    Thermal Comfort Evaluation Within Non-residential Environments: Development of Artificial Neural Network by Using the Adaptive Approach Data☆

    Abstract A new algorithm for the PMV calculation was developed using Artificial Neural Networks. Several experimental investigations were carried out in two classrooms using both Fanger static model and adaptive approaches for the PMV evaluation. The Artificial Neural Network was trained considering a few input parameters; specifically for the network development only the air temperature and relative humidity were considered as experimental data. This algorithm allows to correlate the thermal sensation to both indoor and outdoor factors and it is a useful tool for predicting the PMV index, replacing the traditional methods with less time and cost demanding

    Adhesion GPCRs are widely expressed throughout the subsections of the gastrointestinal tract

    Background: G protein-coupled receptors (GPCRs) represent one of the largest families of transmembrane receptors and the most common drug target. The Adhesion subfamily is the second largest one of GPCRs and its several members are known to mediate neural development and immune system functioning through cell-cell and cell-matrix interactions. The distribution of these receptors has not been characterized in detail in the gastrointestinal (GI) tract. Here we present the first comprehensive anatomical profiling of mRNA expression of all 30 Adhesion GPCRs in the rat GI tract divided into twelve subsegments. Methods: Using RT-qPCR, we studied the expression of Adhesion GPCRs in the esophagus, the corpus and antrum of the stomach, the proximal and distal parts of the duodenum, ileum, jejunum and colon, and the cecum. Results: We found that twenty-one Adhesion GPCRs (70%) had a widespread (expressed in five or more segments) or ubiquitous (expressed in eleven or more segments) distribution, seven (23%) were restricted to a few segments of the GI tract and two were not expressed in any segment. Most notably, almost all Group III members were ubiquitously expressed, while the restricted expression was characteristic for the majority of group VII members, hinting at more specific/localized roles for some of these receptors. Conclusions: Overall, the distribution of Adhesion GPCRs points to their important role in GI tract functioning and defines them as a potentially crucial target for pharmacological interventions. © 2012 Badiali et al.; licensee BioMed Central Ltd

    Thermal Behaviour and Energy Saving Evaluation of Innovative Reinforced Coatings

    Abstract The use of fibre-reinforced interlayer structures as coatings is very spread in building refurbishment, but it is important to find effective solutions both for mechanical and thermal performance. In the present paper the thermal behaviour of innovative fiber-reinforced coatings was investigated in order to find the best solutions in terms of mechanical strength and energy saving in buildings. Four different coatings (with and without the internal reinforced structure) were considered and their thermal conductivities were preliminary investigated by using an innovative measurement apparatus (Small Hot Box). A thermal analysis was carried out for the evaluation of the effects of the coatings on the building envelope, especially the thermal transmittance, calculated for different thickness. Preliminary results showed that 3-4 cm of coatings allowed to reduce thermal transmittance of building envelope by about 60-70%. In order to evaluate the energy saving, one building was considered and simulated by means of Trnsys software: it was chosen for the evaluation of the refurbishment of the building by using the selected solutions. The simulation model was implemented in SketchUp and the energy demand with and without the innovative coatings was simulated; the simulation model was also used for the evaluation of energy saving in different climate conditions. Results show that for two coatings an important reduction of the heat loss through of the building envelope can be obtained

    Health risk assessment of environmental selenium: Emerging evidence and challenges (Review)

    New data have been accumulated in the scientific literature in recent years which allow a more adequate risk assessment of selenium with reference to human health. This new evidence comes from environmental studies, carried out in populations characterized by abnormally high or low selenium intakes, and from high-quality and large randomized controlled trials with selenium recently carried out in the US and in other countries. These trials have consistently shown no beneficial effect on cancer and cardiovascular risk, and have yielded indications of unexpected toxic effects of selenium exposure. Overall, these studies indicate that the minimal amount of environmental selenium which is source of risk to human health is much lower than anticipated on the basis of older studies, since toxic effects were shown at levels of intake as low as around 260 µg/day for organic selenium and around 16 µg/day for inorganic selenium. Conversely, populations with average selenium intake of less than 13-19 µg/day appear to be at risk of a severe cardiomyopathy, Keshan disease. Overall, there is the need to reconsider the selenium standards for dietary intake, drinking water, outdoor and indoor air levels, taking into account the recently discovered adverse health effects of low-dose selenium overexposure, and carefully assessing the significance of selenium-induced proteomic changes

    IGHV mutational status of nodal marginal zone lymphoma by NGS reveals distinct pathogenic pathways with different prognostic implications

    The precise B cell of origin and molecular pathogenesis of nodal marginal zone lymphoma (NMZL) remain poorly defined. To date, due to the rarity of NMZL, the vast majority of already-published studies have been conducted on a limited number of samples and the technical approach to analyze the immunoglobulin genes was of amplifying rearranged variable region genes with the classical direct sequencing of the PCR products followed by cloning. Here, we studied the B cell Ig heavy-chain repertoires by next-generation sequencing (NGS) in 30 NMZL cases. Most of the cases were mutated (20/28; 71.5%) with homologies to the respective germ line genes ranging from 85 to 97, 83%, whereas 8/28 (28.5%) were unmutated. In addition, our results show that NMZL cases have a biased usage of specific immunoglobulin heavy-chain variable (IGHV) region genes. Moreover, we documented intraclonal diversity in all (100%) of the mutated cases and ongoing somatic hypermutations (SHM) have been confirmed by hundreds of reads. We analyzed the mutational pattern to detect and quantify antigen selection pressure and we found a positive selection in 4 cases, whereas in the remaining cases there was an unspecific stimulation. Finally, the disease-specific survival and the progression-free survival were significantly different between cases with mutated and unmutated IGHV genes, pointing out mutational status as a possible new biomarker in NMZL

    Luteinizing Hormone/Human Chorionic Gonadotropin Receptor Immunohistochemical Score Associated with Poor Prognosis in Endometrial Cancer Patients.

