    Effects of Pesticide Temephos on the Liver of Aphanius dispar (Rüppell 1828) (Pisces: Cyprinodontidae): A Microscopic Study

    The effects of  the pesticide temephos, at different concentrations, on the liver of Aphanius dispar, (Rüppell 1828) a cyprinodont fish, have been described. The changes in the liver tissue after exposure to 1 ppm, 5 ppm, 10 ppm and 25 ppm concentrations of temephos have been presented. Light and transmission electron microscopy showed that the degeneration of the liver cells commenced after exposure to 1 ppm and steadily progressed to show maximum changes at 25 ppm. Histopathological changes included the dilation of sinusoids, an increase in the number of lipid droplets and cytoplasmic vacuolation, pyknosis of the nuclei and focal necrosis. These results falsify the claim that temephos is a non-systemic poison

    Disseminated infections due to Immune Reconstitution Inflammatory Syndrome after Highly Active Antiretroviral Therapy-Report of 3 cases from Nigeria

    Immune Reconstitution Inflammatory Syndromes (IRIS) are exaggerated pathological inflammatory reactions occurring after initiation of highly active antiretroviral therapy (HAART) due to exuberant immune responses to occult or apparent opportunistic infections or cancers. In view of paucity of studies from Nigeria, we report 3 cases of IRIS presenting as disseminated infections in HIV-1 infected patients initiating HAART. The first case was a previously healthy female who developed disseminated tuberculosis after 4 weeks of regular HAART. Her HAART regimen was continued and she improved after commencement of anti-tuberculosis drugs, with evidence of progressive increase in CD4 cell count. The second case was a HAART-experienced female who stopped her drugs for 4months. Two months after recommencement of her previous HAART regimen, she developed features of disseminated herpes zoster infection, despite evidence of decrease in viral load by 95%. HAART was continued and she recovered completely after receiving valaciclovir tablets and antibiotics. The third patient was a female student who was commenced HAART on account of chronic cough and weight loss. Three months after regular HAART, she developed features of disseminated Kaposi's sarcoma involving the skin, oropharynx and lungs, despite evidence of 42% increase in CD4 cell count. Unfortunately, she rapidly deteriorated and died during the course of management. Clinicians should be alert to the possibility of IRIS in HIV-infected patients initiated or re-initiated on HAART. There is need for future prospective studies determining risk factors for IRIS in HIV-infected patients from Nigeria

    The home and school environments, physical activity levels, and adiposity indices of school-age children

    The home and school environments as well as physical activity may be linked to the development of childhood obesity. We evaluated the home and school environments (HSEs), physical activity levels (PAL), television viewing (TVV) and their associations with adiposity indices of school-age children. This cross-sectional study included children aged 6-12 years and their parents from Tamale, Ghana. HSEs and TVV were assessed using questionnaires. The physical activity questionnaire for children (PAQ-C) was used to assess children’s PALs. Weight, height and waist circumferences were measured using standard tools. About 45% of children lived within a walking distance to parks or outdoor recreation centres. Majority of the parents considered their neighbourhoods to be safe for children to engage in physical activity. Only 27% of the schools had a food and nutrition policy, and more than 70% had a field for outdoor activities. Children watched TV for an average of 1.7 hours/day. Mean physical activity scores was 2.51. The school-age children had mean (SD) BMI-for-age z-scores was -0.23(1.47). Time spent watching TV or playing video games was associated with children’s BMI-for-age z-scores (β=0.48, p=0.043), BMI (β=2.28 p=0.005), and % body fat (β=3.80, p=0.005). Child’s level of activity was negatively associated with waist circumference (β =-0.65, p<0.001). Lack of nutrition policy in schools was common. TVV hours predisposed children to excess weight whiles physical activity decreased the likelihood of being obese.

