O uso turístico enquanto valorização patrimonial: a importância do planeamento e da gestão
A dissertação apresentada versa sobre a relação entre as atividades relacionadas
com o património e o turismo e a utilização dos recursos patrimoniais através da prática
turística. Primeiramente são abordados os temas em separado, sendo depois estudada a
relação entre ambos.
Na segunda parte é abordada a importância do planeamento e da gestão na realização de
iniciativas que assentem na utilização turística do património. É apresentado um estudo de
caso, Aljezur, a que se segue a caracterização do concelho a vários níveis, é de seguida
apresentada uma análise crítica da forma como estão desenvolvidas as inciativas
relacionadas com o turismo e o património neste destino; ABSTRACT: This dissertation focuses the relation between tourism and heritage and the use
of heritage throw the tourism. On the first part the issues are firstly submitted separately
and then the relation between both.
The second part is about the importance of planning and management of iniciatives about
the touristic use of heritage. In this part are presented a case study about the village of
Aljezur, his main characteristics and a critical analysis about the iniciatives relationed with
tourism and heritage that are developed in this destination
Correlation between peripheral cytopenias and cytogenetic changes in the bone marrow in a paediatric population. Experience of 22 years
The haemogram is the most frequent request and an essential tool in the diagnosis of different pathologies in paediatric age, especially in haematological diseases. Peripheral cytopenias is the first laboratory finding suggestive of haematological disease, such as myelodysplastic syndrome, idiopathic thrombocytopenic purpura, among others. Confirmation of these pathologies should include the study of bone marrow, with analysis by different methodologies, including conventional cytogenetic karyotype analysis.
In this work, we intend to present and establish a correlation between the results obtained by conventional cytogenetics in bone marrow samples and observation of peripheral cytopenias in a paediatric population over 22 years.
A retrospective 22-year series (1995-2017) of 154 bone marrow samples from a paediatric population was analysed, which at the initial diagnosis presented peripheral cytopenia. The samples were process according to the established protocol for chromosome analysis in bone marrow, including cell culture, for each biological product, followed by a cytogenetic study to identify the karyotype.
In the 154 samples analysed with peripheral cytopenias, 31 were bicytopenias, 33 pancytopenias, 21 neutropenias, 11 anaemias and 58 thrombocytopenias, of which 22 were of idiopathic origin. We identify 15 samples with abnormal karyotype, some of which presented a complex karyotype. Samples with abnormal karyotypes had pancytopenia or bicytopenia at the same time.
Peripheral cytopenias are extremely important for suspicion of paediatric haematological diseases, especially in myelodysplastic syndrome. Conventional cytogenetic analysis of the bone marrow plays a fundamental role in the confirmation of these pathologies, their clinical evolution and the choice of proper therapy, However, micro-arrays should be performed with the aim of identifying micro-deletions / duplications or loss of heterozygosity that are characteristic in this group of pathologies.

