    Near-surface common-midpoint seismic data recorded with automatically planted geophones

    This is the publisher's version, also available electronically from "http://onlinelibrary.wiley.com".[1] We introduce the Autojuggie II as a device to speed the emplacement of geophones for near-surface seismic common-midpoint (CMP) surveys. Hydraulic cylinders force rigidly interconnected geophones into the ground simultaneously and automatically. We demonstrate that accurate CMP data can be recorded with geophones planted by this device, and that a CMP stacked section can be processed, from which reliable geologic information can be extracted. To make this demonstration, we compare the stacked section to a coincident and parallel section, whose data was acquired using conventionally hand-planted geophones. The two sections are very similar in amplitude, phase, and frequency. A slight difference in coherency exists in a ∼35-ms reflection; the stack corresponding to the automatically planted geophones shows better coherency relative to the comparison stack. However, the similarity of the sections indicates that accurate CMP data can be recorded using geophones planted by the Autojuggie II

    Cholesterol feeding strongly reduces hepatic VLDL-triglyceride production in mice lacking the liver X receptor alpha

    The oxysterol-activated nuclear receptor liver X receptor alpha (LXR alpha) has been implicated in the control of both cholesterol and fatty acid metabolism. In this study, we have evaluated the effects of excess dietary cholesterol on hepatic cholesterol metabolism, lipogenesis, and VLDL production in homozygous (Lxr alpha(-/-)), heterozygous (Lxr alpha(+/-)), and wild-type mice. Mice were fed either chow or a cholesterol-enriched diet (1%, w/w) for 2 weeks. On the high-cholesterol diet, fractional cholesterol absorption was higher in Lxr alpha(-/-) mice than in controls, leading to delivery of more dietary cholesterol to the liver. Lxr alpha(-/-) mice were not able to induce expression of hepatic Abcg5/Abcg8, and massive accumulation of free cholesterol and cholesteryl esters (CEs) occurred. Interestingly, despite the inability to upregulate Abcg5/Abcg8, the highly increased hepatic free cholesterol content did stimulate biliary cholesterol output in Lxr alpha(-/-) mice. Hepatic cholesterol accumulation was accompanied by decreased hepatic expression of lipogenic genes, probably caused by impaired sterol-regulatory element binding protein 1c processing, lower hepatic triglyceride (TG) contents, strongly reduced plasma TG concentrations (290%), and reduced VLDL-TG production rates (-60%) in Lxr alpha(-/-) mice. VLDL particles were smaller and CE-enriched under these conditions. Lxr alpha deficiency did not affect VLDL formation under chow-fed conditions. Hepatic stearyl coenzyme A desaturase 1 expression was decreased dramatically in Lxr alpha(-/-) mice and did not respond to cholesterol feeding, but fatty acid profiles of liver and VLDL were only slightly different between Lxr alpha(-/-) and wild-type mice.     Intermediate-depth icequakes and harmonic tremor in an Alpine glacier (glacier d 'Argentiere,France): evidence for hydraulic fracturing

    International audienceWe detected several thousand deep englacial icequakes on Glacier d’Argentiere (Mont-Blanc Massif) between 30 March and 3 May 2012. These events have been classified in eight clusters. Inside each cluster, the waveforms are similar for P-waves and S-waves, although the time delay between the P-waves and S-waves vary by up to 0.03 s, indicating an extended source area. Although these events were recorded by a single accelerometer, they were roughly located using a polarization analysis. The deepest events were located at a depth of 130 m, 60 m above the ice/bed interface. The clusters are separated in space. The largest cluster extends over about 100 m. For this cluster, the strike of the rupture plane is nearly parallel to the direction of the open crevasses, and the dip angle is 56◦. Deep icequakes occur in bursts of activity that last for a few hours and are separated by quiet periods. Many events occurred on 28-29 April 2012, during the warmest days, when snow melting was likely important. The distribution of events in time and space obeys a power law, as also observed for earthquakes, but with larger exponents.The polarity of the P-waves for all of the events is consistent with tensile faulting. Finally, between 25 April and 3 May, we observed a gliding harmonic tremor with a fun- damental resonance frequency that varied between 30 Hz and 38 Hz, with additional higher frequency harmonics. During this time we also observed shallow hybrid events with high-frequency onsets and a monochromatic coda. These events might be produced by the propagation of fractures and the subsequent flow of water into the fracture. The strongest resonance was observed just after a strong burst of deep icequakes, and during an un- usually warm period when the snow height decreased by 60 cm in one week. The res- onance frequency shows a succession of several sharp decreases and phases of progressive increases. One of the strongest negative steps of the resonance frequency on 28 April coincides with a burst of deep icequakes. These events appear to be associated with the propagation of fractures, which can explain the decrease in the resonance frequency. Finally, we observed an acceleration of glacier flow on 29 April and suggested that melt-water had reached the ice/bed interface. These observations suggest that deep icequakes are due to hydraulic fracturing, and that they can be used to track fluid flow inside glaciers

