    Maize open-pollinated populations physiological improvement: validating tools for drought response participatory selection

    Participatory selection—exploiting specific adaptation traits to target environments—helps to guarantees yield stability in a changing climate, in particular under low-input or organic production. The purpose of the present study was to identify reliable, low-cost, fast and easy-to-use tools to complement traditional selection for an e ective participatory improvement of maize populations for drought resistance/tolerance. The morphological and eco-physiological responses to progressive water deprivation of four maize open-pollinated populations were assessed in both controlled and field conditions. Thermography and Chl a fluorescence, validated by gas exchange indicated that the best performing populations under water-deficit conditions were ‘Fandango’ and to a less extent ‘Pigarro’ (both from participatory breeding). These populations showed high yield potential under optimal and reduced watering. Under moderate water stress, ‘Bilhó’, originating from an altitude of 800 m, is one of the most resilient populations. The experiments under chamber conditions confirmed the existence of genetic variability within ‘Pigarro’ and ‘Fandango’ for drought response relevant for future populations breeding.     An anti-inflammatory and low fermentable oligo, di, and monosaccharides and polyols diet improved patient reported outcomes in fibromyalgia: A randomized controlled trial

    Background: Fibromyalgia (FM) has been associated with dysbiosis and low-grade inflammation. Studies have reported that diet influences clinical features in FM. Objective: To evaluate the effect of an anti-inflammatory and low fermentable oligo, di, and monosaccharides and polyols (FODMAP) diet on clinical outcomes of patients with FM. Methods: This two arms Randomized Controlled Trial (NCT04007705) included 46 female patients with FM. The intervention group (n = 22) adopted an anti-inflammatory diet for 3 months, excluding gluten, dairy, added sugar, and ultra-processed foods, along with a low FODMAPs diet in the first month. The control group (n = 24) followed general healthy eating recommendations. Both diets were applied by a certified dietitian. Before and after the intervention, participants were assessed regarding pain, fatigue, gastrointestinal symptoms, quality of sleep, and quality of life, through the Revised Fibromyalgia Impact Questionnaire (FIQR), Visual Analogue Pain Scale (VAS), Visual Analog Scale from gastrointestinal symptoms (VAS GI), Brief Pain Inventory (BPI), Pittsburg Sleep Quality Index (PSQI), Fatigue Severity Survey (FSS), and The Short Form Health Survey (SF-36). A blood sample was collected and high-sensitive C-Reactive Protein and Erythrocyte Sedimentation Rate were quantified. Paired Samples t-test/Wilcoxon and independent samples t-test/Mann-Whitney were used to compare variables between groups. Results: After intervention, there was an improvement in intervention group scores of FIQR (p = 0.001), VAS (p = 0.002), BPI (p = 0.011), FSS (p = 0.042), VAS_GI (p = 0.002), PSQI (p = 0.048), and SF36 (p = 0.045) compared to control group. Inflammatory biomarkers (hs-CRP, ESR) did not change in both groups. The intervention was beneficial in the intervention group, regardless of age, disease duration, body mass index variation, and body fat change between baseline and post-intervention. Conclusion: An anti-inflammatory and low-FODMAP diet improved clinical features in patients with FM and may be useful as a complement to pharmacological therapy

    Association Between Memory B-Cells and Clinical and Immunological Features of Primary Sjögren’s Syndrome and Sicca Patients

    B-cells play a pivotal role in primary Sjögren's syndrome (pSS) pathogenesis. We aim to (1) evaluate the distribution of B-lymphocyte subpopulations in pSS and Sicca patients, (2) establish cut-off points that discriminate pSS from controls, (3) evaluate the association between memory B-cells and phenotypic features in pSS. We included 57 pSS patients, 68 Sicca and 24 healthy controls. Circulating B-cells were characterized by flow cytometry as naïve and memory subsets and classified from Bm1 to Bm5. Compared to controls, pSS patients had lower percentages (29.5 vs 44.4%) and absolute numbers (47 vs 106 cells/µl) of memory B-cells. Through ROC curves, a cut-off of ≤ 58 total memory B-cells/µl yielded a specificity of 0.88 and a sensitivity of 0.60 for pSS, and was met by 59.6% of pSS patients, 38.8% of Sicca and 12.5% of controls. A cut-off of < 23.5 Switched-memory B-cells/µl yielded a specificity of 0.88 and a sensitivity of 0.54 and was met by 54.4% of pSS patients, 37.3% of Sicca and 12.5% of controls. In pSS, lower total memory B-cells count was associated with longer disease duration (14.3 vs 8.1 years, p = 0.006) and more active disease profile, as evaluated by the European League Against Rheumatism (EULAR) Sjögren's Syndrome Disease Activity Index (ESSDAI) (3.1 vs 1.4, p = 0.043). Decreased numbers of memory B-cells clearly discriminated pSS from controls and can also have prognostic value. It remains to be clarified whether Sicca patients with decreased memory B-cells represent pSS and if B-cell profiling could help in the diagnosis of pSS.info:eu-repo/semantics/publishedVersio

