    Retentissements psychiques du cancer gynécologique pelvien sur la sexualité féminine

    Pelvic gynaecological cancer is a disease that fits into internal reproductive organs of women : vagina, uterus, cervix and ovaries. These cancers are treated in patients of the study by hysterectomy, extended or not (removal of genitals), chemotherapy, radiotherapy and brachytherapy. It is a castration that these women experience in the reality of a body that suffers. Cancer and treatments are corporal effractions that reiterate a psychic representation of the penetration as the passive reception of the other in oneself. This figure of the foreigner inside the ego summons fantasies, affects and penetrating representations that damage body image, generates passivity images and rekindles passivation experiences. Moreover, this effraction takes place specifically on the intimate sex place, where the growing into a woman and then into a mother incarnates itself, summoning the feminine of the woman in his plurality but also in his connections to feminity. After clearing, in a very broad literary repository, the few studies that put into perspective the effraction of the gynaecological cancer and its treatments with feminine sexuality, we set our own research in the approach of psychoanalytic psychopathology in its relations with psychosomatics. A consistent identification of psychoanalytic theory of the two major fields requested by the subject of the study ensues : feminine sexuality and body in the disease. The original methodology implemented fits into an exploratory process supported by the administration of two projective tests : Rorschach and TAT, along with clinical research interviews. This is a longitudinal methodology at three selected steps of the therapeutic course of these women. The results, treated initially in the singular dimension of the case study are then grouped according to the hypotheses to show the shared adjustments in these patients. The analogy between the situation of the infans and the women under passivity constraints accompanies the description of the three steps of the study. It responds to the hypothesis of a mobilization of psychic modalities specific to corporal effraction treating process which summon, mistreat, wound and remove internal genitalia. The study reveals specifically feminine primitive anxiety such as penetration anxiety, treated by limit modalities, narcissistic and anal around a holed body fantasy and a dangerous interiority. Finally, the study explores the entire set of drives in order to figure out fusion and defusion movements subtending the erotic decathexis of the genital, summoning the feminine in its passive potentialities and accompanying the reorientation of the object-relation under the death drive influence.Le cancer gynécologique pelvien est une pathologie qui se loge dans les organes génitaux internes de la femme : vagin, utérus, col de l'utérus et ovaires. Ces organes malades sont traités chez les patientes de l'étude par hystérectomie plus ou moins élargie (ablation des organes génitaux), chimiothérapie, radiothérapie et curiethérapie. C'est donc une castration que ces femmes subissent dans la réalité d'un corps qui souffre. Cancer et traitements sont autant d'effractions corporelles qui réitèrent une représentation psychique de la pénétration comme de l’accueil passif de l'autre en soi. Cette figure de l'étranger à l’intérieur du moi convoque des fantasmes, affects et représentations pénétrantes qui endommagent l'image du corps, génère des images de passivité et ravive des expériences de passivation. De plus, cette effraction s'opère spécifiquement sur le lieu intime du sexe, là où s'incarne le devenir femme puis le devenir mère, convoquant le féminin de la femme dans sa pluralité, mais aussi dans ses rapports à la féminité. Après avoir dégagé, dans un référentiel littéraire très large, les rares travaux qui mettent en perspective l'effraction d'un cancer gynécologique et ses traitements avec la sexualité des femmes, nous avons situé notre propre recherche dans une approche de psychopathologie psychanalytique et dans ses rapports avec la psychosomatique. S'ensuit un repérage de la théorie psychanalytique conséquent des deux grands champs sollicités par le sujet de l'étude : la sexualité féminine et le corps dans la maladie. La méthodologie originale mise en place s'inscrit dans une démarche exploratoire étayée de la passation de deux tests projectifs : le Rorschach et le TAT, ainsi que d'entretiens cliniques de recherche. Il s'agit d'une méthodologie longitudinale sur trois temps choisis du parcours thérapeutique de ces femmes. Les résultats, traités dans un premier temps dans la dimension singulière de l'étude de cas, sont regroupé ensuite en fonction des hypothèses pour faire apparaître des aménagements communs chez ces patientes. L'analogie entre la situation de l'infans et celle de ces femmes sous contraintes de passivité accompagne la description des trois temps de l'étude. Celle-ci répond à l'hypothèse d'une mobilisation de modalités psychiques spécifiques du traitement de l'effraction corporelle qui convoquent, malmènent, blessent et suppriment les organes génitaux internes. L'étude révèle l'actualisation d'angoisses primitives spécifiquement féminines, telle que l'angoisse de pénétration, traitées par des modalités limites, narcissiques et anales autour d'un fantasme de corps troué et d'une intériorité dangereuse. En définitive, l'étude explore l'ensemble du jeu pulsionnel pour en décrypter les mouvements d’intrication et de désintrication sous-tendant le désinvestissement érotique du génital, convoquant le féminin dans ses potentialités passives et accompagnant l'infléchissement de la relation d'objet sous l'influence de la pulsion de mort

