    Reductions in disease activity in the AMPLE trial: clinical response by baseline disease duration

    Objectives: To evaluate clinical response by baseline disease duration using 2-year data from the AMPLE trial. Methods: Patients were randomised to subcutaneous abatacept 125 mg weekly or adalimumab 40 mg biweekly, with background methotrexate. As part of a post hoc analysis, the achievement of validated definitions of remission (Clinical Disease Activity Index (CDAI) ≤2.8, Simplified Disease Activity Index (SDAI) ≤3.3, Routine Assessment of Patient Index Data 3 (RAPID3) ≤3.0, Boolean score ≤1), low disease activity (CDAI \u3c10, SDAI \u3c11, RAPID3 ≤6.0), Health Assessment Questionnaire-Disability Index response and American College of Rheumatology responses were evaluated by baseline disease duration (≤6 vs \u3e6 months). Disease Activity Score 28 (C-reactive protein) \u3c2.6 or ≤3.2 and radiographic non-progression in patients achieving remission were also evaluated. Results: A total of 646 patients were randomised and treated (abatacept, n=318; adalimumab, n=328). In both treatment groups, comparable responses were achieved in patients with early rheumatoid arthritis (≤6 months) and in those with later disease (\u3e6 months) across multiple clinical measures Conclusions: Abatacept or adalimumab with background methotrexate were associated with similar onset and sustainability of response over 2 years. Patients treated early or later in the disease course achieved comparable clinical responses

    Incorporating costs, thresholds and spatial extents for selecting stream bioindicators in an ecotone between two Brazilian biodiversity hotspots

    Biomonitoring is critical for characterizing and monitoring status, spatial patterns, and long-term trends in the ecological condition of freshwater ecosystems. The selection of cost-effective bioindicators is a critical step in establishing such monitoring programs. Key indicator considerations are a reliable response to anthropogenic disturbances, a high benefit-cost-ratio and sensitivity at multiple spatial extents. We evaluated non-linear responses of Ephemeroptera, Plecoptera and Trichoptera (EPT) and fish to the effects of native vegetation loss within buffers of 100 m and 1000 m and assessed the sampling and processing costs involved for each assemblage. We sampled 37 neotropical stream sites in the Formoso River network, a karstic region of the Bodoquena Plateau, midwest Brazil, lying in the ecotone between the Cerrado and Atlantic Forest biological hotspots. We used TITAN (threshold indicator taxa analysis) to identify six indicator taxa, four EPT genera and two fish species. The four EPT genera had low negative thresholds to native vegetation loss, whereas the two fish species had positive thresholds. Thresholds were lower for the 100 m buffers than the 1000 m buffers for EPT. The most sensitive taxon (Macronema, Trichoptera) had a threshold of 0% native vegetation loss in the 100 m buffers and nearly 40% in the 1000 m buffers. For taxa richness, we found no non-linear response to the effects of native vegetation loss for buffer extent nor assemblage. The total cost for EPT biomonitoring was US3,616;whereasforfish,thetotalwasUS 3,616; whereas for fish, the total was US 1,901. Although fish were less expensive than EPT, they did not respond negatively to native vegetation loss and their positive threshold started at 48%, a level of vegetation loss that was highly disruptive of EPT. Therefore, we do not recommend using fish to monitor the effects of native vegetation loss on headwater streams in the Bodoquena Plateau. Although EPT monitoring costs 52% more than fish assemblage monitoring, it detected earlier impacts of the effects of native vegetation loss on stream biota, especially in the 100 m buffer. Therefore, EPT are more cost-effective early warning indicators for monitoring the effects of native vegetation loss in Bodoquena Plateau headwaters

    Seasonality modulates the direct and indirect influences of forest cover on larval anopheline assemblages in western Amazônia

