    The molecular control of tomato fruit quality traits: the trade off between visual attributes, shelf life and nutritional value

    Tomato (Solanum lycopersicum) is an established model to study fleshy fruit development and ripening and is an important crop in terms of its economic and nutritional value. Tomato fruit quality is a function of metabolite content which is prone to physiological changes related to fruit development and ripening. It has been described some ripening tomato mutants, delayed fruit deterioration (DFD), non-ripening (NOR) and ripening-inhibitor (RIN) which substantially extend “shelf life” in tomato for up to several months when defined in terms of softening, water loss and resistance to postharvest biotic infection. However, it is not known whether this extension in “shelf life” is in fact a desirable objective from the perspective of nutritional quality of the fruits. The aim of this work was to use a metabolomics approach join to genomic tools to characterize compositional changes (sugars, amino acids, organic acids and carotenoids) of non-softening tomato mutants reported (DFD, NOR and RIN) in comparison with the normally softening fruits (Ailsa Craig and M82) during ripening and postharvest shelf-life.     Sugar deficiency causes changes in cuticle permeability and cell wall composition that influence fruit postharvest shelf-life

    The cuticle is a protective layer synthesized by epidermal cells of the plants and consisting of cutin covered and filled by waxes. In tomato (Solanum lycopersicum) fruit, the thick cuticle embedding epidermal cells has crucial roles in the control of pathogens, water loss, cracking, and postharvest shelf-life. Tomato fruits with reduced expression of the tomato gene LIN5 encoding cell wall invertase exhibits decreases transpirational water loss. Transcriptomic, biochemical, histological, and biomechanical analysis identified several unsual features of RNAi-LIN5 cuticles and the data indicate that, perturbation of endogenous fruit sugar levels affects the composition of the tomato cuticle and cell wall architecture which are an integral and regulated part of the ripening program affecting the postharvest shelf-life. A model is proposed in which sugar levels affects the cuticle formation which has a direct effect in softening of intact tomato fruit both directly, by providing a physical support, and indirectly, by regulating water status.University of Málaga, Campus de Excelencia Internacional de Andalucia Tech.     The aim of this work was to investigate the effect of decreased cytosolic phosphoenolpyruvate carboxykinase (PEPCK) and plastidic NADP-dependent malic enzyme (NADP-ME) on tomato (Solanum lycopersicum) ripening. Transgenic tomato plants with strongly reduced levels of PEPCK and plastidic NADP-ME were generated by RNA interference gene silencing under the control of a ripening-specific E8 promoter. While these genetic modifications had relatively little effect on the total fruit yield and size, they had strong effects in fruit metabolism. Both transformants were characterized by lower levels of starch at breaker stage. Analysis of the activation state of ADP-glucose pyrophosphorylase correlated with the decrease of starch in both transformats, which suggest that is due to an altered cellular redox status. Moreover, metabolic profiling and feeding experiments involving positional labelled glucoses of fruits lacking in plastidic NADP-malic enzyme and cytosolic PEPCK activities revealed differential changes in overall respiration rates and tricarboxylic acid (TCA) cycle flux. Inactivation of cytosolic PEPCK affected the respiration rate which suggests that excess of oxaloacetate OAA is converted to aspartate and reintroduced in the TCA via 2-oxoglutarate/glutamate. On the other hand, the plastidic NADP-malic enzyme antisense lines were characterized by no changes in respiration rates and TCA cycle flux and together with an increase of pyruvate kinase and phosphoenolpyruvate carboxylase activities indicates that pyruvate is supply through these enzymes to the TCA cycle. These results are discussed in the context of current models of the importance of malate during tomato fruit ripening

    Nitrogen dioxide concentrations in neighborhoods adjacent to a commercial airport: a land use regression modeling study