    Kv1.3 inhibitors in the treatment of glioma and melanoma

    The groups of Prof.s Szabò and Gulbins demonstrated that the inhibition of the mtKv1.3 by different membrane-permeant specific drugs is able to induce apoptosis in different cancer cell lines in vitro, ex vivo in B-CLL cells and in a in vivo orthotopic mouse melanoma model (Leanza L. et al 2012, Leanza L. et al 2013). Here, I tested different Kv1.3 inhibitors on their ability to influence tumor growth in two types of models, glioma and melanoma. In particular, I investigated whether these drugs are able to induce cell death by depolarizing the mitochondrial membrane, increasing ROS production, eventually leading to cytochrome c release in different cancer cell lines. Thanks to their augmented solubility and mitochondrial targeting ability, the newly synthesized drugs, together with clofazimine, were applied, after a pharmacokinetic analysis, in vivo to a syngeneic glioma model and to the orthotopic melanoma model. Beside the impact on tumor growth, the synergistic effect with ROS level, previously proposed in the in vitro studies, was analyzed. The last part of the study focused on the mechanism of the in vivo action of the precursor and the derivatives in the reduction of melanoma tumor, addressing the influence of these compounds on the immune system.     Analysis of total organic carbon in soil-biochar systems

    Amending agricultural soils with biochar can contribute to negative carbon strategies when the resistance to oxidation of soil carbon is improved (avoided CO2 emission) and plant growth is promoted (increased CO2 fixation). The environmental stability and sequestering capacity of biochar is dependent on the chemical form of carbon and its physical location in the carbonaceous matrix. The addition of biochar in soil increases noticeably the stable carbon pool, while the effect on labile carbon, including polyaromatic structures, is less marked.1 The fertilizing action can be lost if biochar is removed from the cultivated area due to physical processes (vertical transport, lateral export, slacking). Assessing the fate of carbon in the soil requires the use of suitable analytical methods that should be validated for the presence of biochar.     During rain events, pollutants in agricultural soils can be transported from fields to surface and/or groundwater resulting in contamination of streams and rivers. Researchers and farmers must work together to find solutions to ensure the preservation of crop production without jeopardizing water quality or the health of the ecosystem. Establishment of riparian zones may reduce the effects of diffuse discharges of pollutants into waterways. The addition of biochar to soils, particularly in a riparian zones, can reduce the mobility of contaminants and improve removal efficiency due its sorptive capacity.     Multiple Bayesian network meta-analyses to establish therapeutic algorithms for metastatic triple negative breast cancer

    Immunotherapy; PARP inhibitors; PembrolizumabImmunoteràpia; Inhibidors de PARP; PembrolizumabInmunoterapia; Inhibidores de PARP; PembrolizumabMetastatic triple-negative breast cancer (mTNBC) is a poor prognostic disease with limited treatments and uncertain therapeutic algorithms. We performed a systematic review and multiple Bayesian network meta-analyses according to treatment line to establish an optimal therapeutic sequencing strategy for this lethal disease. We included 125 first-line trials (37,812 patients) and 33 s/further-lines trials (11,321 patients). The primary endpoint was progression-free survival (PFS). Secondary endpoints included overall response rates (ORR), overall survival (OS) and safety, for first and further lines, separately. We also estimated separate treatment rankings for the first and subsequent lines according to each endpoint, based on (surface under the cumulative ranking curve) SUCRA values. No first-line treatment was associated with superior PFS and OS than paclitaxel ± bevacizumab. Platinum-based polychemotherapies were generally superior in terms of ORR, at the cost of higher toxicity.. PARP-inhibitors in germline-BRCA1/2-mutant patients, and immunotherapy + chemotherapy in PD-L1-positive mTNBC, performed similar to paclitaxel ± bevacizumab. In PD-L1-positive mTNBC, pembrolizumab + chemotherapy was better than atezolizumab + nab-paclitaxel in terms of OS according to SUCRA values. In second/further-lines, sacituzumab govitecan outperformed all other treatments on all endpoints, followed by PARP-inhibitors in germline-BRCA1/2-mutant tumors. Trastuzumab deruxtecan in HER2-low mTNBC performed similarly and was the best advanced-line treatment in terms of PFS and OS after sacituzumab govitecan, according to SUCRA values. Moreover, comparisons with sacituzumab govitecan, talazoparib and olaparib were not statistically significant. The most effective alternatives or candidates for subsequent lines were represented by nab-paclitaxel (in ORR), capecitabine (in PFS) and eribulin (in PFS and OS)