    Climatic change emphasize the importance of biodiversity maintenance, Suggesting that germplasm adapted to organic, low input, or conventional conditions is needed to face future demands. This Study presents: I - The two steps genesis of the synthetic maize population Fandango, A) NUTICA creation: in 1975, Miguel Mota and Silas Pego, initiated a new type of polycross method involving 77 yellow elite inbred lines (dent and flint; 20% Portuguese and 80% North American germplasm) from the NUMI programme (NUcleo de melhoramento de Milho, Braga, Portugal). These inbreds were intermated in natural isolation and progenies submitted to intensive selection for both parents during continued cycles; B) From NUTICA to Fandango: Tandango was composed of all the crosses that resulted from a North Carolina Design I matting design (1 male crossed with 5 females) applied to NUTICA. II - The diversity evolution of Fandango under a Participatory Breeding project at the Portuguese Sousa Valley region (VASO) initiated in 1985 by Pego, with CIMMYT support. Morphological, fasciation expression, and yield trials were conducted in Portugal (3 locations, 3 years) and in the USA (4 locations, I year) using seeds obtained from five to seven cycles of mass selection (MS). The selection across cycles wits clone by the breeder (until cycle 5) and farmer (before cycle II in present). ANOVA and regression analysis on the rate of direct response to selection were performed when the assumption of normality was positively confirmed. Otherwise the non parametric Multivariate Adaptive Regression Splines (MARS) was performed. Response to mass selection in lowa showed significant decrease in yield, while in Portugal a significant increase for time of silking, plant and ear height, ear diameters 2, 37 4, kernel number, cot) diameters, and rachis was observed. At this location also a significant decrease was observed for thousand kernel weight and ear length. These results showed that mass selection were not effective for significant yield increase, except when considered Lousada with breeder selection (3.09% of gain per cycle per year). Some non-para metric methods (MARS, decision trees and random forests) were used to get insights on the causes that explain yield in Fandango. Kernel weight and ear weight were the most important traits, although row numbers, number of kernels per row, ear length, and ear diameter were also of some importance influencing Fandango yield

    Added value of lymphocyte subpopulations in the classification of Sjögren's syndrome

    Sjögren's Syndrome (SjS) is a chronic systemic immune-mediated inflammatory disease characterized by lymphocytic infiltration and consequent lesion of exocrine glands. SjS diagnosis and classification remains a challenge, especially at SjS onset, when patients may have milder phenotypes of the disease or uncommon presentations. New biomarkers are needed for the classification of SjS, thus, we aimed to evaluate the added-value of lymphocyte subpopulations in discriminating SjS and non-Sjögren Sicca patients. Lymphocyte subsets from 62 SjS and 63 Sicca patients were characterized by flow cytometry. The 2002 AECG and the 2016 ACR/EULAR SjS classification criteria were compared with clinical diagnosis. The added discriminative ability of joining lymphocytic populations to classification criteria was assessed by the area under the Receiver-Operating-Characteristic Curve (AUC). Considering clinical diagnosis as the gold-standard, we obtained an AUC = 0.952 (95% CI: 0.916-0.989) for AECG and an AUC = 0.921 (95% CI: 0.875-0.966) for ACR/EULAR criteria. Adding Tfh and Bm1 subsets to AECG criteria, performance increased, attaining an AUC = 0.985 (95% CI: 0.968-1.000) (p = 0.021). Th1/Breg-like CD24hiCD27+ and switched-memory B-cells maximized the AUC of ACR/EULAR criteria to 0.953 (95% CI: 0.916-0.990) (p = 0.043). Our exploratory study supports the potential use of lymphocyte subpopulations, such as unswitched memory B cells, to improve the performance of classification criteria, since their discriminative ability increases when specific subsets are added to the criteria.info:eu-repo/semantics/publishedVersio

    Reborn from Ashes: The Alvares Project

    The North and Centre of Portugal encompass many productive forest areas that are particularly prone to fires. In the year of 2017, the most destructive fire season in the history of the country devastated extensive areas of land. Somewhere in the middle of this burnt region, the parish of Alvares stands as a representative case of the entire territory: consecutive decades of rural exodus dramatically changed the economic structure in which its population used to subsist. Following this path of abandonment, the exploitation of forest resources also changed noticeably, leading to a new fire regime that is hampering the necessary reforms to sustain proper forest management. The high frequency of devastating fires causes many social and economic damages as well as substantial environmental deterioration and has become incompatible with profitable investments for the typical small landowner. After the fire of 2017, a group of researchers joined the stakeholders within the forestry industry in Alvares to build an ambitious plan to modify the fire regime in an economically sustainable way. In order to do so, their forest planning proposal included the implementation of various barriers to sustain the propagation of fire, together with a diversification of the use of land and of the forest mix. However, their success crucially depends on the economic feasibility of the project for the small landowners and, therefore, new business ideas have to be put into practice to revitalize the region.     Warm Season Grain Legume Landraces From the South of Europe for Germplasm Conservation and Genetic Improvement

    Currently, there is a high concern from consumers regarding food quality, with emphasis on the origin of food sources. We here review the current situation of beans (Phaseolus spp.) and cowpea (Vigna unguiculata (L.) Walp.) landraces in the South of Europe focusing on morpho-agronomic and genetic diversity and physiological adaptation to the different agrosystems, including the symbiotic association with rhizobia. Despite the reduction in the production and consumption of grain legumes in Southern Europe, the variability of common bean, runner bean and cowpea landraces in this region is adequately preserved ex situ in germplasm banks and in breeder collections in Portugal, Spain, Italy and Greece; however, on-farm (in situ) conservation in isolated areas mainly in gardens and small fields for farmers own consumption and local markets is not guaranteed. This variability can be used for the genetic improvement of varieties, which will result in environmental-friendly improved legumes for a sustainable production in the South of Europe as well as in other regions of the World