    Composition of Solid Waste in Al Jabal Al Akhdar, a Mountain Region Undergoing Rapid Urbanization in Northern Oman

    There is a lack of data on the generation and composition of waste in rural areas worldwide. The present study analyzed the composition of solid waste in Al Jabal Al Akhdar, a rural mountain region in the Sultanate of Oman, which is presently experiencing a rapid rate of urbanization due to tourism development. The solid wastes here are generated by the municipality collecting waste from residential, commercial, institutional and recreational areas, the military from a training camp and a few non-governmental private companies from their camps and hotels. The whole load from each of the three sources was manually segregated each month from June 2013 – May 2014. The results indicated that plastic is the dominant category in the wastes collected by the municipality and accounts for 26.7%, followed by paper (17.9%) and then food (14.4%). Food is the dominant category in the wastes collected by the military and private companies and accounts for 36.5% and 45.5% respectively. Management issues associated with solid waste are briefly considered. The study concluded that the municipality should implement an improved system for the collection of plastic waste and initiate a system for recycling it; the military and private companies should reduce the quantities of food waste by improved planning and management of the catering services

    Some Aspects of the Anatomy and Histology of Digestive Tracts in Two Sympatric Species of Freshwater Fishes

    Comparative anatomy and histology of the digestive tracts of two sympatric species of freshwater fish, Aphanius dispar (Cyprinodontidae) and Garra barreimiae (Cyprinidae) are studied. Morphometric measurements of alimentary canal such as length and the number and height of rugae in sections have been made for both species. Relationships between these morphometric characters and the total length of fish have been evaluated. The ratio between the length of alimentary canal and total length of fish in both species reflects their feeding habits. Histology of the ‘stomach’ and ‘intestine’ of these two species as shown by light microscopy has been described and compared. Results of this study are used to discuss the query whether these species have true stomachs

    Soil bacterial flora and enzymatic activities in zinc and lead contaminated soil

    Soil bacterial flora and enzymatic activities in lead and zinc contaminated soil of Ishiagu, Nigeria were investigated. The physicochemical properties measured showed that the mining pit had acidic pH (5.6) which gradually increased till 7.5 in the control. Organic matter was only 2.57mg/g in the pit but gradually reached 7.41mg/g in control. Pb concentration was higher at pit 360.52mg/g, 305.46mg/g at 5m away and lowest at control 36.16mg/g. Zn was 217.47mg/g at the pit, 176.32mg/g at 5m, 106.18mg/g at 10m and only 40.67mg/g at control. This showed a gradual fall away from the pit. Major organisms at the pit were Pseudomonas and Bacillus species (30% each) and Mocrococcus and Chromobacter species (20% each) E. coli, Salmonella and Lactobacillus species, which occurred in the control soil, were absent in the pit soil but occurred at various rates in other soil samples. Bacterial prevalence, diversity, and bioload were all high in the control, followed by 100m away while values decreased significantly towards the pit. Soil enzymatic activities correlated negatively with heavy metal concentration. This showed that the higher the heavy metal concentration the lower the enzymatic activities. Urease, dehydrogenase activity, hydrogen peroxidase and polyphenol oxidase were adversely affected but alkaline phosphatase did not show any significant effect

    A protocol for custom CRISPR Cas9 donor vector construction to truncate genes in mammalian cells using pcDNA3 backbone

    Background Clustered regularly interspaced short palindromic repeat (CRISPR) RNA-guided adaptive immune systems are found in prokaryotes to defend cells from foreign DNA. CRISPR Cas9 systems have been modified and employed as genome editing tools in wide ranging organisms. Here, we provide a detailed protocol to truncate genes in mammalian cells using CRISPR Cas9 editing. We describe custom donor vector construction using Gibson assembly with the commonly utilized pcDNA3 vector as the backbone. Results We describe a step-by-step method to truncate genes of interest in mammalian cell lines using custom-made donor vectors. Our method employs 2 guide RNAs, mutant Cas9D10A nickase (Cas9 = CRISPR associated sequence 9), and a custom-made donor vector for homologous recombination to precisely truncate a gene of interest with a selectable neomycin resistance cassette (NPTII: Neomycin Phosphotransferase II). We provide a detailed protocol on how to design and construct a custom donor vector using Gibson assembly (and the commonly utilized pcDNA3 vector as the backbone) allowing researchers to obtain specific gene modifications of interest (gene truncation, gene deletion, epitope tagging or knock-in mutation). Selection of mutants in mammalian cell lines with G418 (Geneticin) combined with several screening methods: western blot analysis, polymerase chain reaction, and Sanger sequencing resulted in streamlined mutant isolation. Proof of principle experiments were done in several mammalian cell lines. Conclusions Here we describe a detailed protocol to employ CRISPR Cas9 genome editing to truncate genes of interest using the commonly employed expression vector pcDNA3 as the backbone for the donor vector. Providing a detailed protocol for custom donor vector design and construction will enable researchers to develop unique genome editing tools. To date, detailed protocols for CRISPR Cas9 custom donor vector construction are limited (Lee et al. in Sci Rep 5:8572, 2015; Ma et al. in Sci Rep 4:4489, 2014). Custom donor vectors are commercially available, but can be expensive. Our goal is to share this protocol to aid researchers in performing genetic investigations that require custom donor vectors for specialized applications (specific gene truncations, knock-in mutations, and epitope tagging applications)

