    XENO-Free production and recovery of human pluripotent stem cells using synthetic dissolvable microcarriers

    The implementation of scalable culture platforms for the large-scale production of human pluripotent stem cells (hPSC) and their derivatives is mandatory to fulfill the requirement of obtaining large numbers of these cells for cell therapies and other in vitro biomedical applications, such as drug screening, toxicology assays and disease modeling. Recent progress includes the development of chemically-defined culture conditions for manufacturing of hPSC and their derivatives, namely the development of xeno-free microcarrier platforms to meet good manufacturing practice (GMP) quality requirements [1]. One challenge that remains to be addressed is the establishment of a robust, scalable, and cost-effective downstream processing for cell recovery and removal of the microcarriers. Since hPSC have the tendency to create multilayers of cells on the microcarriers, often forming very large cell-microcarrier aggregates, the process of cell recovery can be technically challenging and time consuming. In this work, we developed a robust and efficient platform for large-scale production of hPSC using synthetic dissolvable microcarriers, which can be quickly dissolved by a non-proteolytic enzyme. This allows an easy cell recovery without the need of the microcarrier separation step, facilitating the downstream processing. Moreover, these synthetic microcarriers are sterile and ready-to-use, and are functionalized with the Synthemax® surface, based on a peptide-acrylate matrix designed for long-term support of hESC self-renewal [2]. hPSC were able to attach and grow on the dissolvable microcarriers and the expansion process was evaluated in a scalable stirred culture system. The cells growth performance on these microcarriers was comparable with the ones obtained when culturing hPSCs in non-dissolvable microcarriers (backbone of polystyrene coated with different ECM molecules), being possible to obtain 1.3x106 cells/mL during 5 days. Importantly, hPSCs cultured on these novel microcarriers were efficiently recovered without the need of the filtration step to separate the microcarriers from the cells and maintained their typical colony morphology and pluripotency-associated marker-expression after re-plating on tissue culture plates. Moreover, their potential for spontaneous differentiation into cells of the three embryonic germ layers was demonstrated through formation of embryoid bodies containing cells expressing typical markers of endoderm, ectoderm and mesoderm. These novel synthetic dissolvable microcarriers allow an easy and efficient downstream processing for hPSCs recovery after expansion/differentiation, without compromising the quality of the cells (viability, potency and functionality), which are a major process breakthrough for stem cell manufacturing.     An MCDM approach to the selection of novel technologies for innovative in-vehicle information systems

    Driving a car is a complex skill that includes interacting with multiple systems inside the vehicle. Today’s challenge in the automotive industry is to produce innovative In-Vehicle Information Systems (IVIS) that are pleasant to use and satisfy the costumers’ needs while, simultaneously, maintaining the delicate balance of primary task vs. secondary tasks while driving. The authors report a MCDM approach for rank ordering a large heterogeneous set of human-machine interaction technologies; the final set consisted of hundred and one candidates. They measured candidate technologies on eight qualitative criteria that were defined by domain experts, using a group decision-making approach. The main objective was ordering alternatives by their decision score, not the selection of one or a small set of them. The authors’ approach assisted decision makers in exploring the characteristics of the most promising technologies and they focused on analyzing the technologies in the top quartile, as measured by their MCDM model.     Numerical 3D modeling of heat transfer in human tissues for microwave radiometry monitoring of Brown fat metabolismo

    Background: Brown adipose tissue (BAT) plays an important role in whole body metabolism and could potentially mediate weight gain and insulin sensitivity. Although some imaging techniques allow BAT detection, there are currently no viable methods for continuous acquisition of BAT energy expenditure. We present a non-invasive technique for long term monitoring of BAT metabolism using microwave radiometry. Methods: A multilayer 3D computational model was created in HFSS™ with 1.5 mm skin, 3-10 mm subcutaneous fat, 200 mm muscle and a BAT region (2-6 cm3) located between fat and muscle. Based on this model, a log-spiral antenna was designed and optimized to maximize reception of thermal emissions from the target (BAT). The power absorption patterns calculated in HFSS™ were combined with simulated thermal distributions computed in COMSOL® to predict radiometric signal measured from an ultra-low-noise microwave radiometer. The power received by the antenna was characterized as a function of different levels of BAT metabolism under cold and noradrenergic stimulation. Results: The optimized frequency band was 1.5-2.2 GHz, with averaged antenna efficiency of 19%. The simulated power received by the radiometric antenna increased 2-9 mdBm (noradrenergic stimulus) and 4-15 mdBm (cold stimulus) corresponding to increased 15-fold BAT metabolism. Conclusions: Results demonstrated the ability to detect thermal radiation from small volumes (2-6 cm3) of BAT located up to 12 mm deep and to monitor small changes (0.5°C) in BAT metabolism. As such, the developed miniature radiometric antenna sensor appears suitable for non-invasive long term monitoring of BAT metabolism

