The Protective Effect of L-carnitine on Ionizing Radiation-induced Free Oxygen Radicals
Ionizing radiation is known to generate reactive oxygen species (ROS) that can be removed by antioxidants. L-carnitine, a natural component of mammalian tissue, is a necessary factor in the utilization of long-chain fatty acids to produce energy. Furthermore it has been shown that L-carnitine is an antioxidant which has a scavenger effect on ROS and a stabilizing effect on damaged cell membranes. The aim of the study was to evaluate the potential protective effect of L-carnitine on radiation-induced free radicals in hamsters. Lcarnitine was given by gavage at a dose of 50 mg/kg for 15 consecutive days before irradiation with a single dose of 8 Gy. 24 h after radiation exposure, the hamsters were sacrificed and samples were taken from blood and tissues, and the biochemical and histopatological determinations were carried out. In the irradiated group, there were significant increases in plasma and liver malondialdehyde (MDA) with marked reduction in glutathione (GSH) levels in the liver, compared with controls. In red blood cells, superoxide dismutase (SOD) and catalase activities were also reduced. All these effects were reversed by L-carnitine. In conclusion, L-carnitine with its antioxidant and free radical scavenging properties could play a modulatory role against the cellular damage produced by free radicals induced by ionizing radiation.
Comparison of Protective Effects of Melatonin and Amifostine on Radiation-Induced Renal Oxidative Stress in Rats
ΔΕΝ ΔΙΑΤΙΘΕΤΑΙ ΠΕΡΙΗΨΗIn this study, we aimed to compare the protective effects of melatonin and amifostine on radiation-induced oxidative stress. Fifty female Wistar rats (3-4 months old, weighing 200±25 g) were divided into five groups (with ten rats each) and treated as follows: control (Cont), radiotherapy alone (RT), radiotherapy + amifostine (RT+AMI), radiotherapy + melatonin (RT+MEL), radiotherapy + amifostine + melatonin (RT+AMI+MEL). Rats were irradiated individually with a single dose of 8 Gy and amifostine (200 mg/kg) and melatonin (10 mg/kg) was administered to rats 30 minutes before irradiation. At the end of this follow-up period (72 hours) the rats were sacrificed. Spectrophotometric Analysis has been performed to kidney tissue samples. As a result of statistical comparison between groups after RT, total antioxidant capacity (TAC) decreased, total oxidant status (TOS) and oxidative stress index (OSI) increased, although the statistically significant change was only for OSI (p = 0.030). Addition of AMI or MEL to RT increased TAC and OSI significantly (p = 0.000), but there was no additive effect for TAC and OSI when both drugs were given together (p = 1.000, p = 0.172, respectively). Recurrent NOMO1 gene deletion is a potential clinical marker in early-onset colorectal cancer and is involved in the regulation of cell migration
The incidence of early-onset colorectal cancer (EOCRC; age younger than 50 years) has been progressively increasing over the last decades globally, with causes unexplained. A distinct molecular feature of EOCRC is that compared with cases of late-onset colorectal cancer, in EOCRC cases, there is a higher incidence of Nodal Modulator 1 (NOMO1) somatic deletions. However, the mechanisms of NOMO1 in early-onset colorectal carcinogenesis are currently unknown. In this study, we show that in 30% of EOCRCs with heterozygous deletion of NOMO1, there were pathogenic mutations in this gene, suggesting that NOMO1 can be inactivated by deletion or mutation in EOCRC. To study the role of NOMO1 in EOCRC, CRISPR/cas9 technology was employed to generate NOMO1 knockout HCT-116 (EOCRC) and HS-5 (bone marrow) cell lines. NOMO1 loss in these cell lines did not perturb Nodal pathway signaling nor cell proliferation. Expression microarrays, RNA sequencing, and protein expression analysis by LC–IMS/MS showed that NOMO1 inactivation deregulates other signaling pathways independent of the Nodal pathway, such as epithelial–mesenchymal transition and cell migration. Significantly, NOMO1 loss increased the migration capacity of CRC cells. Additionally, a gut-specific conditional NOMO1 KO mouse model revealed no subsequent tumor development in mice. Overall, these findings suggest that NOMO1 could play a secondary role in early-onset colorectal carcinogenesis because its loss increases the migration capacity of CRC cells. Genome Sequencing and Analysis of a Type A Clostridium perfringens Isolate from a Case of Bovine Clostridial Abomasitis
