Design data for the 3D printer modification to print gels and pastes and the corresponding firmware
In order to deposit gel and paste-like materials, a commercially available HICTOP Prusa i3 plastic 3D printer was modified. The modification included replacing the existing plastic microextruder with a customised 3D printed syringe pump which could hold a syringe containing the printing material. The arrangement also allowed the temperature in the syringe to be controlled. Since the hardware of the printer was changed significantly, a new firmware was loaded on the 3D printer which was customised to enable it to perform its new function.The present data consists of the 3D image files of the syringe pump assembly and instructions on how to assemble the components. It also provides a copy of the modified firmware with a list of the changes made to it.This data will allow the readers to modify a similar type of 3D printer to print pastes and gels. This can be achieved by recreating the entire syringe pump assembly by 3D printing the given 3D image file data. With some changes, these designs can also be adapted to a variety of different printers. Similarly, the given firmware can also be loaded onto a similar type of printer. The list and explanation of the changes made to the firmware also allow such changes to be made to the respective firmwares of a variety of different printers
Building information modelling to cut disruption in housing retrofit
There is a large stock of solid-wall homes in the UK with poor thermal insulation and low energy performance.
Although the UK government has supported efforts to improve these buildings, the identification of appropriate technical solutions that effectively improve the existing stock remains challenging. This research investigates how four dimensional building information modelling (4D BIM) could improve the retrofit of social housing, specifically that of ‘no-fines’ solid-wall homes, through the development of what-if scenarios that enable the analysis of alternative solutions considering costs, energy performance and disruption to users. This paper focuses on the use of 4D building information models to evaluate disruption to end users. The results indicate that the development of such models supports a better understanding of the retrofit process on site. It also supports the definition of production plans with as minimal disruption as possible to users while delivering energy-oriented and cost-effective solutions
ParaVR: A Virtual Reality Training Simulator for Paramedic Skills maintenance

Virtual Reality (VR) technology is emerging as a powerful educational tool which is used in medical training and has potential benefits for paramedic practice education.
Aim
The aim of this paper is to report development of ParaVR, which utilises VR to address skills maintenance for paramedics.
Methods
Computer scientists at the University of Chester and the Welsh Ambulance Services NHS Trust (WAST) developed ParaVR in four stages: 1. Identifying requirements and specifications 2. Alpha version development, 3. Beta version development 4. Management: Development of software, further funding and commercialisation.
Results
Needle Cricothyrotomy and Needle Thoracostomy emerged as candidates for the prototype ParaVR. The Oculus Rift head mounted display (HMD) combined with Novint Falcon haptic device was used, and a virtual environment crafted using 3D modelling software, ported (a computing term meaning transfer (software) from one system or machine to another) onto Oculus Go and Google cardboard VR platform.
Conclusion
VR is an emerging educational tool with the potential to enhance paramedic skills development and maintenance. The ParaVR program is the first step in our development, testing, and scaling up of this technology
In vitro modeling of the neurobiological effects of glucocorticoids: A review
Hypothalamic-pituitary adrenal (HPA)axis dysregulation has long been implicated in stress-related disorders such as major depression and post-traumatic stress disorder. Glucocorticoids (GCs) are released from the adrenal glands as a result of HPA-axis activation. The release of GCs is implicated with several neurobiological changes that are associated with negative consequences of chronic stress and the onset and course of psychiatric disorders. Investigating the underlying neurobiological effects of GCs may help to better understand the pathophysiology of stress-related psychiatric disorders. GCs impact a plethora of neuronal processes at the genetic, epigenetic, cellular, and molecular levels. Given the scarcity and difficulty in accessing human brain samples, 2D and 3D in vitro neuronal cultures are becoming increasingly useful in studying GC effects. In this review, we provide an overview of in vitro studies investigating the effects of GCs on key neuronal processes such as proliferation and survival of progenitor cells, neurogenesis, synaptic plasticity, neuronal activity, inflammation, genetic vulnerability, and epigenetic alterations. Finally, we discuss the challenges in the field and offer suggestions for improving the use of in vitro models to investigate GC effects
The landscape of somatic mutations in infant MLL-rearranged acute lymphoblastic leukemias.
