    A space-time discontinuous Galerkin finite-element discretization of the Euler equations using entropy variables

    A method to numerically solve the Euler equations for fluids with general equations of state is presented. It is based on a formulation solving the conservation equations for either pressure primitive variables or entropy variables, instead of the commonly used conservation variables. We use a space-time discontinuous Galerkin finite-element discretization, which yields a highly local, potentially higher-order scheme. The algorithm is applied to test cases for compressible fluids to demonstrate its capabilities and the performance of the different variable sets

    MicroRNAs dysregulated in multiple sclerosis affect the differentiation of CG-4 cells, an oligodendrocyte progenitor cell line

    IntroductionDemyelination is one of the hallmarks of multiple sclerosis (MS). While remyelination occurs during the disease, it is incomplete from the start and strongly decreases with its progression, mainly due to the harm to oligodendrocyte progenitor cells (OPCs), causing irreversible neurological deficits and contributing to neurodegeneration. Therapeutic strategies promoting remyelination are still very preliminary and lacking within the current treatment panel for MS.MethodsIn a previous study, we identified 21 microRNAs dysregulated mostly in the CSF of relapsing and/or remitting MS patients. In this study we transfected the mimics/inhibitors of several of these microRNAs separately in an OPC cell line, called CG-4. We aimed (1) to phenotypically characterize their effect on OPC differentiation and (2) to identify corroborating potential mRNA targets via immunocytochemistry, RT-qPCR analysis, RNA sequencing, and Gene Ontology enrichment analysis.ResultsWe observed that the majority of 13 transfected microRNA mimics decreased the differentiation of CG-4 cells. We demonstrate, by RNA sequencing and independent RT-qPCR analyses, that miR-33-3p, miR-34c-5p, and miR-124-5p arrest OPC differentiation at a late progenitor stage and miR-145-5p at a premyelinating stage as evidenced by the downregulation of premyelinating oligodendrocyte (OL) [Tcf7l2, Cnp (except for miR-145-5p)] and mature OL (Plp1, Mbp, and Mobp) markers, whereas only miR-214-3p promotes OPC differentiation. We further propose a comprehensive exploration of their change in cell fate through Gene Ontology enrichment analysis. We finally confirm by RT-qPCR analyses the downregulation of several predicted mRNA targets for each microRNA that possibly support their effect on OPC differentiation by very distinctive mechanisms, of which some are still unexplored in OPC/OL physiology.ConclusionmiR-33-3p, miR-34c-5p, and miR-124-5p arrest OPC differentiation at a late progenitor stage and miR-145-5p at a premyelinating stage, whereas miR-214-3p promotes the differentiation of CG-4 cells. We propose several potential mRNA targets and hypothetical mechanisms by which each microRNA exerts its effect. We hereby open new perspectives in the research on OPC differentiation and the pathophysiology of demyelination/remyelination, and possibly even in the search for new remyelinating therapeutic strategies in the scope of MS

    Extensive Spinal Cord Injury following Staphylococcus aureus Septicemia and Meningitis

    Bacterial meningitis is rarely complicated by spinal cord involvement in adults. We report a case of Staphylococcus aureus septicemia complicated by meningitis and extensive spinal cord injury, leading to ascending brain stem necrosis and death. This complication was investigated by magnetic resonance imaging which demonstrated intramedullary hyperintensity on T2-weighted images and by multimodality evoked potentials. Postmortem microscopic examination confirmed that the extensive spinal cord injury was of ischemic origin, caused by diffuse leptomeningitis and endarteritis

    Reversible Cortical Blindness Following Successful Surgical Repair Of Two Stab Wounds In The Heart

    This report describes a case of cortical blindness that followed successful surgical repair of two stab wounds in the heart in a 29-year old Libyan man. The patient presented in a state of pre cardiac arrest (shock and low cardiac output status), following multiple chest stab wounds. Chest tube was immediately inserted. Surgery was urgently performed suturing the two wounds; in the root of the aorta and in the left ventricle, and haemostasis was secured. Cardiac arrest was successfully prevented. The patient recovered smoothly, but 24 hours later he declared total blindness. Ophtalmic and neurological examinations and investigations that included fundoscopy, Electroencephalograms (EEGs) and Computed Tomography Scans revealed no abnormalities, apart from absence of alpha waves in the EEGs. We diagnosed the case as cortical blindness and continued caring for the patient conservatively. Three days later, the patient regained his vision gradually and was discharged on the 7th postoperative day without any remarks

    A regional benthic fauna assessment method for the Southern North Sea using Margalef diversity and reference value modelling