    The importance of thermal torques on the migration of planets growing by pebble accretion

    A key process in planet formation is the exchange of angular momentum between a growing planet and the protoplanetary disc, which makes the planet migrate through the disc. Several works show that in general low-mass and intermediate-mass planets migrate towards the central star, unless corotation torques become dominant. Recently, a new kind of torque, called the thermal torque, was proposed as a new source that can generate outward migration of low-mass planets. While the Lindblad and corotation torques depend mostly on the properties of the protoplanetary disc and on the planet mass, the thermal torque depends also on the luminosity of the planet, arising mainly from the accretion of solids. Thus, the accretion of solids plays an important role not only in the formation of the planet but also in its migration process. In a previous work, we evaluated the thermal torque effects on planetary growth and migration mainly in the planetesimal accretion paradigm. In this new work, we study the role of the thermal torque within the pebble accretion paradigm. Computations are carried out consistently in the framework of a global model of planet formation that includes disc evolution, dust growth and evolution, and pebble formation. We also incorporate updated prescriptions of the thermal torque derived from high-resolution hydrodynamical simulations. Our simulations show that the thermal torque generates extended regions of outward migration in low-viscosity discs.     Evaluation of biological behavior of Toxoplasma gondii atypical isolates # 14 and # 163

    Toxoplasma gondii is an obligate intracellular protozoan parasite capable of infecting warm-blooded animals, including humans. A highly diverse genetic population has been reported in Central and South America, predominating mainly atypical genotypes. Different genotypes showed different biological behavior in mice. The aim of this study was to evaluate the biological behavior of T. gondii isolates obtained from Macropus rufogriseus (TgMr) and Saimiri boliviensis (TgSb) identified as atypical genotypes # 14 and # 163, respectively. Strains RH, ME49 and VEG were used as reference for clonal types I, II and III, respectively. In vitro invasion and replication capacity assays were analyzed at 6 and 18 hpi, respectively. In vivo assay was performed in Swiss mice (n = 30) using 1 × 102 and 1 × 103 parasites/mouse as infective doses (ME49, VEG, TgMr, TgSb and negative control). Morbi-mortality and tissues PCR were assessed. Lymphoproliferation assays were performed and gamma interferon was measured by ELISA. The ME49 strain showed the highest invasion, followed by TgSb and VEG, while RH and TgMr presented the lowest invasions. The RH strain and the TgSb isolate showed more endodyogeny events (fastest doubling times) than VEG and ME49 strains and the TgMr isolate. Both atypical isolates showed high virulence (100% morbi-mortality, at 8–10 dpi) and parasite DNA was detected in all tissue samples. Splenocytes from mice inoculated with TgMr and TgSb registered the highest values of gamma interferon. An in vitro invasion-replication index was established which correlates inversely with virulence in mice. In conclusion, T. gondii atypical isolates # 14 and # 163 showed a different in vitro behavior than clonal strains, with low invasion-replication indexes but being highly virulent in mouse model.Fil: Bernstein, Mariana.     Os desafios da extensão rural no processo de mediação de saberes

    Este ensaio teórico tem por objetivo trazer reflexões sobre o processo de mediação em ações de extensão rural frente a uma realidade multifacetada do rural sob a perspectiva do enfoque agroecológico. Parte-se do profissional de extensão rural como central na discussão. Percebe-se nos últimos anos, a formação pela academia, de um considerável contingente de especialistas para atender demandas complexas, apresentadas pela realidade rural. Portanto, tendo como lócus o órgão oficial de extensão Rural do RS, podese inferir um certo descompasso entre as diretrizes e princípios da Emater/Ascar-RS e a operacionalização das políticas públicas no processo de mediação dos agentes de extensão rural com as populações rurais. Discute-se, neste artigo, como resultados a abordagem do “Novo profissional de Extensão Rural”, considerando até que ponto atualmente tem-se este novo profissional mediando saberes e conhecimentos.This theoretical essay aims to bring reflections on the process of mediation in rural extension actions in a rural multiple reality from the perspective of agroecological approach. Realize in recent years, training by the academy a considerable number of experts to meet complex demands presented by rural reality. Therefore, with the locus the official organ of the RS Rural extension, is possible to infer some gap between the guidelines and principles of Emater/Ascar-RS and the implementation of public policies in the process of mediation of rural extension agents with rural populations.
