    Biochemical, histological and histochemical changes in Aristichthys nobilis Rich. liver exposed to thiamethoxam

    Background and purpose: The aim of the present study was to investigate the effects of the insecticide thiamethoxam on some biochemical, histological and histochemical parameters of bighead carp liver (Aristichthys nobilis), which is an economically important fish species for aquaculture. Materials and methods: Different increasing concentrations of 6.6 mg l-1, 10 mg l-1 and 20 mg l-1 of the test chemical under laboratory conditions were tested for their toxicity on fish for 96. The hepatic activity of the enzymes lactate dehydrogenase (LDH), aspartate aminotransferase (ASAT) and alanine aminotransferase (ALAT) were measured. In addition, the hepatic histological structure was observed for alterations, as well as the lipid content was observed by histochemical staining. Results: It was determined that the enzymatic activity of the exposed fish was increased compared to the control group (p<0.05). Furthermore, the enzymatic activity was increased proportionally to the increasing thiamethoxam concentrations. The histological lesions, which were observed in the liver parenchyma, were degenerative and necrotic. The degenerative alterations were as follows: granular, balloon and fatty degeneration. The necrotic alterations in the fish liver were associated with presence of karyopyknosis, karyorrehsis and karyolysis, respectively. The histological alterations in the liver blood vessels were hyperemia and lymphocyte proliferation. Along with the established histological changes in the liver parenchyma, we found presence of fatty degeneration in the hepatocytes using Sudan III staining on cryostat sections. Conclusions: Our study shows that there is a relation between the concentration of the insecticide and biochemical changes, as well the severity of expression of the histological and histochemical alterations in the bighead carp liver. Overall, such experiments could be successfully applied in research and monitoring programs to study the effects of pesticides on fish

    Effects of alirocumab on types of myocardial infarction: insights from the ODYSSEY OUTCOMES trial

    Aims  The third Universal Definition of Myocardial Infarction (MI) Task Force classified MIs into five types: Type 1, spontaneous; Type 2, related to oxygen supply/demand imbalance; Type 3, fatal without ascertainment of cardiac biomarkers; Type 4, related to percutaneous coronary intervention; and Type 5, related to coronary artery bypass surgery. Low-density lipoprotein cholesterol (LDL-C) reduction with statins and proprotein convertase subtilisin–kexin Type 9 (PCSK9) inhibitors reduces risk of MI, but less is known about effects on types of MI. ODYSSEY OUTCOMES compared the PCSK9 inhibitor alirocumab with placebo in 18 924 patients with recent acute coronary syndrome (ACS) and elevated LDL-C (≄1.8 mmol/L) despite intensive statin therapy. In a pre-specified analysis, we assessed the effects of alirocumab on types of MI. Methods and results  Median follow-up was 2.8 years. Myocardial infarction types were prospectively adjudicated and classified. Of 1860 total MIs, 1223 (65.8%) were adjudicated as Type 1, 386 (20.8%) as Type 2, and 244 (13.1%) as Type 4. Few events were Type 3 (n = 2) or Type 5 (n = 5). Alirocumab reduced first MIs [hazard ratio (HR) 0.85, 95% confidence interval (CI) 0.77–0.95; P = 0.003], with reductions in both Type 1 (HR 0.87, 95% CI 0.77–0.99; P = 0.032) and Type 2 (0.77, 0.61–0.97; P = 0.025), but not Type 4 MI. Conclusion  After ACS, alirocumab added to intensive statin therapy favourably impacted on Type 1 and 2 MIs. The data indicate for the first time that a lipid-lowering therapy can attenuate the risk of Type 2 MI.     Effect of alirocumab on mortality after acute coronary syndromes. An analysis of the ODYSSEY OUTCOMES randomized clinical trial