    Impact of Formulation Conditions on Lipid Nanoparticle Characteristics and Functional Delivery of CRISPR RNP for Gene Knock-Out and Correction

    The CRISPR-Cas9 system is an emerging therapeutic tool with the potential to correct diverse genetic disorders. However, for gene therapy applications, an efficient delivery vehicle is required, capable of delivering the CRISPR-Cas9 components into the cytosol of the intended target cell population. In this study, we optimized the formulation conditions of lipid nanoparticles (LNP) for delivery of ready-made CRISPR-Cas9 ribonucleic protein (RNP). The buffer composition during complexation and relative DOTAP concentrations were varied for LNP encapsulating in-house produced Cas9 RNP alone or Cas9 RNP with additional template DNA for gene correction. The LNP were characterized for size, surface charge, and plasma interaction through asymmetric flow field flow fractionation (AF4). Particles were functionally screened on fluorescent reporter cell lines for gene knock-out and gene correction. This revealed incompatibility of RNP with citrate buffer and PBS. We demonstrated that LNP for gene knock-out did not necessarily require DOTAP, while LNP for gene correction were only active with a low concentration of DOTAP. The AF4 studies additionally revealed that LNP interact with plasma, however, remain stable, whereby HDR template seems to favor stability of LNP. Under optimal formulation conditions, we achieved gene knock-out and gene correction efficiencies as high as 80% and 20%, respectively, at nanomolar concentrations of the CRISPR-Cas9 RNP

    Boron Intake and decreased risk of mortality in kidney transplant recipients

    PURPOSE: In a search for potentially modifiable factors to improve long-term outcome among kidney transplant recipients (KTR), we hypothesized that boron exposure is associated with improved long-term outcome in KTR. METHODS: We determined 24 h urinary boron excretion using inductively coupled plasma mass spectrometry as a measure of boron exposure in 693 stable KTR (57% male, mean age 53y), enrolled in the TransplantLines Food and Nutrition Biobank and Cohort Study. Dietary intake was assessed using validated food-frequency questionnaires. RESULTS: Linear regression analyses showed that dietary intake of fruit, wine and nuts were key determinants of boron excretion. In addition, boron excretion was negatively correlated with homocysteine and inflammatory parameters. In total, 73 (32%), 47 (20%) and 30 (13%) KTR died among the lowest, middle and highest tertiles of 24 h urinary boron excretion, respectively (P(log-rank) < 0.001). Cox regression analyses showed that high boron excretion was strongly associated with lower risk of mortality, independent of age, sex, estimated glomerular filtration rate and history of cardiovascular disease (HR per doubling: 0.51, 95% CI: 0.40 to 0.66, P < 0.001). CONCLUSION: Boron may be an overlooked target to improve long-term survival among KTR and potentially other patients, likely through pathways other than inflammation or the methionine-homocysteine cycle that were previously suggested. Interventional trials are warranted to confirm the potential of dietary boron supplementation in KTR and other patient populations.     Ethnic differences in mental health among incarcerated youths: do Moroccan immigrant boys show less psychopathology than native Dutch boys?

    Psychiatric disorders are highly prevalent among incarcerated youth. However, whereas ethnic minority youths are overrepresented in the juvenile justice system, limited research is available on their mental health. In this study, differences in mental health problems between incarcerated adolescents of native Dutch and Moroccan origin, were examined. Child Behavior Checklist and Youth Self-Report scores were compared between incarcerated adolescents of native Dutch and Moroccan origin. Their scores were also compared to those of native Dutch and Moroccan immigrant youths in the general, non-incarcerated population. Native Dutch incarcerated adolescents showed higher levels of various mental health problems than incarcerated adolescents with a Moroccan background. Compared to the general population, incarcerated youths showed higher levels of mental health problems, but this deviation was much larger for native Dutch than for Moroccan immigrant youths. These ethnic differences in mental health problems could not be explained by ethnic differences in socio-economic background and social desirable answering tendencies. Incarcerated youths of Moroccan origin show less psychopathology than incarcerated native Dutch youths, which might be explained by disparities in sentencing procedures
