    58 pages, 5 figures, 3 tables- The World Economic Forum now ranks biodiversity loss as a top-five risk to the global economy, and the draft post-2020 Global Biodiversity Framework proposes an expansion of conservation areas to 30% of the earth’s surface by 2030 (hereafter the “30% target”), using protected areas (PAs) and other effective area-based conservation measures (OECMs). - Two immediate concerns are how much a 30% target might cost and whether it will cause economic losses to the agriculture, forestry and fisheries sectors. - Conservation areas also generate economic benefits (e.g. revenue from nature tourism and ecosystem services), making PAs/Nature an economic sector in their own right. - If some economic sectors benefit but others experience a loss, high-level policy makers need to know the net impact on the wider economy, as well as on individual sectors.     New insights into the genetic etiology of Alzheimer's disease and related dementias

    Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele

    Multiancestry analysis of the HLA locus in Alzheimer’s and Parkinson’s diseases uncovers a shared adaptive immune response mediated by HLA-DRB1*04 subtypes

    Across multiancestry groups, we analyzed Human Leukocyte Antigen (HLA) associations in over 176,000 individuals with Parkinson’s disease (PD) and Alzheimer’s disease (AD) versus controls. We demonstrate that the two diseases share the same protective association at the HLA locus. HLA-specific fine-mapping showed that hierarchical protective effects of HLA-DRB1*04 subtypes best accounted for the association, strongest with HLA-DRB1*04:04 and HLA-DRB1*04:07, and intermediary with HLA-DRB1*04:01 and HLA-DRB1*04:03. The same signal was associated with decreased neurofibrillary tangles in postmortem brains and was associated with reduced tau levels in cerebrospinal fluid and to a lower extent with increased Aβ42. Protective HLA-DRB1*04 subtypes strongly bound the aggregation-prone tau PHF6 sequence, however only when acetylated at a lysine (K311), a common posttranslational modification central to tau aggregation. An HLA-DRB1*04-mediated adaptive immune response decreases PD and AD risks, potentially by acting against tau, offering the possibility of therapeutic avenues

    Für Patienten, die an chronisch entzündlichen Darmerkrankungen (CED) leiden, gibt es zwar bereits ein Spektrum an Behandlungsmöglichkeiten. Allerdings ist es unabdingbar spezifischere, schneller wirksamere und verträglichere Medikationen zu finden. Morbus Crohn und Colitis ulcerosa Patienten, bei denen bisher keine Therapie zur Remissionserhaltung geführt hat, bekommen somit eine Chance, eine spezifischere Behandlung zu erhalten, die den gewünschten Erfolg erzielen könnte. Die anfänglichen Therapieformen, vor allem die Behandlung mit small molecule drugs wie 5-ASAs, weichen nun immer mehr der Behandlung mit sogenannten Biologika wie Antikörpern oder Antisense-Oligonukleotiden, da solche spezifischer und schneller wirken. Da Morbus Crohn und Colitis ulcerosa Patienten ein stark erhöhtes RNA- und Protein-Level der T-Helferzell-Transkriptionsfaktoren T-bet und GATA-3 aufweisen, sind diese Faktoren potentielle Ziele einer Biologika-vermittelten Therapie. Deshalb soll die Funktion von GATA-3 und T-bet bei der Initiierung und Aufrechterhaltung experimenteller Colitis untersucht werden, um anschließend DNAzym-basierte Therapien, die gegen diese Faktoren gerichtet sind, in diesen Modellen zu analysieren. Diese Analyse soll die Anwendung GATA-3 und T-bet spezifischer Medikamente für die Klinik ermöglichen und verbessern.There is already a range of treatment options available for patients with chronic inflammatory bowel disease (IBD). However, it is essential to find more specific, faster effective and tolerable medications. For Crohn's Disease (CD) and Ulcerative Colitis (UC) Patients, who have not received a therapy, that maintains remission, would this be a chance to receive an innovative treatment that could achieve the desired outcome. The initial forms of therapy, especially the treatment with small molecule drugs such as 5-ASAs, are more and more replaced by so-called biologicals such as antibodies or antisense oligonucleotides, as these operate more specific and faster. Because Crohn's Disease and Ulcerative Colitis patients have greatly increased RNA and protein levels of the T helper cell transcription factors T-bet and GATA-3, these factors are potential targets of a biological-mediated therapy. Therefore, the role of GATA-3 and T-bet in the initiation and maintenance of experimental colitis will be investigated in order to subsequent analysis of DNAzyme-based therapies directed against these factors in the colitis models.