    Background: Previous trials of PCSK9 (proprotein convertase subtilisin-kexin type 9) inhibitors demonstrated reductions in major adverse cardiovascular events, but not death. We assessed the effects of alirocumab on death after index acute coronary syndrome. Methods: ODYSSEY OUTCOMES (Evaluation of Cardiovascular Outcomes After an Acute Coronary Syndrome During Treatment With Alirocumab) was a double-blind, randomized comparison of alirocumab or placebo in 18 924 patients who had an ACS 1 to 12 months previously and elevated atherogenic lipoproteins despite intensive statin therapy. Alirocumab dose was blindly titrated to target achieved low-density lipoprotein cholesterol (LDL-C) between 25 and 50 mg/dL. We examined the effects of treatment on all-cause death and its components, cardiovascular and noncardiovascular death, with log-rank testing. Joint semiparametric models tested associations between nonfatal cardiovascular events and cardiovascular or noncardiovascular death. Results: Median follow-up was 2.8 years. Death occurred in 334 (3.5%) and 392 (4.1%) patients, respectively, in the alirocumab and placebo groups (hazard ratio [HR], 0.85; 95% CI, 0.73 to 0.98; P=0.03, nominal P value). This resulted from nonsignificantly fewer cardiovascular (240 [2.5%] vs 271 [2.9%]; HR, 0.88; 95% CI, 0.74 to 1.05; P=0.15) and noncardiovascular (94 [1.0%] vs 121 [1.3%]; HR, 0.77; 95% CI, 0.59 to 1.01; P=0.06) deaths with alirocumab. In a prespecified analysis of 8242 patients eligible for ≄3 years follow-up, alirocumab reduced death (HR, 0.78; 95% CI, 0.65 to 0.94; P=0.01). Patients with nonfatal cardiovascular events were at increased risk for cardiovascular and noncardiovascular deaths (P<0.0001 for the associations). Alirocumab reduced total nonfatal cardiovascular events (P<0.001) and thereby may have attenuated the number of cardiovascular and noncardiovascular deaths. A post hoc analysis found that, compared to patients with lower LDL-C, patients with baseline LDL-C ≄100 mg/dL (2.59 mmol/L) had a greater absolute risk of death and a larger mortality benefit from alirocumab (HR, 0.71; 95% CI, 0.56 to 0.90; Pinteraction=0.007). In the alirocumab group, all-cause death declined wit h achieved LDL-C at 4 months of treatment, to a level of approximately 30 mg/dL (adjusted P=0.017 for linear trend). Conclusions: Alirocumab added to intensive statin therapy has the potential to reduce death after acute coronary syndrome, particularly if treatment is maintained for ≄3 years, if baseline LDL-C is ≄100 mg/dL, or if achieved LDL-C is low.     Abstract. The study was conducted during the period 2014 – 2016 in the experimental field of Institute of Plant Genetic Resources, Sadovo. The influence of the period of sowing, the predecessor and the fertilization on the elements of productivity and grain yield per hectare in einkorn wheat were studied. It was established that the structural elements of productivity - number of productive tillers, length of the spike, number of spikelets per spike, mass of central spike, number of grains in central spike, weight of the grains from the central spike are with the highest values in plants grown after predecessor peas, sown in autumn and fertilized with extra nitrogen. With least developed productive capabilities are the plant variants sown in spring after sunflower and without additional fertilization with nitrogen. With proved the biggest impact on grain yield per hectare is the date of sowing. The sources of variation: predecessor and combined effect of factors predecessor x fertilization show equal share of influence on the productivity of einkorn. The results from the study give science-based information on the most favorable time of sowing, fertilizing and predecessor in einkorn

    Abstract. The aim of the current study is to trace the changes in the length of the growing season in accordance with the accumulated effective temperatures, rains and plant nutrition during the vegetation period, as well as the influence of these indices on the grain yield. The study was conducted in the period 2015 – 2016 in the experimental field of Institute of Plant Genetic Resources – Sadovo with the local variety B3E0025 from the National Genebank in Sadovo. The experiment was made in block method in four repetitions. Three sowing times were made – autumn, winter and early spring and two options of N fertilization – without additional N and 3 kg/da input of active N substance in tillĐ”ring phase. The beginning of the basic phenophases has been observed. As a result of the study it was established that the vegetation period in T. monococcum L. has 114-238 days’ duration and accumulation of effective temperature of 2266.0- 2714.71°C. The length of the growing season and the necessary accumulated temperature depends on the sowing period, the predecessor, the amount of nutrient substances and soil moisture. The extension of the critical phases of autumn sowing favours the obtaining of higher yield. The significance of the effective temperatures decreases with additional N input

    Pepper (Capsicum annuum var. annuum L.) is one of the most important and wide spread vegetables in the Balkan region. A great number of local populations with specific characteristics for shape, color, taste, biological value and type of use are grown in different Balkan countries together with the main commercial cultivars. Partially the old native forms are not within attention of the sort maintenance selection, but they are preserved in limited area and are valuable genetic resources. A SEE-ERA. NET project started in 2010 setting the task to investigate pepper as target species. The strategic objectives are: i) to collect, describe and explore the existing Capsicum biodiversity in the partner countries (Albania, Bulgaria, Greece, Macedonia and Serbia) related to valuable traits; ii) to create work and basic collections of local accessions and the respective database for more detailed and profound future collaborative investigations and breeding programs. Methods for description and maintenance of the collected material have been standardized in order to create a uniform description of existing local germplasm of pepper. First joint collecting expeditions have been carried out. Local expedition trips for expanding the national collections of old cultivars and domestic landraces have also been organized. Preparation of work and basic collections in the National Seed Genebank, Sadovo, Bulgaria, has started. The data will be publicly launched in the European Search Catalogue for Genetic Resources, EURISCO (www.eurisco.ecpgr.org). The data generated from this study, knowledge and collections will serve as a solid basis for effective preservation, management and exploitation of Balkan pepper germplasm biodiversity. The discovery of new sources of genetic variation and identification of accessions with valuable traits as pest resistance will be used in future pepper-breeding programs
