data fitted Langmuir, Freundlich and Temkin adsorption isotherms, in each case, the Langmuir model appears to have better regression coefficients than the Freundlich and Temkin. Thermodynamics investigation showed that Gibb’s free energy change (ΔG) was negative indicating that the adsorption of Mn(II) and Co(II) ions by maize cob activated carbon were feasible and spontaneous. The positive value of enthalpy change (ΔH) implies that the reaction was endothermic while positive value of entropy change (ΔS) implies an irregular increase in the randomness at the solid/solution interface of the adsorbent during the adsorption process.Keywords: Adsorption, Heavy metals, Kinetics, Thermodynamic

    Organic photovoltaics: The current challenges

    Organic photovoltaics are remarkably close to reaching a landmark power conversion efficiency of 20%. Given the current urgent concerns regarding climate change, research into renewable energy solutions is crucially important. In this perspective article, we highlight several key aspects of organic photovoltaics, ranging from fundamental understanding to implementation, that need to be addressed to ensure the success of this promising technology. We cover the intriguing ability of some acceptors to undergo efficient charge photogeneration in the absence of an energetic driving force and the effects of the resulting state hybridization. We explore one of the primary loss mechanisms of organic photovoltaics - non-radiative voltage losses - and the influence of the energy gap law. Triplet states are becoming increasingly relevant owing to their presence in even the most efficient non-fullerene blends, and we assess their role as both a loss mechanism and a potential strategy to enhance efficiency. Finally, two ways in which the implementation of organic photovoltaics can be simplified are addressed. The standard bulk heterojunction architecture could be superseded by either single material photovoltaics or sequentially deposited heterojunctions, and the attributes of both are considered. While several important challenges still lie ahead for organic photovoltaics, their future is, indeed, bright

    Bacterial diversity in the intestinal tract of the funguscultivating termite Macrotermes michaelseni (Sjöstedt)

    Microorganisms in the intestinal tracts of termites play a crucial role in the nutritional physiology of termites. The bacterial diversity in the fungus-cultivating Macrotermes michaelseni was examined usingboth molecular and culture dependent methods. Total DNA was extracted from the gut of the termite and 16S rRNA genes were amplified using bacterial specific primers. Representatives from forty-one (41) RFLP patterns from a total of one hundred and two (102) clones were sequenced. Most of the clones were affiliated with 3 main groups of the domain Bacteria: Cytophaga-Flexibacter-Bacteriodes(73), Proteobacteria (13), and the low G+C content Gram-positive bacteria (9). Two RFLPs related to planctomycetes, but deeper branching than known members of the phylum, were detected. In addition, 1 and 2 RFLPs represented the spirochetes and TM7-OP11 groups, respectively. In studies using culture dependent techniques, most of the isolates obtained belonged to the Gram-positive bacteriawith a high G+C content. However, only one of the clones represented Gram-positive bacteria with High G+C content. These results show a high bacterial diversity in the intestinal microbiota of M. michaelseni, which continues to escape cultivation. As is the case in other termites many of the clones represent previously uncultured bacteria. The fact that most of the clones clustered with clones from Macrotermes gilvus provides further support for the hypothesis that microorganisms in intestinal tracts of termites have co-evolved with their hosts

    The antiproliferative drug doxorubicin inhibits liver fibrosis in bile duct-ligated rats and can be selectively delivered to hepatic stellate cells in vivo

    Hepatic stellate cell (HSC) proliferation is a key event in liver fibrosis; therefore, pharmacological intervention with antiproliferative drugs may result in antifibrotic effects. In this article, the antiproliferative effect of three cytostatic drugs was tested in cultured rat HSC. Subsequently, the antifibrotic potential of the most potent drug was evaluated in vivo. As a strategy to overcome drug-related toxicity, we additionally studied how to deliver this drug specifically to HSC by conjugating it to the HSC-selective drug carrier mannose-6-phosphate-modified human serum albumin (M6PHSA). We investigated the effect of cisplatin, chlorambucil, and doxorubicin (DOX) on 5-bromo-2'-deoxyuridine incorporation in cultured HSC and found DOX to be the most potent drug. Treatment of bile duct-ligated (BDL) rats with daily i.v. injections of 0.35 mg/kg DOX from day 3 to 10 after BDL reduced alpha-smooth muscle actin-stained area in liver sections from 8.5 +/- 0.8 to 5.1 +/- 0.9% (P +/- 0.01) and collagen-stained area from 13.1 +/- 1.3 to 8.9 +/- 1.5% (P +/- 0.05). DOX was coupled to M6PHSA, and the organ distribution of this construct (M6PHSA-DOX) was investigated. Twenty minutes after i.v. administration, 50 +/- 6% of the dose was present in the livers, and colocalization of M6PHSA-DOX with HSC markers was observed. In addition, in vitro studies showed selective binding of M6PHSA-DOX to activated HSC. Moreover, M6PHSA-DOX strongly attenuated HSC proliferation in vitro, indicating that active drug is released after uptake of the conjugate. DOX inhibits liver fibrosis in BDL rats, and HSC-selective targeting of this drug is possible. This may offer perspectives for the application of antiproliferative drugs for antifibrotic purposes