    On the bioactivity of Echinacea purpurea extracts to modulate the production of inflammatory mediators

    Inflammatory diseases are the focus of several clinical studies, due to limitations and serious side effects of available therapies. Plant-based drugs (e.g., salicylic acid, morphine) have become landmarks in the pharmaceutical field. Therefore, we investigated the immunomodulatory effects of flowers, leaves, and roots from Echinacea purpurea. Ethanolic (EE) and dichloromethanolic extracts (DE) were obtained using the Accelerated Solvent Extractor and aqueous extracts (AE) were prepared under stirring. Their chemical fingerprint was evaluated by liquid chromatography–high resolution mass spectrometry (LC-HRMS). The pro- and anti-inflammatory effects, as well as the reduction in intracellular reactive oxygen and nitrogen species (ROS/RNS), of the different extracts were evaluated using non-stimulated and lipopolysaccharide-stimulated macrophages. Interestingly, AE were able to stimulate macrophages to produce pro-inflammatory cytokines (tumor necrosis factor -TNF-α, interleukin -IL-1β, and IL-6), and to generate ROS/RNS. Conversely, under an inflammatory scenario, all extracts reduced the amount of pro-inflammatory mediators. DE, alkylamides-enriched extracts, showed the strongest anti-inflammatory activity. Moreover, E. purpurea extracts demonstrated generally a more robust anti-inflammatory activity than clinically used anti-inflammatory drugs (dexamethasone, diclofenac, salicylic acid, and celecoxib).     Bioactive lipids of seaweeds from the Portuguese north coast: health benefits versus potential contamination

    The total lipid content and lipidic profile of seaweeds harvested in the North Coast and purchased in Portugal were determined in this paper. The amount of total lipids in the different species of seaweeds varied between 0.7 ± 0.1% (Chondrus crispus) and 3.8 ± 0.6% (Ulva spp.). Regarding the fatty acid content, polyunsaturated fatty acids (PUFA) ranged between 0–35%, with Ulva spp. presenting the highest amount; monounsaturated fatty acids (MUFA) varied between 19 and 67%; and saturated fatty acids (SFA) were predominant in C. crispus (45–78%) and Gracilaria spp. (36–79%). Concerning the nutritional indices, the atherogenicity index (AI) was between 0.4–3.2, the thrombogenicity index (TI) ranged from 0.04 to 1.95, except for Gracilaria spp., which had a TI of 7.6, and the hypocholesterolemic/hypercholesterolemic ratio (HH) values ranged between 0.88–4.21, except for Gracilaria spp., which exhibited values between 0.22–9.26. The n6/n3 ratio was below 1 for most of the species evaluated, except for Ascophyllum nodosum, which presented a higher value, although below 2. Considering the PUFA/SFA ratio, seaweeds presented values between 0.11–1.02. The polycyclic aromatic hydrocarbons (PAHs) and aliphatic hydrocarbons (AHCs) contamination of seaweeds under study was also quantified, the values found being much lower than the maximum levels recommended for foodstuff.info:eu-repo/semantics/publishedVersio

    Boat noise impacts Lusitanian toadfish breeding males and reproductive outcome

    Anthropogenic noise is a growing threat to marine organisms, including fish. Yet very few studies have addressed the impact of anthropogenic noise on fish reproduction, especially in situ. In this study, we investigated the impacts of boat noise exposure in the reproductive success of wild Lusitanian toadfish (Halobatrachus didactylus), a species that relies on advertisement calls for mate attraction, using behavioural, physiological and reproductive endpoints. Two sets of artificial nests were deployed in the Tagus estuary and exposed to either ambient sound or boat noise during their breeding season. Toadfish males spontaneously used these nests to breed. We inspected nests for occupation and the presence of eggs in six spring low tides (in two years) and assessed male vocal activity and stress responses. Boat noise did not affect nest occupation by males but impacted reproductive success by decreasing the likelihood of receiving eggs, decreasing the number of live eggs and increasing the number of dead eggs, compared to control males. Treatment males also showed depressed vocal activity and slightly higher cortisol levels. The assessment of oxidative stress and energy metabolism-related biomarkers revealed no oxidative damage in noise exposed males despite having lower antioxidant responses and pointed towards a decrease in the activity levels of energy metabolism-related biomarkers. These results suggest that males exposed to boat noise depressed their metabolism and their activity (such as parental care and mate attraction) to cope with an acoustic stressor, consistent with a freezing defensive response/behaviour. Together, our study demonstrates that boat noise has severe impacts on reproductive fitness in Lusitanian toadfish. We argue that, at least fishes that cannot easily avoid noise sources due to their dependence on specific spawning sites, may incur in significant direct fitness costs due to chronic noise exposure.Fundação para a Ciência e Tecnologia - FCTinfo:eu-repo/semantics/publishedVersio

