    Identificação de raças fisiológicas de Hemileia vstatrix Berk. & Br. no Estado do Espírito Santo, Brasil.

    Cento e trinta e nove isolados de Hemileia vastatrix foram coletados de plantas de cultivares de Coffea canephora e Coffea arabica, nos estados do Espírito Santo e Minas Gerais. Os isolados foram inoculados em plantas do cultivar Catuaí Vermelho LCH 2077-2-5-44 em condição de casa de vegetação, de junho de 1995 a julho de 1997. A identificação de raças foi acompanhada pela inoculação de isolados de H. vastatrix em clones diferenciadores, mantidos em casa de vegetação no Departamento de Fitopatologia da Universidade Federal de Viçosa. Raças de H. vastatrix foram determinada segundo D’Oliveira (1954, 1957). A raça II foi identificada em todos os 96 isolados de C. canephora do estado do Espírito Santo

    Foram avaliados, quanto aos tipos de reação às raças I, II, III e XIII de Hemileia vastatrix, 33 clones de Coffea canephora componentes de variedades clonais, sendo nove com ciclo de maturação precoce da variedade EMCAPA 8111; treze com ciclo de maturação intermediário da variedade EMCAPA 8121 e onze de ciclo de maturação tardio da variedade EMCAPA 8131. Empregou-se escala de notas de 1 a 6, sendo que graus médios de doença menores que 4 corresponderam a reação de resistência e maiores ou igual a 4, de suscetibilidade. Utilizou-se o intervalo de confiança do grau médio de doença como medida de estabilidade de reação dos clones em relação às raças inoculadas. Os clones 154 (precoce), 132, 149 e 201 (intermediários), e, 100 e 143 (tardios) mostraram-se resistentes às quatro raças do patógeno. Entretanto, observou-se variabilidade de reação entre repetições de 12 clones analisados. Os resultados mostraram que as três variedades clonais são constituídas por clones resistentes e suscetíveis às quatro raças do patógeno, indicando a necessidade de estudos mais detalhados, em relação à obtenção de clones com maior nível e durabilidade de resistência à H. vastatrix

    Using 3D organotypic rat brain cell cultures in aggregates we recently identified 2-methylcitrate (2-MCA) as the main toxic metabolite for developing brain cells in methylmalonic aciduria. Exposure to 2-MCA triggered morphological changes and apoptosis of brain cells. This was accompanied by increased ammonium and decreased glutamine levels. However, the sequence and causal relationship between these phenomena remained unclear. To understand the sequence and time course of pathogenic events, we exposed 3D rat brain cell aggregates to different concentrations of 2-MCA (0.1, 0.33 and 1.0mM) from day in vitro (DIV) 11 to 14. Aggregates were harvested at different time points from DIV 12 to 19. We compared the effects of a single dose of 1mM 2-MCA administered on DIV 11 to the effects of repeated doses of 1mM 2-MCA. Pan-caspase inhibitors Z-VAD FMK or Q-VD-OPh were used to block apoptosis. Ammonium accumulation in the culture medium started within few hours after the first 2-MCA exposure. Morphological changes of the developing brain cells were already visible after 17h. The highest rate of cleaved caspase-3 was observed after 72h. A dose-response relationship was observed for all effects. Surprisingly, a single dose of 1mM 2-MCA was sufficient to induce all of the biochemical and morphological changes in this model. 2-MCA-induced ammonium accumulation and morphological changes were not prevented by concomitant treatment of the cultures with pan-caspase inhibitors Z-VAD FMK or Q-VD-OPh: ammonium increased rapidly after a single 1mM 2-MCA administration even after apoptosis blockade. We conclude that following exposure to 2-MCA, ammonium production in brain cell cultures is an early phenomenon, preceding cell degeneration and apoptosis, and may actually be the cause of the other changes observed. The fact that a single dose of 1mM 2-MCA is sufficient to induce deleterious effects over several days highlights the potential damaging effects of even short-lasting metabolic decompensations in children affected by methylmalonic aciduria

    Bionanoconjugates of tyrosinase and peptide-derivatised gold nanoparticles for biosensing of phenolic compounds