    BACKGROUND: There is growing concern in communities surrounding airports regarding the contribution of various emission sources (such as aircraft and ground support equipment) to nearby ambient concentrations. We used extensive monitoring of nitrogen dioxide (NO(2)) in neighborhoods surrounding T.F. Green Airport in Warwick, RI, and land-use regression (LUR) modeling techniques to determine the impact of proximity to the airport and local traffic on these concentrations. METHODS: Palmes diffusion tube samplers were deployed along the airport's fence line and within surrounding neighborhoods for one to two weeks. In total, 644 measurements were collected over three sampling campaigns (October 2007, March 2008 and June 2008) and each sampling location was geocoded. GIS-based variables were created as proxies for local traffic and airport activity. A forward stepwise regression methodology was employed to create general linear models (GLMs) of NO(2 )variability near the airport. The effect of local meteorology on associations with GIS-based variables was also explored. RESULTS: Higher concentrations of NO(2 )were seen near the airport terminal, entrance roads to the terminal, and near major roads, with qualitatively consistent spatial patterns between seasons. In our final multivariate model (R(2 )= 0.32), the local influences of highways and arterial/collector roads were statistically significant, as were local traffic density and distance to the airport terminal (all p < 0.001). Local meteorology did not significantly affect associations with principal GIS variables, and the regression model structure was robust to various model-building approaches. CONCLUSION: Our study has shown that there are clear local variations in NO(2 )in the neighborhoods that surround an urban airport, which are spatially consistent across seasons. LUR modeling demonstrated a strong influence of local traffic, except the smallest roads that predominate in residential areas, as well as proximity to the airport terminal

    Chemical Characterization and Source Apportionment of Household Fine Particulate Matter in Rural, Peri-urban, and Urban West Africa

    Household air pollution in sub-Saharan Africa and other developing regions is an important cause of disease burden. Little is known about the chemical composition and sources of household air pollution in sub-Saharan Africa, and how they differ between rural and urban homes. We analyzed the chemical composition and sources of fine particles (PM2.5) in household cooking areas of multiple neighborhoods in Accra, Ghana, and in peri-urban (Banjul) and rural (Basse) areas in The Gambia. In Accra, biomass burning accounted for 39–62% of total PM2.5 mass in the cooking area in different neighborhoods; the absolute contributions were 10–45 μg/m3. Road dust and vehicle emissions comprised 12–33% of PM2.5 mass. Solid waste burning was also a significant contributor to household PM2.5 in a low-income neighborhood but not for those living in better-off areas. In Banjul and Basse, biomass burning was the single dominant source of cooking-area PM2.5, accounting for 74–87% of its total mass; the relative and absolute contributions of biomass smoke to PM2.5 mass were larger in households that used firewood than in those using charcoal, reaching as high as 463 μg/m3 in Basse homes that used firewood for cooking. Our findings demonstrate the need for policies that enhance access to cleaner fuels in both rural and urban areas, and for controlling traffic emissions in cities in sub-Saharan Africa

    We consider an infinite homogeneous tree V endowed with the usual metric d defined on graphs and a weighted measure μ. The metric measure space (V, d, μ) is nondoubling and of exponential growth, hence the classical theory of Hardy spaces does not apply in this setting. We construct an atomic Hardy space H1(μ) on (V, d, μ) and investigate some of its properties, focusing in particular on real interpolation properties and on boundedness of singular integrals on H1(μ)