    The aims of this study are to develop an optimized method for regional benthic fauna assessment of the Southern North Sea which (a) is sensitive and precise (quantified as the slope and the R2 value of the pressure-impact relationships, respectively) for the anthropogenic pressures bottom fishing and organic enrichment, (b) is suitable for estimating and modelling reference values, (c) is transparent, (d) can be efficiently applied using dedicated software; and to apply this method to benthic data from the Southern North Sea. Margalef diversity appeared to be the best performing benthic index regarding these criteria, even better than several Multi-Metric Indices (MMIs) containing e.g. AMBI (AZTI Marine Biotic Index) and ITI (Infaunal Trophic Index). Therefore, this relatively simple and very practical index, including a new reference value estimation and modelling method, and BENMMI software were selected as a common OSPAR (Oslo Paris convention) method for the benthic fauna assessment of the Southern North Sea. This method was applied to benthic fauna data from the Southern North Sea collected during the period 2010–2015. The results in general show lower normalized Margalef values in coastal areas, and higher normalized Margalef values in deeper offshore areas. The following benthic indices were compared in this study: species richness, Margalef diversity, SNA index, Shannon index, PIE index, AMBI, ITI. For each assessment area, the least disturbed benthic dataset was selected as an adjacent 6 year period with, on average, the highest Margalef diversity values. For these datasets, the reference values were primarily set as the 99th percentile values of the respective indices. This procedure results in the highest stable reference values that are not outliers. In addition, a variable percentile method was developed, in which the percentile value is adjusted to the average bottom fishing pressure (according to data from the International Council for the Exploration of the Sea, ICES) in the period 2009–2013. The adjusted percentile values were set by expert judgement, at 75th (low fishing pressure), 95th (medium fishing pressure) and 99th (high fishing pressure) percentile. The estimated reference values for Margalef diversity correlate quite well with the median depth of the assessment areas using a sigmoid model (pseudo-R2 = 0.86). This relationship between depth and Margalef diversity was used to estimate reference values in case an assessment area had insufficient benthic data .For testing the effects of bottom fishing pressure, normalized index values (NIV; index value divided by reference value) were used. The rationale for using NIVs is the assumption that, although a certain level of bottom fishing pressure will have a larger absolute effect on more biodiverse benthic communities in deeper waters than on more robust and less biodiverse coastal benthic communities, the relative effects (tested using NIVs) are comparable. A clear exponentially decreasing relationship (R2 = 0.26–0.27, p 2 cm, respectively) and normalized Margalef diversity values, with an asymptotic normalized Margalef value of 0.45 at a subsurface fishing activity >2.3 sweeps/year. This asymptotic value is predominantly found in coastal waters, and probably shows that the naturally more robust coastal benthic communities have been transformed into resilient benthic communities, which rapidly recover from increasing fishing pressure

    We present the description and the first results of a new project devoted to the search for extremely metal-deficient blue compact/HII-galaxies (BCGs) and to the creation of a well selected large BCG sample with strong emission lines. Such galaxies should be suitable for reliable determination of their oxygen abundance through the measurement of the faint [OIII]4363A line. The goals of the project are two-fold: a) to discover a significant number of new extremely metal-poor galaxies (Z <= 1/20 Zo), and b) to study the metallicity distribution of local BCGs. Selection of candidates for follow-up slit spectroscopy is performed on the database of objective prism spectra of the Hamburg Quasar Survey. The sky region is limited by delta >= 0 deg. and b^ii <= -30 deg. In this paper we present the results of the follow-up spectroscopy conducted with the Russian 6m telescope. The list of observed candidates contained 52 objects, of which 46 were confirmed as strong-lined BCGs (EW([OIII]5007) >= 100 A). The remaining five lower excitation ELGs include three BCGs, and two galaxies classified as SBN (Starburst Nucleus) and DANS (Dwarf Amorphous Nucleus Starburst). One object is identified as a quasar with a strong Ly_alpha emission line near 5000 A (z~3). We provide a list with coordinates, measured radial velocities, B-magnitudes, equivalent widths EW([OIII]5007) and EW(H_beta) and for the 46 strong-lined BCGs the derived oxygen abundances 12+log(O/H). The abundances range between 7.42 and 8.4 (corresponding to metallicities between 1/30 and 1/3 Zo).     A framework for a European network for a systematic environmental impact assessment of genetically modified organisms (GMO)

    The assessment of the impacts of growing genetically modified (GM) crops remains a major political and scientific challenge in Europe. Concerns have been raised by the evidence of adverse and unexpected environmental effects and differing opinions on the outcomes of environmental risk assessments (ERA). The current regulatory system is hampered by insufficiently developed methods for GM crop safety testing and introduction studies. Improvement to the regulatory system needs to address the lack of well designed GM crop monitoring frameworks, professional and financial conflicts of interest within the ERA research and testing community, weaknesses in consideration of stakeholder interests and specific regional conditions, and the lack of comprehensive assessments that address the environmental and socio economic risk assessment interface. To address these challenges, we propose a European Network for systematic GMO impact assessment (ENSyGMO) with the aim directly to enhance ERA and post-market environmental monitoring (PMEM) of GM crops, to harmonize and ultimately secure the long-term socio-political impact of the ERA process and the PMEM in the EU. These goals would be achieved with a multi-dimensional and multi-sector approach to GM crop impact assessment, targeting the variability and complexity of the EU agro-environment and the relationship with relevant socio-economic factors. Specifically, we propose to develop and apply methodologies for both indicator and field site selection for GM crop ERA and PMEM, embedded in an EU-wide typology of agro-environments. These methodologies should be applied in a pan-European field testing network using GM crops. The design of the field experiments and the sampling methodology at these field sites should follow specific hypotheses on GM crop effects and use state-of-the art sampling, statistics and modelling approaches. To address public concerns and create confidence in the ENSyGMO results, actors with relevant specialist knowledge from various sectors should be involved