    Echinacea purpurea fractions represent promising plant-based anti-inflammatory formulations

    Echinacea purpurea is traditionally used in the treatment of inflammatory diseases. Therefore, we investigated the anti-inflammatory capacity of E. purpurea dichloromethanolic (DE) and ethanolic extracts obtained from flowers and roots (R). To identify the class of compounds responsible for the strongest bioactivity, the extracts were fractionated into phenol/carboxylic acid (F1) and alkylamide fraction (F2). The chemical fingerprint of bioactive compounds in the fractions was evaluated by LC-HRMS. E. purpurea extracts and fractions significantly reduced pro-inflammatory cytokines (interleukin 6 and/or tumor necrosis factor) and reactive oxygen and nitrogen species (ROS/RNS) production by lipopolysaccharide-stimulated primary human monocyte-derived macrophages. Dichloromethanolic extract obtained from roots (DE-R) demonstrated the strongest anti-inflammatory activity. Moreover, fractions exhibited greater anti-inflammatory activity than whole extract. Indeed, alkylamides must be the main compounds responsible for the anti-inflammatory activity of extracts; thus, the fractions presenting high content of these compounds presented greater bioactivity. It was demonstrated that alkylamides exert their anti-inflammatory activity through the downregulation of the phosphorylation of p38, ERK 1/2, STAT 3, and/or NF-κB signaling pathways, and/or downregulation of cyclooxygenase 2 expression. E. purpurea extracts and fractions, mainly DE-R-F2, are promising and powerful plant-based anti-inflammatory formulations that can be further used as a basis for the treatment of inflammatory diseases.This work was financially supported by the FCT to the Ph.D. grants of S.F.V.     Amyloid precursor protein interaction network in human testis: sentinel proteins for male reproduction

    Background Amyloid precursor protein (APP) is widely recognized for playing a central role in Alzheimer's disease pathogenesis. Although APP is expressed in several tissues outside the human central nervous system, the functions of APP and its family members in other tissues are still poorly understood. APP is involved in several biological functions which might be potentially important for male fertility, such as cell adhesion, cell motility, signaling, and apoptosis. Furthermore, APP superfamily members are known to be associated with fertility. Knowledge on the protein networks of APP in human testis and spermatozoa will shed light on the function of APP in the male reproductive system. Results We performed a Yeast Two-Hybrid screen and a database search to study the interaction network of APP in human testis and sperm. To gain insights into the role of APP superfamily members in fertility, the study was extended to APP-like protein 2 (APLP2). We analyzed several topological properties of the APP interaction network and the biological and physiological properties of the proteins in the APP interaction network were also specified by gene ontologyand pathways analyses. We classified significant features related to the human male reproduction for the APP interacting proteins and identified modules of proteins with similar functional roles which may show cooperative behavior for male fertility. Conclusions The present work provides the first report on the APP interactome in human testis. Our approach allowed the identification of novel interactions and recognition of key APP interacting proteins for male reproduction, particularly in sperm-oocyte interaction.publishe

    Nuclear Medicine Technologists Basic and Advanced Practice: What Does it Really Means?

    Nuclear Medicine is an independent medical specialty for almost fifty years. If that is quite clear and widely accepted, being in the basis and the justification for specialization between the distinct professionals involved, in many countries there is one exception: the Technologists. Since, in some countries, Nuclear Medicine is part of Medical Imaging area (which could include different specialties such as Radiology or Ultrasonography and, in a more restrict number of countries, even Radiotherapy), the “versatility” of Technologists might be somehow appreciated by some of the parts involved. Nevertheless, there are critical issues that need to be addressed - like the ability and capacity to perform distinct Practices at a certain level of autonomy and responsibility - and the final positions might be very distinct from one to another country