Clostridium perfringens is a common inhabitant of the avian and mammalian gastrointestinal tracts and can behave commensally or pathogenically. Some enteric diseases caused by type A C. perfringens, including bovine clostridial abomasitis, remain poorly understood. To investigate the potential basis of virulence in strains causing this disease, we sequenced the genome of a type A C. perfringens isolate (strain F262) from a case of bovine clostridial abomasitis. The ∼3.34 Mbp chromosome of C. perfringens F262 is predicted to contain 3163 protein-coding genes, 76 tRNA genes, and an integrated plasmid sequence, Cfrag (∼18 kb). In addition, sequences of two complete circular plasmids, pF262C (4.8 kb) and pF262D (9.1 kb), and two incomplete plasmid fragments, pF262A (48.5 kb) and pF262B (50.0 kb), were identified. Comparison of the chromosome sequence of C. perfringens F262 to complete C. perfringens chromosomes, plasmids and phages revealed 261 unique genes. No novel toxin genes related to previously described clostridial toxins were identified: 60% of the 261 unique genes were hypothetical proteins. There was a two base pair deletion in virS, a gene reported to encode the main sensor kinase involved in virulence gene activation. Despite this frameshift mutation, C. perfringens F262 expressed perfringolysin O, alpha-toxin and the beta2-toxin, suggesting that another regulation system might contribute to the pathogenicity of this strain. Ancient origin of the biosynthesis of lignin precursors
BACKGROUND: Lignin plays an important role in plant structural support and water transport, and is considered one of the hallmarks of land plants. The recent discovery of lignin or its precursors in various algae has raised questions on the evolution of its biosynthetic pathway, which could be much more ancient than previously thought. To determine the taxonomic distribution of the lignin biosynthesis genes, we screened all publicly available genomes of algae and their closest non-photosynthetic relatives, as well as representative land plants. We also performed phylogenetic analysis of these genes to decipher the evolution and origin(s) of lignin biosynthesis. RESULTS: Enzymes involved in making p-coumaryl alcohol, the simplest lignin monomer, are found in a variety of photosynthetic eukaryotes, including diatoms, dinoflagellates, haptophytes, cryptophytes as well as green and red algae. Phylogenetic analysis of these enzymes suggests that they are ancient and spread to some secondarily photosynthetic lineages when they acquired red and/or green algal endosymbionts. In some cases, one or more of these enzymes was likely acquired through lateral gene transfer (LGT) from bacteria. CONCLUSIONS: Genes associated with p-coumaryl alcohol biosynthesis are likely to have evolved long before the transition of photosynthetic eukaryotes to land. The original function of this lignin precursor is therefore unlikely to have been related to water transport. In coal opencast production, with the rise of surface water and ground water level large or small ponds are composed. The most important environmental problems of these ponds are low pH (acidic characteristic) and high metal concentrations (Fe, Mn, Al, Cu, Pb, Zn etc.) of these ponds, besides the sulfide minerals containing (S04) and the waste materials. These ponds needed to be rehabilitated for is one the sustainability of natural resources. In this study, the average pH values 6.22-7.79, turbidity (NTU) 0.63-6.71, sulphate content 840-1720 mg/L, KOI 2.27-61.5mg/L and electrical conductivity 1.72 -2.71 mS/cm have been measured during the monitoring study of three different lignite opencast mine post-production lakes of the TKI -GELI and YLI.