    Multi-wavelength polarimetric study towards the open cluster NGC 1893

    We present multi-wavelength linear polarimetric observations for 44 stars of the NGC 1893 young open cluster region along with V-band polarimetric observations of stars of other four open clusters located between l ~160 to ~175 degree. We found evidence for the presence of two dust layers located at a distance of ~170 pc and ~360 pc. The dust layers produce a polarization Pv ~2.2%. It is evident from the clusters studied in the present work that, in the Galactic longitude range l ~160 to 175 degree and within the Galactic plane (|b| < 2 degree), the polarization angles remain almost constant, with a mean ~163 degree and a dispersion of 6 degree. The small dispersion in polarization angle could be due to the presence of uniform dust layer beyond 1 kpc. Present observations reveal that in case of NGC 1893, the foreground two dust layers, in addition to the intracluster medium, seems to be responsible for the polarization effects. It is also found that towards the direction of NGC 1893, the dust layer that exists between 2-3 kpc has a negligible contribution towards the total observed polarization. The weighted mean for percentage of polarization (Pmax) and the wavelength at maximum polarization ({\lambda}max) are found to be 2.59 \pm 0.02% and 0.55 \pm 0.01 \mum respectively. The estimated mean value of {\lambda}max indicates that the average size of the dust grains within the cluster is similar to that in the general interstellar medium. The spatial variation of the polarization is found to decrease towards the outer region of the cluster. In the present work, we support the notion, as already has been shown in previous studies, that polarimetry, in combination with (U-B)/(B-V) colour-colour diagram, is a useful tool for identifying non-members in a cluster.Comment: 36 pages, 12 figures, 10 tables, accepted for the publication in MNRA

    Three-dimensional pattern formation, multiple homogeneous soft modes, and nonlinear dielectric electroconvection

    Patterns forming spontaneously in extended, three-dimensional, dissipative systems are likely to excite several homogeneous soft modes (\approx hydrodynamic modes) of the underlying physical system, much more than quasi one- and two-dimensional patterns are. The reason is the lack of damping boundaries. This paper compares two analytic techniques to derive the patten dynamics from hydrodynamics, which are usually equivalent but lead to different results when applied to multiple homogeneous soft modes. Dielectric electroconvection in nematic liquid crystals is introduced as a model for three-dimensional pattern formation. The 3D pattern dynamics including soft modes are derived. For slabs of large but finite thickness the description is reduced further to a two-dimensional one. It is argued that the range of validity of 2D descriptions is limited to a very small region above threshold. The transition from 2D to 3D pattern dynamics is discussed. Experimentally testable predictions for the stable range of ideal patterns and the electric Nusselt numbers are made. For most results analytic approximations in terms of material parameters are given.Comment: 29 pages, 2 figure

    Exposure to Inhalable, Respirable, and Ultrafine Particles in Welding Fume

    This investigation aims to explore determinants of exposure to particle size-specific welding fume. Area sampling of ultrafine particles (UFP) was performed at 33 worksites in parallel with the collection of respirable particles. Personal sampling of respirable and inhalable particles was carried out in the breathing zone of 241 welders. Median mass concentrations were 2.48 mg m−3 for inhalable and 1.29 mg m−3 for respirable particles when excluding 26 users of powered air-purifying respirators (PAPRs). Mass concentrations were highest when flux-cored arc welding (FCAW) with gas was applied (median of inhalable particles: 11.6 mg m−3). Measurements of particles were frequently below the limit of detection (LOD), especially inside PAPRs or during tungsten inert gas welding (TIG). However, TIG generated a high number of small particles, including UFP. We imputed measurements <LOD from the regression equation with manganese to estimate determinants of the exposure to welding fume. Concentrations were mainly predicted by the welding process and were significantly higher when local exhaust ventilation (LEV) was inefficient or when welding was performed in confined spaces. Substitution of high-emission techniques like FCAW, efficient LEV, and using PAPRs where applicable can reduce exposure to welding fume. However, harmonizing the different exposure metrics for UFP (as particle counts) and for the respirable or inhalable fraction of the welding fume (expressed as their mass) remains challenging