    Bionanoconjugates of the enzyme tyrosinase (TYR) and gold nanoparticles (AuNPs) functionalised with a peptide (CALNN) were produced in solution and characterised. The formation of stable TYR-AuNP:CALNN bionanoconjugates (BNCs) was supported by a decrease of the surface charge of the BNCs as determined by zeta-potential and an increase in hydrodynamic diameter as determined by Dynamic Light Scattering (DLS). UV/Vis studies of pH-induced aggregation revealed distinct protonation patterns for the BNCs when compared with AuNP:CALNN alone, further substantiating BNC formation. Activity studies of the BNCs for the reduction of di-phenols in solution indicated that TYR not only remains active after conjugation, but interestingly its activity in the BNCs is higher than for the free enzyme. In conclusion, AuNP:CALNN can provide a suitable platform for the immobilisation of TYR, leading to BNCs with increased enzyme activity and a wider pH working range, with promising uses in electrochemical biosensors for the detection of mono- and di-phenolic compounds

    SALO, a novel classical pathway complement inhibitor from saliva of the sand fly Lutzomyia longipalpis.

    Blood-feeding insects inject potent salivary components including complement inhibitors into their host's skin to acquire a blood meal. Sand fly saliva was shown to inhibit the classical pathway of complement; however, the molecular identity of the inhibitor remains unknown. Here, we identified SALO as the classical pathway complement inhibitor. SALO, an 11 kDa protein, has no homology to proteins of any other organism apart from New World sand flies. rSALO anti-complement activity has the same chromatographic properties as the Lu. longipalpis salivary gland homogenate (SGH)counterparts and anti-rSALO antibodies blocked the classical pathway complement activity of rSALO and SGH. Both rSALO and SGH inhibited C4b deposition and cleavage of C4. rSALO, however, did not inhibit the protease activity of C1s nor the enzymatic activity of factor Xa, uPA, thrombin, kallikrein, trypsin and plasmin. Importantly, rSALO did not inhibit the alternative or the lectin pathway of complement. In conclusion our data shows that SALO is a specific classical pathway complement inhibitor present in the saliva of Lu. longipalpis. Importantly, due to its small size and specificity, SALO may offer a therapeutic alternative for complement classical pathway-mediated pathogenic effects in human diseases

    MEMS Deformable Mirrors for Space-Based High-Contrast Imaging

    Micro-Electro-Mechanical Systems (MEMS) Deformable Mirrors (DMs) enable precise wavefront control for optical systems. This technology can be used to meet the extreme wavefront control requirements for high contrast imaging of exoplanets with coronagraph instruments. MEMS DM technology is being demonstrated and developed in preparation for future exoplanet high contrast imaging space telescopes, including the Wide Field Infrared Survey Telescope (WFIRST) mission which supported the development of a 2040 actuator MEMS DM. In this paper, we discuss ground testing results and several projects which demonstrate the operation of MEMS DMs in the space environment. The missions include the Planet Imaging Concept Testbed Using a Recoverable Experiment (PICTURE) sounding rocket (launched 2011), the Planet Imaging Coronagraphic Technology Using a Reconfigurable Experimental Base (PICTURE-B) sounding rocket (launched 2015), the Planetary Imaging Concept Testbed Using a Recoverable Experiment - Coronagraph (PICTURE-C) high altitude balloon (expected launch 2019), the High Contrast Imaging Balloon System (HiCIBaS) high altitude balloon (launched 2018), and the Deformable Mirror Demonstration Mission (DeMi) CubeSat mission (expected launch late 2019). We summarize results from the previously flown missions and objectives for the missions that are next on the pad. PICTURE had technical difficulties with the sounding rocket telemetry system. PICTURE-B demonstrated functionality at >100 km altitude after the payload experienced 12-g RMS (Vehicle Level 2) test and sounding rocket launch loads. The PICTURE-C balloon aims to demonstrate 10(-7) contrast using a vector vortex coronagraph, image plane wavefront sensor, and a 952 actuator MEMS DM. The HiClBaS flight experienced a DM cabling issue, but the 37-segment hexagonal piston-tip-tilt DM is operational post-flight.     High-pT pi^zero Production with Respect to the Reaction Plane in Au + Au Collisions at sqrt(s_NN) = 200 GeV

