    Distinct mechanisms mediate X chromosome dosage compensation in Anopheles and Drosophila

    Sex chromosomes induce potentially deleterious gene expression imbalances that are frequently corrected by dosage compensation (DC). Three distinct molecular strategies to achieve DC have been previously described in nematodes, fruit flies, and mammals. Is this a consequence of distinct genomes, functional or ecological constraints, or random initial commitment to an evolutionary trajectory? Here, we study DC in the malaria mosquito Anopheles gambiae. The Anopheles and Drosophila X chromosomes evolved independently but share a high degree of homology. We find that Anopheles achieves DC by a mechanism distinct from the Drosophila MSL complex–histone H4 lysine 16 acetylation pathway. CRISPR knockout of Anopheles msl-2 leads to embryonic lethality in both sexes. Transcriptome analyses indicate that this phenotype is not a consequence of defective X chromosome DC. By immunofluorescence and ChIP, H4K16ac does not preferentially enrich on the male X. Instead, the mosquito MSL pathway regulates conserved developmental genes. We conclude that a novel mechanism confers X chromosome up-regulation in Anopheles. Our findings highlight the pluralism of gene-dosage buffering mechanisms even under similar genomic and functional constraints

    Free flap head and neck reconstruction in the elderly: What is the impact on quality of life?

    Morphofunctional reconstruction is a pivotal aspect in the surgery of head and neck neoplasms: Nowadays, microvascular free flap surgery represents the gold standard. In choosing the surgical technique, the effects on residual quality of life, especially in elderly people, usually considered more fragile and so often excluded from microsurgical procedures, must be taken into account. This multicentre study evaluated the quality of life index in patients more than 75 years of age and who underwent to head and neck microsurgical reconstruction. Data from patients aged > 75 years at the time of major head and neck reconstruction conducted with free flaps between 1 January 2005 and 30 June 2015 were analysed retrospectively. We administered the Italian version of Quality of Life questionnaire SF-36, at least 24 months after surgery. Results were compared to those for the general Italian population of the same age. We enrolled 39 patients with an average age of 80.6 years. The results did not differ significantly from the reference population. The international literature has already shown that chronologic age is not a valid parameter to determine the surgical treatment modality. Even considering the quality of residual life, our study supports the indication for free-flap reconstruction of head and neck defects in the elderly, confirming its effectiveness in this population

    Patterns of Brain Structural Changes in First-Contact, Antipsychotic Drug-NaĂŻve Patients with Schizophrenia

    BACKGROUND AND PURPOSE: Previous studies have suggested that structural changes do occur in the brain of patients with schizophrenia compared with healthy control participants. However, findings from such studies are inconclusive, probably because of the different methodologic approaches, the clinical heterogeneity of patient samples, and also the fact that patients enrolled were treated with antipsychotic drugs. The aim of this study was to investigate brain GM volumes and intrinsic structural WM changes in first-contact, antipsychotic drug-naive patients with schizophrenia. MATERIALS AND METHODS: A total of 43 first-contact, drug-naive, patients with schizophrenia and 17 age-matched control participants were studied. All participants underwent T1-weighted MR imaging and DTI scans. Voxel-based morphometry and tract-based spatial statistics were used to compare GM volumes and WM DTI metrics between groups. MR imaging measures were correlated with the duration of the untreated psychosis and the clinical positive and negative symptoms. RESULTS: Compared with control participants, patients with schizophrenia showed smaller volumes of the temporal, parietal, and occipital GM, and a pattern of decreased mean diffusivity and increased fractional anisotropy in the brain stem and cerebellum bilaterally, interhemispheric and cortico-cortical connections bilaterally, and right anterior and posterior limb of the internal capsule. In patients, decreased mean diffusivity and increased fractional anisotropy in several brain regions were related to a longer duration of the untreated psychosis and the severity of positive symptoms. CONCLUSIONS: First-contact, drug-naive, patients with schizophrenia present with volumetric and DTI changes, which correlated with their clinical features. This study increases our knowledge on the neural networks involved in the pathophysiologic mechanisms of schizophrenia

    Physical Investigation of the Potentially Hazardous Asteroid (144898) 2004 VD17

    In this paper we present the observational campaign carried out at ESO NTT and VLT in April and May 2006 to investigate the nature and the structure of the Near Earth Object (144898) 2004 VD17. In spite of a great quantity of dynamical information, according to which it will have a close approach with the Earth in the next century, the physical properties of this asteroid are largely unknown. We performed visible and near--infrared photometry and spectroscopy, as well as polarimetric observations. Polarimetric and spectroscopic data allowed us to classify 2004 VD17 as an E-type asteroid. A good agreement was also found with the spectrum of the aubrite meteorite Mayo Belwa. On the basis of the polarimetric albedo (p_v=0.45) and of photometric data, we estimated a diameter of about 320 m and a rotational period of about 2 hours. The analysis of the results obtained by our complete survey have shown that (144898) 2004 VD17 is a peculiar NEO, since it is close to the breakup limits for fast rotator asteroids, as defined by Pravec and Harris (2000). These results suggest that a more robust structure must be expected, as a fractured monolith or a rubble pile in a "strength regime" (Holsapple 2002).Comment: 32 pages, 7 figure, paper accepted for publication in Icaru

    Pet dogs' behavior when the owner and an unfamiliar person attend to a faux rival

    While dog owners ascribe different emotions to their pets, including jealousy, research on secondary emotions in nonhuman animals is very limited and, so far, only one study has investigated jealousy in dogs (Canis familiaris). This work explores jealousy in dogs one step further. We conducted two studies adapting a procedure devised to assess jealousy in human infants. In each study 36 adult dogs were exposed to a situation in which their owner and a stranger ignored them while directing positive attention towards three different objects: a book, a puppet and a fake dog (Study 1: furry; Study 2: plastic). Overall, the results of both studies do not provide evidence that the behavioral responses of our dogs were triggered by jealousy: we did not find a clear indication that the fake dogs were perceived as real social rivals, neither the furry nor the plastic one. Indeed, dogs exhibited a higher interest (i.e. look at, interact with) towards the fake dogs, but differences in the behavior towards the fake dog and the puppet only emerged in Study 2. In addition, many of the behaviors (protest, stress, attention seeking, aggression) that are considered distinctive features of jealousy were not expressed or were expressed to a limited extent, revealing that dogs did not actively try to regain their owner's attention or interfere with the interaction between the owner and the faux rival. Finally, a differentiated response towards the attachment figure (the owner) and the unfamiliar person (the stranger) did not emerge. Differently from what reported in human infants, dogs' behavior towards the attachment figure and the stranger interacting with the potential competitor (in this case, the fake dog) did not significantly differ: in both studies dogs paid attention to the owner and the stranger manipulating the fake dog to the same extent. In conclusion, we do not exclude that dogs could possess a rudimentary form of jealousy, but we suggest that research on this topic should require the use of a real social interloper (conspecific or human) and more naturalistic procedures