    Serious concerns have arisen regarding urbanization processes in western Amazônia, which result in the creation of artificial habitats, promoting the colonization of malaria vectors. We used structural equation modelling to investigate direct and indirect effects of forest cover on larval habitats and anopheline assemblages in different seasons. We found 3474 larvae in the dry season and 6603 in the rainy season, totalling ten species and confirming the presence of malaria vectors across all sites. Forest cover had direct and indirect (through limnological variables) effects on the composition of larval anopheline assemblages in the rainy season. However, during the dry season, forest cover directly affected larval distribution and habitat variables (with no indirect affects). Additionally, artificial larval habitats promote ideal conditions for malaria vectors in Amazonia, mainly during the rainy season, with positive consequences for anopheline assemblages. Therefore, the application of integrated management can be carried out during both seasons. However, we suggest that the dry season is the optimal time because larval habitats are more limited, smaller in volume and more accessible for applying vector control techniques

    Longitudinal study of repeated sprint performance in youth soccer players of contrasting skeletal maturity status

    The purpose of the study was to evaluate the developmental changes in performance in a repeated-sprint ability (RSA) test in young soccer players of contrasting maturity status. A total of 83 regional level Portuguese youth soccer players, aged 11-13 years at baseline was assessed annually. Stature, body mass, 7x34.2-m sprint protocol (25-s active recovery), 20-m multi-stage continuous shuttle endurance run and counter-movement jump (CMJ) without the use of the arms were measured. Fat-free mass (FFM) was determined by age and gender-specific formulas. Developmental changes in total sprint time across ages were predicted using multilevel modeling. Corresponding measurements were performed on an independent cross-sectional sub-sample of 52 youth soccer players 11-17 years to evaluate the predictive model. CA, CA(2), maturational status (SA-CA), body size (mass and stature), FFM, aerobic endurance, lower limb explosive strength and annual volume training significantly improved the statistical fit of the RSA multilevel model. In 'late' maturing athletes, the best model for predicting change in RSA was expressed by the following equation: 86.54 - 2.87 x CA + 0.05 x CA(2) - 0.25 x FFM + 0.15 x body mass + 0.05 x stature - 0.05 x aerobic endurance - 0.09 x lower limb explosive strength - 0.01 x annual volume training. The best fitting models for players who were 'on time' and 'early' maturing were identical to the best model for late maturing players, less 0.64 seconds and 1.74 seconds, respectively. Multilevel modeling provided performance curves that permitted the prediction of individual RSA performance across adolescent years in regional level soccer players

    Multivariate relationships among morphology, fitness and motor coordination in prepubertal girls

    Motor coordination and physical fitness are multidimensional concepts which cannot be reduced to a single variable. This study evaluated multivariate relationships among morphology, physical fitness and motor coordination in 74 pre-pubertal girls 8.0-8.9 years of age. Data included body dimensions, eight fitness items and four motor coordination tasks (KTK battery). Maturity status was estimated as percentage of predicted mature stature attained at the time of observation. Canonical correlation analysis was used to examine the relationships between multivariate domains. Significant pairs of linear functions between indicators of morphology and fitness (r(c) = 0.778, Wilks' Lambda = 0.175), and between fitness and motor coordination (r(c) = 0.765, Wilks' Lambda = 0.289) were identified. Girls who were lighter and had a lower waist-to-stature ratio and % fat mass attained better scores in the endurance run, sit-ups and standing long jump tests, but poorer performances in hand grip strength and 2-kg ball throw. Better fitness test scores were also associated with better motor coordination scores. Relationships between body size and estimated fatness with motor fitness suggested an inverse relationship that was particularly evident in performance items that required the displacement of the body through space, while motor coordination was more closely related with fitness than with somatic variables

    The UA9 experimental layout

    The UA9 experimental equipment was installed in the CERN-SPS in March '09 with the aim of investigating crystal assisted collimation in coasting mode. Its basic layout comprises silicon bent crystals acting as primary collimators mounted inside two vacuum vessels. A movable 60 cm long block of tungsten located downstream at about 90 degrees phase advance intercepts the deflected beam. Scintillators, Gas Electron Multiplier chambers and other beam loss monitors measure nuclear loss rates induced by the interaction of the beam halo in the crystal. Roman pots are installed in the path of the deflected particles and are equipped with a Medipix detector to reconstruct the transverse distribution of the impinging beam. Finally UA9 takes advantage of an LHC-collimator prototype installed close to the Roman pot to help in setting the beam conditions and to analyze the efficiency to deflect the beam.     Parkin is activated by PINK1-dependent phosphorylation of ubiquitin at Serine<sup>65</sup>