Fanconi Anemia in Portugal: retrospective study of 34 years of investigation at National Health Institute (1980-2014)
A Anemia de Fanconi (AF) é uma doença recessiva rara, com uma frequência
estimada de 4 a 7 por 1 000 000 de nascimentos. Caraterizase
por malformações congénitas, falência medular e hipersensibilidade
a agentes clastogénicos de DNA. Devido à grande complexidade desta
patologia a primeira abordagem de diagnóstico, consiste na análise da
instabilidade cromossómica, após cultura celular com estimulação com
agentes clastogénicos diepoxibutano (DEB) ou mitomicina C (MMC). Realizou-
se um estudo retrospetivo de 34 anos (1980-2014) em 243 amostras
com suspeita de AF e de 25 amostras de familiares de doentes de
AF, num total de 268 amostras. Nas 243 amostras suspeitas de Anemia
de Fanconi, foram identificadas 37 com AF. A idade média ao diagnóstico
foi de 7 anos, existindo um ligeiro predomínio da incidência no sexo
feminino (59%). Uma amostra foi classificada como AF(-/+). Nos familiares
de doentes com AF foram identificados 2 casos positivos, o que
perfaz 39 amostras de AF positivas. Em quatro das amostras AF negativas,
observaram-se cariotipos anormais. Estes resultados não permitem
estimar uma frequência de doentes de AF em Portugal, uma vez que
não englobam indivíduos de todas as regiões portuguesas, mas permitem
uma estimativa da frequência espectável.Fanconi anemia (FA) is a rare recessive disease with an estimated frequency
from 4 to 7 per 1 000 000 live births. It is characterized by congenital
malformations, bone marrow failure and hypersensitivity to DNA
clastogenic agents. Due to the great complexity of the pathology the first
diagnostic approach is the analysis of chromosomal instability in cultured
cells after stimulation with the clastogenic agents diepoxybutane (DEB)
or mitomycin C (MMC). We performed a retrospective study of 34 years
(1980-2014) in 243 samples suspected of FA and 25 samples from relatives
of FA patients, totalizing 268 samples. In 243 suspected samples
37 were confirmed to be FA. The average age at diagnosis was 7 years,
with a slight predominant incidence in females (59%). One sample was
classified as FA (-/+). Among the patients’ relatives, 2 positive FA cases
were identified totaling 39 positive samples. In four negative samples of
suspected FA, abnormal karyotypes were observed. Since these samples
are not representative of individuals from all Portuguese regions, the data
only allow estimating the frequency of FA in Portugal
Toward unified molecular surveillance of RSV: A proposal for genotype definition
Background: Human respiratory syncytial virus (RSV) is classified into antigenic subgroups A and B. Thirteen genotypes have been defined for RSV-A and 20 for RSV-B, without any consensus on genotype definition. Methods: We evaluated clustering of RSV sequences published in GenBank until February 2018 to define genotypes by using maximum likelihood and Bayesian phylogenetic analyses and average p-distances. Results: We compared the patterns of sequence clustering of complete genomes; the three surface glycoproteins genes (SH, G, and F, single and concatenated); the ectodomain and the 2nd hypervariable region of G gene. Although complete genome analysis achieved the best resolution, the F, G, and G-ectodomain phylogenies showed similar topologies with statistical support comparable to complete genome. Based on the widespread geographic representation and large number of available G-ectodomain sequences, this region was chosen as the minimum region suitable for RSV genotyping. A genotype was defined as a monophyletic cluster of sequences with high statistical support (≥80% bootstrap and ≥0.8 posterior probability), with an intragenotype p-distance ≤0.03 for both subgroups and an intergenotype p-distance ≥0.09 for RSV-A and ≥0.05 for RSV-B. In this work, the number of genotypes was reduced from 13 to three for RSV-A (GA1-GA3) and from 20 to seven for RSV-B (GB1-GB7). Within these, two additional levels of classification were defined: subgenotypes and lineages. Signature amino acid substitutions to complement this classification were also identified. Conclusions: We propose an objective protocol for RSV genotyping suitable for adoption as an international standard to support the global expansion of RSV molecular surveillance.Fil: Goya, Stephanie. Gestão Documental na Câmara Municipal de Lisboa