    The aim of this study was to develop a scoring system of the immunohistochemical (IHC) expression of luteinizing hormone/human chorionic gonadotropin receptor (LHCG-R) in endometrial cancer (EC) patients. Nonconsecutive hysterectomy specimens containing EC collected from April 2013 to October 2015 were selected. Hematoxylin-eosin stained sections from each case were reviewed and representative sections from each tumor were selected. IHC staining was performed for the detection of LHCG-R. The percentage of stained cells and the staining intensity were assessed in order to develop an immunohistochemical score. Moreover, we examined the correlation of the score with grading and lymphovascular space invasion (LVSI). There was a statistically significant positive correlation between grading and IHC scoring (p=0.01) and a statistically significant positive correlation between LVSI and IHC score (p<0.01). In conclusion, we suggest that the immunohistochemical score presented here could be used as a marker of bad prognosis of EC patients. Nevertheless, further studies are needed in order to validate it. The study was registered in the Careggi Hospital public trials registry with the following number: 2013/0011391

    Factors influencing weight loss practices in Italian boxers: A cluster analysis

It is common practice in combat sports that athletes rapidly lose body weight before a match, by applying different practices—some safer and others possibly dangerous. The factors behind the choice of practices utilised have not been fully studied. This study aimed to investigate the weight loss strategies used by Italian boxers and to look at the difference between higher and lower risk practice adaptors. A modified version of a validated questionnaire has been sent to 164 amateur (88%) and professional (12%) boxers by email. A heatmap with hierarchical clustering was used to explore the presence of subgroups. Weight loss strategies were used by 88% of the athletes. Two clusters were found, defined by the severity of weight loss behaviours. Professional fighters, high-level athletes and females were more represented in Cluster 2, the one with more severe weight-loss practices. These athletes were characterised by a higher weight loss magnitude and frequency throughout the season and reported being more influenced by physicians and nutritionists, compared with the boxers in Cluster 1. Not all the weight loss practices are used with the same frequency by all boxers. The level of the athlete and the boxing style have an influence on the weight-cutting practices

    CCAP regulates feeding behavior via the NPF pathway in Drosophila adults

    The intake of macronutrients is crucial for the fitness of any animal and is mainly regulated by peripheral signals to the brain. How the brain receives and translates these peripheral signals or how these interactions lead to changes in feeding behavior is not well-understood. We discovered that 2 crustacean cardioactive peptide (CCAP)-expressing neurons in Drosophila adults regulate feeding behavior and metabolism. Notably, loss of CCAP, or knocking down the CCAP receptor (CCAP-R) in 2 dorsal median neurons, inhibits the release of neuropeptide F (NPF), which regulates feeding behavior. Furthermore, under starvation conditions, flies normally have an increased sensitivity to sugar; however, loss of CCAP, or CCAP-R in 2 dorsal median NPF neurons, inhibited sugar sensitivity in satiated and starved flies. Separate from its regulation of NPF signaling, the CCAP peptide also regulates triglyceride levels. Additionally, genetic and optogenetic studies demonstrate that CCAP signaling is necessary and sufficient to stimulate a reflexive feeding behavior, the proboscis extension reflex (PER), elicited when external food cues are interpreted as palatable. Dopaminergic signaling was also sufficient to induce a PER. On the other hand, although necessary, NPF neurons were not able to induce a PER. These data illustrate that the CCAP peptide is a central regulator of feeding behavior and metabolism in adult flies, and that NPF neurons have an important regulatory role within this system

    Burkitt lymphoma with a granulomatous reaction: an M1/Th1-polarised microenvironment is associated with controlled growth and spontaneous regression

    Aims: Burkitt lymphoma (BL) is an aggressive B-cell lymphoma that, in some instances, may show a granulomatous reaction associated with a favourable prognosis and occasional spontaneous regression. In the present study, we aimed to define the tumour microenvironment (TME) in four such cases, two of which regressed spontaneously. Methods and results: All cases showed aggregates of tumour cells with the typical morphology, molecular cytogenetics and immunophenotype of BL surrounded by a florid epithelioid granulomatous reaction. All four cases were Epstein–Barr virus (EBV)-positive with type I latency. Investigation of the TME showed similar features in all four cases. The analysis revealed a proinflammatory response triggered by Th1 lymphocytes and M1 polarised macrophages encircling the neoplastic cells with a peculiar topographic distribution. Conclusions: Our data provide an in-vivo picture of the role that specific immune cell subsets might play during the early phase of BL, which may be capable of maintaining the tumour in a self-limited state or inducing its regression. These novel results may provide insights into new potential therapeutic avenues in EBV-positive BL patients in the era of cellular immunotherapy