    We have previously reported that the Parkinson's disease-associated kinase PINK1 (PTEN-induced putative kinase 1) is activated by mitochondrial depolarization and stimulates the Parkin E3 ligase by phosphorylating Ser(65) within its Ubl (ubiquitin-like) domain. Using phosphoproteomic analysis, we identified a novel ubiquitin phosphopeptide phosphorylated at Ser(65) that was enriched 14-fold in HEK (human embryonic kidney)-293 cells overexpressing wild-type PINK1 stimulated with the mitochondrial uncoupling agent CCCP (carbonyl cyanide m-chlorophenylhydrazone), to activate PINK1, compared with cells expressing kinase-inactive PINK1. Ser(65) in ubiquitin lies in a similar motif to Ser(65) in the Ubl domain of Parkin. Remarkably, PlNK1 directly phosphorylates Ser(65) of ubiquitin in vitro. We undertook a series of experiments that provide striking evidence that Ser(65)-phosphorylated ubiquitin (ubiquitin(Phospho-Ser65)) functions as a critical activator of Parkin. First, we demonstrate that a fragment of Parkin lacking the Ubl domain encompassing Ser(65) (Delta Ubl-Parkin) is robustly activated by ubiquitin(Phospho-Ser65), but not by non-phosphorylated ubiquitin. Secondly, we find that the isolated Parkin Ubl domain phosphorylated at Ser(65) (Ubl(phospho-Ser65)) can also activate Delta Ubl-Parkin similarly to ubiquitin(PhosPh-Ser65). Thirdly, we establish that ubiquitin(PhosPh-Ser65), but not non-phosphorylated ubiquitin or Ubl(PhosPh-Ser65) activates full-length wild-type Parkin as well as the non-phosphorylatable S65A Parkin mutant. Fourthly, we provide evidence that optimal activation of full-length Parkin E3 ligase is dependent on PINK1-mediated phosphorylation of both Parkin at Ser(65) and ubiquitin at Ser(65), since only mutation of both proteins at Ser(65) completely abolishes Parkin activation. In conclusion, the findings of the present study reveal that PINK1 controls Parkin E3 ligase activity not only by phosphorylating Parkin at Ser(65), but also by phosphorylating ubiquitin at Ser(65). We propose that phosphorylation of Parkin at Ser(65) serves to prime the E3 ligase enzyme for activation by ubiquitin(PhosPh-Ser65), suggesting that small molecules that mimic ubiquitin(PhosPh-Ser65) could hold promise as novel therapies for Parkinson's disease

    A Compilation of Global Bio-Optical In Situ Data for Ocean-Colour Satellite Applications

    A compiled set of in situ data is important to evaluate the quality of ocean-colour satellite-data records. Here we describe the data compiled for the validation of the ocean-colour products from the ESA Ocean Colour Climate Change Initiative (OC-CCI). The data were acquired from several sources (MOBY, BOUSSOLE, AERONET-OC, SeaBASS, NOMAD, MERMAID, AMT, ICES, HOT, GeP&CO), span between 1997 and 2012, and have a global distribution. Observations of the following variables were compiled: spectral remote-sensing reflectances, concentrations of chlorophyll a, spectral inherent optical properties and spectral diffuse attenuation coefficients. The data were from multi-project archives acquired via the open internet services or from individual projects, acquired directly from data providers. Methodologies were implemented for homogenisation, quality control and merging of all data. No changes were made to the original data, other than averaging of observations that were close in time and space, elimination of some points after quality control and conversion to a standard format. The final result is a merged table designed for validation of satellite-derived ocean-colour products and available in text format. Metadata of each in situ measurement (original source, cruise or experiment, principal investigator) were preserved throughout the work and made available in the final table. Using all the data in a validation exercise increases the number of matchups and enhances the representativeness of different marine regimes. By making available the metadata, it is also possible to analyse each set of data separately. The compiled data are available at doi:10.1594/PANGAEA.854832 (Valente et al., 2015)