    Exposure to flame retardant chemicals on commercial airplanes

    Background: Flame retardant chemicals are used in materials on airplanes to slow the propagation of fire. These chemicals migrate from their source products and can be found in the dust of airplanes, creating the potential for exposure. Methods: To characterize exposure to flame retardant chemicals in airplane dust, we collected dust samples from locations inside 19 commercial airplanes parked overnight at airport gates. In addition, hand-wipe samples were also collected from 9 flight attendants and 1 passenger who had just taken a cross-country (USA) flight. The samples were analyzed for a suite of flame retardant chemicals. To identify the possible sources for the brominated flame retardants, we used a portable XRF analyzer to quantify bromine concentrations in materials inside the airplanes. Results: A wide range of flame retardant compounds were detected in 100% of the dust samples collected from airplanes, including BDEs 47, 99, 153, 183 and 209, tris(1,3-dichloro-isopropyl)phosphate (TDCPP), hexabromocyclododecane (HBCD) and bis-(2-ethylhexyl)-tetrabromo-phthalate (TBPH). Airplane dust contained elevated concentrations of BDE 209 (GM: 500 ug/g; range: 2,600 ug/g) relative to other indoor environments, such as residential and commercial buildings, and the hands of participants after a cross-country flight contained elevated BDE 209 concentrations relative to the general population. TDCPP, a known carcinogen that was removed from use in children’s pajamas in the 1970’s although still used today in other consumer products, was detected on 100% of airplanes in concentrations similar to those found in residential and commercial locations. Conclusion: This study adds to the limited body of knowledge regarding exposure to flame retardants on commercial aircraft, an environment long hypothesized to be at risk for maximum exposures due to strict flame retardant standards for aircraft materials. Our findings indicate that flame retardants are widely used in many airplane components and all airplane types, as expected. Most flame retardants, including TDCPP, were detected in 100% of dust samples collected from the airplanes. The concentrations of BDE 209 were elevated by orders of magnitude relative to residential and office environments

    An evidence map of psychosocial interventions for the earliest stages of bipolar disorder.

    Depression, schizophrenia, and bipolar disorder are three of the four most burdensome problems in people aged under 25 years. In psychosis and depression, psychological interventions are effective, low-risk, and high-benefit approaches for patients at high risk of first-episode or early-onset disorders. We review the use of psychological interventions for early-stage bipolar disorder in patients aged 15-25 years. Because previous systematic reviews had struggled to identify information about this emerging sphere of research, we used evidence mapping to help us identify the extent, distribution, and methodological quality of evidence because the gold standard approaches were only slightly informative or appropriate. This strategy identified 29 studies in three target groups: ten studies in populations at high risk for bipolar disorder, five studies in patients with a first episode, and 14 studies in patients with early-onset bipolar disorder. Of the 20 completed studies, eight studies were randomised trials, but only two had sample sizes of more than 100 individuals. The main interventions used were family, cognitive behavioural, and interpersonal therapies. Only behavioural family therapies were tested across all of our three target groups. Although the available interventions were well adapted to the level of maturity and social environment of young people, few interventions target specific developmental psychological or physiological processes (eg, ruminative response style or delayed sleep phase), or offer detailed strategies for the management of substance use or physical health

    Relationship between resilience, optimism, and burnout in Pan-American athletes

    Aim: A series of knowledge has been developed on burnout syndrome in the sports context that has allowed to generate a solid theoretical structure that requires new contributions to delve into those aspects that have been less addressed, as is the case of optimism and resilience when it comes to linking them to the syndrome in top-performance athletes. For this purpose, the Burnout Inventory for Athletes (IBD-r), the Resilience Scale adapted to Spanish, and the Life Orientation Scale-Revised (LOT-R) for optimism were used. Methods: From this perspective, the study was approached with 121 Uruguayan athletes who participated in the last Pan-American Games, assuming practically all the subjects who have this level of sport (142) considering gender and type of sport. Results: The results indicate that 67% of the sample presents burnout symptoms for the emotional exhaustion dimension, and moderate resilience and optimism. Although there are no significant differences attributable to gender or type of sport, this contribution allows us to continue focusing on future work and further analysis. The type of sport presented statistically significant differences in relation to the personal competence dimension and the type of sport. Conclusion: Resilience and optimism obtained a clear influence on the occurrence of burnout, in a statistically negative sense, showing themselves as interesting prevention strategies for future lines of research, where it is essential to design interventions that teach emotional skills to manage adversity and prevent burnout.
