    Inconsistencies in the red blood cell membrane proteome analysis: generation of a database for research and diagnostic applications.

    Based on recent results, the determination of the easily accessible red blood cell (RBC) membrane proteins may provide new diagnostic possibilities for assessing mutations, polymorphisms or regulatory alterations in diseases. However, the analysis of the current mass spectrometry-based proteomics datasets and other major databases indicates inconsistencies-the results show large scattering and only a limited overlap for the identified RBC membrane proteins. Here, we applied membrane-specific proteomics studies in human RBC, compared these results with the data in the literature, and generated a comprehensive and expandable database using all available data sources. The integrated web database now refers to proteomic, genetic and medical databases as well, and contains an unexpected large number of validated membrane proteins previously thought to be specific for other tissues and/or related to major human diseases. Since the determination of protein expression in RBC provides a method to indicate pathological alterations, our database should facilitate the development of RBC membrane biomarker platforms and provide a unique resource to aid related further research and diagnostics

    OPTIMAL PITCH MAP GENERATION FOR SCANNING PITCH DESIGN IN SELECTIVE SAMPLING

    The reverse engineering process represents one of the best known methodologies for creating three-dimensional (3D) virtual models starting from physical ones. Even if in the last few years its usage has significantly increased, the remarkable involvement of the operator has until now represented a significant constraint for its growth. Having regard to the fact that this process, and in particular its first step (that is the acquisition phase), strongly depends on the operator's ability and expertise, this paper aims at proposing a strategy for automatically supporting an "optimal" acquisition phase. Moreover, the acquisition phase represents the only moment in which there is a direct contact between the virtual model and the physical model. For this reason, designing an "optimal" acquisition phase will provide as output an efficient set of morphological data, which will turn out to be extremely useful for the following reverse engineering passages (pre-processing, segmentation, fitting, …). This scenario drives the researcher to use a selectivesampling plan, whose grid dimensions are correlated with the complexity of the local surface region analyzed, instead of a constant one. As a consequence, this work proposes a complete operative strategy which, starting from a first raw preliminary acquisition, will provide a new selectivesampling plan during the acquisition phase, in order to allow a deeper and more efficient new scansion.     Signaling via PI3K/FOXO1A pathway modulates formation and survival of human embryonic stem cell-derived endothelial cells

    Vascular derivatives of human embryonic stem cells (hESC) are being developed as sources of tissue-specific cells for organ regeneration. However, identity of developmental pathways that modulate the specification of endothelial cells is not known yet. We studied phosphatidylinositol 3-kinase (PI3K)-Forkhead box O transcription factor 1A (FOXO1A) pathways during differentiation of hESC toward endothelial lineage and on proliferation, maturation, and cell death of hESC-derived endothelial cells (hESC-EC). During differentiation of hESC, expression of FOXO1A transcription factor was linked to the expression of a cluster of angiogenesis- and vascular remodeling-related genes. PI3K inhibitor LY294002 activated FOXO1A and induced formation of CD31(+) hESC-EC. In contrast, differentiating hESC with silenced FOXO1A by small interfering RNA (siRNA) showed lower mRNA levels of CD31 and angiopoietin2. LY294002 decreased proliferative activity of purified hESC-EC, while FOXO1A siRNA increased their proliferation. LY294002 inhibits migration and tube formation of hESC-EC; in contrast, FOXO1A siRNA increased in vitro tube formation activity of hESC-EC. After in vivo conditioning of cells in athymic nude rats, cells retain their low FOXO1A expression levels. PI3K/FOXO1A pathway is important for function and survival of hESC-EC and in the regulation of endothelial cell fate. Understanding these properties of hESC-EC may help in future applications for treatment of injured organs

    No effect of 24 h severe energy restriction on appetite regulation and ad libitum energy intake in overweight and obese males