    Psychometric validity of the distress thermometer and problem check list in ART-na\uefve HIV infected patients in Northern Nigeria

    Background: HIV diagnosis comes with a lot of worry and distress. Ability to objectively estimate this distress by non-psychiatrist will enhance early detection of psychological distress for intervention. Objectives: To investigate the validity of the Distress Thermometer (DT) and its problem checklist in achieving early detection of mental distress among ART-na\uefve HIV infected patient. Materials and Methods: A total of 90 ART-na\uefve HIV infected patients completed the DT and its problem check list, Hospital Anxiety Depression Scale (HADS), Oslo Social Support Scale and the 14-item Resilience Scale. Results: The DT was positively correlated with all the measures of distress and reversely correlated with all the positive wellness in this study. The correlations were only significant for the negative measures of psychological wellness. The internal consistency of the DT\u2019s problem list overall and sub-categories were within acceptable range (i.e. \u3b1 > 0.50). The Receiver Operating Characteristic (ROC) curves and Area Under the Curves (AUC) analysis were significant and found the DT and Problem List to respectively differentiate between cases of distress, anxiety and depression. The DT\u2019s cut-off was >5.0 with AUC range (0.754 \u2013 0.709); sensitivity range (81.0% \u2013 70.4%); specificity range (68.3% \u2013 65.2%) for distress, anxiety and depression as determined by HADS. And the Problem List cut-off was >6.0 with AUC range (0.854 \u2013 0.821); sensitivity range (90.5% - 85.7%); specificity range (68.3% - 65.2%) for distress, anxiety and depression as measured by HADS. Conclusion: The DT and Problem List were found to be valid measures of distress in ART-na\uefve HIV infected patients

    Central-West Siberian-breeding Bar-tailed Godwits (<i>Limosa lapponica</i>) segregate in two morphologically distinct flyway populations

    Long-distance migratory species often include multiple breeding populations, with distinct migration routes, wintering areas and annual-cycle timing. Detailed knowledge on population structure and migratory connectivity provides the basis for studies on the evolution of migration strategies and for species conservation. Currently, five subspecies of Bar-tailed Godwits Limosa lapponica have been described. However, with two apparently separate breeding and wintering areas, the taxonomic status of the subspecies L. l. taymyrensis remains unclear. Here we compare taymyrensis Bar-tailed Godwits wintering in the Middle East and West Africa, respectively, with respect to migration behaviour, breeding area, morphology and population genetic differentation in mitochondrial DNA. By tracking 52 individuals from wintering and staging areas over multiple years, we show that Bar-tailed Godwits wintering in the Middle East bred on the northern West-Siberian Plain (n = 19), while birds from West Africa bred further east, mostly on the Taimyr Peninsula (n = 12). The two groups differed significantly in body size and shape, and also in the timing of both northward and southward migrations. However, they were not genetically differentiated, indicating that the phenotypic (i.e. geographical, morphological and phenological) differences arose either very recently or without current reproductive isolation. We conclude that the taymyrensis taxon consists of two distinct populations with mostly non-overlapping flyways, which warrant treatment as separate taxonomic units. We propose to distinguish a more narrowly defined taymyrensis subspecies (i.e. the Bar-tailed Godwits wintering in West Africa and breeding on Taimyr), from a new subspecies (i.e. the birds wintering in the Middle East and breeding on the northern West-Siberian Plain)