Infant acute lymphoblastic leukemia (ALL) with MLL rearrangements (MLL-R) represents a distinct leukemia with a poor prognosis. To define its mutational landscape, we performed whole-genome, exome, RNA and targeted DNA sequencing on 65 infants (47 MLL-R and 18 non-MLL-R cases) and 20 older children (MLL-R cases) with leukemia. Our data show that infant MLL-R ALL has one of the lowest frequencies of somatic mutations of any sequenced cancer, with the predominant leukemic clone carrying a mean of 1.3 non-silent mutations. Despite this paucity of mutations, we detected activating mutations in kinase-PI3K-RAS signaling pathway components in 47% of cases. Surprisingly, these mutations were often subclonal and were frequently lost at relapse. In contrast to infant cases, MLL-R leukemia in older children had more somatic mutations (mean of 6.5 mutations/case versus 1.3 mutations/case, P = 7.15 × 10(-5)) and had frequent mutations (45%) in epigenetic regulators, a category of genes that, with the exception of MLL, was rarely mutated in infant MLL-R ALL
Deregulation of DUX4 and ERG in acute lymphoblastic leukemia
Chromosomal rearrangements deregulating hematopoietic transcription factors are common in acute lymphoblastic leukemia (ALL).1,2 Here, we show that deregulation of the homeobox transcription factor gene DUX4 and the ETS transcription factor gene ERG are hallmarks of a subtype of B-progenitor ALL that comprises up to 7% of B-ALL. DUX4 rearrangement and overexpression was present in all cases, and was accompanied by transcriptional deregulation of ERG, expression of a novel ERG isoform, ERGalt, and frequent ERG deletion. ERGalt utilizes a non-canonical first exon whose transcription was initiated by DUX4 binding. ERGalt retains the DNA-binding and transactivating domains of ERG, but inhibits wild-type ERG transcriptional activity and is transforming. These results illustrate a unique paradigm of transcription factor deregulation in leukemia, in which DUX4 deregulation results in loss-of-function of ERG, either by deletion or induction of expression of an isoform that is a dominant negative inhibitor of wild type ERG function
Characterization of behavioral, signaling and cytokine alterations in a rat neurodevelopmental model for schizophrenia, and their reversal by the 5-HT₆ receptor antagonist SB-399885
Post-weaning social isolation of rats produces neuroanatomical, neurochemical and behavioral alterations resembling some core features of schizophrenia. This study examined the ability of the 5-HT₆ receptor antagonist SB-399885 to reverse isolation-induced cognitive deficits, then investigated alterations in hippocampal cell proliferation and hippocampal and frontal cortical expression of selected intracellular signaling molecules and cytokines. Male Lister-hooded rats (weaned on post-natal day 21-24 and housed individually or in groups of 3-4) received six i.p. injections of vehicle (1% Tween 80, 1 mL/kg) or SB-399885 (5 or 10 mg/kg) over a two week period starting 40 days post-weaning, on the days that locomotor activity, novel object discrimination (NOD), pre-pulse inhibition of acoustic startle and acquisition, retention and extinction of a conditioned freezing response (CFR) were assessed. Tissue was collected 24 h after the final injection for immunohistochemistry, reverse-phase protein microarray and western blotting. Isolation rearing impaired NOD and cue-mediated CFR, decreased cell proliferation within the dentate gyrus, and elevated hippocampal TNFα levels and Cdc42 expression. SB-399885 reversed the NOD deficit and partially normalized CFR and cell proliferation. These effects were accompanied by altered expression of several members of the c-Jun N-terminal Kinase (JNK) and p38 MAPK signaling pathways (including TAK1, MKK4 and STAT3). Although JNK and p38 themselves were unaltered at this time point hippocampal TAK1 expression and phosphorylation correlated with visual recognition memory in the NOD task. Continued use of this neurodevelopmental model could further elucidate the neurobiology of schizophrenia and aid assessment of novel therapies for drug-resistant cognitive symptoms
Study of Severe Nutritional Anaemia in Children 1-5 years of Age: Can We Do Better?
Introduction: Nutritional anemia in young children is one of the most prevalent health hazard in developing countries like India, despite of various policy initiated since long. Present study was done to know correlated factors, clinical presentations, type of severe nutritional anemia in children 1-5 years of age.
Methods: All admitted children between 1-5 years of age having severe nutritional anemia (Hb level <7 gm%)were enrolled in study. Detailed demographic profile, nutritional status, clinical presentation was recorded. Anemia were classified According to haematological & morphological profile, serum iron and B12 levels. Final outcome was observed.
Results: Total 160 patients were enrolled. we have observed that age<3 years, lower socio economic class and malnutrition were significantly associated with severe anemia (p value<0.001). Only 13.75% patients presented with sign and symptoms of anemia per se.136(88%) patients were diagnosed having IDA and rest were having B12 deficiency or mixed deficiency.
Conclusion: Severe anemia is a hidden threat in younger children and it needs to be screened and early intervened by strong implementation of different supplementation programme and we can do much better to decrease morbidity and mortality due to same