    Background/Objectives: Long-term success of weight loss diets might depend on how the appetite regulatory system responds to energy restriction (ER). This study determined the effect of 24 h severe ER on subjective and hormonal appetite regulation, subsequent ad libitum energy intake and metabolism. Subjects/Methods: In randomised order, eight overweight or obese males consumed a 24 h diet containing either 100% (12105 (1174 kJ; energy balance; EB) or 25% (3039 (295) kJ; ER) of estimated daily energy requirements (EER). An individualised standard breakfast containing 25% of EER (3216 (341) kJ) was consumed the following morning and resting energy expenditure, substrate utilisation and plasma concentrations of acylated ghrelin, glucagon-like peptide-1 (GLP-17–36), glucose-dependant insulinotropic peptide (GIP1–42), glucose, insulin and non-esterified fatty acid (NEFA) were determined for 4 h after breakfast. Ad libitum energy intake was assessed in the laboratory on day 2 and via food records on day 3. Subjective appetite was assessed throughout. Results: Energy intake was not different between trials for day 2 (EB: 14946 (1272) kJ; ER: 15251 (2114) kJ; P=0.623), day 3 (EB: 10580 (2457) kJ; 10812 (4357) kJ; P=0.832) or day 2 and 3 combined (P=0.693). Subjective appetite was increased during ER on day 1 (P0.381). Acylated ghrelin, GLP-17–36 and insulin were not different between trials (P>0.104). Post-breakfast area under the curve (AUC) for NEFA (P<0.05) and GIP1–42 (P<0.01) were greater during ER compared with EB. Fat oxidation was greater (P<0.01) and carbohydrate oxidation was lower (P<0.01) during ER, but energy expenditure was not different between trials (P=0.158). Conclusions: These results suggest that 24 h severe ER does not affect appetite regulation or energy intake in the subsequent 48 h. This style of dieting may be conducive to maintenance of a negative EB by limiting compensatory eating behaviour, and therefore may assist with weight loss

    Research into health disparities has long recognized the importance of residential mobility as a crucial factor in determining health outcomes. However, a lack of connectivity between the health and mobility literatures has led to a stagnation of theory and application on the health side, which lacks the detail and temporal perspectives now seen as critical to understanding residential mobility decisions.     The decline and rise of neighbourhoods: the importance of neighbourhood governance

    There is a substantial literature on the explanation of neighbourhood change. Most of this literature concentrates on identifying factors and developments behind processes of decline. This paper reviews the literature, focusing on the identification of patterns of neighbourhood change, and argues that the concept of neighbourhood governance is a missing link in attempts to explain these patterns. Including neighbourhood governance in the explanations of neighbourhood change and decline will produce better explanatory models and, finally, a better view about what is actually steering neighbourhood change

    Do ecosystem insecurity and social vulnerability lead to failure of water security?

    Achieving water security for humans and ecosystems is a pervasive challenge globally. Extensive areas of the Americas are at significant risk of water insecurity, resulting from global-change processes coupled with regional and local impacts. Drought, flooding, and water quality challenges pose significant threats, while at the same time, rapid urban expansion, competing water demands, river modifications, and expanding global markets for water-intensive agricultural products drive water insecurity. This paper takes a social-ecological systems perspective, aiming to identify examples and pathways towards resilient ecosystems and social development. It draws on lessons from two science-policy network projects, one focusing on water scarcity in arid and semi-arid regions of Argentina, Chile, Brazil, Peru, Mexico and the United States; and the second addressing river and lake basins as sentinels of climate variability and human effects on water quantity and quality in Canada, the United States, Argentina, Colombia, Uruguay and Chile. Together, these ?complementary contrasts? provide an analytical basis to empirically examine stakeholder engagement, knowledge co-production and science-policy interaction supporting decision-making to achieve water security. The paper identifies four tenets for decision-making based on water-security-focused global-change science in the Americas: 1) Decision makers should focus on protecting ecosystems because water security (along with food and energy security) depend on them; 2) Water-use and allocation decisions ought to be made considering future environmental and societal vulnerabilities, especially climate projections; 3) Holistic approaches (at basin or other appropriate levels) are best suited to ensure social-ecological system resilience and reduce vulnerability; and 4) It is essential to support local/traditional livelihoods, and underserved populations to achieve equitable water security and ecosystem resilience.Fil: Scott, Christopher A..