    Measurements of the azimuthal anisotropy of high-\pT neutral pion neutral pion production in Au+Au collisions at sqrt(s_NN) = 200 GeV by the PHENIX experiment are presented. The data included in this paper were collected during the 2004 RHIC running period and represent approximately an order of magnitude increase in the number of analyzed events relative to previously published results. Azimuthal angle distributions of pi^0s detected in the PHENIX electromagnetic calorimeters are measured relative to the reaction plane determined event-by-event using the forward and backward beam-beam counters. Amplitudes of the second Fourier component (v_2) of the angular distributions are presented as a function of pi^0 transverse momentum p_T for different bins in collision centrality. Measured reaction plane dependent pi^0 yields are used to determine the azimuthal dependence of the pi^0 suppression as a function of p_T, R_AA (Delta phi,p_T). A jet-quenching motivated geometric analysis is presented that attempts to simultaneously describe the centrality dependence and reaction plane angle dependence of the pi^0 suppression in terms of the path lengths of hypothetical parent partons in the medium. This set of results allows for a detailed examination of the influence of geometry in the collision region, and of the interplay between collective flow and jet-quenching effects along the azimuthal axis.Comment: 344 authors, 35 pages text, RevTeX-4, 24 figures, 8 tables.     Transverse-energy distributions at midrapidity in pp++pp, dd++Au, and Au++Au collisions at sNN=62.4\sqrt{s_{_{NN}}}=62.4--200~GeV and implications for particle-production models

    Measurements of the midrapidity transverse energy distribution, d\Et/d\eta, are presented for pp++pp, dd++Au, and Au++Au collisions at sNN=200\sqrt{s_{_{NN}}}=200 GeV and additionally for Au++Au collisions at sNN=62.4\sqrt{s_{_{NN}}}=62.4 and 130 GeV. The d\Et/d\eta distributions are first compared with the number of nucleon participants NpartN_{\rm part}, number of binary collisions NcollN_{\rm coll}, and number of constituent-quark participants NqpN_{qp} calculated from a Glauber model based on the nuclear geometry. For Au++Au, \mean{d\Et/d\eta}/N_{\rm part} increases with NpartN_{\rm part}, while \mean{d\Et/d\eta}/N_{qp} is approximately constant for all three energies. This indicates that the two component ansatz, dET/dη(1x)Npart/2+xNcolldE_{T}/d\eta \propto (1-x) N_{\rm part}/2 + x N_{\rm coll}, which has been used to represent ETE_T distributions, is simply a proxy for NqpN_{qp}, and that the NcollN_{\rm coll} term does not represent a hard-scattering component in ETE_T distributions. The dET/dηdE_{T}/d\eta distributions of Au++Au and dd++Au are then calculated from the measured pp++pp ETE_T distribution using two models that both reproduce the Au++Au data. However, while the number-of-constituent-quark-participant model agrees well with the dd++Au data, the additive-quark model does not.Comment: 391 authors, 24 pages, 19 figures, and 15 Tables.     Particle-species dependent modification of jet-induced correlations in Au+Au collisions at sqrt(s_NN) = 200 GeV

    We report PHENIX measurements of the correlation of a trigger hadron at intermediate transverse momentum (2.5<p_{T,trig}<4 GeV/c), with associated mesons or baryons at lower p_{T,assoc}, in Au+Au collisions at sqrt(s_NN) = 200 GeV. The jet correlations for both baryons and mesons show similar shape alterations as a function of centrality, characteristic of strong modification of the away-side jet. The ratio of jet-associated baryons to mesons for this jet increases with centrality and p_{T,assoc} and, in the most central collisions, reaches a value similar to that for inclusive measurements. This trend is incompatible with in-vacuum fragmentation, but could be due to jet-like contributions from correlated soft partons which recombine upon hadronization.Comment: 344 authors, 4 pages text, RevTeX, 4 figures.     Systematic study of charged-pion and kaon femtoscopy in Au++Au collisions at sNN\sqrt{s_{_{NN}}}=200 GeV

    We present a systematic study of charged pion and kaon interferometry in Au++Au collisions at sNN\sqrt{s_{_{NN}}}=200 GeV. The kaon mean source radii are found to be larger than pion radii in the outward and longitudinal directions for the same transverse mass; this difference increases for more central collisions. The azimuthal-angle dependence of the radii was measured with respect to the second-order event plane and similar oscillations of the source radii were found for pions and kaons. Hydrodynamic models qualitatively describe the similar oscillations of the mean source radii for pions and kaons, but they do not fully describe the transverse-mass dependence of the oscillations.Comment: 499 authors, 27 pages, 13 figures, and 11 tables. v2 is the version accepted for publication in Phys.