A Modernização Administrativa nos Serviços Públicos é actualmente um imperativo ao desenvolvimento das instituições e à eficaz gestão dos seus recursos. A necessidade surge quando se constata a ineficácia de procedimentos administrativos, lentidão na resposta ao público, aumento da máquina burocrática, sistemas informáticos desactualizados e falta de normalização de documentos.Atenta às novas necessidades, a Câmara Municipal de Lisboa, com a participação activa da Divisão de Gestão de Arquivos inicia um trabalho de caracterização funcional de cada serviço produtor, do seu sistema de arquivo e das séries documentais produzidas.Este projecto tem como objectivos, o descongestionamento da massa documental, a sistematização da organização de documentos e um maior controlo sobre os mesmos, a eficácia na recuperação da informação, a aceleração na tomada de decisões, a promoção de estabilidade, continuidade e eficácia administrativa, a optimização da utilização do espaço, equipamento e dos recursos humanos.Pretende-se então elaborar uma arquitectura de sistema informático comum a toda a Câmara Municipal, desenvolvida pelo Departamento de Modernização Administrativa e Gestão de Informação, estrutura essa que funcionará de modo a impor uma normalização de documentos, que consequentemente facilitará a sua avaliação e destino final.Esta primeira fase, de levantamento, análise e diagnóstico desenvolve-se com trabalho de campo, assente em inquéritos às estruturas da autarquia, com a finalidade de conceber, executar e coordenar medidas de apoio e de cooperação entre Serviços que simplifiquem metodologias de trabalho e prestem um melhor Serviço Público.Os resultados, para além da aplicabilidade funcional e da modernização de sistemas informáticos, tem também em vista a revisão do Regulamento Arquivístico, de forma a adaptá-lo às realidades de uma Câmara Municipal cada vez mais complexa e específica na sua actividade
Consolidación de los estudios migratorios en la Patagonia Sur. Trayectoria de un grupo interdisciplinar: Capítulo 3
This paper gives an account of the action carried out within the framework of the research project migratory trajectories of chileans residing in Río Gallegos, Argentine southern Patagonia end, of the extension project Tales of Chileanness in Río Gallegos, both based at the National University of Austral Patagonia, those that were presented at the Second Scientific Meeting “Functioning dynamics of research/extension teams in critical contexts. Vicissitudes in times of pandemic COVID 19”; on axis 2: subjects, subjectivities and migratory movements. The writing exposes the development of the general and specificobjectives of these projects in the context of the pandemic, isolation and social distancing, witch led to the readaptation of the methodology originally proposed. Particularly, the objective of the article is to give an account of the progress of the research and extensionwork carried out by the group, from an interdisciplinary approach, exposing the background and trajectories that, in the field, the members of the group have traveled.El presente trabajo da cuenta de las acciones realizadas en el marco del proyecto de investigación Trayectorias migratorias de chilenos residentes en Río Gallegos, Patagonia Austral Argentina y, del proyecto de extensión Relatos de la chilenidad en Río Gallegos, ambos radicados en la Universidad Nacional de la Patagonia Austral, las que fueron expuestas en la Segunda Reunión Científica “Dinámicas de funcionamiento de equipos de investigación/extensión en contextos críticos. Vicisitudes en tiempos de pandemia COVID 19”; en el eje 2: sujetos, subjetividades y movimientos migratorios. El escrito expone el desarrollo de los objetivos generales y específicos de estos proyectos en el contexto de pandemia, de aislamiento y distanciamiento social, que llevó a la readaptación de la metodología planteada originalmente. Evaluation of bone turnover markers and serum minerals variations for predicting fracture healing versus non-union processes in adult sheep as a model for orthopedic research
Bone turnover markers (BTMs) have been considered as an auxiliary method of following the fracture healing process and for early prediction of impaired bone healing. A better understanding of the potential of BTMs in this application could allow for earlier interventions and improved patient care. The aim of this study with a large animal experimental model was to assess the variation of bone formation markers SOH namely the total alkaline phosphatase (ALP) and its bone-specific isoform (BALP), serum concentration of intact osteocalcin (OC), N-terminal propeptide type III procollagen (PIIINP) and of bone resorption markers â  namely tartrate resistant acid phosphatase (TRAP) and deoxypyridinoline crosslink (DPD) during the first stages of a normal fracture healing process and of a segmental critical size defect (CSD), which progresses to a non-union process. Thirty healthy female sheep (Portuguese Churra-da-Terra- Quente breed), approximately 4-years-old, were enrolled in this study. Jugular venous blood samples were collected pre-operatively and at 1, 2, 3, 4, 6, 8, 10 and 12 post-operative weeks. The animals of the CSD group showed significant lower serum levels of BALP, OC and significant higher serum PIIINP levels at early stages of the fracture healing process, compared with animals that progressed in a normal fracture healing process. Serum BALP, OC and PIIINP levels could be useful as non-invasive auxiliary tools with other complementary methods for predicting the outcome of traumatic bone fractures.Cristina P.
